4.Clinical detection and modulation strategy of lung cancer multidrug resistance with membrane transport proteins as the target

The major problem in lung cancer chemotherapy is the emergence of inherent and acquired drug resistance of the cancer cells. The cancer becomes resistant not only to the drugs used initially, but also to those to which it has not yet been exposed, This condition is known as multidrug resistance (MDR). MDR has been found to be related to three members of the ABC-superfamily of membrane transport proteins (transporters) and one member of the non-ABC-superfamily of transporter. The formers are P-glycoprotein (P-gp), multidrug resistance-associated protein (MRP), and breast cancer resistance pro-tein (BCRP). The latter is lung resistance-related protein (LRP). These findings provided new molecular targets to predict drug resistance, and an atraumatic detection method by using Tc-99m methoxyisobutyl isonitrile ( 99m Tc-MIBI ) SPECT has been developed for predicting MDR in lung cancer. Based on these transporters, different strategies are tried attempting to reverse drug resistance in lung cancer. Encouraging results have been acquired, but further research is necessary.

AIM To investigate the effects of levo phenoprolamine hydrochloride [ levo 1 (2,6 dimethylphenoxy) 2 (3,4 dimethoxyphenyl ethylamino) propane hydrochloride] [ l DDPH([?]D 25 1 08)]on experimental arrhythmias. on experimental arrhythmias. Apolipoprotein M (apoM) is a novel apolipoprotein discovered in 1999 and belongs tothe lipocalin protein superfamily.The APOM gene is mainly expressed in the liver and kidney,Where it is highly regulated by transcription factors,cytokines,hormones etc.Series of studies have shown that apoM plays an important role in lipoprotein metabolism especially in high-density lipoprotein (HDL).Since the relationship between HDL and atherosclerosis (AS) has been well known,apoM is supposed to be of potential fuctional importance in AS.Here we review the progress made in understanding the characteristics of apoM and its relationship to cardiovascular disease.
7.The expression of CIP2A and c-Myc and their correlation analysis in cervical carcinoma tissues

Objective To investigate the expression of CIP2A and c‐Myc and their correlation in cervical carcinoma .Methods We detected CIP2A and c‐Myc expression using immunohistochemistry in 72 samples of cervical carcinoma and 12 samples of normal cervix tissues .Correlation between proteins and clinicopathologic features and relation between CIP2A and c‐Myc expression were analyzed .Results The positive expression of CIP2A and c‐Myc in cervical carcinoma tissues were 52 .8% and 56 .9% ,respec‐tively .While their positive expression in normal cervix tissues were 8 .3% and 25 .0% ,respectively .The differences had statistical significance(χ2 =8 .169 ,P=0 .004 ;χ2 =4 .208 ,P=0 .040 ,respectively ) .Clinicopathological analysis suggested that CIP2A and c‐Myc protein expression were associated with histopathological differentiation and clinical stage(P<0 .05) in cervical carcinoma ,but the protein expression was not related to age ,lymph node metastasis and pathological type .CIP2A was significantly positive correla‐ted with c‐Myc protein in cervical carcinoma(r=0 .673 ,P=0 .001) .Conclusion The high‐expression of CIP2A is correlated with malignant clinicopathologic characteristics ,and CIP2A is positively associated with c‐Myc ,suggesting that CIP2A may promote tumor initiation and development through maintaining c‐Myc protein in cervical carcinoma .

Objective To study the effect of self-modified Guth & Samaha's myosin-ATPase staining method on the classification of SD rat skeletal muscle fiber types.Methods 8 ?m-thick cryostat sections from rat chest muscles and biceps brachii muscles were transfected with cryostat.Myosin-ATPase staining was carried out according to the following procedure: ① Fixing sections for 5 min in fixative solution containing 40 g/L paraformaldehyde;② Rinsing slides in Tris-rinse solution and then preincubating them in alkaline preincubation solution for 15 min;③ Rinsing slides in Tris-rinse solution twice and then incubating them for 60 min in incubation solution;④ Washing slides for three times in 10 g/L CaCl2 and then placing them in 20 g/L CoCl2 for 3 min;⑤ Washing sides in distal water and then placing them in 10 g/L(NH4)2S for 3 min and;⑥ Washing slides in running tap water for 3 min,dehydrating in graded ethanol,clearing in xylene and mounting in neutral balsam.Results After being stained by modified myosin-ATPase staining method,both chest muscle and biceps brachii muscle samples from SD rats could be clearly identified as type Ⅰ fibers and type Ⅱfibers as the fibers of type Ⅰ were stained white while the fibers of type Ⅱ were stained brown.Conclusion Modified myosin-ATPase staining method is a simple and effective way for muscle fiber type classification and can be applied in skeletal muscle related study.
