1.Protection of heme oxygenase-1 against hepatic ischemia reperfusion injury in cirrhotic rats

Objective To study the proctective effect of heme oxygeanse-1(HO-1) on cirrhotic rats after hepatic ischemia reperfusion injury.Methods All the rats were randomly divided into 5 groups as follows: normal group(N),liver cirrhosis group(LC),sham operation group(S), ischemia reperfusion group(I/R),and I/R+hemin group.Except for N group,the other groups were injected with 40% CCl4 twice a week.After 11 weeks,the model of liver cirrhosis was formed.The segmental(70%) hepatic ischemia reperfusion operation was carried out one week later.The animals were sacrificed 6h after reperfusion.Blood was collected to measure the liver function,antioxidative ability,NF-?B and caspase-3,and HO-1 expressions were studied by immunohistochemistry method. Results Administration of hemin to induce high HO-1 expression could lessen hepatic injury,increase MnSOD enzyme level,and decrease caspase-3 and NF-?B expresssion.Conclusion HO-1 plays an important role in protecting liver cirrhosis against ischemia reperfusion injury by increasing MnSOD enzyme level and decreasing expression of caspase-3 and NF-?B.
2.Quantitative biomechanical relationship between the anterior cruciate ligament deficiency and the posterior horn of medial meniscus

Objective To establish a quantitative biomechanical relationship between the anterior cruciate ligament deficiency and the posterior horn of medial meniscus. Methods 10 human cadaveric knees were examined using an in vitro knee-testing apparatus. In response to a combined 134 N anterior and 200 N axial compressive tibial load, the in situ forces in the posterior horn of medial meniscus were measured. Testing was performed on 10 knees at multiple angles of flexion (30?, 60?, 90?) before and after resection of anterior cruciate ligament. Results The resultant forces on the posterior horn of medial meniscus were as follows: intact ACL knees, (22.8?11.5) N in 30? flexion, (27.1?16.3) N in 60? flexion, (26.7?14.5) N in 90? flexion. Study the infect of child anorexia granule on serum ghrelin and leptin of anorexia children and its clinical efficacy. Selected 81 cases of anorexia children aged 1-6 years old into treatment group (42 cases) and control group (39 cases), in addition, 30 case healthy children as healthy control group. The control group children were treated with domperidone suspension 0.3 mg x kg(-1) x d(-1), tid, orally 30 minutes before meals. Treatment group were treated with child anorexia granule, 1-3 years 1 package, bid; 4-6 years 1 package, tid; po, 4 weeks as a course of treatment. Study the change of serum ghrelin and leptin before and after therapy. The study demonstrates that before treatment, the serum ghrelin level of disease group was lower than healthy group (P < 0.01), and the serum leptin level was higher than healthy group (P < 0.01). After treatment, the serum ghrelin level both increase, and the serum leptin decline. And the change of treatment group was significantly different with control group (P < 0.01). And the clinical effective rate are 95.23% and 74.35% (P < 0.01). After 6 months of follow-up visit, the children weight significantly increase in treatment group (P < 0.01). Results indicate that child anorexia granule can facilitate secretion of ghrelin, and inhibit secretion of leptin, so as to work up an appetite. And the molecular mechanism is its infect on serum ghrelin, leptin.

4.DETERMINATION OF FLAVONOIDS IN HAWTHORN FRUITS AND THEIR ANTIOXIDATION EFFECT

Objective: To extract and determine the flavonoids of hawthorn fruits and their antioxidative effect. Method: The ultrasonic method was used for extracting flavonoids. The content of flavonoids was determined by spectrophotometry, and the antioxidation effect was determined by flow-injection chemiluminescence.. Results and Conclution: The recovery rate was 96%~105% and the coefficient of variation was 0.14 % by spectrophotometry. Hawthorn fruits had high antioxidative effect, showing dose-response relation.
5.The effect of rALR on the proliferation and IL-2 production of spleen mononuclear cells of rat in vitro

Objective: To study the possible role and way of ALR in the immune regulation in vitro. Methods: The proliferation of spleen mononuclear cells of rat was detected with 3H-TdR method in vitro under the following different treatments:(1)the administration of the different concentration of rALR with 5 ?g/ml ConA at same time;(2)the addition of 30 ?g/ml ALR after 5 ?g/ml ConA pretreatment in 10 h and 32 h, respectively;(3)the addition of 5 ?g/ml ConA after 30 ?g/ml ALR pretreatment until 30 h. Cyclosporin A and the supernatant from cultures of yeast were used as positive and negative controls, respectively. The IL-2 levels in the supernatants from the mononuclear cells by various treatments were detected with RIA test kit. Results: rALR inhibited the proliferation and the production of IL-2 of the mononuclear cells from rat spleen stimulated by ConA dose-dependently. The mononuclear cell proliferations were still inhibited by 30 ?g/ml rALR after stimulated by ConA for 10 h or 32 h, but the pretreatment of 30 ?g/ml rALR for 30 h had no influence on the reactivity of mononuclear cell to ConA compared with control. Conclusion: rALR could inhibit in dose-dependent way the proliferation of mononuclear cells from rat spleen stimulated by ConA in vitro, but it couldn’t influence the rest mononuclear cells. This suggests that the rest mononuclear cells might not express the receptor of ALR.

OBJECTIVE:To prepare Lornoxicam for injection and to study its stability.METHODS:The formular and pre paration technology of Lornoxicam for injection was optimized with the index of contents,the related substances and pH value;3batches of samples'stability was investigated.RESULTS:The best formula was found to be the following:4.0g of lornoxicam,4.0g of propylene glycol,100g of mannitol and sufficient quantum of1mol/L caustic soda,3batches of sample preparations man ufactured under this formula were found to be stable in quality in the accelerated,room temperature storage test under the condition of commercial packing.CONCLUSION:The formula design was reasonable and the preparation technology was feasible.
7.Two new sesquiterpenoids from basidiomycete Tyromyces chioneus.

Two new sesquiterpenoids, named as tyromols A and B (1 and 2), were isolated from cultures of basidiomycete Tyromyces chioneus, along with two previously reported 15-hydroxy-6 α, 12-epoxy-7β, 10αH, 11βH-spiroax-4-ene (3) and agripilol C (4). Compounds 1-4 were separated and purified by silica gel, RP-18, Sephadex LH-20 column chromatography. Their structures were elucidated on the basis of extensive spectroscopic analysis including IR, MS, 1D and 2D NMR experiments.

9.Application of low molecular weight heparin in induced hemodialysis during acute renal failure

Objective To observe the anticoagulant effect of low molecular weight heparin on induced hemodialysis in patients with acute renal failure.Method One hundred and eight patients with acute renal failure treated with induced hemodialysis were randomly divided into low molecular weight heparin(LMWH)group and unfractionated heparin(UFH)group.A bolus disc of UFH was given at first and then maintained by continuous infusion in UFH group,whereas a single bolus dose of LMWH with 2000AFXa IU to 4000AFXa IU in LMWH group.Results Anticoagulant effect between LMWH and UFH did not show significant discrepancy during induced hemodialysis.The bleeding from internal jugular vein catheter increased in the UFH group much more than that in the UFH group was significantly higher than that in the LMWH group.Anti-FXa blood levels were significantly higher in LMWH group than in UFH group.Conclusions LMWH has minor influence on aPTT and TT,while its anticoagulation effect approximates to that of UFH.LMWH represents a realistic alternative agent UFH in acute renal failure induced hemodialysis.
