Objective To study the value of prealbumin(PAB) ,C reactive protein(CRP) and apolipoprotein A1(Apo A1) in e-valuating the prognosis of patients with severe pneumonia .Methods 63 cases of patients with severe pneumonia were selected ,and the fasting serum level of prealbumin ,C-reactive protein and apolipoprotein A1 were detected in 24 hours at the admission day ,and APACHE Ⅱ score was calcumulated .The patients were divided into group A (APACHEⅡscore <20) and group B(APACHEⅡscore≥20) ,with the incidence of MODS and mortality rate compared .The patients were divided into non-MODS group and MODS group .In addition ,the patients were divided into survival group and death group according to the prognosis .PAB ,CRP and Apo A1 in each group were compared .Results The incidence of MODS and mortality rate in group B (57 .9% ,47 .4% ) were higher than that in group A(24 .0% ,16 .0% )(P<0 .01 ,P<0 .05) .PAB and Apo A1 of patients in MODS group[(134 .13 ± 36 .20)mg/L , (0 .62 ± 0 .21)g/L] and death group[(129 .05 ± 52 .24)mg/L ,(0 .76 ± 0 .29)g/L] were respectively lower than that in non-MODS group[(215 .03 ± 72 .08)mg/L ,(1 .06 ± 0 .39)g/L] and survival group[(185 .52 ± 57 .63)mg/L ,(1 .15 ± 0 .36)g/L](P<0 .05) , while the CRP in MODS group(102 .37 ± 35 .65)mg/L and death group(96 .37 ± 34 .72)mg/L were higher than that in non-MODS group(69 .68 ± 32 .92)mg/L and survival group(62 .94 ± 38 .36)mg/L (P<0 .05) .Conclusion The measurement of PAB ,CRP and Apo A1 are valuable to evaluate the prognosis of the patients with severe pneumonia .
3.Influence of Bacillus Calmette-Guerin Intervention on Stem Cell Factor and Macrophage Colony-Stimulating Factor in Bronchoaveolar Lavage Fluid of Asthmatic Mouse

Objective To observe the expressions of stem cell factor(SCF) and macrophage colony-stimulating factor(M-CSF) in bronchoaveolar lavage fluid(BALF) of asthmatic mouse,and the effect of bacillus calmette-guerin(BCG) intervention on them.Methods Thirty Kunming rats were randomly divided into asthmatic group,BCG intervention group and control group,each group had 10 rats.Mice were sensitized and challenged with ovalbumin (OVA) to establish asthmatic model.Asthmatic mice were injected intradermally with BCG on 7,3,1 d before sensitization.After 24 h of last challenge,BALF were collected.The total cells and eosinophils(EOS) were counted in the BALF.The SCF and M-CSF levels of BALF were determined by enzyme-linked immunosorbent assay(ELISA).Results The number of total cells and EOS [(27.06?4.25)?107 L-1,(7.58?1.30)?107 L-1]in BALF in asthmatic mice were more than those in control group[(4.93?1.43)?107 L-1,0](Pa0.05).The M-CSF level also increased noticeably in asthmatic mice[(204.30?10.39) ng/L] compared with the control group[(181.33?8.63) ng/L](P

6.Role of RANTES in evolution of alloreactive CD4+ memory T cells adoptive transfer-induced cardiac allograft acute rejection

