To elucidate the role of reactive carbonyl compounds in the pathogenesis of vascular complication in patients with chronic renal failure or diabetes. Vascular endothelial cells (VECs) were isolated from human umbilical vein and cultured with 3-deoxyglucosone (3-DG) or methylglyoxal(MGO) in vitro. Expression of ICAM-1 and VCAM-1 on the surface of VECs was detected by flow cytometer. The results showed that up-regulation of expression of ICAM-1 and VCAM-1 was observed when either 3-DG or MGO was added into the cultures, which was inhibited by a carbonyl compounds scavanger, aminoguanidine. These data suggested that accumulation of reactive carbonyl compounds in patients with chronic renal failure or diabetes might be involved in the mechanism of arteriosclerosis.

3.Effect of transcutaneous electrical stimulation on the gene expression of MyoD from denervated skeletal muscle of rats

BACKGROUND: MyoD expression of skeletal muscle of rats increases distinctly in the earlier period of denervation, which can greatly postpone denervated skeletal muscle atrophy. The clinical test testifies that electrical stimulation is an effective method to cure denervated muscle atrophy. But the influence of electrical stimulation on MyoD expression during denervated muscle atrophy is still unproved. OBJECTIVE: To discuss the influences of electrical stimulation on the gene expression of MyoD of skeletal muscle of rats. DESIGN, TIME AND SETTING: Randomized control animal experiment was performed in the Animal Experimental Center of Shanxi Medical University between July and November in 2008. MATERIALS: A total of 36 healthy Sprague-Duwley rats of either gender were divided randomly into three groups, control group, denervation group and electrical stimulation group. Each group contained 12 rats. METHODS: Standard models of right sciatic nerve dissociation and gastrocnemius denervation were established in right limb of each rat in denervation group and electrical stimulation group. Thirty-minute electrical stimulation was given to the denervated gastrocnemius muscle of each rat of electrical stimulation group once a day. The rats were executed at 2, 7, 14, 28 days after denervation to dissect and dissociate their gastrocnemius muscles of the right lower limbs. MAIN OUTCOME MEASURES: MyoD mRNA expression was detected by reverse transcription polymerase chain reaction, and MyoD protein level by immunohistochemistry. 4.Myogenic regulatory factor Myf-5 expression at different periods of denervated skeletal muscle atrophy

BACKGROUND: Myogenic regulatory factors Myf-5 is an importance gene involved in muscle occurrence process to adjust and control, start and maintain the skeleton muscle cell growth. It has a possible relation with denervated skeletal muscle atrophy. OBJECTIVE: To detect the expression of Myf-5 gene in different skeletal muscle sitas and different periods following denervatad skeletal muscle atrophy.DESIGN, TIME AND SE'I-rlNG: Randomized control animal experiment was performed in the Shanxi Medical University between March and April in 2008. MATERIALS: A total of 24 Sprague-Dudley male rats was selected and divided randomly into four groups, with six in each, Le. sham operated group (innervations), denervated 2 d group, denarvated 7 d group and denervatad 28 d group. METHODS: Sham operations were made only for the rats in the sham operated group and sciatic nerve were not cut off. Sciatic nerves were cut off more than one centimeter at the mid-level of their right lower limb for the rats in denervated groups. The rats were executed by vertebrae dislocation method at 2, 7, 28 days after denervation. The skeletal muscles of the right lower limb (tibialis anterior, soleus, gastrocnemius and plantar) were dissected and dissociated. MAIN OUTCOME MEASURES: Myf-5 mRNA expression was detected by reverse transcription-polymerase chain reaction. Immunohistochemical stain (ABC method) with polyclonal antibody against Myf-5 protein was performed and the gray value was counted.RESULTS: Expressions of Myf-5 mRNA in denervatad skeletal muscle were up-regulated at 2, 7, 28 days in the eady stage of denervation (P < 0.05). The number of Myf-5 antibody positive-stained cell nuclei was the most in satellite cells at 28 days following denervation. CONCLUSION: At the early stage of denervated skeletal muscle atrophy in SD rats, expressions of Myf-5 in different skeletal muscles are all up-regulated. Denervation of rat skeletal muscle raises the Myf-5 expression in satellites cells.
5.Imaging measurement for internal fixation design of axial lumbosacral vertebral fusion viaposterior rectal space

