1.Mechanism Study of Kang’ai Fangyi Tablets in Inhibiting Angiogenesis of 4T1 Breast Cancer

Objective To study the angiogenesis-inhibitory mechanism of Kang’ai Fangyi Tablets ( KFT) , a Chinese compound recipe with the action of inhibiting cancer metastasis, for 4T1 breast cancer. Methods BALB/c mice were divided into blank control group, model group, Cytoxan ( CTX, 0.04 g·kg-1·d-1) group, and KFT ( 5.2 g·kg-1·d-1) group. Mice model of 4T1 breast cancer was established. Except that the blank control group and model group were given the saline, the mice in the medication groups were given the corresponding medicine. After medication for 4 weeks, the mice were executed, and then we calculated the mass of tumor, the inhibition rate of tumor mass, and the number of lung metastatic nodules. The number of microvessel and expression of vascular endothelial growth factor (VEGF) were measured by immunohistochemical method. Results Compared with the model group, mice tumor mass was decreased ( P<0.05) , the number of pulmonary metastatic nodules was reduced ( P<0.05) , and the number of tumor microvessel and VEGF expression in the isolated tumor mass showed the decreasing trend in KFT group. Conclusion KFT can inhibit 4T1 breast cancer growth and metastasis by reducing the tumor microvessel formation and by affecting the expression of VEGF.
2.Calculation of the surface density of the RGD-containing peptide on allogenic bone using isotopic tracing technique

Objective To investigate the feasibility of determining the surface density of RGD-containing peptide on allogenic bone by isotopic tracing technique using RGD peptide labelled with 125Ⅰ,and the impact of the input concentration of RGD-containing peptide on the surface density,and to obtain the history between the surface density and the input concentration of RGD-containing peptide.Methods The synthetic RGD-containing peptide was labelled with 125Ⅰ,and the specific radioactivity was calculated.The reactive solutions of RGD-containing peptide with the radioactive 125Ⅰ-RGD as a probe were prepared at the input concentrations of 0.01 mg/ml,0.10 mg/ml,0.50 mg/ml,1.00 mg/ml,2.00 mg/ml,4.00 mg/ml.Using EDC as the cross-linking agent,the reaction was carried out by placing the allogenic bone pieces into the reactive solutions of RGD-containing peptide with different input concentrations.After the reaction,the surface density of RGD-containing peptide grafted onto the allogenic bone pieces was calculated by evaluating the radioactivity and the surface area of the bone pieces.The impact of the input concentration of RGD-containing peptide on the surface density was measured and the curve was ascertained.Results After measuring in the radiodensity γ-counter,the result showed the RGD peptides have been marked with 125Ⅰ successfully.The allogenic bone pieces were radioactive after the reaction,which demonstrated that the RGD-containing peptide had been grafted onto the surface of bone pieces successfully.It was also found that with the increasing of input concentration,the surface density raised.Conclusion The surface density of RGD-containing peptide is related to its input concentration.With the increasing of input concentration,the surface density raises to the saturation value gradually.
3.Expression of Cyclin D1 and BRAF (V600 E) mutation in papillary thyroid carcinoma and their correlation with clinicopathologic features

Purpose To investigate the correlation of the expression of Cyclin D1 and BRAF ( V600E) mutation in papillary thyroid carcinoma ( PTC) and their clinical significance, and to analyze their role in the pathogenesis of PTC. Methods The expression of CK34βE12 and MC protein was detected by immunohistochemical EnVision method in 52 cases of PTC and 52 cases of thyroid benign lesions (25 cases of nodular goiters, 15 cases of Hashimoto’s thyroiditis, 12 cases of follicular adenoma), and the mutation status of BRAF gene protein in above specimens were detected by polymerase chain reaction and DNA sequencing. Expression of Cyclin D1 and BRAF (V600E) mutation in papillary thyroid carcinoma and their correlation clinicopathologic features were also analyzed. Results The positive rate of Cyclin D1 in 88 cases of PTC was 84. 6%,and the immunoreactivity score (4. 6 ± 2. 4), compared with thyroid be-nign lesions (1. 3 ± 1. 6), was statistically significant difference (t=8. 525, P<0. 01). Expression of Cyclin D1 was closely related with lymph node metastasis, capsular invasion, tumor stage Ⅲ+Ⅳ and BRAF (V600E) mutation (P<0. 05), but not related with age, gender, number and diameter of tumor nodules. In 52 cases of PTC BRAF gene mutation rate was 63. 5%, but in thyroid benign lesions mutation rate was 0, with significant difference (P<0. 01). The BRAF mutation is closely related with lymph node metastasis, capsular invasion and tumor stage Ⅲ+Ⅳ(P<0. 05), but not related with age, gender, number and diameter of tumor nodules. The expression of Cyclin D1 in BRAF (V600E) mutation group was stronger, the positive expression rate was significantly higher than that of wild group (100% vs 57. 9%), and expression of the mean score was higher than that of the wild group (5. 7 ± 1. 6 vs 4. 0 ± 2. 5);the comparison between the two groups showed statistical difference (t=2. 652, P<0. 05). Conclusions The expression of Cyclin D1 and BRAF (V600E) mutation are positively related, which are also closely related with the occurrence and development of PTC, and can be used as an index to predict the invasion and metastasis in PTC.

