1.The Research Status of Anti-tumor Effects of Artemisinin and its Derivatives

Artemisinin is a traditional anti-malarial drug. In recent years, many researches indicated that artemisinin and its derivatives have obvious anti-tumor effects. Its mechanisms of anti-tumor lies in cytotoxicity, direct destruction of tumor cells due to oxidative stress reaction, inducing apoptosis, inhibiting of tumor cell proliferation, restraining tumor angiogenesis and performing immunological regulation. Compared with the traditional anti-cancer drugs, artemisinin can selectively inhibit tumor cells and manifestating no cross-resistance with traditional chemotherapeutic agents. Besides it can reverse the phenomenon of multi-drug resistant of tumor cells. Therefore, artemisinin, as an new type of anti-cancer drug, will be applied to a wide range of clinical practice.
2.Comparative Study on MSCT-3D and COPT in Stomatology

Purpose:To explore the value of MSCT-3D in stomatology through comparing with COPT.Materials and Methods:Both MSCT 117 patients and COPT 117 patients in stomatology were selected at a particular period,COPT imaging was using computed post-processing,MSCT imaging was processed according to 3D information through work station,then to analyze the imaging results according to metal tooth、damaged tooth and broken tooth,later to compare the photograph 、the technique of computed post-processing、the photograph condition and exposure of MSCT and COPT.Results:Normal COPT patients were 9 cases ( 0.24% ),normal teeth were 3160 (84.4% ),unusual teeth were 551 (15.6%),normal MSCT-3D patients were llcases ( 0.29% ),normal teeth were 3138 (83.8% ),and unusual teeth were 606 (16.2% ).COPT belonged to COPT photograph,the emphasis was 2D,MSCT-3D belonged to spiral photograph,the emphasis was 3D,and MSCT imaging could be processed according to 3D reformation.The 3D effect of MSCT was batter than that of COPT.The radiation dosage was much higher than that of COPT.Conclusions: COPT could get 2D effects: MSCT-3D could improve 3D results on 2D basis,but the radiation dosage of MSCT is much higher than that of COPT in stomatology.
4.Clinical significance of CD4+ CD25+ regulatory T cells and transforming growth factor beta-1 in the elderly patients with hepatocellular carcinoma

Objective To evaluate the changes of CD4+ CD25+ regulatory cells (Treg) in peripheral blood and the serum levels of transforming growth factor beta-1 (TGF-β1)in elderly patients with hepatocellular carcinoma. Methods The ratios of Treg in the peripheral blood of patients aged over 80 years from 22 hepatocellular carcinoma,26 metastatic liver cancer,20 healthy controls,were determined by flow cytometry.Meanwhile,the serum levels of TGF-β1 were detected by ELISA.Results The ratio of Treg to total CD; T cells in the peripheral blood [(9.71±3.23)% vs.(5.81±1.18)%,P<0.01]and the serum levels of TGF-β1 [(78.10±29.41)ng/L vs.(7.78± 3.54) ng/L,P<0.01]of elderly patients with hepatocellular carcinoma were significantly higher than those in the healthy controls.Meanwhile,the ratio of Treg to total CD[ T cells in the peripheral blood [(9.71±3.23)% vs.(7.36±2.07) %,P<0.05]and the serum levels of TGF-β1[(78.10± 29.41 )ng/L vs.(19.33± 10.90) ng/L,P< 0.01 ]in the patients with hepatocellular carcinoma was increased as compared with those in metastatic liver cancer (P<0.05 or P<0.01).The correlation indicated that the ratio of Treg to total CD4+ T cells in the peripheral blood were positively related with TGF-β1 levels and tumor clinical stage(r=0.698 and 0.782,P< 0.01 ),but negatively with Karnofsky performance status score(KPS) (r=-0.643,P<0.01). Conclusions The ratio of Treg to total CD4+ T cells in the peripheral blood from elderly hepatocellular carcinoma is increased and correlated with TGF-βl level,tumor clinical stage and KPS.It might helpful to determine the prognosis of elderly hepatocellular carcinoma by detecting the ratio of Treg to total CD4+ T cells in the peripheral blood.
5.Study on Quality Standard of Naru-3 Cataplasm

Objective To establish the quality standard of Naru-3 Cataplasm. Method The contents of Radix aconiti kusnezoffii were determined by UV-vis, and meaconitine was determined by HPLC. Results The linear range of meaconitine was 0.129 5~2.074 0 ?g. The quality standard of Naru-3 Cataplasm was established. Conclusion The method can be used for quality control of Naru-3 Cataplasm.
6.Correlation of height change of intervertebral space of fused segment with axial synptoms after anterior cervical fusion

