Objective To purify rabbit anti-hRBBP10 polyclonal antibody.Methods The recombinant fusion protein PTC-hRBBP10 was expressed in the E.coli and was purified through amylose resin chromatography column and superose 12 gel fitration .The purified PTC-hRBBP10 was coupled to the NHS-activated sepharoseTM to prepare affinity chromatography column to purify rabbit anti-hRBBP10 polyclonal antibody. Results ① PTC-hRBBP10 was expressed and purified successfully with relative molecular mass(Mr) of 80?103 and its purity could reach about 95%.② Purified Rabbit anti-hRBBP10 polyclonal antibody could bind Specifically to PTC-hRBBP10.Conclusions With better specificity, The purified rabbit anti-hRBBP10 polyclonal antibody provide condition for studying the function of the protein, and helps to develop new techniques of tumor diagnosis and treatment.

Objective To prepare rabbit anti-human RBBP10 polyclonal antibody and then character them.Methods New Zealand rabbit was immunized with RBBP10 protein,Polyclonal antiserum was analyzed by ELISA and Western blot.Results The titer of the antiserum obtained in this experiment determined by ELISA was up to 1∶16000 and the sensitivity was at least 10?g/?l.Western blot results show the antibody has high specificity.Conclusions Preparation the Rabbit anti-human RBBP10 Polyclonal antibody in this method is successfully,The expression of the RBBP10 protein in tumor can be detected in immunohistochemistry method,it is useful.

Objective To assessment short-term prognosis in patients with acute on chronic liver failure , several scoring systems were compared. Methods Two hundred and sixteen patients with acute on chronic liver failure were divided into survival group and death group according to the results of 90 days after admission.CTP , MELD,APACHEⅡ, SOFA and SMSVH score were calculated.After ROC curves were performed ,the areas under the curves of these scoring systems were compared. Results The areas under the ROC curves of MELD, APACHEⅡ, SOFA, CTP and SMSVH were 0.88, 0.76, 0.89,0.79and 0.69,respectively. The areas under the curves of SOFA and MELD were larger than the APACHEⅡ, CTP and SMSVH (P<0.05). There was no difference between the SOFA and MELD (P>0.05). The area under the curve of CTP was larger than the APACHEⅡ, but there was no statistically significant difference (P > 0.05). The area under the curve of SMSVH were less than 0.7. Conclusions The SOFA, MELD,CTP and APACHEⅡcan predict the short-term prognosis of acute on chronic liver failure. The SOFA and MELD are the best scoring systems.CTP,APACHEⅡ are better than SMSVH. SMSVH fail to predict the prognosis of acute on chronic liver failure.

Objective To investigate the associations of peripheral CD4+ CD25 + CD127low/-regulatory T cells (Treg) with HBV viral load and liver pathology in hepatitis B virus (HBV) carriers.Methods Forty six chronic HBV carriers admitted in the first hospital of Jiaxing during October 2012 and February 2014,and 23 healthy subjects (controls) were enrolled in the study.CD4+ CD25 + CD127low/-Treg in peripheral blood of the two groups were detected by flow cytometry.Ultrasound-guided liver biopsies were performed in chronic HBV carriers and HBV DNA load was determined by real-time PCR method.Independent samples t test was used for the comparison between groups,and Spearman rank correlation or Pearson linear correlation analyses were performed.Results The frequency of the peripheral CD4+ CD25+CD127low/-Treg in 46 HBV carriers was (5.11 ±1.47)％,which was significantly higher than that in healthy controls [(3.46 ± 1.23) ％,t =4.629,P ＜ 0.01].The HBV DNA load in HBV carriers was (6.21 ±1.98)lg copies/mL,which was positively correlated with the CD4+ CD25+ CD127low/-Treg level (r =0.405,P ＜ 0.01).Among 46 HBV carriers,21 (45.65％) were of inflammation grade 2 or above,and 16 (34.78％) were of fibrosis stage 2 or above.Peripheral CD4+ CD25+ CD127low/-Treg level was negatively correlated with inflammation and fibrosis in HBV carriers (r =-0.343 and-0.452,P ＜ 0.05).Conclusion CD4 + CD25 + CD127low/-Treg may be associated with the chronicity of HBV infection and the degree of liver damage.

