1.Sufentanil combined with umbilical cord blood stem cells protects against myocardial infarction

BACKGROUND:In recent years, delayed protection of drug pretreatment has become a hot spot in the field of pretreatment, and sufentanil is a non-selective opioid receptor agonist that plays a significant myocardial protective role.
OBJECTIVE:To investigate the protective effect of sufentanil combined with umbilical cord blood stem cel transplantation on injured myocardium after myocardial infarction.
METHODS:Myocardial infarction model by ligating the left anterior descending coronary artery (ischemia 30 minutes and reperfusion 180 minutes) was made in 90 healthy adult Wistar rats, and then these rat models were randomly divided into ischemia-reperfusion (IR) group, cel transplantation group, combined treatment group (n=30/group). Rats in the IR group were given intraperitoneal y intravenous injection of 1 mL normal saline 5 minutes before IR;rats in the cel transplantation group were given intraperitoneal y intravenous injection of 1 mL umbilical cord blood stem cel suspension 5 minutes prior to IR;and rats in the combined treatment group were given intraperitoneal y intravenous injection of 10μg/kg sufentanil 10 minutes before IR and 1 mL umbilical cord blood stem cel suspension 5 minutes prior to IR. Two weeks later, related indicators were detected.
RESULTS AND CONCLUSION:Compared with the IR group, myocardial infarct size was reduced significantly in the combined treatment group, fol owed by the cel transplantation group (P<0.05). Lowest levels of serum creatine kinase, lactate dehydrogenase and troponin I were obtained in the combined treatment group fol owed by the cel transplantation group and IR group, while highest level of nitric oxide was observed in the combined treatment group fol owed by the cel transplantation group and IR group (P<0.05). Highest expression of Caspase-3 protein was recorded in the combined treatment group, fol owed by the cel transplantation group and IR group (P<0.05). Significantly increased left ventricular diastolic blood pressure and decreased left ventricular end diastolic pressure were found in the two cel transplantation groups compared with the IR group (P<0.05). Moreover, pathological injury of the myocardial tissues was mitigated in the cel transplantation group and significantly relived in the combined treatment group compared with the IR group. In conclusion, the combined use of sufentanil and umbilical cord blood mesenchymal stem cel transplantation can reduce the degree of myocardial infarction and protect against myocardial injury.
2.Effect of cimetidine on proliferation and apoptosis of human colon cancer cell LoVo

Objective:To investigate the regulatory effect of cimetidine on proliferation and apoptosis of colon cancer cells LoVo in vitro.Methods:Colon cancer cell line LoVo originated from human colon carcinoma was cultured in vitro.Cell proliferation was measured by MTT colorimetric assay.Cell cycle and apoptotic percentage were detected by flow cytometry (FCM),morphological changes of apoptotic cell were observed by light and electron microscopy,and apoptotic cells were quantitatively determined by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL).In addition,expression of intracellular bcl-2 protein was analysed by flow cytometric indirect immunofluorescent technique.Results:Cimetidine (1?10 -2 mol/L~3?10 -2 mol/L) inhibited LoVo cells growth significantly in a dose-dependent manner.After 48 hours of exposure to cimetidine(1?10 -2 mol/L),the enhanced degree of apoptotic percentage was 32.7%.There were many morphologic features of apoptosis,including cell shrinkage,nuclear condensation,DNA fragmentation and formation of apoptotic bodies;apoptotic cells were also statistically significant than those of control group( P
3.Evaluation of CAP test system for allergen-specific IgE assay

Objective To evaluate the validity and predictive value of CAP specific IgE test system.Methods Five allergen-specific IgEs,i.e.,specific IgE to mites,mugwort pollens,hop pollens,cat epithelium and dander,and Alternaria alternate,were tested in selected randomly allergic and non-allergic patients by using CAP system.Results In 957 tests the sensitivity of specific IgE to the 5 allergens was 99.0%,97.9%,91.7%,93.5% and 85.5%,and the specificity was 96.2%,95.8%,93.0%,97.0% and 91.7%.The positive and negative predictive values were greater than 94% and 80% respectively.Conclusion CAP system is of great potential value for diagnosis of allergens.
4.Biological activity of components, protein concentration of althernaria alternate lysates by different methods

