1.Effects of Oral Administration and Intraperitoneal Injection of Total Polysaccharide of Sijunzi Decoction on Mouse Immune Function

Objective: To study difference of the immune response between oral administration and intraperitoneal injection of total polysaccharide of Sijunzi decoction (SJTP) in mice. Methods: The immunosuppressed mice model was established by intraperitoneal injection of cyclophosphamide (CY). The effects of SJTP on the weight of immune organs,hemolysin response and delayed type hypersensitivity (DTH) were observed,and the influences on normal mouse spleen lymphocytic proliferation of mice were also studied. Results:SJTP can remarkably counteract the immunosuppression induced by CY, decrease the weight loss immune organs,increase the hemolysin response and DTH,and promote the proliferation of lymphocytes.The effect of oral administration is superior to intrapertoneal injection.Conclusion:SJTP can improve the humoral and cellalar immunity of mice.The immune activities resulting from oral administration of SJTP have some special characteristics,and its possible mechanism is also discussed.
2.Study on the correlation between contrast-enhanced ultrasonographic characteristics and microvessel density in breast cancer

Objective To investigate the relation of contrast-enhanced ultrasound ( CEUS ) characteristics and microvessel density ( MVD ) in the breast cancer .Methods From October 2010 to February 2012, 45 cases of patients with breast cancer were studied in Yantai Yuhuangding Hospital , Affiliated to Qingdao University Medical College .All lesions were examined by CEUS before surgery .The blood perfusion parameters such as rising time (RT),peak intensity(PI),time to peak(TTP),wash-in slope (WIS) and mean transit time ( MTT) were obtained by time-intensity curve ( TIC).Immunohistochemical staining for anti-factor CD34 was performed on surgery specimen and the MVD was evaluated .The CEUS characteristics and blood perfusion parameters between different MVD groups of breast cancer were compared.Results In 45 cases of breast cancer,mean MVD was(47.6 ±14.2)/high power field(HPD). Twenty-one cases(46.7%) were classified as high MVD group(MVD>48/HPD) and 24 cases(53.3%) were classified as lower MVD group ( MVD≤48/HPD) .Besides two cases without contrast agent perfusion in CEUS imaging, blood perfusion was observed in 43 cases (95.6%).Heterogeneous enhancement was observed in 25 cases(55.6%).Local blood perfusion defect was observed in 27 cases(60.0%).Irregular shape was observed in 37 cases(82.2%).Centripetal enhancement was observed in 25 cases(55.6%). Penetrating surrounding vessels was observed in 32 cases(71.1%).Poorly-defined margin was observed in 34 cases(75.6%).Compared with the surrounding normal breast tissue ,RT and TTP of center region of neoplasms was shorter[(9.3 ±3.3)s vs (11.1 ±3.7)s and (25.3 ±5.9)s vs (27.5 ±6.4)s],PI was higher[(12.1 ±4.6)dB vs (9.2 ±2.8)dB],WIS was higher(1.0 ±0.4 vs 0.8 ±0.3) and differences were significant(t =-3.001, -4.785,6.987 and 5.438,all P <0.05).Compared with center region of neoplasms,TTP of periphery region of neoplasms was shorter [(22.2 ±6.0)s vs (25.3 ±5.9)s],PI was higher[(15.4 ±5.1)dB vs (12.1 ±4.6)dB],WIS was larger(1.3 ±0.5 vs 1.0 ±0.4) and differences were significant(t=-2.839,3.194 and 3.151,all P<0.05).The detection rate of blood perfusion defect and heterogeneous enhancement was higher in the high-MVD group than in the low-MVD group(χ2 =4.0179 and 7.2024,both P <0.05).While the enhancement shape,margin and penetrating vessels showed no statistical difference between the two groups .Breast neoplasms in the high-MVD group had higher PI than those in the low-MVD group[(18.2 ±5.6)dB vs (12.9 ±3.1)dB,t=-3.738,P<0.05].While the RT, TTP and WIS showed no statistical difference between the two groups ( t=-0.798,-0.760 and -0.378, all P>0.05).Conclusion CEUS characteristics of breast lesions were associated with MVD ,which may reflect the microvessel distributional characteristics of neoplasm and may be one of bases used to evaluate neoplasm angiogenesis .
3.Three different strategies of urine drainage following hypospadias surgery: clinical nursing and observation.