[Objective] To investigate the height change of intervertebral space after anterior cervical fusion and its relation to neck axial symptoms.[Method]Totally 122 patients receiving anterior cervical fusion were followed up for 2 to 5 years,average 3.9 years.The height change of intervertebral space of the fused segment on X-rays were evaluated before,3 months and 2 years after operation.All patients were classified into 3 groups by means of the postoperative height change of intervertebral space.The axial symptoms after operation were observed and analyzed during follow-up.[Result]The incidence of neck axial symptoms was 54.55%,25.84% and 45.45%,respectively.There were significant differences in statistical analysis in group A group B,group B to group C(x2=7.18,P0.0125).[Conclusion]There is significant relation between height change of intervertebral space and neck axial symptoms after anterior cervical fusion.

GDF-15,a distant member of the TGF-superfamily,is identified as an apoptotic accelerat- ing,anti-tumorigenesis and nerve2nutritional factor in varied injures and tumors and has cardioprotective ac- tivity.The characteristics and roles of GDF-15 gene/protein and antibodies are expounded besides the rela- tionship between GDF-15 serum level/genetypes and CRC.It is also discussed here that some antitumorigenic substances inducing GDF-15 in CRC tissues and CRC cells.
8.Cloning and Fusion Expression of the gene encoding part of HTLV envelope protein gp21

Objective The gene encoding part of HTLV(Human Tcell leukemia viruses) envelope protein gp21 was cloned and expressed in order to prepare diagnostic agent for detecting HTLV.Methods The gene encoding part of HTLV envelope protein gp21 was obtained by combining four oligonucleotides into a long DNA fragment,then cloned and subcloned into Tvector and pGEX2T respectively.Expression was performed under induction in a fusion way that target protein gene was inserted downstream the carrying protein gene of pGEX2T which makes purification of target protein convenient.At last,Target protein was purified by affinity chromatography in glutathione sepharose 4B column and its antigenicity was confirmed by immunoblotting.Results and conclusion Sequence analysis showed it was feasible to obtain a target gene by ligating short oligonucleotides;As expected,target protein was expressed which exhibited strong antigenicity after being subcloned into fusion expression vector pGEX2T.
9.Study of transcription and cleavage in vitro of HCV with HCV specific hammerhead ribozyme

Objective To study the effect of specific hammerhead ribozyme (Rz) to hepatitis C virus (HCV) in vitro. Methods Rz 1 Rz 2 were designed to cleavage at 5′-NCR nucleotide positions under 136~160 and 313~337, Rz 3 was designed to cleavage at C region nucleotide position under 373~388. As a control, cleavage deficient Rzm that have A→G point mutations in the catalytic loop of the hammerhead domain. 32P-labeled transcript of target HCV RNA was incubated with gel-purified Rz ( Rz 1, Rz 2, Rz 3 and Rzm ) respectively at different concentration based on specified condition and autoradiographed after denaturing gel-electrophoresis. Results Except Rzm, Rz 1 Rz 2 Rz 3 were active at 37 ℃ and more so at higher concentration, and more so with cleavage site nearly to the HCV initial code. Conclusions The HCV specific hammerhead ribozyme can be designed in vitro, further study about cleavage in vitro and in vivo will continue.
10.Sequence Analysis of Coat Protein Gene of Cucumber Mosaic Virus Isolate Infecting Basella rubra L.

Cucumber mosaic virus was detected from infected Basella rubra L. with the indirect enzyme-linked immunosorbent assay. Total RNA was extracted from infected leaves and the cDNAs of coat protein gene of CMV-Ba were obtained by RT-PCR. The amplified cDNA fragments were then cloned into pMD 18-T vector and sequenced,the result showed that the CP gene was 657 nucleotides in length. This sequence was aligned with the obtained CP gene and some CMV strains or isolates of subgroup Ⅰ and subgroup Ⅱ in GenBank using DNA MAN software. The results showed that CMV-Ba shared 90.9%~93.8% and 76.1%~76.9% identity with the known CP genes of subgroup Ⅰ and Ⅱ respectively in nucleotide level,on the other hand,amino acids deduced from CMV-Ba CP gene shared 92.7%~97.7% and 72.4%~78.1% identity with the known CP protein of subgroup Ⅰ and Ⅱ,respectively. This suggested that CMV-Ba CP gene belongs to CMV subgroupⅠ.
