1.miR-21 influences growth of glioma cells by targeting FasL gene

AIM:ToinvestigatetheregulationofmiR-21onFasLexpressionanditseffectonthegrowthand apoptosis in glioma cells , and to evaluate the molecular mechanism .METHODS:Differential expression levels of miR-21 in human glioma U251 cells were achieved by transfecting with miR-21 mimics, miR-21 inhibitor or scramble .The viability and apoptosis of U251 cells were detected by CCK-8 assay and flow cytometry with Annexin V-FITC/PI double staining. The recombination vector pmirGLO-FasL was constructed .Dual-luciferase reporter experiment was performed to validate the target genes of miR-21.The expression vector pcDNA3.1-FasL was also constructed , and the biological activity and regula-tory role of miR-21 in U251 cell apoptosis were analyzed by a restore experiment .RESULTS:Exogenous overexpression of miR-21 increased the viability and decreased the apoptosis of U 251 cells ( P<0.05 ) , while miR-21 inhibitors generated the opposite results (P<0.05).Dual-luciferase reporter assay and restore experiment revealed that miR-21 negatively reg-ulated the expression of FasL gene which was regarded as the target gene , thus decreasing the apoptosis of U 251 cells. CONCLUSION:miR-21 increases the viability of glioma U251 cells, in which FasL may be one of the target genes .
2.Epithelial-mesenchymal transition and stemness of cancer cells

Epithelial-mesenchymal transition (EMT) is an important biological process in embryonic development and tumori-genesis. In this process, epithelial tumor cells can obtain mesenchymal phenotypes and promote tumor invasion and metastasis. EMT is controlled by multiple transcription factors, signal pathways, and microRNAs. EMT and cancer stem cells are closely related. EMT also promotes the self-renewal ability of tumor cells and the overexpression of EMT-related markers in tumor cells with stemness. Some mi-croRNAs can control EMT process and stemness. In this study, recent developments on EMT and cancer stem cells in tumor metastasis were summarized to provide new insights into target therapy of tumor metastasis and recurrence.
3.Feasibility and influence factors of a new silver salt method for determination of arsenic in drinking water

ObjectiveTo investigate the feasibility and influence factors of a new silver salt method for determination of arsenic in drinking water. Methods Arsenic was determined at different reaction temperature,acidity, specification and addition of zinc granular, fill weight of lead acetate cotton, and the effect of these factors on assay results was observed. Arsenic in drinking water was determined in accordance with standard test methods (GB/T 5750.6) of the new silver method. Results The recoveries of the arsenic were 75.3% - 93.6%(F =9.21,P < 0.01 ) with reaction temperature at 10 - 30 ℃, and the best reaction temperature was 25 ℃. The recoveries of the arsenic ranged from 80.3% - 91.6%(F =4.67, P< 0.05) when sulfuric acid was 3.0 - 6.0 ml and the best value was 5.0 ml. The recoveries of the honeycomb structural zinc granular(diameter 0.2 - 0.3, 0.3 - 0.4 mm) and button zinc granular were 87.2%, 90.7% and 83.0%, respectively; the best specification was 0.3 - 0.4 mm. The recoveries of zinc granular weight(3 - 5 g ) were 74.6% - 91.7%, respectively; the best was 5 g. The recoveries ranged from 79.6% - 91.3% with fill weight of lead acetate cotton at 25 - 100 mg, the best fill weight was 50 mg.The recoveries of the arsenic were 90.7%, 92.5%, 81.5% and 74.2% with lead acetate cotton length at 0.5 - 2.0 cm,and the best loose-tight degree was 1.0 cm. The stability time of arsine solution was 5.0, 4.0, 2.5 h with corresponding temperature at 20, 25, 30 ℃, respectively. ConclusionsIn order to ensure precision and accuracy of the measurement, it is necessary to control reaction temperature, acidity, specification and addition of zinc granular free from arsenic, fill weight of lead acetate cotton and loose-tight degree in the reaction system.
4.Intragastric administration of Radix Paeoniae Rubra Extracts versus Taohong Siwu Tang for the serum concentration of peonifiorin in mice

