3.Measurement and analysis of acupuncture manipulation.
Chinese Acupuncture & Moxibustion 2007;27(11):829-831
Acupuncture manipulation is an important part of the theory of acupuncture and moxibustion, and it is a basic skill for acupuncture physicians. In order to further disclose the essence and character of acupuncture manipulation, the scholars collect in time acupuncture manipulations by the multi-subject crossing methods for qualitative, quantitative analysis and study, and make its analysis of dynamics and kinematics with the riper engineering. However, the current study and analysis on acupuncture manipulation is not deep and not systemic, so the thesis proposes a research way which should create a database of the acupuncture manipulation, so as to provide reliable evidences for studies of correlativity between acupuncture manipulation and acupuncture effects, and establish the theoretical basis for the study of the quantification and standardization of acupuncture manipulation.

Powder X-ray diffraction (PXRD) technology combined with cluster analysis method was used to classify 75 batches of crystalline ceftriaxone sodium into subtypes, the crystalline characteristics of each subtype were measured with scanning electron microscope (SEM). By comparing some parameters of these subtypes correlated to crystallization process of ceftriaxone sodium, such as salification rate, water content in different subtypes, as well as by studying different lattice stabilities, different compatibilities with rubber closures during accelerated stability tests, the key point to improve the quality of domestic ceftriaxone sodium was disclosed. The results of this paper indicated that the fine structure of the products could be controlled well by improving the salification and crystallization process. As a result, the subtype II of ceftriaxone sodium with high stability can be produced.

6.Effects of N-acyl glucosamines on GABA uptake activity of GABA transporter 1
Journal of China Pharmaceutical University 2016;47(2):228-234
In order to screen the inhibitors of GAT1 protein, the effects of synthetic N-acylglucosamines on the GABA uptake activity of GAT1 were examined in the HEK-293 cell model stably expressing GAT1 by performing GABA uptake assay. And since the N-linked oligosaccharides, especially their terminal sialic acid of GAT1 are necessary for the function of GAT1, their effects on the modification of N-glycans of GAT1 were also determined by the quantitative analysis of the sialic acid of GAT1. The results showed that 10 mmol /L 3-O-methyl-N-acetylglucosamine(3-O-Met-GlcNAc)reduced the GABA uptake activity of GAT1 to 53%(P< 0. 01)through inhibiting the sialylation of GAT1(66. 8%). And the GABA uptake activities were decreased to 54%, 63%, 63% and 67%(P< 0. 01), respectively, by the treatments of 10 mmol /L N-propionylglucosamine(GlcNProp), N-hexanoylglucosamine(GlcNHex), N-cycloproylgormylglucosamine(GlcNCyclo)and N-acetamidoacetylglucosamine(GlcNAc-acetamido)through inhibiting N-glycan trimming. These results indicate that the analogues of glucosamines have great potential in the development of inhibitors of GAT1 activity or sialic acid biosynthesis.

Abstract: Objective To understand the prevalence and influencing factors of major chronic diseases among Kirgiz residents in Aheqi County, Xinjiang, and to provide theoretical basis for the prevention and control of chronic diseases in this area. Methods The data of residents in Aheqi County, Kizilsu Kirgiz Autonomous Prefecture, Xinjiang in 2020 for health checkup were selected, and the permanent residents of Kirgiz nationality aged ≥18 were selected as the survey objects, and the prevalence of major chronic diseases and their influencing factors were analyzed by Logistic regression model. Results A total of 21 935 patients were enrolled, and 10 602 patients suffered from chronic diseases with a prevalence rate of 48.33%, including 4 929 cases of hypertension with a prevalence rate of 22.47%, 4 238 cases of obesity with a prevalence rate of 19.32%, 3 610 cases of dyslipidemia with a prevalence rate of 16.46%, 1 632 cases of anemia with a prevalence of 7.44%, and 1 236 cases of type 2 diabetes with a prevalence of 5.63%. The prevalence of hypertension, obesity, dyslipidemia and type 2 diabetes increased with age, while the prevalence of anemia decreased with age (P<0.01). The prevalence of hypertension dyslipidemia and type 2 diabetes was higher in males than in females, and the prevalence of obesity and anemia was higher in females than in males, with statistical significance (P<0.01). Logistic regression analysis showed that 30-<40 years old (OR=1.836, 95%CI: 1.565-2.034), 40-<50 years old (OR=2.916, 95%CI: 2.613-3.255), 50-<60 years old (OR=5.244, 95%CI: 4.651-5.913), 60 years old and above (OR=10.866, 95%CI: 9.533-12.385), government personnel (OR=1.789, 95%CI: 1.405~2.279), professional and technical personnel (OR=1.774, 95%CI:1.372-2.295), the office staff (OR=1.923, 95%CI: 1.418-2.607) occasional alcohol consumption (OR=1.157, 95%CI: 1.055-1.270) and occasional exercise (OR=1.498, 95%CI: 1.238-1.812) were risk factors for chronic disease. Conclusion The prevalence of chronic diseases among residents in Aheqi County of Xinjiang is at a low level. Local health institutions have strengthened health education and publicity on multiple chronic diseases and the prevention and control of anemia among young and middle-aged women.

OBJECTIVEThe present study is concerning qualitative and quantitative detection of Poria cocos quality based on FT-near infrared (FT-NIR) spectroscopy combined with chemometrics.
METHODThe Poria cocos polysaccharides contents were determined by UV. Transmission mode was used in the collection of NIR spectral samples. The pretreatment method was first derivation and vector normalization. Then principal component analysis (PCA) was used to build classification model and partial least square (PLS) to build the calibration model.
RESULTThe results showed that conventional criteria such as the R, root mean square error of calibration (RMSEC), and the root mean square error of prediction (RMSEP) are 0.944 0, 0.072 1 and 0.076 2, respectively. The misclassified sample is 0 using the qualitative model built by PCA.
CONCLUSIONThe prediction models based on NIR have a better performance with high precision, good stability and adaptability and can be used to predict the polysaccharose content of Poria cocos rapidly, which can provide a fast approach to discriminate the different parts of Poria cocos.

OBJECTIVETo investigate the mechanism of senescence delay of human diploid fibroblast (2BS) and protecting telomere length by epimedium flavonoids (EF).
METHODSThe drug sera of EF were used to treat the 2BS. The population doublings of 2BS cells were observed, the mRNA expression of p16 gene were determined by fluorescence real-time quantitative RT-PCR, the telomerase activation of 2BS cells were determined by TRAP-Hyb, the total retinoblastoma (Rb) and phosphorated Rb protein content were detected by ELISA, the telomere length of 2BS cells were determined by telomere restriction fragment (TRF) Southern blot assay.
RESULTSEF could significantly extend the population doublings of 2BS cells, the expression of p16 mRNA was decreased and the content of phosphorated Rb protein were increased by EF. The telomere lengthening of 2BS cells were improved by EF, but the telomerase was not activated.
CONCLUSIONIn senescence human fibroblasts 2BS cells, p16 gene mRNA expression increased, content of phosphorated Rb protein decreased and the telomere length of 2BS shortened, EF might delay the aging of cells through inhibiting the p16 gene expression, promoting the production of phosphorated Rb protein and to protect the length of telomere, but not activating the telomerase.