Objective To observe the intluence ot chemokine RANTES influence on cardiac allograft acute rejection caused by alloreactive memory CD4+ T cells (Tm) adoptive transfer.Methods Heterotopic heart transplantation (HTx) from Balb/c donors to C57BL/6 recipients was performed by anastomosis of the vessels of the neck.Mice undergoing heterotopic heart transplantation received either adoptive transfer of 1 × 106 CD4+ Tm from the spleen of alloantigen-primed C57BL/6 mice or no cells (control group).After the cardiac transplantation,the mean survival time (MST),mean histologic rank of rejection,relative gene expression and serum concentration of RANTES in the cardiac grafts.Results (1) The percentage of CD4+ Tm was 26.83% at the spleen of alloantigenprimed mice; (2) The MST was 5.17 ± 0.17 days in the CD4+ Tm+ HTx group versus 7.76 ± 0.21 days at the HTx group (control group) (P<0.01); (3) The histological tests revealed that mean histologic rank of rejection activity in the sections of cardiac allografts on the day 5 post grafting was grade 3.92 ± 0.08 in the HTx+ CD4+Tm group versus grade 2.67 ± 0.14 in HTx group (P<0.01) ;(4) The relative gene expression level of RANTES was 2.6 ± 0.21 in the CD4+ Tm + HTx group,significantly higher than in the control group (P<0.01) ; (5) The serum concentration of RANTES in the CD4+ Tm+ HTx group was 223.6 ± 16.79 pg/mL,higher than in the control group (120.7 ±9.47 pg/mL,P<0.01).Conclusion Alloreactive CD4+ Tm contribute to the increased expression and secretion of RANTES,and cardiac allograft acute rejection was more extensive in the CD4 + Tm + HTx group.
7.Effect of "Shuanggu Yitong" acupuncture therapy on interleukin-6 of experimental rats with postmenopausal osteoporosis

Objective:To study the mechanisms that acupuncture therapy of "Shuanggu Yitong" in treating postmenopausal osteoporosis(PMO).Methods:60female SD rats aged 6 months were resected bilateral ovaries and feeded for 90 days in order to make the model of experimental osteoporosis.Then,they were divided into 5 groups such as control group,regular acupoints treated group,"Shuanggu" group,"Yitong" group,"Shuanggu Yitong" group,with 12 rats each group,and the control group without any treatment;Pishu(BL 20),Weishu(BL 21),Shenshu(BL 23),Qihaishu(BL 24) was selected for the regular acupoints treated group;Guanyuan(RN 4),Zusanli(ST 36) for the "Shuanggu" group;and Shenshu(BL 23),Geshu(BL 17),Dazhu(BL 11) for the "Yitong" group;Guanyuan(RN 4),Zusanli(ST 36),Shenshu(BL 23),Geshu(BL 17),Dazhu(BL 11)for the "Shuanggu Yitong" group.Electroacupuncture therapy was given once a day,15 minutes each time,the needles were retained for 10 minutes.Then these rats rested for 1 day after consecutive therapy for 6 days.The therapy lasted for 12 weeks.RT-PCR was used to examine the expression level of interleukin-6(IL-6)mRNA in bone tissue.Results:Compared with control group,the expression level of IL-6mRNA in all acupuncture therapy groups decreased(P0.05).Conclusion:Acupuncture can reduce the expression level of IL-6mRNA in osteoporosis model rats without ovaries and "Shuanggu Yitong" acupuncture therapy is superior to other regular acupuncture therapy.

Objective To observe the expressions of the chemokine receptor CXCR3 and its ligand IFN-?-inducible protein-10(IP-10) in lung tissue of asthmatic model mice and to explore further the effect of dexamethasone(DEX)and bacillus Calmette-Guerin vaccine(BCG)on the expressions of CXCR3 and IP-10.Methods Forty Kunming mice were randomly divided into 4 groups:asthmatic group,DEX group,BCG group and control group,10 in each group.Mice were sensitized and challenged with ovalbumin (OVA) to establish asthmatic models.Ten mice in DEX group were administrated DEX (2 mg/kg) by abdomen injection 30 min before challenge (DEX group).Ten mice in BCG group were injected BCG(0.025 mg) intradermally 7,3,and 1 days before sensitization.The mice in control group mice were treated with 9 g/L saline instead of OVA.Mice of each group were executed 24 hours after the final challenge.Their lung tissue were paraffin embedded,sliced and stained by HE.The expressions of CXCR3 and IP-10 protein in lung tissue of mice were detected by immunohistochemistry.Results Distinct differences were discovered in the expressions of CXCR3(F=4.602 P=0.008) and IP-10(F=4.207 P=0.012) among the 4 groups.The expressions of CXCR3 and IP-10 in lung tissue of mice in asthmatic group were significantly higher than those in control group (Pa=0.002),while that in DEX group were significantly lower than that in asthmatic group (P=0.029,0.019).There were no significant difference between the BCG group and asthmatic group.Conclusions The chemokine receptor CXCR3 and its ligand IP-10 play an important roles in mechanisms in the pathogenesis of asthma.DEX and BCG can interfere the expressions of CXCR3 and IP-10 in varying degress.
