1.Characteristics in frequency and phenotype of myeloid and plasmacytoid dendritic cells in chronic HCV infection

Objective To explore the frequencies and phenotype of myeloid and plasmacytoid dendritic cells (mDC and pDC) in chronic HCV infection and to investigate the relationships between DC frequencies and HCV viral load and serum ALT level. Methods PBMC were isolated from chronic HCV infected patients and healthy control. Multi-color flow cytometry was used to analyze the frequencies and surface marker expression on mDC and pDC. The relationship between DC frequencies and viral load and ALT level was also calculated. Results In comparison with healthy control, frequencies of mDC and pDC in chronic HCV infection were significantly decreased (0. 37 ± 0. 19 and 0. 19 ± 0. 12 vs 0. 51 ± 0. 18 and 0. 29 ± 0.13, P<0.05). The frequency of mDC was negatively correlated with HCV viral load (r= -0.5878, P < 0. 0001 ) and serum ALT level ( r = - 0. 4628 , P = 0. 003 ). Both costimulatory markers ( HLA-DR, CD83, CD86, and CD40) and coinhibitory marker (PD-L1) expression on mDC and pDC in HCV infection were increased (P<0.01 for costimulatory marker, P<0.05 or F<0.01 for coinhibitory marker). Conclusion The frequencies of mDC and pDC in chronic HCV infection were decreased, while the expression of costimulatory markers and coinhibitory marker were increased or not decreased in HCV infection. The decreased frequency of mDC was probably related to persistance of HCV infection.
3.Clinical study of Osteoset~ synthetic bone transplants and internel fixations on calcaneus fracture

[Objective]To evaluate curative effects of Osteoset~ synthetic bone transplants and internel fixations on calcaneus fracture.[Method]Twenty-three patients with calcaneus fracture,who were treated by Osteoset~ synthetic bone transplants and internel fixation,were followed up by plain X-Rays and JOA functional questionnaires.Fracture patterns were based on Sanders methods.[Result]A series of 23 patients,7 of which were sanders Ⅱ,15 were sanders Ⅲ,3 were sanders Ⅳ,were treated with ORIF and Osteoset~ implants.The preoperative Blher's angle was 4.8 degrees(standard deviation 6.6 degrees).Blher's angle increased to a mean of 26.8 degrees(SD 8.6 degrees) postoperatively.In the first 3 months after surgery,a mean decrease in the Blher's angle of 0.4 degrees(SD 2.27 degrees) occurred.One month after surgery osteoset synthetic bone transplants,which was replaced by new bone,were gradually absorbed and degraded.JOA questionnaires suggested that the whole effective rate was 92 percent.[Conclusion]Osteoset synthetic bone transplants and internel fixations has good therapeutic effects on patients with comminuted calcaneus fracture.Osteoset~ synthetic bone has the same effects as autogenous bone.
4.Determination of Five Residual Solvents in Vidarabine Monophosphate by Headspace Gas Chromatography

OBJECTIVE:To establish a method for the determination of ethanol,acetonitrile,dichloromethane,ethyl acetate, pyridine in vidarabine monophosphate. METHODS:Headspace GC was performed on the column of Agilent DB-624,programmed temperature,inlet temperature was 200 ℃,the detector was flame ionization detector,detecting temperature was 250 ℃,nitrogen was carrier gas,flow rate was 3 ml/min,split ratio was 1∶1,the top bottles equilibrium temperature was 100 ℃,and equilibrium time was 45 min,injection volume was 1 ml. external standard was used for quantitative analysis. RESULTS:The peaks of five re-sidual solvents could be completely separated from the other peaks respectively,The linear rang was 24.7-296.3 μg/ml for ethanol (r=0.999 6)、1.9-23.2 μg/ml for acetonitrile(r=0.999 0),2.8-33.6 μg/ml for dichloromethane(r=0.998 0),24.7-295.9 μg/ml for ethyl acetate(r=0.999 5),1.0-11.9 μg/ml for pyridine(r=0.998 6);RSDs of precision and reproducibility tests were lower than 4.35%;recoveries were 102.4%(RSD=2.0%,n=9)、102.1%(RSD=3.4%,n=9)、105.5%(RSD=4.8%,n=9)、100.3%(RSD=4.8%, n=9)、98.3%(RSD=4.0%,n=9). The minimum quantifation limit was 0.304 4-0.988 0 μg/ml and the minimum detection limit was 0.101 5-0.329 3 μg/ml. CONCLUSIONS:The method is simple,accurate and reproducible,and can be used for the determination of residual solvents in vidarabine monophosphate.
5.Effect of “Shuangguyitong” Acupuncture Therapy on Serum Interleukin-6 of STZ-induced Diabetic Rats