BACKGROUND: The minimally invasive axial lumbar interbody approach (AxiaLIF) for L4–S1 fusion has been applied in America and Europe, and has obtained satisfactory curative efficacy. Because of significant anatomical differences between Chinese and Europeans and Americans, whether AxiaLIF is appropriate for Chinese remains unclear. Moreover,there are some problems in the application of AxiaLIF, so how to optimize AxiaLIF is a key to its promotion in China.OBJECTIVE: To provide anatomical data for the design of axial screws suitable for Chinese through measuring the mid axial line of the lateral lumbar radiograph and cross sections of L5 and S1 on lumbar CT in normal Chinese population.METHODS: The lateral lumbar radiographs from Chinese healthy population were selected, including 35 males and 30 females, the axial height of S1, the disc distance between L5 and S1, and the axial height of L5 were measured so as to provide anatomical data for designing the length of the axial screw. The transverse and sagittal diameters of L5 and S1 in the lumbar CT of 26 adult healthy males and 24 healthy females were measured to provide anatomical data for designing the diameter of the axial screw.RESULTS AND CONCLUSION: (1) The axial height of S1 in males and females was (26.76±3.94) mm and (22.91±2.91) mm, respectively (P < 0.05). The disc distance between L5 and S1 was (12.62±1.90) mm for males and (11.92±1.78) mm for females (P > 0.05). The axial height of L5 was (29.12±2.18) mm for males and (26.91±2.47) mm for females (P <0.05). (2) The transverse diameter of S1 was (49.14±4.14) mm for males and (46.11±4.44) mm for females (P < 0.05).The transverse diameter of L5 was (41.34±4.31) mm for males and (43.12±3.71) mm for females (P < 0.05). The sagittal diameter of L5 was (34.48±2.32) mm for males and (33.03±3.48) mm for females, and the sagittal diameter of S1 was (35.65±4.28) mm for males and (33.53±3.26) for females, (both P > 0.05). (3) That is to say, this study provides the anatomical data for designing the axial screws suitable for the lumbar fusion of Chinese by measuring the mid axial line of the lateral lumbar radiographs and the cross sections of L5 and S1 on lumbar CT. The image measurement method can be used to analyze the preoperative images of the patients to predict the feasibility of the surgical approach and pre-select the internal fixation model for personalized screw positioning.
6.Interaction of novel gene AngRem104 and Bardet-Biedl syndrome 2 protein in mammalian cells

Objective To screen for proteins interacting with novel gene AngRem104 and to identify the putative interaction of novel gene AngRem104 and Bardet-Bied1 syndrome 2 (BBS2) protein in mammalian cells. Methods The yeast strain AH109 was transformed with AngRem104pGBKT7/c-myc and yeast-mating was utilized to screen for interacting proteins with AngRem104 in pretransformed human kidney cDNA library. The human embryonic kidney (HEK 293T) cells were transformed with two recombined plasmids,AngRem104-pcDNA3.1/V5-His and BBS2-pCMV/c-myc. Mouse anti-human V5 monoclonal antibody and mouse anti-human c-myc monoclonal antibody were used in immunoprecipitation and immunoblot analysis, respectively. Results Seven proteins that interact with AngRem104, including BBS2 were identified. The AngRem104-V5 and the BBS2-c-myc fusion protein were detected respectively in the immunoprecipitation by anti-c-myc and anti-V5 antibody. Conclusion The novel gene AngRem104 may interact with BBS2 protein in mammalian cells,which provides insights as to the function exploration of novel gene AngRem104 and the pathogenesis investigation of Bardet-Biedl syndrome.
7.The investigation on occupational stress of radiation workers