Outcomes for patients with hemophilia have improved dramatically over the past 50 years.With the increased availability of safe clotting factor concentrates,the primary focus in clinical management is now the prevention of long-term complications,most notably the debilitating hemophilic arthropathy that is associated with severe disease.Definitive evidence of improved clinical results from primary prophylaxis started in young patients-with severe hemophilia A and a minimal bleeding history is presented.Furthermore,recent studies showing benefits for initiating prophylaxis in older adolescents and adults with established joint disease are examined.lnhibitors to factor Ⅷ are the most problematic complication of factor replacement therapy.Patient-specific and treatment-related factors that contribute to the risk of inhibitor formation are discussed and controversies and clinical evidence related to approaches for tolerance induction are reviewed.Immune tolerance induction is the proven method for eradication of inhibitors.This article reviews latest research presented at the 55th ASH annual meeting on the hemophilias and their clinical management.
5.Research on Clinical Resistance and Aerolysin Genes of Aeromonas Hydrophila

Objective To study Aeromonas hydrophila infection and the clinical status of the major causative factor Aerolysin gene.Methods Clinical isolates of Aeromonas hydrophila was collected from 2012 to 2013 year in People’s Hospital of Shenzhen Longhua Branch.Its identification and antibiotic susceptibility testing was analyzed by VITEK 2 compact.Clinical resistance rates and distribution was analyzed by WHONET5.6 software.Polymerase chain reaction (PCR)was used to de-tect the Aerolysin gene form in chromosomes and plasmids extracted genome.Results The clinical total of 48 isolated Aero-monas hydrophila ,distributed in 16 clinical samples of sputum,blood 11,10 secretions,urine 7 and stool 4.Distribution in ward decentralized,the central tendency was not detected.Drug resistance rates to ampicillin,ampicillin/sulbactam and ce-fazolin reached more than 80%,while amikacin,cefepime,levofloxacin,piperacillin/tazobactam and imipenem was low 10%or less.43.7% strains carried the Aerolysin gene.39.5% of Aerolysin gene was found on chromosome genome.Conclusion Aeromonas hydrophila clinical infection existed in dispersed form,most of which carried Aerolysin gene in chromosome ge-nome.Aeromonas hydrophila had serious resistance to penicillins and first generation cephalosporin,but to broad-spectrum drugs maintaining high sensitivity.Precaution of Aeromonas hydrophila ,as an important condition pathogenic bacteria,is some significant for preventing it’s proliferation of drug-resistant strains.

Chelation therapy could remove transfusional iron burden. Three chelators are currently available, including deferoxamine, deferiprone, and deferasirox, which can be used as monotherapy or in combination.Several factors must be considered in the design of optimal and individualized chelation regimens,which include chelator availability and its properties,degree of organ-specific iron loading,ongoing transfusional iron burden, and patient preference. Comparative effectiveness trials may help to determine the ideal strategy. This article reviews latest research presented at the 53rd annual meeting of the American Society of Hematology (ASH) on the use of iron chelators.

8.Advances in Study on Hedgehog Signaling Pathway in Pathogenesis of Barrett’s Esophagus

Hedgehog( Hh)signaling pathway is evolutionarily conserved in vertebrates,its excessive activation is associated with abnormal cell differentiation, over proliferation, apoptosis resistance and promotion of invasion and metastasis of tumor cells. Hh signaling pathway involves in the formation and maintenance of esophageal columnar epithelium in embryonic stage,however,undetectable or barely expressed in matured esophageal squamous epithelium. Studies have shown that esophageal Hh signaling pathway can be activated by gastric acid and bile salts. Aberrant activation of Hh signaling pathway can cause the gradual transition of squamous epithelium to columnar epithelium and intestinal-type epithelium,ultimately induces the occurrence of Barrett’s esophagus( BE). Therefore,targeted inhibiting the Hh signaling pathway may be a new strategy for the treatment of BE. This article reviewed the advances in study on Hh signaling pathway in the pathogenesis of BE.
9.Effect of initial periodontal therapy on the eradication of gastric Helicobacter pylori

Objective: To investigate whether periodontitis might adversely affect the outcome of eradication of gastric Helicobacter pylori(Hp) . Methods: Ninety one patients with gastric Helicobacter pylori and periodontitis were enrolled. The patients were treated by medicine therapy, meanwhile 33 of them were treated by initial periodontal therapy. After treatment, the effect of initial periodontal therapy on the eradication of gastric( Hp ) was examined with 13 C urea breath test. The correlation between the gastric eradication rate and periodontitis was analyzed. Objective:To investigate involvement of the Ras/Erk signal pathway in activation of human ??T cells stimulated by Mycobacterium tuberculosis low molecular peptide antigen(Mtb-Ag).Methods:PBMC were isolated from peripheral blood of healthy subjects and simulated in vitro with Mtb-Ag,CD3mAb or PMA and inomysin(IM).CD69 expression of total T cells and ??T cells were measured by flow cytometry at different hours after stimulation, and the number of expanded ??T cells were counted after 10 days of culture. PD98059,a specific inhibitor for Erk pathway was used to treat PBMC for 30 min before stimulation.Results:The proportions of CD69 expressing cells in total T cells and ??T cells were same as 99% at 6 h after stimulation of PBMC with PMA +IM and similar about 55% at 24 h after stimulation with CD3 mAb,whereas those at 24 h stimulation of PBMC with Mtb-Ag were 75.2% and 16.0%,respectively.CD69 expression and proliferation of ??T cells activated by Mtb-Ag were markedly inhibited by pretreatment of PBMC with PD98059.Conclusion:Mtb-Ag is a specific stimulator for activation of ??T cells, and Ras/Erk signal transduction pathway involves in the activation of ??T cells.
