2.Immunophenotypic Features of NK/T Cell Lymphoma in Children and Its Association with Epstein-Barr Virus

Objective To study the immunophenotypic features of NK/T cell lymphoma in children and its association with Epstein-Barr virus (EBV) infection. Methods Five cases of children′s NK/T cell lymphoma were studied. CD45RO, CD3?, CD56, CD20, TIA-1 and granzyme B were detected by immunohistochemistry staining for investigating immunophenotype. The expression of EBV-latent membrane protein (LMP-1) were detected by immunohistochemistry. EBV-encoded RNA (EBER1/2) were detected by in situ hybridization (ISH). Results CD45RO, CD3?, TIA-1 and granzyme B were positive in 5 cases, CD56 was positive in 2 cases, while CD20 negative in 5 cases.EBER1/2 positive in 4 cases and LMP-1 positive in 3 cases.Conclusions NK/T cell lymphoma in children is strong associated with EBV infection,and EBV infection may play an important role in the pathogenesis of children NK/T cell lymphoma.
3.Construction and functional identification of rat Hes1 adenovirus expression vector

Objective to construct the high titers rat Hes1 adenovirus expression vector (Ad-Hes1).Methods With the rat cDNA as a template,the Hes1 fragment was amplified by PCR,which constructed pShuttle-CMV-Hes1 shuttle plasmid by directly clone.Based on pShuttle-CMV-Hes1,pAdeno-Hes1 virus plasmid was constructed,pAdeno-Hes1 was transfected into 293 cells to package Ad-Hes1,virus titers were determined by modified TCID50.Hes1 was detected by Western blot after Ad-Hes1 infected with H9c2 myocardial cells.Results pShuttle-CMV-Hes1 shuttle plasmid and pAdeno-Hes1 plasmid were constructed successfully,with a general titer of 1.6 × 1011 PFU,Ad-Hes1 can be expressed in H9c2 myocardial cells,and its MOI value was 30.Conclusion Ad-Hes1 is successfully constructedand packaged,thus provide basis for further research on the protection effect of Hes1 on myocardium.
4.Effect of smoke water and distillation liquid on the seed germination and seedling growth of Trichosathes kirilowii.

Smoke water and distillation liquid were used to treat the seeds of Trichosathes kirilowii and to study the effects of smoke water and distillation liquid on the seed germination and seedling growth of T. kirilowii. The results showed that germination rate, germination index and germination vigor of T. kirilowii all were significantly improved with the treatment of SW and DL treatment. The activity of α-amylase were significantly increased with the treatment of SW and DL at 1:2,000. SW and DL treatment showed no significant effects on the activity of SOD. The activity of POD were markedly enhanced under the treatment of SW (1:000) and DL (1:2,000). CAT activity were increased with the treatment of SW and DL at 1:2,000 while were inhibited by SW and DL at 1:500. Seedling height and root length were increased with the treatment of SW and DL (1:1,000, 1:2,000). SW and DL treaments improved the content of chlorophyll, and moreover with the concentration of SW and DL, the stimulatory were also increased. This work demonstrated that smoke water and diatillation liquid at 1:2,000 could stimulate the seed germination and seedling growth of T. kirilowii, and it provided the references for the study of seed germination technology.

5.Meta-analysis of tacrolimus and cyclophosphamide in the treatment of lupus nephritis

Objective To evaluate the clinical efficacy of tacrolimus and cyclophosphamide (CTX) on lupus nephritis.Methods The clinical trials on treatment of lupus nephritis with tacrolimus and CTX published before October 2012 were searched at Cochrane library,PubMed,OVID,Wanfang database,Chinese Journal full-text Database,Chongqing Weipu Database by using the methods of Cochrane systematic review.At the same time the information from related journals,professional data and network were hand-searched.The homogeneous evaluation was performed by Meta-analysis.Statistical analysis of clinical data was performed by using Stata11 software.Results A total of 4783 reports were found,while only 9 papers (8 randomized controlled trials and 1 cohort study) met the inclusion criteria.Tacrolimus group got better complete remission ratio (Z =4.05,P < 0.01),similar partial remission ratio (Z =0.44,P =0.661),and better overall remission ratio (Z =4.29,P < 0.01) as compared with CTX group.There were no significant differences between tacrolimus group and CTX group in the incidence of infection (Z=1.75,P=0.081),renal damage (Z=0.88,P=0.38) and abnormal glucose metabolism (Z =1.91,P =0.070).Side effects such as liver function impairment (Z =2.65,P<0.01),gastrointestinal discomfort (Z=2.31,P<0.05),menstrual disorders (Z=3.88,P<0.01),bone marrow suppression (Z =3.29,P < 0.01) in tacrolimus group were lower than those in CTX group.Conclusions Compared with CTX,tacrolimus has better complete remission ratio and overall remission ratio,with less side-effects in the treatment of lupus nephritis.However,large scale,multicenter,well-designed clinical trials should be adopted to further confirm the conclusions.

