1.Dynamic stuey on intracellular free calcium,ATP level and membrane Ca 2+-Mg2+ATPase activity of erythrocyte in patients with acute cereb ral infarction

Objective To explore the dynamic alteration of intracellular free calcium concentration([ Ca2+]i),ATP level and membrane Ca2+-Mg2+ATPase acti vity of erythrocyte in the patients with acute cerebral infarction(CI).Methods we examined [Ca2+]i,ATP level and membrane Ca2+-Mg2+ATPase activity of erythrocyte in 30 patients with acute CI and 28 health controls by Fluorescence Activated Cell Sorter. Results [Ca2+]i in erythrocyte increased significantly in CI group(P<0.01),while the ATP level and membrane Ca2+-Mg 2+ATPase activity were lower than the controls(P<0.05,P<0.0 01).The above result was more remarkable in the elderly group than the young one .The dynamic alteration of [Ca2+]i in erythrocyte increased obv iously during 1~2 days after the attack,and reached the peak in 3~7 day s,it decreased slowly to the slightly low level at the beginning of th e attack in about two weeks,but it was still higher than the controls.The dynamic alteration of ATP level and Ca2+-Mg2+ATPase activit y after acute CI,it decreased significantly during 1~2 days after the at tack,and reached the lowest in 3~4 days.this status could last about one week.Then both of them increased slightly. There was remarkable negative correlation betwe en RBC [Ca2+]i and ATP level or membrance Ca2+-Mg2+ A TPase activity (r=-0.904,r=-0.978,P<0.05).There was positive correl ation between ATP lev el and membrane Ca2+-Mg2+ATPase activity(r=0.835,P<0.05 ).Conclusion There was calcium overload [Ca2+]i in th e intracellular of erythrocyte in acute CI,ATP level and membrane Ca2+ -Mg2+ATPase activity of erythrocyte CI was involved in the pathologi cal course of calcium overload,and related to the age.
2.Effects of cryopreserved neural stem cell transplantation on rat axonal regeneration after spinal cord injury:Retrograde tracing observation

AIM:There are some reports about the treatment of spinal cord injury with neural stem cells(NSCs) transplantation,but the ideas about the transplantation time and patterns and detection indexes are still different.In this study,the effects of cryopreserved NSC transplantation on the axonal regeneration after the spinal cord injury in rats were observed.METHODS:The experiment was carried out in the experimental animal center of China Medical University from June 2005 to June 2006.①Thirty-six adult Wistar rats,either male or female and 250-300 g,were provided by the experimental animal department of China Medical University.Neural stem cells were isolated from 10 neonate rats and cultured.All treatments for animals were accorded with the animal ethical criteria.②NSCs at logarithmic phase were cryopreserved at-70 ℃ for 2 weeks and labeled with 5-bromodeoxyuridine(Brdu) after rewarming.After the models of spinal cord injury were established,the NSCs were transplanted into the injured site immediately.Thirty-six rats were randomly divided into NSC transplantation group,DMEM solution group,and control group.③NSC survival and migration were detected by immunohistochemistry,and the reconstruction of spinal cord were detected by horseradish peroxidase(HRP) staining.RESULTS:Of the 36 spinal injured model rats,4 died of overdose of anesthesia,and 5 died of infection,but all were supplemented.Brdu-labeled positive NSCs were detected in the injured spinal cord after transplantation on the 7th day,and increased on the 14th day,then gradually decreased since the 28th day till disappeared.The number of HRP positive cells in transplantation group was significantly higher than DMEM solution.CONCLUSION:Cryopreserved NSCs can survive in the injured site after transplantation and promote the reconstruction of axoplasm pathway of injured spinal cord.

5.Repair of spinal cord injuries in mice using peripheral nerve and activited macrophages transplantation

[Objective]To explore the effect of spinal cord injury(SCI) repair by peripheral nerve combined with activitied macrophages graft.[Method]Fistly the preparation of the activated macrophages and fabrication of the peripheral nerve were done.81 BALB/C mice were grouped in A,B,C,D groups randomly after their spinal cord were hemi-resected.20 mice of group A were dealed with activated macrophages graft;20 of group B were dealed with peripheral nerve graft;21 of group C were transplanted both two graftes;20 of group D were contral group.Each of the 4 groups was respectively divided into 3 subgroups randomly.7 of subgroup 1 were investigated by their restoration of the motor function and improvement of the somatosensory evoked potential(SEP).7 of subgroup 2 were tabken for histological examination.The spinal cords were fixed and cut into longitudinal frozen sections,and were immuneostained and stained with hematoxylin and eosin(HE).The numbers of cells or neurofilaments were counted under microscope.6 of subgroup 3(7 of C3)were used as complement ones.All the results were recorded from 1 to 12 weeks postoperative and analyzed statistically using the SPSS 12.0 software package.[Result]The BBB scale and the SEP wave:there was a significant difference between group C1 and the other three groups,there was a significant difference between group A1,B1 and D1,too(ONE WAY ANOVA,P
6.Surgical treatment of old thoraco-lumbar vertebral fracture with kyphosis and spinal cord injury