Objective To investigate the effect of “Shuangguyitong acupuncture therapy on serum interleukin-6 of streptozotocin (STZ) induced diabetic rats. Methods Among seventy-three 180~220 g Wistar rats, thirteen were assigned as normal control group (group A) and sixty were induced diabetes by injecting STZ into celiac cavity. The diabetic rats were randomly divided into model group(group B), “Yitong” treatment group (group C), “Shuangguyitong” acupuncture group (group D) and common acupuncture group (group E). 6.Direct Molecular Analysis of Garlic Using Internal Extractive Electrospray Ionization Mass Spectrometry

Theapplicationofinternalextractiveelectrosprayionizationmassspectrometry(iEESI-MS)was extended to direct molecular analysis of garlic tissues. By obviating time-consuming sample preparations, fragile active garlic substances such as organosulfur compounds ( e. g. , alliin, allicin ) were successfully detected and identified via collision-induced dissociation ( CID) , together with amino acids ( e. g. , arginine) and saccharides ( glucose, polysaccharides) . Mass spectral fingerprints of different kinds of garlic cloves, as well as various post-treatment ones, were further processed via principal component analysis ( PCA) to better visualize the differences. Our experimental results indicated that iEESI-MS allowed rapid recognition of metabolic changes in the garlic tissue subject to various external stimuli. The merits included simplicity of analysis, high speed ( less than 2 min per sample ) , good specificity, and minimal disturbance to the bioactivity of analytes.
7.Clinical research of integrated traditional Chinese medicine in the treatment of infertility resulted from ovulation dysfunction

Objective To observe the clinical efficacy of integrated traditional Chinese and western medicine in the treatment of infertility resulted from ovulation dysfunction.Methods 106 patients with ovulation dysfunction were divided into observation group(n =56)and control group(n =50)in stratified randomization.Both two groups were given clomiphine to induce ovulation.Futhermore,the observation group was added Chinese herb recipe.Both two groups were treated for 3 menstruations.The basic body temperature,follicle development,endometrium depth, ovulation and pregnancy were observed.The serum levels of E2,LH and FSH were detected.Results There was no difference in the basic body temperature between the two groups(P >0.05).After treatment,the basic body tempera-ture of the observation group was obviously superior to that of the control group(χ2 =0.627,P =0.049),there was no difference in the endometrium depth between the two groups(P >0.05).After treatment,the endometrium in the observation group was significantly better than before treatment and that in the control group(t =5.132,P <0.05). After treatment,the serum levels of E2 ,LH and FSH in the observation group were all improved more obviously than those in the control group(t =6.489,5.167,6.035,all P <0.05).The efficacy of the two groups was higher than that of the control group(χ2 =6.049,P =0.048).Conclusion The method of integrated traditional Chinese and western medicine can promote the development of follicle and endometrium,improve levels of E2,LH and FSH,enhance ovula-tion rate and pregnancy rate in the treatment of infertility resulted from ovulation dysfunction,which is worthy of clini-cal promoting.
8.Effect of brain tissue extract after acupuncture preconditioning on cerebral ischemia-reperfusion injury in rats