9.Survivin expression in midline T-cell lymphoma in relation to Epstin-Barr virus infection.

To investigate the expression of survivin gene and its relationship with Epstin-Barr virus (EBV) infection in midline T-cell lymphoma (MTL), immunohistochemistry staining method was used to examine the expression of survivin and EBV-latent membrane protein (LMP-1) in the 41 cases. In situ hybridization (ISH) was used to detect EBV-encoded RNA (EBER1/2). The results showed that the expression of survivin was positive in 26 cases of midline T-cell lymphoma, but no positive was detected in 10 cases of reactive lymphoid tissues. The positive expression ratio of survivin was 12.5% in cases of MTL with low grade of malignancy, and was 75.76% in cases of MTL with middle and high grades of malignancy, the significant difference was found between these two groups (chi(2) = 8.55, P < 0.01). Positive expression ratios of EBER1/2 and LMP-1 were 70.73% and 41.46% respectively. Survivin expression was not significantly different between EBER1/2 positive and negative cases (P > 0.05). It is concluded that survivin expression is up-regulated in MTL, and survivin positive expression rate is associated with the degree of malignancy. Survivin may play a role in the pathogenesis of the MTL by influencing cell apoptosis. EBV infection is not significantly associated with survivin expression in the MTL.

10.Interaction of fluorine, calcium and iodine on weight of rats.

OBJECTIVETo study the interaction of fluorine (F), calcium (Ca) and iodine (I) on body weight of rats.
METHODSOne-month-old Wistar rats were randomly divided into 8 groups: moderate concentrations of F, Ca, I (group 1); moderate concentrations of F, I and high concentration of Ca (group 2); moderate concentrations of Ca, I and high concentration of F (group 3); moderate concentration of I and high concentrations of F, Ca (group 4); moderate concentrations of F, Ca and low concentration of I (group 5); moderate concentration of F,high concentration of Ca and low concentration of I (group 6); moderate concentration of Ca, high concentration of F and low concentration of I (group 7); high concentrations of F, Ca and low concentration of I (group 8) based on 2 x 2 x 2 factorial design. The moderate concentration of F was 90 microg/d and the high concentration of F was 2700 microg/d. The moderate Ca concentration was 13 mg/d and the high Ca concentration was 260 mg/d. The moderate concentration of I was 3.5 microg/d and the low concentration of I was 0.23 microg/d. After twenty weeks, body weight was measured.
RESULTSAccording to the results of factorial ANOVA, significant interaction effects of F with Ca were found (F = 5.933, P = 0.017). The empty body weight was measured at the end of the fifth month. When both iodine and fluorine were at the optimal level, the weight of group 2 [(262.5 +/- 47.1) g] and group 1 [(307.9 +/- 55.0) g] showed significant difference (t = 4.24, P < 0.05). When both iodine and fluorine were at low level, the weight of group 6 [(248.8 +/- 30.0) g] and group 5 [(293.3 +/- 19.7) g] showed significant difference (t = 4.16, P < 0.05). Animals with optimal iodine and calcium [(269.3 +/- 27.3) g] showed significant difference compared to the weight of low level iodine and optimal fluorine [(307.9 +/- 55.0) g]. When the low level iodine and optimal calcium were applied, weight of group 7 [(261.9 +/- 31.3) g] and group 5 [(293.3 +/- 19.7) g] showed significant difference. (t = 2.94, P < 0.05).
CONCLUSIONInteraction effects of F with Ca were found on body weight in rats.