BACKGROUND: Taohong Siwu Tang is made up of Taoren, Honghua,Danggui, Chishao and Chuangxiong. Paeoniflorin is one of the major effective components in Radix Paeoniae Rubra Extracts and Taohong Siwu Tang, and it can promote blood circulation and remove blood stasis and has the effect of anti-acute myocardial ischemia.OBJECTIVE: To compare the serum concentration of paeoniflorin in mice after intragastric administration (i.g.) of Radix Paeoniae Rubra extracts and Taohong Siwu Tang.DESIGE: Rndomized controlled observation.SETTING: Department of Pharmacy and Department of Basic Medicine,Hebei North University.MATERIALS: The trial was performed in the first laboratory of Department of Pharmacy, Hebei North University during February to June 2005.Eighty involved Kunming mice of either gender and of clean grade, with body mass of 18 to 22 g, were purchased from Experimental Animal Center, Institute of Genetics and Developmental Biology, Chinese Academy of Sciences. Taoren, Honghua, Danggui, Chishao, Shengdi and Chuanxiong 6 Chinese herbs were all purchased from Zhangjiakou Institute of Traditional Chinese Medicine.METHODS: ①Blank serum was isolated from eight mice which were fasted for 12 hours. Control sample solution of paeoniflorin was added into blank serum. Acetonitrile was used to deposit protein, the solution was centrifugated and isolated ,and 20μL supernatant was taken to obtain regression equation. ② Seventy-two Kunming mice were randomized into Radix Paeoniae Rubra extracts group and Taohong Siwu Tang group, with 36 mice in each. After being fasted for 12 hours, the mice in each group were intragastric administration of Radix Paeoniae Rubra extracts and Taohong Siwu Tang, respectively and the concentration of paeoniflorin was 300 mg/kg.1 mL blood was taken respectively at 15, 30, 60, 90, 120 and 180 minutes after intragastric administration and prepared into serum sample (6 mice at each time point in each group). The time to peak and difference in peak concentration of serum paeoniflorin were compared between intragastric administration of Radix Paeoniae Rubra extracts and Taohong Siwu Tang.The concentration of paeoniflorin was measured with high-performance liquid chromatography (HPLC). Chromatographic condition: chromatographic column YWG-C18 (250×4.6 mm,10 μm). The mobile phase was methanolKH2PO4 (0.05 mol/L) (38∶62) at a flow rate of 1.0 mL/min. The detective wavelength was set at 230 nm. The column temperature was 25 ℃.MAIN OUTCOME MEASURES: Serum concentration of paeoniflorin after administration of Radix Paeoniae Rubra extracts and Taohong Siwu Tang in mice.RESULTS: Seventy-two mice were used for Radix Paeoniae Rubra extr acts and Taohong Siwu Tang tests, and all of them entered the stage of result analysis without deletion. ① Chromatographic isolation results: Paeoniflorin could be well isolated from other components. The linear range for detection of paeoniflorin was 5.40-646.0 μg/mL. The mean recovery rate was 93.6% and the limit of detection was determined as 1.08 mg/L. ②Comparison of concentration of serum paeoniflorin of mice between two groups: The concentration of serum paeoniflorin in the Radix Paeoniae Rubra extracts group and Taohong Siwu Tang group reached the peak at 30 and 60 minutes after intragastric administration, respectively. The peak concentration of paeoniflorin of Taohong Siwu Tang group was lower than that of Radix Paeoniae Rubraextracts group [(36.27±5.72) mg/L vs.(46.82±5.29) mg/L, P < 0.01 )].CONCLUSION: The concentration of serum paeoniflorin is significantly decreased after intragastric administration. It was further confirmed the decreased concentration of the crude paeonifiorin in the mice sera might result from promotion of its metabolic conversion by other chemical components in the complex prescription. This result needs further investigation.
5.Effects of Rhamnocitrin in Oxytropis Falcata on Nonspecial Immune Function of ;Immunosuppressive Mice

Objective To study action mechanism of rhamnocitrin in Oxytropis Falcata on enhance the immune function. Methods Kunming mice were randomly divided into blank control group, model group, positive control group, traditional Chinese medicine high, medium, and low dose groups. All treatment groups received gavage with relevant medicine for 10 days. From the 8th day, blank control group received intraperitoneal injection with normal saline, while other groups received intraperitoneal injection with cyclophosphamide for 3 days. Through macrophages carbon clearance test, macrophage phagocytosis of chicken erythrocytes test, and immunosuppressive mice organ index detection, effects of rhamnocitrin in Oxytropis Falcata on nonspecific immune function of immunosuppressive mice were observed. Results Compared with model group and blank control group, traditional Chinese medicine high, medium, and low dose groups could improve the phagocytic activity of immunosuppressive mice, clearance index, and phagocytic coefficient (P<0.05), and increase the organ index of the thymus and spleen. Compared with the positive control group, the efficacy of traditional Chinese medicine high, medium, and low dose groups was low. Conclusion Rhamnocitrin in Oxytropis Falcata can enhance the immune function, improve the formation of spleen cells of mice serum hemolysin of chicken red blood cell immune.

