Objective To investigate the method of surgical treatment of mixed type Takayasu′s arteritis(TA).Methods Clinical data of 12 cases of TA were retrospectively analyzed.Results All 12 patients were treated with operations,including arterial bypass with prosthesis-graft in 5 cases;percutaneous transluminal angioplasty (PTA) and intraluminal stent implantation in 3 cases;combination of the two methods in 4 cases.All patients were followed up for 3 to 60 months.10 cases of organ ischemia or renal hypertension were very well controlled,2 cases showed restenosis of renal artery and one of them died from cerebrovascular accident.Conclusions Combination of arterial bypass,PTA and intraluminal stent impantation can control the ischemia of the vital organs and reduce the blood pressure of renal hypertension in patients with TA.

Objective To investigate the significance of K-ras gene status and ras protein expression in immunophenotypic classification of gastric signet ring cell carcinoma.Methods The expression of ras protein in 180 cases of gastric signet-ring cell carcinoma was detected by tissue microarray immunohistochemistry.Meanwhile,the mutation in codon 12,13 of K-ras gene was determined by using PCR-based DNA direct sequencing analysis.Results The rate of ras protein expression was 27.8％.The rate of ras protein expression in intestinal phenotype was significantly higher than those in gastric and gastrointestinal phenotypes(P<0.05).The rate of ras protein expression in cases with lymph node metastasis was significantly higher than those in cases without nodal involvement(P<0.05).The rate of ras protein expression was significantly higher in cases with deeper invasion(P<0.05).The frequency of K-ras gene mutation was 22(12.2％).All of them were found in codon 12.The types of mutation included GGT→AGT(1 case),GGT→TGT(1 case),GGT→GCT(2 cases),GGT→GTT(8 cases)and GGT→GAT(10 cases).K-ras mutation was significantly associated with intestinal phenotype(P<0.05).The rates of ras protein expression in cases with mutational type of K-ras gene was higher than those in cases with wild type(P<0.05).The ras protein expression was positively associated with K-ras gene mutation(r=0.61,P<0.05).Conclusion The ras protein expression is correlated with nodal involvement and invasion.K-ras gene mutation and expression of ras protein is related to phenotypic classification,and they might influence the phenotypic transformation in gastric signet ring cell carcinoma.

In order to explore the effects of testicular infection of murine cytomegalovirus (MCMV) on mature sperm viability at different periods following MCMV inoculation in mice, 91 BALB/c mice without MCMV infection were randomly divided into two groups: an experimental group (n = 56) and a control group (n = 35). The mice in the experimental group were treated by inoculating MCMV intratesticularly, while those in the controlled group were directly inoculated with DMEM without MCMV. The mice in both groups were sacrificed separately on the day 1, 1. 5, 2, 4, 6, 9 and 14 post-inoculation (D1) 1. 5, 2, 4, 6, 9 and 14 PI). The MCMV M83 mRNA gene was detected in the testis by in situ hybridization (ISH) with MCMV late-mRNA probe labeled with digoxin. Sperm viability of mature sperm in the epididymis cauda was measured. The results demonstrated the positive signal of ISH of MCMV was found mainly in the cytoplasm of the testicular interstitial cells and spermatogenic cells in the experimental group. Compared with that in the controlled group, the sperm viability in the experimental group was decreased significantly on D1 PI and D1.5 PI (P < 0.05). No statistically significant difference in the sperm viability was found after D2 PI between two groups (P > 0.05). This suggested that sperm viability in mice might be descended significantly shortly after MCMV infection and might return to normal with time, indicating that MCMV acute infection might temporarily degrade sperm quality and influence procreation transiently.

Eight-year medical education is to train medical personnel with high research abi lity and innovative ability, while the experimental teaching, as an important part of physiology teaching, plays an important role in training students in observation, problem-solving skills and practical ability. This article mainly discusses our exploration and practice of teaching in physiology experiment for eight-year program medical students from teaching contents and organization of class teaching, in order to fully increase the studying enthusiasms and meet the scientific mind of these students.

