Objective To investigate the effects and the possible mechanisms of berberine in human cervical cancer Hela cell line in vitro. Methods Cell growth rate was determined with MTT assay. TUNEL was used to examine cell apoptos rate. The expression of COX-2 and Bcl-2 proteins were detected by immunocytochemistry method. Results Berberine induced cytotoxicity and apoptosis in human cervical carcinoma Hela cells in time-dependent and dose-dependent manner. Berberine could down-regulated the Bcl-2 protein expression of Hela cells,but had no significant influence in the expression of COX-2 protein. Conclusion Berberine can inhibit Hela cell growth of cervical carcinoma and induce apoptosis in vitro. Down-regulation of the Bcl-2 protein expression may be involved in berberine-induced apoptosis of HeLa cells.

OBJECTIVE To analyze the hospital infection-related factors of infant incubator and their countermeasures.METHODS The hospital infections cases with the use of infant incubators from Jan 2005 to Sept 2008 were retrospectively analyzed.RESULTS From the 205 cases of children with the use of infant incubator,14 cases were infected,the hospital infection rate was 6.82%.Their risk factors were weak immunity,poor hygiene concepts,not strictly executing the hand disinfection systems and hygiene norms,unsatisfactory basic care and high humidity in incubators.CONCLUSIONS To strictly execute the disinfection system and hand hygiene norms,rationally use the antibiotics and to strengthen the basic care and care management can reduce the incidence of hospital infections.

BACKGROUND:Ligustrazine hydrochloride which promotes nerve repair can be applied to the treatment of nervous system injury. OBJECTIVE:To investigate the effect of ligustrazine hydrochloride combined with bone marrow mesenchymal stem cels transplantation on electrophysiological property and hindlimb function of rats with spinal cord injury. METHODS:T9 spinal cord transection injury models were made in rats using Alen’s method, and then rat models were randomized into three groups: rats in control group received tail vein injection of culture solution; rats in cel transplantation group underwent bone marrow mesenchymal stem cel transplantationvia the tail vein; rats in combined group were subjected to the tail vein injection of ligustrazine hydrochloride and bone marrow mesenchymal stem cels that lasted for 4 hours. At 1, 2, 4, 6, 8 weeks after modeling, Basso, Beattie and Bresnahan scores and modified Tarlov scores were used to detect the motor function of rats. At 72 hours after modeling, RT-PCR method was used to detect the expression of Bcl-2 and basic fibroblast growth factor around the injured region. At 4 weeks after modeling, somatosensory and motor evoked potentials were measured for evaluation of neurophysiological recovery. At 8 weeks after modeling, horseradish peroxidase tracer was used to assess the regeneration of rat spinal cord nerve fibers; PKH-26 labeling was used to observe the survival and migration of transplanted cels. RESULTS AND CONCLUSION:At 1, 2, 4, 6, 8 weeks after modeling, Basso, Beattie and Bresnahan scores and modified Tarlov scores were significantly higher in the combined group than the cel transplantation folowed by the control group (P < 0.05). At 72 hours after modeling, the expression of Bcl-2 and basic fibroblast growth factor around the injured region was significantly higher in the combined group than the cel transplantation group and control group (P < 0.05). At 4 weeks after modeling, the latencies of somatosensory and motor evoked potentials were ranked as folows: combined group < cel transplantation group < control group (P < 0.05); the amplitudes of somatosensory and motor evoked potentials were ranked as folows: combined group > cel transplantation group > control group (P < 0.05). At 8 weeks after modeling, horseradish peroxidase-labeled pyramidal cels in the cel transplantation group and combined group showed apparent crossing signs; the number of PKH-26-positive cels and horseradish peroxidase-positive cels was the most in the combined group folowed by the cel transplantation group, and was the least in the control group (P < 0.05). These findings indicate that ligustrazine hydrochloride combined with bone marrow mesenchymal stem cels transplantation can facilitate nerve cel regeneration, promote the expression of Bcl-2 and basic fibroblast growth factor, and improve motor function in rats after spinal cord injury. Objective: We studied the differences in intelligence,self-concept and behavior problem between girls with true precocious puberty and the normal girls.Methods: 35 girls with true precocious puberty aged 8 to 10 years was put into the research group;another 35 girls was put into the normal group(n=35).Both the research group and normal group completed the Piers-Harris Children's Self-concept Scale(PHCSS) and Raven's Standard Progressive Matrices in order to evaluate their levels of self-concept and intelligence respectively.Their parents completed a Child Behavior Check-list(CBCL) and a questionnaire on background status.Results: ①Children in the research group had significantly higher intellectual level than those in the control group.②The PHCSS yielded significantly lower scores of total self-concept,behavior and somatic appearance in the research group than those of the control group.③Higher scores of internal behavior problems,external behavior problems,depression,poor social communication,somatic complaints,delinquent behavior and aggressive behavior at CBCL showed in the research group than those of the control group.Conclusion: ①The level of intelligence of the children in research group was higher than that of children in the control group.②Girls with true precocious puberty showed more behavior problems and lower level of self-concept than those in the control group.

Objective: To identify the related psychosocial risk factors of emotional disorders in children. Methods:To use case-control approach in which. Diagnosis was made by clinical interview according to ICD-10 criteria. Eighty eight cases and controls separately filled out general condition inventory. The results were put into Logistic regression model for analysis. Results: The children with timid personality, without kindergarten education, or with parents who were administrative or technical personnel, were apt to have emotional disorders. The children who were usually counseled by their mothers had less emotional disorders than those were beaten. Conclusion: The emotional disorders were the results of multiple factors. Prevention of children's emotional disorders should be focused on the children's personality and family education.

