1.Model of immune-mediated inner ear disease and efficacy of dexamethasone by systemic application or round window administration
Journal of Shanghai Jiaotong University(Medical Science) 2006;0(01):-
Objective To establish a model of immune-mediated inner ear disease(IMIED) and to compare the efficacy of dexamethasone between systemic application and round window adminstration. Methods The guinea pigs were injected with keyhole limpet hemocyanin(KLH) mixed with complete Freund's adjuvant subcutaneouly in the back, and were boosted with KLH mixed with incomplete Freund's adjuvant two weeks later.Meanwhile,a small piece of gelatin sponge full of KLH was delivered on the round window through operation. Thus established a model of IMIED.Micro-osmotic pumps were used to deliver dexamethasone through the round window(PBS control group also set),and systemic application was implemented by intraperitoneal injection. The efficacy was compared by recording auditory brain-stem response(ABR) before and after the treatment. Results None of the 6 guinea pigs in the control group experienced significant hearing loss, while 22 of the 39 guinea pigs in the experiment group suffered from hearing loss beyond 10 dB.Seventeen guinea pigs with hearing loss no less than 15 dB were divided into three groups randomly and were given different treatment.Those(n=6) treated by local application through the round window enjoyed the efficacy and the mean ABR threshold decreased 13.3 dB.Four out of the 6 treated by systemic application enjoyed the efficacy and the mean ABR threshold decreased 13.7 dB.No efficacy was observed in the PBS group. Conclusion The model of IMIED can be successfully induce by KLH through the round window, and dexamethasone administered through the round window is as efficacious as systemic application.

[Objective]To examine the effects of fast setting calcium phosphate scaffold on human mesenchymal stem cells(MSCs) adhesion and proliferation.[Method]Scaffolds were self-hardened in molds.Three ml human bone marrow aspirates, taken form the iliac crest of normal donors were suspended in low glucose DMEM supplemented with 10% fetal bovine serum.Atfer 72 hours the nonadherent cells were removed by changing medium, and the adherent layer cultured until it reached 80% conference.Cultured cells at passage 3 were used for the experiments.MSCs were seeded on calcium phosphate scaffold. The cell-scaffold constructs were cultured with complete media for 1 week.Scanning electron microscope(SEM)and the terazolium-based colorimetric assay(MTT test)examinations were performed.[Result]When cells were cultured in scaffolds in vitro, the number of cells on scaffold increased with time.SEM showed that the scaffold had a three-dimensional interconnected pore structure. The cells appeared to infiltrate into the macropores of the scaffold and the extracellular matrix components were observed.[Conclusion]This study demonstrates that the fast setting calcium phosphate scaffold has good biocompatibility, cells are able to adhere and proliferate on scaffold specimens, and it is a good substrate candidate for tissue engineered bone substitute.

There have been many methods to prepare fluoroquinolones-loaded sustained-release chitosan microspheres, including precipitation, emulsion crosslinking, spray-drying, and ionic gelation. Each of them has its own merits and disadvantages, but the mechanisms underlying drug delivery are basically similar. The prepared drug microspheres can enhance the capacity of anti-fluoroquinolones-resistant bacteria and are primarily used in the anti-inflammation and anti-infection. This paper introduces the preparation methods and sustained drug delivery mechanisms of fluoroquinolones-loaded sustained-release chitosan microspheres and investigates drug delivery characteristics and applications by analyzing, sorting, and summarizing related manuscripts published recently. Fluoroquinolones-loaded sustained-release chitosan microspheres show satisfactory sustained-release and anti-bacterial properties. Therefore, it is of important scientific research and clinical application significance to develop fluoroquinolones-loaded sustained-release nanometer-sized chitosan microspheres.

Chitosan microsphere containing fluoroquinolones is a kind of novel sustained-released antibacterial agent that possesses the antibacterial and bacteriostatic property of chitosan as well as bactericidal properties of fluoroquinolones. Loading fluoroquinolones in the chitosan microsphere can greatly enhance its antibacterial, bactericidal, anti-inflammation and anti-infection characteristics. This paper reviewes the recent study on different preparation methods of chitosan mierosphere containing fluoroqninolones and discusses its applications.

Npu DnaE intein was used to produce some large proteins, which were difficult to obtain through conventional expression systems.A T7 expression system was described,by which the gene of T7 RNA polymerase is split into two pieces, and each piece fuses with Npu DnaE N-and C-terminal sequences respectively.Functional T7 RNA polymerase is created by mixing the two kinds of fusion constructs in vitro. The approach of split intein-mediated production of large proteins,in theory, readily generalizable to the purification of other large, cytotoxic or membrane proteins.

China Biotechnology 2006;0(03):-
Lactic acid bacteria (LAB) are traditional dairy strains which have a long history of safe use. They are a kind of food-grade microorganisms that can be used in industry to produce lots of food and drink, such as yoghourt. In the last two decades the physiology and genetics of these bacteria have been thoroughly studied. Because of their genetic accessibility and easy to handle, LAB, in addition to their traditional applications, have been extensively developed and used for the expression of heterologous genes. So they have an important foreground in agricultural, medical and many other fields. People have developed a series of food-grade gene expression systems of LAB. LAB, especially their model strain Lactococcus lactis and their most useful food-grade induced expression system--nisin controlled gene expression system NICE and its food-grade inducer nisin, food-grade host, expressing vectors and applications in expressing heterologous proteins of NICE will be introduced.

Objective To investigate in vitro anti-hydatid efficacy on Echinococcus granulosus protoscolex(EgPSC) by using albendazole sulfoxide(ASOX) and its two enantiomeric antipodes, L-ASOX and D-ASOX.Methods Eg protoscoleces were divided into eight groups and cultured in the DMEM culture media under two concentrations(50 ?g/ml and 100 ?g/ml) of ASOX, L-ASOX and D-ASOX respectively. The appropriate controls included(i) a culture containing an equal amount of DMSO and(ii) a culture medium alone. The mortality of EgPSC in each group was daily counted until 100% EgPSC death in some groups. Results Significant difference of EgPSC mortality was found among the three drugs with various concentrations compared to control group(P0.05), but between ASOX group and L-ASOX group(P
