1.Research on quality management practices of software about medical apparatus and instruments

Objective: To explore and practice the method of quality management of software about medical apparatus and instruments so as to ensure the quality of software. Methods:The quality management system of software developed from medical X-ray imaging equipment was constructed through two aspects included target management (quality measurement of qualitative and quantitative were adopted to ensure the quality of software could achieve predictive target) and process management(included the development management of Scrum mode, change management of process and version control based on Git technique. Results: After this method of quality management of software about medical apparatus and instruments was implemented, the software quality and satisfaction of customer were obviously enhanced. Conclusion: Through the control and management of two aspects, the target for ensuring and enhancing quality of software was achieved.
3.Application study of PET-CT image fusion in conformal radiotherapy of non-small cell lung cancer

The application of PET-CT could not only play a part in the diagnosis of clinical stage for NSCLC on PET, but also make full use of the discrimination of anatomical structure on CT. The precision of target contouring could be increased and the normal tissue and risk organ are also protected efficiently.
4.The variation of N400 event-related potentials during language cognition of different paradigms in stroke patients

Objective To explore the cognition processes of language in stroke patients by characterizing the differences between the event-related potentials (ERPs) N400 evoked bypicture-word andsentence-wordparadigms.Methods Twenty patients with a single brain lesion resulting from a first episode of stroke were tested along with 20 normal controls.Their N400 ERPs were tested using pictures and sentences as activating stimuli and phrases composed of nouns and verbs as the target stimuli.The N400 potentials were recorded from electrodes located at Fz,Pz,T3 and T4 positions of the International 10-20 system.The N400 latencies and amplitudes and the behavioral responses of the experimental and control groups were compared.Results The latencies were longer and the amplitudes were lower in response to sentence activation than to picture activation.The differences in terms of amplitude were the greatest with N400 recorded from Pz.The responses to the target stimuli were not significantly different with regard to the types of stimuli and recording location.The average response time was longer and the error rate was higher in response to sentence activation than to picture activation.Conclusion Multiple accesses,activating stimuli and integrating processes are involved in language cognition.The brain is more sensitive and accurate in response to picture stimuli in language cognition.The response to sentence stimuli is slower,and the error rate is higher.This suggests that it is better to usc varied prime stimuli in teaching children languages.
5.The application of polypropylene mesh & plug in hernia repair.

Objective To approach the advantages and summarize the experience of mesh & plug tension-free hernia repair. MethodsThe mesh & plug tension-free hernia repair was administered in 102 patients of groin hernia with the BardR Mesh & PerfixR plug offered by Bard Company in U. S. A. The operation time,postoperative pain,patient's ability recovery, complications and recurrence rate were followed up. ResultsThe average operation time was 31.5minutes. The patients could move 4~6 hours after operation. Postoperative pain lasted about 2~3 days. One had retention of urine and two had wound subcutaneous hydrops. No wound infection. Nine had feeling of foreign matter in the groin region. Only one had recurrence in 89 patients who were followed-up. ConclusionBardR Mesh & PerfixR Plug has good tissue-compatibility,no rejection,and capability of anti-infection. It is an ideal material for hernia repair. The mesh & plug tension-free hernia repair is easy to be administered, with minor lesion, quicker recovery and lower recurrence. And the operation indication can be widened. It is a most advanced method in treating groin hernia.
6.Construction and expression of mouse-human chimeric anti-human melanoma antibody

Objective:To construct eukaryotic expression vector for anti-human melanoma chimeric antibody,and express the antibody in eukaryotic cells.Methods:Variable regions of the murine antibody were cloned from the anti-human melanoma hybridoma cell line HB8459,HB8760,Mel3,and was inserted them into the chimeric antibody eukaryotic expression vector pMH-CA.The constructed vector was transfected into 293T,the chimeric antibody was expressed,as detected by RT-PCR,ELISA,and Western blot.Results:Eukaryotic expression vectors of the chimeric antibody against human melanoma were successfully constructed,and expressed in eukaryotic cells.The expression of chimeric gene was confirmed by RT-PCR,specificity of the chimeric antibody was proved by ELISA and Western blot.Conclusion:Anti-human melanoma chimeric antibody was expressed successfully.
7.Inhibitory effect of rapamycin on the proliferation of Rhesus retinal vascular endothelial cells