BACKGROUND: According to the thought "prevention of diseases", a conception of "strengthening the vital by acupuncture preconditioning (AP)" is suggested recently.OBJECTIVE: To investigate the effect of brain tissue extract after AP on cerebral ischemia-reperfusion injury.DESIGN: Random control experiment.SETTING: Institute of Acupuncture & Moxibustion and Massage, and Department of Anatomy and Embryology, Hubei College of Traditional Chinese Medicine.MATERIALS: Totally 102 adult Wistar rats were selected during the experiment, which was completed in the Institute of Acupuncture & Moxibustion and Massage of Hubei College of Traditional Chinese Medicine from September 2003 to July 2004. Among them, 20 rats were used to prepare cerebral tissue extract, and another 82 were used in the subsequent experiment.METHODS: The brain tissue extract was obtained from the rats which were given electroacupuncture at Shenshu (BL-23) and Baihui (DU-20).Totally 82 rats were randomly divided into 6 groups. Five rats in blank control group were taken as blank control, 15 in sham-operation control group were performed with sham operation, 16 in cerebral ischemia-reperfusion control group with cerebral ischemia-reperfusion, 16 in saline control group with the injection of saline intravenously firstly and then cerebral ischemia-reperfusion modeling, 15 in normal cerebral tissue extract control group with the injection of normal cerebral tissue extract intravenously firstly and then cerebral ischemia-reperfusion modeling, and 15 in AP cerebral tissue extract group with the injection of cerebral tissue extract intravenously firstly and then cerebral ischemia-reperfusion modeling.Intravenous injection was performed 2 hours and 1 hour before cerebral ischemic modeling, and each rat was injected twice with 1 mL/time. Brain tissue of the rats was taken ont 1, 3, 7 days after reperfusion respectively (or each group was divided into 3 subgroups with 5 rats in each) except those in blank control group. The blain tissue of rat in each group was selected at the relevant time points, and embedded with paraffin and cut into pieces. Cerebral histopathology was observed under the light scope (× 400)and the survival neurons were counted whose area was layer y of region Ⅰ in parietal cortex (inner cone).cortex.RESULTS: Two rats died during the experiment in cerebral ischemiareperfusion control group and saline control group respectively. Another Except blank control group and sham operation group, the brain sections of different time points in other groups showed scattering ischemic anoxic Count of survival neurons in layer Ⅴ of area I in parietal cortex: One day after reperfusion, survival nerve density of the brain ischemic reperfusion model group [(338.8±31.2)/mm2] was significantly lower than that of blank control group [(753.4±60.8)/mm2] (F=129.36, P < 0.05); degeneration of the nerves became worse after reperfusion for 3 days and 7 days, but with no significant difference (F=1.76, P > 0.05). There was no significant difference between the saline control group, normal brain tissue extract group and brain ischemic reperfusion model group at different time points (F=1.76, P > 0.05). Survival neuron density in group of brain tissue extract after AP at the three time points was significantly higher than that in brain ischemic reperfusion model group [(438.1±41.0), (338.8±31.2)/mm2,(296.4±27.1), (124.8±13.4)/mm2; (269.5±30.4), (1.324±0.157)/mm2;F=129.36, P < 0.05].CONCLUSION: Injection of the brain tissue extract after AP at Shenshu (BL-23) and Baihui (DU-20) into the celiac cavity of rats could obviously reduce the subsequent neuron loss induced by brain isehemia-reperfusion and protect brain from ischemia-reperfusion injury.
9.An analysis of 49 cases of patients with sysmetic lupus erythematosus and pregnancy

Objective To investigate the pregnancy opportunity and outcome of patients with sysmetic lupus erythematosus (SLE).Methods We carried out a retrospective analysis of the clinical information of patients who suffered from SLE complicated with pregnancy and were hospitalized to People's Hospital of Peking University from December of 1992 to February of 2012.Chi-square test were used for statistical analysis.Results Forty-nine cases of patients with SLE complicated with pregnancy had 52 pregnancies in total.In 27 cases of planned pregnancies,24 cases(89% )resulted in live births and 5 cases( 18% ) had flares during pregnancy.Of the 24 live births,3 cases( 12%)were premature deliveries and 4 cases(17%) had low birth weight infants.In 25 cases of unplanned pregnancies,12 cases (48%) resulted in live births and 20 cases (80%) had flares during pregnancy.Of the 12 live births,6 cases(50% ) were premature deliveries and 6 cases (50%) were low birth weight infants.The patients in the planned pregnancy group tended to have flares during pregnancy more frequently than those in the unplanned group (P<0.01),the former were more likely to have live births than the latter (P<0.01) and were less likely to have premature delivery (P<0.05).Conclusion Patients with SLE should have planned pregnancy and need collaborative supervision of both rheumatology and immunology department and obstetric department to improve live birth rate,decrease premature delivery and the activity of SLE during pregnancy.