Observe and study the morphological changes and effects on yoleperoxidase (MPO) of traditional Chinese medical complex Intestine-clearing Suppository (ICS) in preventing and treating ulcerative colitis (UC).Method.. Divide the experimental rats randomly into 6 groups, i.e., blank, salicylazosulfapyridine (SASP), model, high, middle and low dosage of ICS, except the blank group, makeother rats experimental UC model with albumen immunologic method of rabbit colon mucosa, further divide them randomly into 2 experimental groups, i. e. , prevention and treatment, afterwards at 2nd~21st day, give prevention group the medicine, 22nd~35th day, treatment group, then decapitate them and take the colon tissue, and observe it morphologically under optical microscope, measure active MPO, analyse statastic results. Result: Through general morphological observation ,we found there were serious congestion ,edema, mucosa erosion, many large ulcers in the rats of model group; less serious mucosa erosion and quantity of ulcers in SASP and low-dosage ICS groups: small ulcers, not serious mucosa erosion, low degree of congestion and edema in high-and middle-dosage ICS groups; the scores of inflammatory injury of ICS groups were markedly lower than that of model (P＜0. 01), meanwhile, the preventive and reating effectiveness of high-and middle-dosage groups was better than that of SASP (P＜0.05). Under the optical microscope, there were large area of mucosa coloboma in the rats of model group, the ulcer penetrated to muscular layer and serous coat; in SASP group and low-dosage ICS group, there were erosive mucosa and inflammatory cell infiltration, granulation on the base; in middle-and high-dosage groups, large part of colon mucosa recovered, granulation formed on the base, ulcers were obviously less than that in model group (P <＜0.01); there had marked difference compared with that in SASP group (P＜0.05). The MPO active values of high-and middle-dosage groups were markedly lower than that of model group (P ＜ 0.01). Conclusion: Traditional Chinese complex ICS had good prevention and treatment to rats UC, itseffectiveness was positively correlated with dosage and better than that of SASP suppository.

Colorectal cancer remains a major threat to people’s health around the world. Researchers have paid more and more attention to colorectal cancer epigenetics. From two main aspects of colorectal cancer epigenetics: DNA methylation and histone modiifcation, this article analyzes the similarities and differences between patients with colorectal cancer in Eastern and Western countries. This review brielfy introduces epigenetic modiifcation of genes that were used to be biomarkers and therapeutic targets. Although there are some common features of colorectal cancer in the world, analysis has showed that some obvious epigenetic differences do exist in different races. For example, it had been conifrmed in the studies that there are differences in speciifc gene methylation, histone modiifcation sites and the degree of methylation and acetylation among countries, which provide the basis for speciifc diagnosis, treatment and prognosis of colorectal cancer in different ethnic groups. With improved research methods and increased sample size, more and more special molecular targets of colorectal cancer tissues will be found, and then personalized therapy for colorectal cancer can be achieved.

Objective To explore the basic medical mechanism of XinBao pill on electrophysiological characteristics of hyperpolarization-activated cyclic nucleotide-gated channel 4 (HCN4), and illustrate the mechanism of its therapeutical effect on bradycardia.Methods Human HCN4 mRNA was injected into the Xenopus laevis oocytes, after incubated for 2 ~3 days, channel current properties of HCN4 perfused with 40 mg/L XinBao Pill were observed by double electrode voltage clamp technique.Results At-90mV test potential, compared with control group (no XinBao pill), HCN4 channel peak current and tail current in 40 mg/L XinBao pill group had obvious changes, and V1/2 from ( -103.61 ±3.57)mV to ( -106.42 ±5.33)mV in XinBao pill group, from( -81.11 ±4.26)mV to( -86.36 ±7.44)mV in control group.The values of k from (15.15 ±2.23)mV to (17.33 ±3.58) mV in XinBao pill group, from(11.78 ±0.85)mV to(12.39 ±1.51)mV in control group(n=10).At test potential -90 mV, 40 mg/L XinBao pill perfusion fluid decreased the instantaneous current of(0.15 ±0.24)%, the EC50 was (30.8 ±4.8)mg/L (n=8).At test potential-140 mV~-100 mV level, 40 mg/L XinBao pill group increased the channel activation time constant compared with control group[(226.73 ±31.36)ms vs(143.67 ± 21.44)ms;-140 mV,n=10,P<0.05].40 mg/L XinBao pill group increased the channel deactivation time constant compared with control group [(1293.53 ±95.02)ms vs (647.12 ±61.35)ms;-140 mV,n=10,P<0.05].Conclusion The XinBao pill enhances the instantaneous current of HCN4 in a concentration-dependent manner, and extents channel activation and deactivation processes.
