Objective To prevent the prevalence of drug resistance viruses,we analyzed drug resistance mutations of HIV-1 among HIV-1 infected individuals in seven provinces of China. Methods We acquired 718 HIV-1 infected patients' treatment and compliance from questionnaires, amplified HIV-1 pol genes by RT-PCR and nest-PCR,sequenced RT and protease regions and run clustal with the subtype B consensus amino acid sequence in Stanford HIV drug resistance sequence database. Results The viral load in treated patients was obviously lower than that in untreated patients(P

Percutaneous vertebroplasty has many advantages such as minimal trauma, simple mode of operation, rapid and correct therapeutic effect, high safety, and wide indications. Therefore it has been generally paid close attention and accepted. Percutaneous vertebroplasty has conspicuous curative effect, especially in vertebral compression fracture. Polymethyl methacrylate is the most used filling materials at present; There is not significant difference between unipedicular and bipedicular vertebroplasty; Using vertebral body venography and appropriate dense bone cement can reduce leakage of bone cement; Bolster for self-replacement can rebound vertebral body height, and balloon kyphoplasty and sky bone expander kyphoplasty can be avoided; In order to reduce refracture of vertebral body, anti-osteoporosis drugs should be used in the treatment of percutaneous vertebroplasty.

Journal of Applied Clinical Pediatrics 2004;0(07):-
Objective To evaluate the relationship between children interleukin-1 receptor antagonist gene(IL-1RN) polymorphisms and susceptibility to febrile seizures(FS).Methods Matching case-control study was carried out, blood samples from 52 patients with FS and 53 healthy children were collected.Genomic DNA was extracted and examined by polymerase chain reaction(PCR) amplification for judging IL-1RN genetype. Results Only Ⅰ/Ⅰ,Ⅰ/Ⅱ,Ⅱ/Ⅳ genetypes of IL-1RN were found in the surveyed Chinese population of Han nationality in Guangdong Province. No significant differences of these genotypes were observed in patients and controls. But the allele frequencise of Ⅰ,Ⅱand Ⅳ in patients were 95.2%,3.8% and 1.0%,respectively,while these frequencise in controls were 84.9%,10.4% and 4.7%,respectively.The allele frequency of IL-1RNⅠin patients was obviously higher than that in controls(P

AIM: To analyze the difference of endonuclease domain containing 1 (ENDOD1) expression be-tween benign prostatic hyperplasia ( BPH ) tissues and prostate cancer ( PCa ) tissues and to investigate the effect of ENDOD1 on the biological function of human prostate cancer cells .METHODS: The BPH samples ( n=20 ) and PCa samples (n=21) were processed and analyzed according to the instruction of immunohistochemical (IHC) staining. The mRNA and protein levels of ENDOD 1 in the normal prostate epithelial cells and prostate cancer cells were evaluated by RT -qPCR and Western blot , respectively .The recombinant plasmids pCMV-N-Flag-ENDOD1 was constructed and was trans-fected into the human prostate cancer cells .The proliferation , apoptosis , migration and invasion abilities of the prostate cancer cells were evaluated by MTT assay , flow cytometry, Transwell migration and Matrigel invasion assays , respectively. RESULTS:The analysis of variance of the immunoreactivity score showed that PCa tissues with high Gleason score dis -played significantly lower ENDOD1expression than that with low Gleason score and BPH (P<0.05).The expression of ENDOD1 at mRNA and protein levels in PC3 cells and DU145 cells was significantly lower than that in the LNCap cells (P<0.05).The proliferation of DU145 transfected with ENDOD1 was inhibited. The flow cytometry indicated that ENDOD1 over-expression in the DU145 cells resulted in a notable increase in G0/G1 phase arrest (P<0.05), but the ap-optotic rates showed no statistical difference .The results of Transwell assay showed that migration and invasion abilities of the cells were also inhibited after transfection with over-expressing ENDOD1 plasmid (P<0.05).CONCLUSION: The expression of ENDOD1 significantly decreased in prostate cancer with high Gleaon score .Meanwhile, the ENDOD1 is spe-cifically down-regulated in androgen independent prostate cancer (AIPC) cell lines.Over-expression of ENDOD1 remark-ably inhibits the proliferation , migration and invasion abilities of AIPC .

The nuclear transcription factor nuclear factor erythroid 2-related factor 2 (NRF2) plays a crucial role in maintaining cellular redox homeostasis. The aberrant NRF2 signaling confers enhanced antioxidant capacity, which is linked to tumor progression and therapeutic resistance. The current study investigates the biological effects and molecular mechanism of tribbles homolog 3 (TRIB3), a stress-induced protein, in regulating cell survival and apoptosis in lung cancer. This study first performed the RNA sequencing data analysis with 576 lung adenocarcinoma patients from the cancer genome atlas (TCGA) database. The NRF2- antioxidant response element (ARE) signature was enriched in patients with high TRIB3 expression. Dual-luciferase reporter assay and real-time quantitative polymerase chain reaction (PCR) were used to confirm the effect of TRIB3 on the kelch-like ECH-associated protein-1 (KEAP1)-NRF2 pathway. Journal of Forensic Medicine 2015;31(2):129-131
Species identification of biological samples is widely used in such fields as forensic science and food industry. A variety of accurate and reliable methods have been developed in recent years. The current review shows common target genes and screening criteria suitable for species identification, and described various DNA-based molecular biology methods about species identification. Additionally, it discusses the future development of species identification combined with real-time PCR and sequencing technologies.

Aim To explore the effects of anti-HER-2 chimeric antibody chA 21 on proliferation and apoptosis of SKBR3 cells. Methods MTT colorometric assay,HE staining, transmission electron microscopy, flow cytometry, and TUNEL were used to study the proliferation inhibition and apoptosis induction of SKBR3 cells by chA 21 in vitro. Results Proliferative inhibition rates and apoptotic index of SKBR3 cells were increased in a dose and time dependent manner after exposure to chA21(0.2~5.4 mg?L~(-1)).Conclusion chA 21 could remarkably inhibit proliferation of SKBR3 cells in vitro and apoptosis induction may be one of its main mechanisms.