Gastric cancer is the leading cause of death of patients with malignancy and the most common malignancy in China.Stomach is also a secretory organ,thus the carcinogenesis of human stomach always accompanied neuroendocrine differentiation.There is not a explicit definition of neuroendocrine differentiation so far,and the influence of which in gastric patients prognosis is in dispute.With the deep intensive research of gastric adenocarcinoma,diagnosis and treatment in gastric adenocarcinoma with neuroendocrine differentiation becomes increasingly concerned.

8.Relationship between nutritional risk and cognitive score in patients with acute stroke

Objective To investigate the relationship between nutritional risk and cognitive score in pa-tients with acute stroke .Methods From January 2015 to December 2015, 160 inpatients with acute stroke were observed and divided into two groups according to Nutritional Risk Screening 2002 (NRS 2002) scores.All the patients were on natural diets and did not receive enteral or parenteral nutrition support .Cognitive functions were evaluated using the Mini-Mental State Examination ( MMSE) within 24 hours and 6 months after the acute cere-bral artery diseases .Logistic regression analysis was performed to investigate the effects of nutritional risks on the cognition.Results Totally 145 patients entered the final analysis , among whom 72 had nutritional risks and 73 had no nutritional risks .The average ages of patients with nutritional risks were significantly older than those without nutritional risks [ (67.04 ±8.95) years vs.(63.10 ±9.12) years, P=0.027], along with significantly higher incidence of diabetes (75.0%vs.49.3%, P=0.001).The lesion locations showed no statistically signif-icant difference between two groups ( frontal lobe 24.5%vs.17.5%, P=0.670;parietal lobe and occipital lobe 40.8%vs.26.3%, P=0.398;basal ganglia region 51.0%vs.49.1%, P=0.490;brain stem and cerebellum 53.0%vs.42.1%, P=0.777).Compared to the patients without nutritional risks , the MMSE scores of the pa-tients with nutritional risks were lower within 24 hours, but without statistical significance (27.45 ±2.21 vs. 28.09 ±1.27, P=0.209);however, six-month follow-up showed that the MMSE scores were significantly differ-ent between two groups (24.70 ±2.16 vs.27.43 ±2.32, P=0.036).Univariate Logistic regression analysis was used to evaluate factors associated with cognitive impairment (MMSE<27).NRS 2002, ages, diabetes mellitus history, and body mass index entered in the Logistic regression analysis .In all patients, ages and NRS 2002 had a positive correlation with cognitive impairment (P=0.000, OR=1.156;P=0.004, OR=2.341).Conclusion The cognitive function in stroke patients with nutritional risks is worse than in patients without nutritional risks .
9.Misdiagnosis of Old Ectopic Pregnancy:Report of Two Cases

[Summary] From February 2008 to June 2008, two cases of old ectopic pregnancy were misdiagnosed: Case 1 was misdiagnosed as gestational trophoblastic disease (GTD), and Case 2 was misdiagnosed as cornual pregnancy.In order to further confirm the diagnosis, two cases were all given laparoscopic exploration.Surgery confirmed interstitial chronic ectopic pregnancy, leading to a resection of the fallopian tubes and mass lesions.Embryo tissue was pathologically confirmed postoperatively.The diagnosis of old ectopic pregnancy is difficult.Serum hCG determination is often found no positive, and ultrasound examination has no specificity.The clinical images are varied.Detailed medical history inquiry and attention to differential diagnosis are key for the diagnosis of old ectopic pregnancy.
10.Quantitative biomechanical relationship between the anterior cruciate ligament deficiency and the posterior horn of medial meniscus

Objective To establish a quantitative biomechanical relationship between the anterior cruciate ligament deficiency and the posterior horn of medial meniscus. Methods 10 human cadaveric knees were examined using an in vitro knee-testing apparatus. In response to a combined 134 N anterior and 200 N axial compressive tibial load, the in situ forces in the posterior horn of medial meniscus were measured. Testing was performed on 10 knees at multiple angles of flexion (30?, 60?, 90?) before and after resection of anterior cruciate ligament. Results The resultant forces on the posterior horn of medial meniscus were as follows: intact ACL knees, (22.8?11.5) N in 30? flexion, (27.1?16.3) N in 60? flexion, (26.7?14.5) N in 90? flexion.
