1.AnaIysis of thicknesses change of macuIa centraI fovea and retinaI nerve fiber Iayer in chiIdren with ametropic ambIyopia

· AlM: To investigate the thicknesses change of macula central fovea(MFT) and retinal nerve fiber layer (RNFL) in children with ametropic amblyopia and normal children.
· METHODS: The medical records of 56 children with ametropic amblyopia and 56 normal children in our hospital during the same period were retrospectively analyzed. Optical coherence tomography ( OCT ) was used to measure the MFT and RNFL.The MFT and RNFL change in two groups were statistically evaluated.
· RESULTS: The sex, age and axial length were no significant difference between amblyopia group and control group (P>0.05).The MFT in amblyopia group and control group were 135.15±15.15μm, 136.14±14.26μm respectively (P>0.05).The RNFL in amblyopia group and control group were 117.02 ±8.97μm, 110.43 ±7.94μm respectively, and there was significant difference ( P<0.05).There was negative correlation between RNFL and axial length in control group(r=-0.36, P<0.05), but there was no significant correlation in amblyopiagroup ( r =0.11, P>0.05).
·CONCLUSlON:There are no significant changes in MFT between children with ametropic amblyopia and normal children, but RNFL is thicker in amblyopia group than that in control group, and there is no correlation with axial length.

The application of PET-CT could not only play a part in the diagnosis of clinical stage for NSCLC on PET, but also make full use of the discrimination of anatomical structure on CT. The precision of target contouring could be increased and the normal tissue and risk organ are also protected efficiently.
3.Analysis Clinical Drug Use of Amikacin in a Hosiptal

OBJECTIVE:To reference clinical utilization of amikacin of a hospital.METHODS:By retrospective survey,the clinical information of 30 inpatients who were treated with amikacin and received plasma concentration monitoring from Apr.2006 to Feb.2010 were analyzed statistically in respect of primary disease,results of pathogen culture and drug sensitivity test,dosages and usage,plasma concentration,clinical efficacy,adverse drug reactions,etc.RESULTS:Clinical response rate was 55.2% and dosage of amikacin was small as well as measured value and reference value of plasma concentration.No adverse drug reaction was observed.CONCLUSION:The proportion of amikacin should be increased and plasma concentration monitoring of amikacin should be strengthened.Individual regimen is realized to improve effectiveness and safety of drug use.
4.Study of rehabilitation treatment on old patients with stroke at restoration stage

ObjectiveTo discuss the main point of rehabilitation treatment on old patients with stroke at restoration stage.Methods26 cases in hospital patients were performed a 8-week (5 days/week) rehabilitation program. Before and after rehabilitation treatment evaluations of locomotion function, muscle tone, range of movement of joints, ability of balance, ambulation and activities of daily living for affected limbs were performed and compared each other. Resultse grade of function and muscle tone of affected limbs had no distinct changes, while the locomotion function and ability of activities of daily living improved significantly(P<0.01). Conclusions Rehabilitation treatment for old patients with stroke at restoration stage is different from the acute stage. In the former case it is focused on facilitating the potential function, while in the latter it is emphasized on preventing and curing disuse syndrome and improving maladaptive behaviors.

6.A more accurate and efficient fluorescent probe of Caspase-8 activity based on flow cytometric fluorescence resonance energy transfer.

As a classic fluorescent detect technique, fluorescence resonance energy transfer (FRET) has been widely used in biological researches. Researchers have developed a series of fluorescence detect probes which were based on FRET. Caspase family plays an important role in apoptosis pathway, especially Caspase-8 which located, at the initial of death receptor mediated apoptosis pathway, whose its activation can trigger subsequent precaspases' activation and lead to apoptosis. So it is of great significance to detect the activation of Caspase-8 in apoptosis assay. In this study, a fluorescent probe based on FRET has been designed which can detect the activity change of Caspase-8 in cells. To identify the effectiveness and specificity of the probe, we measure the Caspase-8 activity under the Caspase-8 specifically activated apoptosis inducer RGD-TRAIL with the flow cytometry FRET detection platform. The results show that the probe can respond to the activity change of Caspase-8 in apoptotic cells, and the change can be quantified rapidly by flow cytometry. The study provides a more efficient and convenient detection method of Caspase-8 activity in living cells.

7. Protective effect of U83836E on ischemia-reperfusion injury in isolated rat heart

OBJECTIVE: To study the protective effect of U83836E (lazaroids) on ischemia-reperfusion injury (IRI) in isolated rat heart. METHODS: The isolated rat heart models of IRI were developed by linking the isolated rat heart to Langendorff perfusion system. U83836E was injected in different time, the rat heart function of ischemia-reperfusion was observed. Lactate dehydrogenase (LDH) and the cardiac treponin(cTnT) content in the outflow of coronary artery, and heat shock protein70 (HSP70) of myocardial tissue were measured. RESULTS: Compared with model group of ischemia-reperfusion, in the presence of U83836E, left ventricuIar systolic pressure(LVSP), the maximum rise rate of left ventricular pressure(+dp/dtmax) were increased (P<0.01), left ventricular end diastolic pressure(LVEDP)was depressed(P<0.05), the maximum fall rate left of ventricular pressure (-dp/dtmax) was accelerated (P<0.01). The content of LDH and cTnT in effluent samples of U83836E group(A group or B group) were lower than that in model group(P<0.01), the expression of HSP70 in myocardial tissue was increased(P<0.01). CONCLUSION: U83836E has protective effect on myocardial ischemia-reperfusion injury in isolated rat heart.
8. Effects of HepaCAM gene transfection on invasiveness of renal cancer cells

Objective: This study was to evaluate the effects of hepaCAM (hepatocyte cell adhesion molecule) on invasiveness of human renal cell carcinoma (RCC) cell line 786-0 and explore the underlying mechanism. Methods: A recombinant plasmid containing hepaCAM cDNA (hepaCAM-pEGFPN2) was transfected into 786-0 cells, then the positive cell clones were selected with G418 and amplified. The 786-0 cells transfected with pEGFPN2 and untreated 786-0 cells were as controls. The mRNA expressions of hepaCAM and matrix metalloproteinase (MMP)-2 and MMP-9 before and after stable transfection are detected by reverse transcriptase-polymerase chain reaction (RT-PCR) and real-time fluorescence PCR. The activities of MMP-2 and MMP-9 were analyzed by gelatin zymography. The invasiveness of 786-0 cells was determined by Transwell migration assay in vitro. Results: The mRNA transcription of hepaCAM was significantly increased in 786-0 cells after stable transfection of hepaCAM (P <0.01). The mRNA expressions and activities of MMP-2 and MMP-9 in 786-0 cells were markedly inhibited by hepaCAM (P <0.05). The transmembrane migration capacity of cells was significantly reduced after stable transfection of hepaCAM (P <0.01). Conclusion: HepaCAM inhibits the activities of gelatinase by down-regulating the expressions of MMP-2 and MMP-9, thereby preventing the invasiveness of renal cell carcinoma 786-0 cells.