Objective To find the best methods to make the extraction of alternaria alternate. Methods Alternaria alternate was harvested from air and identified, cultured at 26 ℃ for four weeks. Protein was extracted by different methods and its concentration was determined using the Bradford assay, its bioactivity was tested by RAST inhibition, and residual proteins were analyzed by SDS-PAGE.Results The extraction methods like grinder(a), grinder plus ultrasonic(b), ground with liquid nitrogen(c) and liquid nitrogen plus ultrasonic(d) were applied in the extraction procedure. The concentrations of protein extractions were as follows (g/mL): a was 0.44?0.04;b was 0.75?0.03;c was 0.72?0.03 and d; 1.29?0.05. From SDS-PAGE, we found that the most component in the extraction was that obtained by liquid nitrogen plus ultrasonic. From RAST, 50% inhibition was as follows(g/mL): a was 8.5, b was 9.4, c was 7.0,d was 3.7. Conclusion Ground with liquid nitrogen plus disintegration by ultrasonic was the most effective method of extraction.
5.Concentration of airborne pollen in Beijing city with burkard sampler

Objective:To investigate the airborne pollen concentration in Beijing city during autumn and to grade the Artemisia pollen concentration .Method:Burkard volumetric trap was used to sample the airborne pollen in Beijing city from August 1st 2007 to October 10th 2007. Patients only allergic to mugwort pollen were followed up, and their symptom scores and the mugwort pollen concentration were analyzed statistically.Result:①Mugwort and Humulus(including Cannabis Sativa L) were the main airborne pollen during August and September in Beijing city, accounting for 31% and 51%,respectively; ②Mugwort pollen season was from 8th August to 8th October;③The daily peak concentration of Mugwort pollen was 267 g/m~3,with an average of 71 g/m~3,the daily peak concentration of Humulus pollen was 672 g/m~3,with an average of 124 g/m3;④88.5% out-patients suffering from hay fever or asthma during Autumn were allergic to mugwort, 28.2% allergic to Humulus;⑤the grading of airborne Mugwort pollen concentration: 0~14 g/m~3 as grade 0, 15~40 g/m~3 as grade Ⅰ, 41~116 g/m~3 as grade Ⅱ,more than 117 g/m~3 as grade Ⅲ.Conclusion:It was the first time that Burkard volumetric sampler was employed in monitoring Mugwort and Humulus concentration in Beijing city. Average concentration of mugwort pollen was about 100 g/m~3, and concentration grade Ⅲ of mugwort pollen was distributed during the last ten days of August and the beginning of September.