Objective To explore the effects of different extracts of modified Siwu Siteng Decoction (TFT-Ⅰ, TFT-Ⅱ) on rat models with hyperuricemia. Methods Rat model with hyperuricemia was induced by hypoxanthine and oxonic acid potassium salt. Seventy rats were randomly divided into 7 groups:normal group, model group, allopurinol group, TFT-Ⅰ high and low dose groups, TFT-Ⅱhigh and low dose groups. Each dose group was given lavage with related medicine, and blank group and model group were given lavage with distilled water for one week, respectively. Uric acid in serum (SUA) was measured through fully automatic biochemical analyzer, and xanthine oxidase (XOD) activity in serum and liver tissue was measured through colorimetric method. Results Compared with model group, TFT-Ⅰ and TFT-Ⅱ high dose groups significantly reduced the level of SUA, serum and liver XOD activity (P<0.05), with significant statistical difference between the two groups (P<0.05). TFT-Ⅰ and TFT-Ⅱ low dose groups had no significant effects on SUA, and XOD activity in serum and liver (P>0.05). Conclusion Alcohol soluble ingredients of astragalus membranaceus and Siwu decoction in modified Siwu Siteng Decoction can effectively reduce the level of blood uric acid.

AIM:To explore the ability of different group B streptococci ( GBS) strains on inducing platelet activation.METHODS:Six strains of GBS, separated from the septic patients with thrombocytopenia, were used as the inducers.Light transmission aggregometry was used to measure platelet aggregation.Scanning electron microscopy ( SEM) was performed to investigate the interaction of platelets with bacteria.The expression of platelet CD62P, Toll-like receptor 2 ( TLR2) and TLR4 was determined by flow cytometry and Western blotting.Furthermore, the activity of platelet TLR2 (or TLR4) was blocked by anti-TLR2 (or anti-TLR4) monoclonal antibody, and the platelet aggregation induced by GBS was detected.RESULTS:Only 3 of 6 GBS strains isolated from the septic patients induced platelet aggregation and up-regulated the expression of CD62P and TLR2 in the platelets (P<0.05), but not TLR4.Incubation with anti-TLR2 anti-body, but not anti-TLR4 antibody, significantly blocked platelet aggregation induced by GBS.CONCLUSION:Some GBS strains from the patients are able to trigger platelet activation in vitro, and platelet TLR2 may play an important role in the interaction between GBS and platelets.

Objective To investigate the clinical characteristics,treatment,and prognostic factors for Langerhans cell histiocytosis (LCH) in children.Methods A retrospective review of patients diagnosed as LCH was carried out between January 2007 and December 2012 in Beijing Children's Hospital,Capital Medical University.Target patients were divided into multi-organ high-risk groups (Group Ⅰ),multi-organ low-risk groups (Group Ⅱ),single-organ groups (GroupⅢ),and the corresponding intensity of chemotherapy was given.SPSS 17.0 statistical software was used to analyze the findings.The correlations among the affected organ,the early treatment response and the prognosis were analyzed.Results A total of 217 patients were analyzed including 127 boys and 90 girls with ratio of 1.4 ∶ 1.0 and a median age of 36 months (ranged from 2 months to 14 years) on the diagnosis of LCH,and there were 132 cases (60.8％) in group Ⅰ,33 cases (15.3％) in group Ⅱ and 52 cases (23.9％) in group Ⅲ.The median age on diagnosis was 20 months in group Ⅰ,42 months in group Ⅱ and 72 months in group Ⅲ.The most frequently affected organ was the bone (176 cases,81.2％).Among 217 patients,55 cases (25.3％) had recurrence and 12 cases died.The rate of 3-year overall survival was expected to be 90.78％.The rate of 3-year event free survival was expected to be 76.5％.Myelosuppression,liver function damage and infection were the most common side effects due to chemotherapy with the percentages of 48.4％ (105 cases),24.0％ (52 cases) and 12.4％ (27 cases).Risk organs involvement and no-response to initial therapy (after 6 weeks) indicated a worse prognosis (x2 =10.60,12.84,P =0.017,0.001).Conclusions Incidence of LCH in boys is slightly higher than girls in children.Peak age at onset of LCH in children is 1-3 years old.Bone is the most frequent involved organ.Involvement of risk organs and no-response to initial therapy are key factors in determining worse prognosis.A rescue therapy should be introduced earlier in these patients.

Objective To investigate of the change of peripheral blood lymphocyte subsets and blood cell antibodies in aplas‐tic anemia and its clinical significance .Methods The lymphocyte subsets CD3+ ,CD3+ CD4+ ,CD3+ CD8+ ,CD4+ /CD8+ ,CD3-CD19+ ,CD3-CD16+CD56+ from 25 aplastic anemia(including SAA 7 cases ,NSAA18 cases) ,and hematocyte antibody detection (granulocyte ,erythrocyte ,platelet cytomembrane) and 20 controls were examined by flow cytometry .Results CD4+ T lymphocyte percentage ,CD4/CD8 ratio ,the percentage of CD56+ in aplastic anemia patients significantly decreased(P<0 .05) .Blood cell anti‐bodies (platelet antibody ,erythrocyte antibody ,granulocyte antibody) were significantly higher than the control group (P<0 .05) . Conclusion Aplastic anemia cellular immune function in patients is abnormal ,and antibody positive rate is high ,and it prompt that humoral immunity is also abnormal .