Objective To study the relationship between occupational stress and metal health of radiation workers.Methods Totally 220 radiation workers were investigated by Chinese perceived stress scales questionnaire,Chinese health questionnaire-12,as well as questionnaire related to radiology.Results The stress score of radiation worker with senior title was 18.18 ± 6.12,which was the lowest of all 220 participants.There was positive relationship between stress score and health score (r =0.484).19.1% of the workers sometimes felt stressed due to the danger of work.Conclusions There are some significantly correlation between occupational stress and metal disorder.More attention should be paid on the occupational stress.Positive measures such as decompression and psychological intervention will be helpful to protect physical and mental health of radiation workers.
8.Efficacy of subcutaneous administration of gonadotropin-releasing hormone agonist on idiopathic central precocious puberty.

In order to assess the feasibility of subcutaneous administration of Triptorelin with 6-week intervals for the suppression of pituitary-gonadal axis and changes of clinical signs in girls with idiopathic central precocious puberty (ICPP), 46 girls with ICPP were treated with GnRHa. Triptorelin (Decapeptyl, 3.75 mg) was administered subcutaneously (SC) at 6-weeks intervals or intramuscularly (IM) at 4-weeks intervals randomly for more than 12 months consecutively. During GnRHa therapy, clinical parameters and laboratory data, including height, weight, pubertal stage, bone age, uterine volume and ovarian size, serum levels of luteinizing hormone (LH), follicle stimulating hormone (FSH) and estradiol (E2), were monitored and analyzed. It was found that both treatment regimes led to regression of precocious puberty and reversal of secondary sexual characteristics. Breast developments regressed. Uterine volume was decreased after treatment, but there was no statistically significant difference. Mean ovarian volume did not change significantly during treatment. The height velocity was decreased significantly from 6.3+/-1.4 cm/year to 5.8+/-1.2 cm/year in group SC and 6.7+/-1.3 cm/year to 5.4+/-1.0 cm/year in group IM, respectively. The rate of bone maturation was reduced significantly during treatment. The ratio of deltaBA/deltaCA was 1.2+/-0.2 or 1.3+/-0.3 at the onset of therapy and decreased significantly after the treatment to 0.7+/-0.2 or 0.9+/-0.1, respectively. The predicted adult height was increased significantly and progressively during therapy. The levels of serum LH, FSH and E2 returned to the prepubertal condition. No significant side effects of therapy were noted. The most common side effect during SC treatment was that a non-irritating, 1 cm in diameter mass was palpated at the site of subcutaneous injection in the abdominal wall of patients, which disappeared after 6-12 weeks. Two girls had minimal withdrawal vaginal bleeding episodes after the first injection. It was concluded that both IM and SC triptorelin administrations were clinically effective. They induce profound suppression of hypothalamic-pituitary-gonadal axis while stabilizing height velocity, slowing bone maturation and increasing predicted adult height. These results suggest that subcutaneous injection of triptorelin in 6-weeks intervals at a dosage of 3.75 mg be a safe and acceptable regimen for ICPP

10.Influence of users' cognition on information retrieval behaviors in ubiquitous knowledge environment

Users' cognition and information retrieval are faced with great challenges in ubiquitous knowledge environ-ment. The influence of cognitive factors such as personality traits, cognitive style and ability, knowledge structure and ascribed methods on users' information retrieval behaviors was thus analyzed. It was pointed out that different cognitive factors could influence the retrieval, judgment and absorption of its results,users' satisfaction and continuous use in ubiquitous knowledge environment,with suggestions put forward from the aspects of information retrieval education, information service and information users for improving the users ' knowledge system in knowledge assimilating process by working out effective retrieval strategies using the advantages of cognition.
