3.Optimum of Submerged Fermentation Parameters for Antrodia camphorata

Optimal parameters of submerged fermentation for Antrodia camphorata were studied.According to the yield of Intracellular triter- pene,the optimal fermentation parameters were obtained as follows:40g/Lglucose,6 g/L soybean,1 g/L K_2HPO_4,0.5 g/L MgSO_4 and 100mg/L VB_1.The optimum cultural period was 6 d at 26℃in 250mL shake flasks at 100r/min.The best aeration ratio was 1:2.5(medi- um volume:flask volume).The inoculation volume was 20%.The intracellular triterpene of Antrodia camphorata was obtained(about 15.25mg/100mL) in the optimum culture.
4.Construction of gene library of human-originated single-chain variable fragment antibody associated with lung adenocarcinoma

Objective:To construct human single-chain variable fragment (scFv) antibodies gene library associated with lung adenocarcinoma. Methods:Total RNA was extracted from the lymphatic tissue near the lung adenocarcinoma.VH and VL fragments were amplified with RT-PCR,the purified VH and VL fragments were used to produce scFv fragment by splicing-overlap-extension PCR. The scFv gene was cloned in pCANTAB-5E Phasmid,then transferred to Ecoli TG1.The insertion ratio of antibody gene was identified with RT-PCR,the positive enzyme-cutting products were identified with 1.5% agarose gel electrophoresis. Results:Clear 28 S and 18 S strips were found in total RNA;The size of VH is about 370 bp,VL is about 350 bp.The size of scFv is about 750 bp.After transformation into Ecoli TG1,2.2?107 CFU/?g ampicillin resistant bacteria colonies grow after overnight culture,and the positive insert ratio was 83.3%. Conclusion:Human single-chain variable fragment(scFv) antibodies gene associated with lung adenocarcinoma was constructed successfully. It could provide bases to further screen the antibody library construction.

Objective:Retrospectively evaluate the clinical value of 99mTc-Sc lymphoscintigraphy in sentinel lymph node(SLN)location in breast cancer,and to offer accurate orientation for sentinel lymph node biopsy(SLNB).Methods:Fifty breast cancer patients underwent 99mTc-SC lymphoscintigraphy,the outcomes were compared with?detection probe.Results:SLN was found in 45 of 50 cases(90.00%),the number of SLN was 1.60 in every case averagely.41 patients were found in concordance with?detection probe,4 cases were not detected by?probe.The accuracy,specificity,false negative rate and false positive rate of SLN to predict axillary lymph node status were 93.33%,100%,6.67%,0%.Conclusion:99mTc-SC lymphoscintigraphy can exactly predict axillary lymph node status and guide?detection probe and SLNB.

Objective:To compare the trends and clinical features of common respiratory tract viral infection in hospitalized children in Chongqing Chindren's hospital.Methods:Retrospective analysis of 2 365 cases of respiratory tract viral infection from July 2002 to January 2007,was made using direct immunofluorescence for seven common respiratory viruses in nasopharyngeal secretions.The more common respiratory virus clinical manifestations were analyzed.Results:There were certain relationships between the virus detection rate and gender,age and season.Infants asthma attack was related to Parainfluenza(PIV)3.Respriatory syncytin virus(RSV) and PIV3 were the main pathogens of recurrent respiratory tract viral infection,and also pathogens of the common respiratory tract viral infection when children had basic diseases,such as congenital heart disease,congenital tracheal softening airway development with deformities,and so on.Adenovirus(ADV) could cause early childhood asthma.Conclusion:RSV and PIV3 are the major pathogenic viruses in seven common viruses causing infants recurrent wheezing in Chongqing.It is well known RSV is related to asthma in infants,and PIV3 also has some relevance with asthma in infants.ADV is also an important respiratory tract viral infection pathogen in Chongqing,but there are no serious clinical manifestations.ADV can also cause acute asthma attack in early childhood.IV-A is the main pathogen for upper respiratory tract infection.[
10.A study on the mRNA expression of the glucose transporter gene in the diabetic rats

The study was proceeded on the mRNA expression of the GULT1 and GLUT4 gene in the alloxan diabetic rats by the technique of dot-blot hybridization analysis. The results showed that the levels of the GLUT4 mRNA expression were reduced in diabetic rats while increased in insulin treated diabetic rats. This suggests that the reduced GLUT4 mRNA expression which could affect the quantity of its posttranslational products may be one of the causes of the insulin resistance.