OBJECTIVETo study 3 different strategies of urine drainage following hypospadias urethroplasty, the clinical nursing in their application, and their effects.
METHODSWe retrospectively analyzed the clinical data of 595 cases of hypospadias treated by urethroplasty. After surgery, 133 of the patients underwent urine drainage by suprapubic cystostomy (group A), 202 by urethral stent- tube indwelling (group B), and 260 by early initiative micturition with the urethral stent-tube (group C). All the patients received routine postoperative nursing care required for hypospadias repair.
RESULTSOperations were successfully completed in all the cases. Group C showed a remarkably shorter hospital stay and lower incidence rates of urinary fistula and urethral stricture than groups A and B (P<0.05), but there were no significant differences in the three indexes between A and B (P<0.05).
CONCLUSIONFor urine drainage following hypospadias repair, early initiative micturition with the urethral stent-tube can significantly reduce postoperative complications, decrease difficulties and workload of nursing care, and shorten the hospital stay of the patient.

4.Mechanism of GLI3 gene transcription regulation in idiopathic congenital talipes equinovarus.

OBJECTIVETo investigate the mechanism of transcription regulation of GLI3 gene in idiopathic congenital talipes equinovarus.
METHODSpGL3-Gli3 luciferase report vectors were constructed, and the activity of Gli3 promoter was explored. A P-Match software was used to analyze the sequence upstream of the transcription start site of rat Gli3 gene, which was subsequently verified with chromatin immunoprecipitation assay (CHIP) and electrophoretic mobility shift assay (EMSA). Expression of the Gli3 gene was analyzed in L6 cells transfected with Hoxd13 small interference RNA(siRNA) and Hoxd13 expression vectors.
RESULTSThe 5' region of rat Gli3 gene contains two potential binding sites for the Hoxd13 protein. CHIP and EMSA assays both confirmed that Hoxd13 can directly bind with site 2. As shown in L6 cells, expression of Gli3 may be enhanced with silencing of Hoxd13, whilst exogenous expression of Hoxd13 can down-regulate transcription of Gli3.
CONCLUSIONHoxd13 can directly regulate the expression of Gli3 gene through a Hoxd13 binding site in the limb of rat embryo.

5.Effects of sodium butyrate on growth, apoptosis and telomerase activity in Hep-2 cells.

OBJECTIVETo study the effects of sodium butyrate (SB) on growth, apoptosis and telomerase activity in Hep-2 cells.
METHODSGrowth inhibition effect of SB on Hep-2 cells was assessed by methyl thiazolyl tetrazolium (MTT) assay. Morphological alterations were observed by electronic microscope. Cell apoptosis was confirmed by terminal deoxynucleotidyl transferase-mediated dUTP nick-end-labeling (TUNEL) method, DNA fragmentation and flow cytometry (FCM). Cell cycle was analyzed by FCM. Telomerase activity was examined by telomeric repeat amplification protocol (TRAP)-silver staining. The expression status of telomerase subunits was analyzed by reverse transcription-polymerase chain reaction (RT-PCR).
RESULTSA time-and dose-dependent inhibition was detected in cells treated with SB. Typical morphological changes of apoptotic cells were observed under electronic microscopy. The characteristic DNA fragmentation of apoptotic cells was detected by agarose gel electrophoresis. Apoptosis and the changes of cell cycle were confirmed by TUNEL method and FCM. The apoptosis indexes of the cells before treatment and at 72 h after SB (2.5 mmol/L) treatment were 2.27 +/- 1.18 and 33.50 +/- 2.75 respectively, the apoptosis rates were 2. 86% and 31. 28% respectively, the proportion of the cells at G0/G1 stage were 50.38% and 70.88% respectively, the proportion of the cells at S stage were 27.40% and 8.20% respectively, and the proliferation indexes of the cells were 49.62% and 29.12% respectively. Telomerase activity and expression level of human telomerase reverse transcriptase (hTERT), the key subunit of telomerase, decreased after SB treatment. No significant changes were observed in the expression of human telomerase RNA (hTR) and human telomerase associated protein (hTP1), the other two subunit of telomerase.
CONCLUSIONSB could inhibit growth of Hep-2 cells and induce apoptosis in the cells, and inhibit telomerase activity by decrease expression level of hTERT.

6.Short and long-term efficacy of focused ultrasound therapy for vulva dystrophy

7.Small interfering RNA targeting C-erbB-2 gene increases the radiosensitivity of lung adenocarcinoma cells in vitro.

