2.Influence of diabetes on the protective effect of ischemic preconditioning on ischemic reperfused rat heart

Objective To explore the influence of diabetes on the protective effect of ischemic preconditioning(IPC) on ischemic-reperfused(IR) myocardium of rat.Methods Sixty male SD rats(30 with diabetes and 30 without diabetes) were divided into six groups(10 each): non-diabetic rats control group(group A),non-diabetic rats IR group(group B),non-diabetic rats IPC group(group C),diabetic rats control group(group D),diabetic rats IR group(group E) and diabetic rats IPC group(group F).The isolated heart perfusion Langendorff models were established after rats were sacrificed.Isolated hearts of the two control groups were undergone a 90min perfusion without any other intervention;those of the two IR groups were undergone a 30min equilibration period,a 30min ischemia and a 30min reperfusion;those of the two IPC groups were undergone a 10min equilibration,then elicited by two cycles of 5min ischemia interspersed with 5min reperfusion prior to 30min ischemia and a 30min reperfusion.The recovery rates of the left ventricular function,such as cardiac output(CO) and left ventricular developed pressure(LVDP),and the maximum upstroke and decay velocities of left ventricular pressure(?dp/dtmax) were recorded.The activity of creatine kinase(CK) in coronary outflow,contents of malonyldialdehyde(MDA) and superoxide dismutase(SOD) in myocardium were detected,and myocardial water ratio were assessed.Results In non-diabetic rats IPC group,the activity of CK,content of MDA and water ratio of myocardium declined significantly,while the content of SOD increased and the recovery rates of CO,LVDP and ?dp/dtmax rose significantly compared with that in non-diabetic rats IR group(P0.05).Conclusion Diabetes may inhibit the protective effect of ischemic preconditioning on ischemic reperfused rat heart.

Objective In this study,we determined whether or not cancer-reactive cytotoxic T-lymphocytes(CTLs)could be induced from peripheral blood mononuclear cells(PBMCs)of colon cancer patients by in vitro stimulation with a PAP peptide.Methods The PBMCs of HLA-A24+ colon cancer patients and healthy donors were stimulated with a PAP213-221 peptide in vitro.The PAP-specific IFN-? was measured by ELISA.The cytotoxicity of CTLs were examined by 51Cr release assay.Results IFN-? produced by the PBMCs of 3 of 5 colon cancer patients and 1 of 8 healthy donors after incubation with PAP213-221 peptide pulsed C1R-A2402 cells were significantly higher then that after incubation with HIV peptide pulsed C1R-A2402 cells(P
4.System Design of a Medical Constnt Temperature Flushing Pump.

The flushing pump which is applied to clean operative wound has no temperature controlling function up to now, and doctors have to prepare the flushing fluid that has previously been warmed. The flushing pump system with medical constant temperature designed in our laboratory can absorb flushing fluid at the room temperature, and then eject flushing fluid with the temperature in accordance with the requirements of operations at a controlled constant flow rate. The system combines flow rate control with temperature control functions. The flushing pump system includes flushing part, temperature controlling part, key inputting part, liquid crystal displaying part and exceptional situation monitoring part. The present paper introduces the design method and principle of each part of the system at first, and then gives the debug method of all the system parameters. Finally the paper discusses the performance of the system according to the result of the experiment.

5.Research on data acquisition of portable uroflowmeter and accuracy of measurement.

This paper presents the preliminary design of data acquisition system of a portable uroflowmeter. The system uses double-hole cantilever pressure sensor. The signal is transferred to ATmega644PA microprogrammed control unit (MCU), converted by A/D (analog to digital) convertor. Then the further data are processed and get the corresponding relationship of weight-time and two curves of urine flow and urinary flow rate. In the measurement accuracy of the device about urine flow, two factors about the placement and height of the data acquisition are analyzed to show the accuracy of the equipment through the Origin 8.0 data analysis software. The design is characterized by low cost and high speed of data collection, real-time, high accuracy.