OBJECTIVETo study the hemodynamics of the pulmonary artery (PA) in pulmonary artery hypertension (PAH).

METHODSWith combined clinical hemodynamic measurements and CT data, the 3D model of PA and its branches was reconstructed to obtain the real 3D geometric structure of PA and its branches. Computational fluid dynamics (CFD) simulation was carried out for the reconstructed 3D geometric model of the PA and its branches with Womersley velocity as main pulmonary entrance conditions; then hemodynamic parameters of the PA and its branches in cardiac cycle were calculated and the changes of flow field, pressure, wall shear stress (WSS) at different times, and its impact on physiological processes were analyzed.

RESULTSimulation showed that the pressure of the main PA and the branches of the left and right PA was higher in the systolic period than that in the diastolic period; especially, the that of right PA and its interlobular artery was significantly higher than that of the left. The blood flow velocity of the main PA and its branches of right and left PA was significant different, the velocity was much higher at the distal of the right PA than that at the left. High WSS field formed at the proximal of the right PA before the branches during the systolic.

CONCLUSIONThe right PA at the proximal and lower lobe artery are affected by much larger pressure and change firstly, so the change of morphological and functional of theses segments will be have more important value to prompt existence of the early PAH.

OBJECTIVETo evaluate the preliminary curative effect of interspinous injections for the diagnosis and treatment of back pain caused by lumbar kissing spine (Baastrup's disease) under fluoroscopically guiding.

METHODSFrom November 2011 to March 2013,17 patients with back pain caused by Baastrup's disease were treated with fluoroscopically-guided interspinous injections, including 7 males and 10 females with an average age of 49.6 years old ranging from 40 to 71 years old; the duration of the disease ranged from 2 to 5 years with a mean of 3.7 years. The visual analogue scale (VAS) and the lumbar segments range of motion (ROM) was analyzed at pre-operation, 2 days, 3 months and final followed-up after operation, the effects were evaluated with modified Macnab standard.

RESULTSAll patients were follow-up from 6 to 10 months with an average of 7.6 months. The pre-operative VAS was 6.41 +/- 0.94, the postoperative VAS at different time points improved significantly comparing with pre-operation,and the differences were statistically significant (P < 0.01). There was no significant difference in VAS at different time points after operation (P > 0.05). The ROM of operated segment and adjacent segment was (4.88 +/- 0.86) degrees and (6.82 +/- 0.73) degrees respectively at pre-operation. The postoperative operated segment ROM at different time points improved significantly comparing with pre-operation, and the differences were statistically significant (P < 0.05). Compared with pre-operation, there was no significant difference in adjacent segment ROM at different time points after operation (P > 0.05). According to modified Macnab, the result was excellent in 6 cases, good in 7 cases, fair in 3 cases and poor in 1 case.

CONCLUSIONFluoroscopically-guided interspinous injections is an effective method for the diagnosis and treatment of Baastrup's disease. The method has advantages of simple operation, minimal invasion and safety, satisfactory short-term and medium-term therapeutic effect; it can also effectively lessen the pain of lumbar and back.

Syndrome of resistance to thyroid hormone(RTH)is a rare hereditary thyroid disease with various clinical manifestations and laboratory findings. RTH could be misdiagnosed and mistreated, resulting in aggravation of the disease. We reviewed the medical records of a patient with RTH over the past six years. In addition, we provided a summary of latest progress for RTH to help the clinicians to improve the understanding of the disease.

OBJECTIVETo observe the therapeutic effect of transcutaneous point electric stimulation on periarthritis of shoulder at different stages and compare with electroacupuncture.

METHODSThree hundred and sixty cases of periarthritis of shoulder at different stages were divided into a treatment group (n=186) treated with transcutaneous point electric stimulation, and a control group (n=174) treated with electroacupuncture. Same acupoints were selected in the two groups.

RESULTSThe total effective rate of transcutaneous point electric stimulation was 96.6% at the adhesion prophase and 96.9% at the adhesive stage, but electroacupuncture stimulation was 93.5% and 97.9%, respectively, with no significant differences between the two groups; transcutaneous point electric stimulation not only could relieve pain, but also significantly improve dysfunction of shoulder joints at the adhesive stage.

CONCLUSIONTranscutaneous point electric stimulation is an effective and convenient therapy for periarthritis of shoulder.