OBJECTIVETo investigate the effect of small interfering RNA (siRNA) targeting C-erbb-2 oncogene on the radiosensitivity of C-erbb-2-overexpressing lung adenocarcinoma cell line.
METHODSFour pairs of siRNA targeting the coding sequence of C-erbb-2 mRNA were synthesized and their interference effects were evaluated using quantitative real-time fluorescence PCR. The siRNA with the best interference effects was transfected into Calu-3 cells, which were then exposed to 2 or 5 Gy irradiation, with the cells with transfection or irradiation alone as the control groups. The cell apoptosis after the treatment was detected using annexin V-FITC Kit.
RESULTSThe apoptosis rate of the Calu cells was 7.767-/+0.551 in the blank group, 14.400-/+1.114 in the interference group, 11.867-/+0.737 in 2 Gy irradition group, 23.000-/+1.664 in 2 Gy irradiation + interference group, 16.100-/+0.624 in 5 Gy irradiation group, and 27.900-/+1.709 in 5 Gy irradiation+interference group.
CONCLUSIONThe siRNA targeting C-erbb-2 gene can enhance the radiosensitivity of Calu-3 cells to gamma-ray and increase their apoptosis rate following gamma-ray exposure in vitro.

8.Possible role of GLI3 gene in the pathogenesis of idiopathic congenital talipes equinovarus.

OBJECTIVETo investigate the relationship between GLI3 gene and pathogenesis of idiopathic congenital talipes equinovarus (ICTEV).
METHODPotential mutations in the coding region of GLI3 were detected among 84 patients with ICTEV by denaturing gradient electrophoresis. Expression of GLI3 in the ICTEV patients' disease tissues was assessed by reverse transcription PCR. Following generation of rat model for ICTEV, mRNA and protein levels of GLI3 were evaluated by real-time PCR and immunohistochemistry and Western blotting.
RESULTSNo mutation was found in exons 1 - 8 and 13 of GLI3 gene among the 84 ICTEV patients. No expression of GLI3 gene was detected in the flexor hallucis longus of ICTEV patients or normal controls. Expression of Gli3, in terms of both mRNA and protein, was stronger in the hindlimb of ICTEV rat embryos compared with normal controls.
CONCLUSIONMutation in the coding region of GLI3 may not be responsible for the occurrence of ICTEV. However, there may still be connection between abnormal expression of the gene and pathogenesis of ICTEV.

9.Analysis on the nursing needs and influencing factors of the elderly in penury countries

0bjective To explore the nursing needs of the elders in penury countries and the influencing factors in order to provide suggestions for the improvement of QOL of the elder,and to reach health aging.Methods A total of 623 elders were selected from 8 villages of two penury countries,and the two was by random sampling from five penury counties of Lvliang City.Their nursing needs and influencing factors were assessed by interview and questionnaires.Results The score of nursing needs in penury countries was at a high level,sickness prevention was(3.40 4±0.56),health promotion was(3.30±0.70),nursing of disease was (3.56 4±0.52).r111e main influencing factors were illness,educational background,marital status.sex and average income per month.Conclusions We should aim directly at their features and take effective measures from society-family.individual level to satisfy their nursing needs.
10.Comparison of the curative effect between delayed PCI and medical therapy on ST-segment elevation acute myocardial infarction

Objective To investigate and compare the curative effect between delayed percutaneous coronary intervention (PCI) for patients with acute myocardial infarction presenting 12-24 hours from symptom onset and medical therapy on acute myocardial infarction patients presenting with ST-segment elevation (STEMI). Methods Using a prospective,open,parallel,controlled research approach,186 patients with STEMI were divided into delayed PCI group(n=89),which received PCI within 12-24 hours after STEMI and medical therapy group(n=97),which received medical therapy after STEMI. All patients were followed up 1-6 months with average follow-up (5.6 ± 1.4) months. Data of hospitalization period, the cardiac structures detected by echocardiography such as left atrial diameter (LAD), left ventricular diastolic diameter(LVDd),left ventricular ejection fraction LVEF,left ventricular fractional shortening(LVFS),composite end point events and major adverse cardiac events(MACE)were compared between the two groups.Results Compared with medical therapy group, the hospitalization cycle was significantly shorter in delayed PCI group. Data of the LAD and LVDd were significantly decreased,but LVEF and LVFS were increased in delayed PCI group compared with those of medical therapy group at 30 d and 6-month follow-up. The incidence of MACE and composite end point events were significantly less in delayed PCI group than those of medical therapy group (P<0.05). Conclusion Delayed PCI treatment can decrease the time of hospital stay and decrease the incidence rates of MACE and composite end point events,and improve left ventricular function and prognosis of patients.