[Objective]To explore the surgical method and clinical effects for the treatment of old thoraco-lumbar vertebrral fracture with kyphosis and spinal cord injury[Method]Thirty-six patients with old thoraco-lumbar vertebral fracture with kyphosis and spinal cord injury from 1998 to 2003,all had undergone anterior decompression through posterior median approach.[Result]All patients were followed-up.Kyphosis was rectified well.The Frankle classification of all but 3 patients with complete paraplagia increased one or more degrees after operation.[Conclusion]Operation through posterior median approach can achieve anterior decompression effectively.
7.Effects of Total Glucosides of Paeony on the Expression of CD4 + T Lymphocytes and IL - 6 in Rats with Chronic Abacterial Prostatitis

OBJECTIVE:To investigate the effects of total glucosides of paeony(TGP) on the expressions of CD4+ T lymphocytes and IL- 6 in prostatic tissues of rats with chronic abacterial prostatitis(CAP) and to study its action mechanism on CAP.METHODS:36 SD rats were randomly divided into three groups:CAP control group(n = 12 rats),low dose CAP + TGP group(n = 12) and high dose CAP + TGP group(n = 12).The expressions of CD4+ T lympholeukocytes and IL- 6 in prostatic tissues of rats were detected by immunohistochemistry after medication were investigated,and the changes in histopathology of prostatic tissues.RESULTS:Significant inflammatory condition was noted in CAP control group;improvement of glandular structure and infiltration of small number of inflammatory cells were noted for low dose TGP group.In high dose TGP group,inflammation was abated markedly and there was diffused distribution of a few number of inflammatory cells. 8.Clinical research of Finasteride in treating hemospermia 30 cases

Objective To evaluate the efficiency of Finasteride in treating inflammatory hemospermia and approach the mechanism of action. Methods 56 cases of hemospermia were randomly divided into two groups. The experimental group (n = 30) was treated with Finasteride combined with antibiotic for 1 month, while the control group(n=26) was treated only with antibiotics for 1 month. The expression of VEGF in sperma was de-tected by enzyme linked immunosorbent assay (ELISA) in pretherapy and post-treatment. Results Hemospermia disappeared in 28 cases of experiment group(28/30, 93.3%) and in 17 cases of control group(15/30,57.7%),with significant difference. The expression of VEGF degrade in post-treatment. Conclusions Hemospermia can be treated more effective with finastefide combined with antibiotics. The mechanisms may be associated with Finas-teride can reduce VEGF synthesis and inhibit the microvascular formation.
9.Intravenous infusion of dexmedetomidine to prevent irritation and cough causing by sufentanil

ObjectiveTo observe whether if intravenous infusion of dexmedetomidine in advance can prevent irritation and cough causing by sufentanil.MethodsNinety patients who performed excision of intracranial tumor were divided into observation group and control group by random digits table with 45 cases each.Patients in observation group were intravenously infused 0.5 μg/kg dexmedetomidine for 10 minutes before induction.Patients in control group were intravenously infused 0.9% sodium chloride for 10 minutes before induction.All the patients were intravenously infused 0.3 μg/kg sufentanil after taking drugs of load dosage in 3 seconds,and the incidence of irritation and cough in 1 minute was observed.The mean arterial pressure and heart rate were recorded before infusing dexmedetomidine or sodium chloride(T0),at the end of infusing dexmedetomidine or sodium chloride (T1),before intubation (T2),1 minute after intubation (T3).ResultsThe incidence of irritation and cough,moderate-severe irritation and cough in observation group [ 24.4% ( 11/45 ),11.1% (5/45) ] were lower than those in control group [ 48.9% (22/45),22.2% ( 10/45 ) ]with significant differences(P< 0.01 ).The mean arterial pressure in control group at T2 was lower than that in observation group [ (71.2 ± 3.5) mm Hg( 1 mm Hg =0.133 kPa) vs.(84.7 ± 4.1 ) mm Hg] with significant difference (P < 0.05 ).The mean arterial pressure in control group at T3 was higher than that in observation group [(96.7 ± 6.4) mm Hg vs.(83.1 ± 5.2) mm Hg] with significant difference(P< 0.05).The heart rate at T1,T2,T3 in observation group was lower than that in control group [ (53.2 ± 4.7 ) beats/min vs.(70.4 ± 6.2 )beats/min,(56.3 ± 3.1 ) beats/min vs. (64.7 ± 3.7) beats/min,(59.1 ± 4.8) beats/min vs. (81.5 ± 6.1 )beats/min],and there were significant differences(P <0.05).Conclusions Intravenous infusion of dexmedetomidine (0.5 μg/kg) in advance can effectively prevent irritation and cough causing by sufentanil,and can enhance the stability of circulation during intubation.
