Objective To examine the expression of macrophage migration inhibitory factor(MIF)in lung tissues of rats with experimental pulmonary fibrosis and to discuss its effects on pulmonary fibrosis.Methods36 male SD rats were randomly divided into two groups:the pulmonary fibrosis group and the control group.Rat pulmonary fibrosis was induced by an intratracheal injection of bleomycin.The following tests were undedaken: HE and Masson staining of lung section,immunohistochemical staining of protein of MIF in the lung,RT-PCR of MIFmRNA in the lung.ResultsCompared with the control group,the mRNA and protein expression of lung MIF on day 7 to 28 in pulmonary fibrosis increased and reached the peak on day 14 after an intratracheal injection of bleomycin(P

Traditional Chinese medicine (TCM)has certain advantages in the treatment of chronic liver diseases,especially chronic inflam-matory injury and liver fibrosis.However,the studies on medicinal components and action mechanism of TCM prescriptions are confronted with a great challenge.The computational pharmacology methods used in recent years,mainly including virtual screening technique,omics technologies,and network pharmacology approaches,as well as their application in TCM research,are presented;given the previous re-search work,the progress in research on active ingredients and action mechanism of TCM prescriptions using computational pharmacology methods is reviewed.It is thought that computational pharmacology techniques combined with conventional molecular biological and pharma-cological methods are an important research strategy for the material basis and action mechanism of TCMprescriptions for treating chronic liv-er diseases.

7.Specific anti-leukemic cell effect mediated by dendritic cells pulsed with acute myelogenous leukemia antigen in vitro

Objective To explore the effective method for in vitro culture of the dendritic cells(DCs) and the specific anti-leukemic cell effect mediated by dendritic cells pulsed with acute myelogenous leukemia antigen. Methods Bone marrow mononucleas cells (BMMNCs) isolated from AML patients were induced to undergo differentiation with 500 ng/ml A23187 and pulsed with AML antigen. After 96 h, DCs and T cells were co-cultured for 5 to 7 days. The cytotoxic activity of cytotoxic T lymphocyte(CTL) to AML were detected with MTT colorimetry. Results BMMNCs isolated from AML patients treated with 100 ng/ml rhGM-CSF in combination with 500 ng/ml A23187 for 96 h exhibited typical morphology of DCs with rapidly increased expression of CD1a and CD83 (P<0.01). Dendritic cells pulsed with acid eluted tumor antigen peptides group displayed the strongest cytotoxic activity of CTL to AML. There was a significant difference between two groups(P<0.01). Conclusion BMMNCs isolated from AML patients can be successfully induced to DCs with rhGM-CSF and A23187 and dendritic cells pulsed with acid eluted tumor antigen peptides group had the strongest cytotoxic activity of CTL to AML.
8.Preparation and dissolution rate determination of oridonin solid dispersion

Objective Oridonin is a poorly water soluble drug with low bioavailability.The study aimed to prepared improve the dissolution rate of oridonin by preparation of oridonin solid dispersion with poloxamer. Methods The oridonin solid dispersion was prepared by dissolvent method with poloxamer188 as the carrier and determined by DSC and powder X-ray diffraction.The dissolu-tion rates in different pH dissolution mediums of pure drug, physical mixture and solid dispersion were determined by HPLC method. Results Oridonin exists as an amorphous state in the solid dispersion.Compared with the pure drug and physical mixture, the orido-nin solid dispersion was improved greatly without the pH influence on the drug release.The dissolution rate of oridonin solid dispersion in water, pH 6.8 PBS and pH 1.2 HCl were (92.6 ±4.2)%, (93.1 ±3.5)%and (94.4 ±2.9)% respectively. Conclusion Taking inpoloxamer188 as the carrier, the oridonin solid dispersion has successfully developed its dissolution rate.

10.Application of magnetic resonance imaging in diagnosis and treatment of pelvic neoplasms

Objective To study the value of magnetic resonance imaging(MRI) in the diagnosis and treatment of pelvic neoplasms. Methods The patients who were suspected of malignant pelvic neoplasms were performed MRI.The surgical approach was determined according to the MRI results.The MRI results were compared with operation findings and pathological reports. Results The MRI results were consistent with operation findings and pathological reports in 22 cases highly suspected of pelvic neoplasms and 15 cases with abnormal endometrial echo.Therapeutic regimen was designed for 5 cases with uterine cervix cancer according to MRI stages,and favourable surgical curative effects were obtained. Conclusion MRI provides a reliable guideline for surgical approach and may improve the outcomes.