Background The signal pathway of mammalian target of rapamycin (mTOR) plays an important role in the regulation of cell cycle.Rapamycin(RAPA) can inhibit the proliferation of cells by regulating of cell cycle.Objective This study was to investigate the effect of RAPA on the proliferation of Rhesus retinal vascular endothelial cells (RF/6A).Methods RF/6A cell strains were cultured in vitro,and PBS,10 μg/L RAPA or 5 μg/L RAPA was added into the medium respectively.The expression of cyclin D1 protein in the RF/6A cells (absorbancy) were detected by Western blot.The cell cycle distribution after RAPA action was analyzed by flow cytometry.Matrigel was used in endothelial-cell tube formation to evaluate the effect of RAPA on angiogenesis.Results Western blot assay showed that the expressions of cyclin D z protein were 0.92±0.04,0.58±0.02 and 0.73±0.02 in the PBS group,10 μg/L RAPA group and 5 μg/L RAPA group,showing a significant difference among the three groups (F =246.320,P =0.000),and the relative expressing level of cyclin D1 protein in the l0 μg/L RAPA group and 5 μg/L RAPA group was significantly lower than that of the PBS group (both at P<0.05).The proportion of G0/G1 cells were (42.13±0.57)%,(65.15±0.64)% and (54.09± 0.78)% respectively in the PBS group,10 μg/L RAPA group and 5 μg/L RAPA group,which was significantly different among the three groups (F=887.815,P=0.000).The number of endothelial-cell tubes were (9.67 ± 1.53)/field,(4.33 ± 0.58)/field and (6.33 ±0.58)/field respectively in the PBS group,10 μg/L RAPA group and 5 μg/L RAPA group,and the number of endothelial-cell tubes in 10 μg/L RAPA group and 5 μg/L RAPA group was less than that in the PBS group,with a significant difference among the three groups (F =21.778,P =0.002).Conclusions RAPA can inhibit the proliferation of RF/6A cells in vitro by down-regulating the expression of cyclin D1.
8.Corneal histopathology changes of the soft contact lens long-time wearers

AIM:To investigate the corneal histopathology changes induced by soft contact lenses ( SCL) at diffent time.
METHODS: Central and peripheral cornea structures of myopic patients wearing SCL (150 cases) and the control goups ( 150 cases ) were observed with confocal microscopy. The examinations were performed 1, 3, 5, 8 and 10a after contact lenses wearing respectively.
RESULTS: After wearing the contact lenses for 5a, the cell density of corneal surface epithelial was lower in SCL wearers compared with control goups (P<0. 05) and the corneal vesicle and corneal gutata might be occasionally discovered. The density of Langerhans cells both in the central and peripheral cornea was higher in SCL group ( P<0. 05 ), and the tortuosity of the nerve fibers was significantly increased in the eye wearing SCL compared with control group. The densities of anterior stromal cells were lower while the microdot densities in the stroma were higher in SCL group than that in control group ( P<0.05 ) . The polymorphology of corneal endothelial cells was more obvious in SCL group, but the densities of endothelial cells were similar between two groups ( P>0.05). The SCL wearers showed a thinner thickness of central corneal epithelium than control group (P<0. 05).CONCLUSION: The corneal morphologic changes are more obvious with the longer time, and the changes is not ignored.
10.Study on HLA-DRB1 shared epitope in patients with IgA mesangial proliferative glomerulonephritis

Objective:To explore the clinical correlation of HLA-DRB1 shared epitope with IgA mesangial proliferative glomer-ulonephritis( IgA MsPGN).Methods:The renal function were examined routinely,and HLA-DRB1 shared epitope were determined by PCR in a total of 164 patients with IgA MsPGN and 164 healthy subjects.The renal function and HLA-DRB1 shared epitope were compared in patients with IgA MsPGN and healthy subjects,and clinical correlation of HLA-DRB1 shared epitope and renal function were analyzed.Results:(1)24 h proteinuria,serum creatinine and serum urea nitrogen were significantly different in patients with IgA MsPGN and healthy subjects(P<0.01);(2)The high-frequency gene of HLA-DRB1 in patients with IgA MsPGN were DRB1*04, DRB1*07,DRB1*09,DRB1*11,DRB1*14 and DRB1*15;(3) The distribution frequency of DRB1*09 and DRB1*11 in patients with IgA MsPGN increased significantly(P<0.01 or P<0.05);(4)24 h proteinuria,serum creatinine and serum urea nitrogen in patients with IgA MsPGN with DRB1*09 and DRB1*11 were significantly different compared with those in patients with IgA MsPGN with other HLA-DRB1 shared epitope(P<0.01).Conclusion:HLA-DRB1 shared epitope is related to IgA MsPGN,DRB1*09 and DRB1*11 can increase the risk of IgA MsPGN,and related to the severity of glomerulonephritis.