7.Coping style and psychological adaptation of college students

BACKGROUND: At present, it is affirmed that coping style is the important mediator between stress and health both on mental and body. The coping theory is descripted as three orientations: trait view, contextual view and interaction person-environment view. The conception and classification of the coping styles still remains to be explored. OBJECTIVE: To probe into the dimensions of coping styles, the characteristics of different coping styles and the correlation between coping styles and psychological adaptation among college students.DESTGN: Questionnaire investigation.SETTING: School of Psychology, Beijing Normal University.PARTICIPANTS: Altogether 343 college students were randomly chosen from Tianjin University, Tianjin University of Commerce, Tianjin Normal University and Tianjin University of Finance and Economics. The college students, including 161 males and 182 females, were aged (19±2) years, ranging from 19 to 22 years. According to body health examniation records of them, all the college students had no serious chronic or acute illness. Written informed consents were obtained from all the college students.METHODS: The college students were measured collectively in their own classroom respectively from September to December in 1999. There were totally 16 class units. Altogether 422 copies of questionnaire files were sent out. Each copy includes Coping Styles Scales, Adolescent Psychological Adaptation Scales, and SCL-90.The questionnaires were collected once the college students completed. Copying styles were assessed with Coping Style Scale revised by Liang Baoyong, which includes 20 items. Each item gives one kind of coping strategies in daily life. For example, "To tell myself,my trouble is nothing matter When comparing with others, it is no worthy to be annoyant". The college students should be asked to answer on the 5-point Likert scale. These 20 items covered majority of coping styles which were used by adolescents. Two indexes were used as the results of psychological adaptation: one is Adolescent Psychological Adaptation Scale developed by Chen Huichang, including 20 items. Scores were collected on the 5-point Likert scale in which high scores mean better adaptation. Another result variable is the amount of SCL-90. High scores mean more symptoms with mental or physical state which stands for lower adaptation level. All the assessment data were input into Foxbase data-base, then were analyzed using factor analysis and cluster analysis in order to attain different kinds of college students coping styles. Correlation analysis was used to examine the correlation between coping style and adaptation. MAIN OUTCOME MEASURES: Coping styles of college students and correlation between different coping styles and adaptation results.RESULTS: A total of 422 copies of questionnaires were sent out, and 362 copies were drawn back, 19 copies were invalid questionnaires due to either there were missing answers or the answer had reflective tendency. At last, 343 sets of data entered the analysis procedure. ① According to factor analysis results, the main coping styles used by college students were F1 (seeking support), F2(positive thinking), F3(cognitive adjustment), F4(passive blame oneself), F5(facing reality), F6(avoidance), F7(emotional catharsis).There were three main coping styles from cluster analysis, that was, A (positive confrontation), B (passive avoidance) and C(passive catharsis).② Those who were more adaptive used A coping style more frequently, and those who had lower adaptation level used B or C coping style more frequently. But few students used C coping style. A coping style was significantly negatively correlated with mental or physical symptoms (r= -0.258,P< 0.01 ),but significantly positively correlated with adaptation level (r =0.467, P < 0.01);But both B and C coping styles were significantly positively correlated with mental or physical symptoms (r=0.338, 0.364, P < 0.01 ), B coping style was significantly negatively correlated with psychological adaptation(r =-0.140, P < 0.05). It seemed that C coping style had no correlation with long-term adaptation. A and C coping styles were two dependent styles respectively. In addition, different levels of stress were significantly correlated with three styles. Few A coping styles were used under high level of stress, but more B and C coping styles did.CONCLUSION: Coping style is complex. Use of different copying styles is significantly correlated with psychological adaptation levels. But the direction of correlation is effected by the factors, such as individuals, index of results and degree of stress.
8.Expression and significance of PTTG and Ki67 in fibroadenomatosis and fibroadenomas

Objective To explore the relationship between pituitary tumor transforming gene(PTIG) and Ki67 expression and their significance in multiple adolescent breast fibroadenomas (fibroadenomatosis)and single persons(fibroadenomas) and their clinical value.Methods A total of 53 fibroadenomatosis and 160 fibroadenomas specimens in our hospital were collected.All the samples were fixed with 10% formalin and then embedded with paraffin.The protein expression of PTTG and Ki67 was detected by immunohistochemical staining.The relation between expression of PTTG and Ki67 in fibroadenomatosis was analyzed.Results The positive rate of PTTG and Ki67 was higher in fibroadenomatosis than in fibroadenomas (P < 0.005).Conclusions High expression of PTTG and Ki67 in fibroadenomatosis leads to the proliferation of tumor cell and upregulates tumor angiogenesis.High expression of PTTG and Ki67 may play important roles in tumorigenesis and pathological process of fibroadenomatosis.

10.Processing of realgar and re-evaluation on medicinal efficacy and toxicity

Objective: To prepare the processing solution of realgar by bacteria and to explore the change of pharmacodynamic action and toxicity. Methods: The processing solutions of realgar were prepared using the strain thiobacillus ferroxidans, the realgar powder processed by normal method was selected as control. The effects on RPMI8226 cell proliferation of them were investigated by methyl thiazolyl terazolium (MTT),cell cycle distribution was determined by FCM in RPMI8226 cell. The distribution of arsenic concentration in blood and tissues including heart, liver, spleen, lung, and kidney in the samples were determined by Inductively Coupled Plasma-atomic Emission Spectrometry (ICP-AES). Results: Compared with control group, the solubility rate and the induced apoptotic rate increased, and dosage significantly reduced, while arsenic was hardly found in the tissue in processing group by bacteria. Conclusions: The processing group by bacteria may be enhanced medicinal efficacy and inhibited the toxicity in realgar. It’s intending to provide new approaches for scientific processing realgar.