6.Correlations of ambulatory pulse pressure to large artery elasticity and vascular endothelial function in patients with primary hypertension

BACKGROUND: Ambulatory blood pressure monitoring can sensitively and objectively reflect blood pressure level, which is closely related to target organ damage and disease prognosis. In hypertension, vascular endothelial damage is the most common lesion to target organs. There is little known about how ambulatory pulse pressure correlates to large artery elasticity and vascular endothelial function. OBJECTIVE: To investigate changes of large artery elasticity and of vascular endothelial function in patients with primary hypertension using an automatic pulse wave velocity determinator and ultrasound techniques, and to analyze the correlation of ambulatory pulse pressure to large artery elasticity and vascular endothelial function.DESIGN: A non-randomized concurrent control clinical observation. SETTING: Diagnosis and Treatment Center for Coronary Heart Disease, the 305 Hospital of Chinese PLA. PARTICIPANTS: A total of 156 inpatients and/or outpatients, who were recently confirmed with primary hypertension, were recruited for this study between June 2005 and April 2007. Patients consisted of 114 males and 42 females. All patients averaged 56 ± 4 years of age (range: 40-75). Inclusive criteria: Corresponding to diagnostic standards for preventing and treating hypertension instituted in 2004 by Chinese scholars. Confirmed as primary hypertension within 1 month. Not receiving any blood pressure lowering, hypolipidemic or nitrate-like drug treatments. Written informed consents for laboratory measurements were obtained from all subjects. The study was approved by the hospital's Ethics Committee. METHODS: According to the mean pulse pressure over 24 hours, all patients were assigned into 3 groups: Group A (mean pulse pressure < 40 mm Hg, n=92), group B (40 mm Hg ≤ mean pulse pressure < 60 mm Hg, n=39) and group C (mean pulse pressure > 60 mm Hg, n=25). In each group, daytime pulse pressure and night-time pulse pressure, as well as 24-hour mean pulse pressure were measured using a non-invasive portable ambulatory blood pressure monitor (ABPM-04, Meditech Inc, USA). Carotid-femoral and carotid-radial arterial pulse wave velocities were measured using an automatic pulse wave velocity determinator to evaluate large artery dilation. Blood flow mediated and nitroglycerin-dependent dilatation of the brachial artery was determined using a high-resolution ultrasound technique to evaluate vascular endothelial function. MAIN OUTCOME MEASURES: Correlations of ambulatory pulse pressure to large artery dilation and arterial endothelial function. RESULTS: All 156 patients were included in the final analysis. Correlation of ambulatory pulse pressure to large artery dilation: Carotid-femoral arterial pulse wave velocity was significantly positively correlated to daytime pulse pressure, night-time pulse pressure and 24-hour mean pulse pressure, with coefficient of partial correlation being 0.310, 0.281 and 0.303, respectively, P < 0.01). There were no significant correlations of carotid-radial arterial pulse wave velocity to daytime pulse pressure, night-time pulse pressure or 24-hour pulse pressure (P > 0.05). Correlation of ambulatory pulse pressure to arterial endothelial function: There was a linear relationship between ambulatory pulse pressure and blood flow-mediated blood vessel dilatation values. Linear correlation analysis was performed, taking ambulatory pulse pressure as an independent variable, and endothelial-dependent dilatation as a dependent variable. Results demonstrated that blood flow-mediated blood vessel dilatation was significantly negatively correlated to daytime pulse pressure, night-time pulse pressure and 24- hour mean pulse pressure (r = -0.684, -0.597, -0.668, P < 0.01). There was no correlation of ambulatory pulse pressure to non-endothelial-dependent blood vessel dilatation. CONCLUSION: Ambulatory pulse pressure increase is closely related to large artery elasticity decrease and injury to endothelial function in patients with primary hypertension.
7.Isolation and identification of Ralstonia mannitolilytica strains from a patient with septicaemia and analysis of their drug resistance genes

Objective To isolate and identify the pathogenic bacteria from peripheral blood of a patient with septicemia of unknown etiology and to analyze their drug resistance genes.Methods Two peripheral blood samples were collected from the patient after having fever.Several assays including culturing bacteria on blood agar plates by using streaking technique,Gram-staining of bacterial colonies and microscopic observation,VITEK 2-compact automatic bacterium detection and analysis system as well as a sequencing analysis of the 16s rRNA gene were performed to identify the bacterial pathogens in blood samples.Microdilution test was performed to detect the drug susceptibilities of isolated bacteria to antibiotics.Confirmatory tests were performed to detect the production of β-lactamase and extended spectrum β-lactamase by the isolated strains and the phenotypes of AmpC enzyme and carbapenemase.PCR was used to identify the β-lactamase-encoding genes in the isolated strains by using the primers of 19 common β-lactamase-,AmpC enzyme-and carbapenemase-encoding genes in Enterobacteriaceae strains and the primers of 21 annotated gene sequences encoding the β-lactamase of a Ralstonia mannitolilytica strain.The PCR products were sequenced and analyzed after T-A cloning.Results Ralstonia mannitolilytica strains were isolated from the two peripheral blood samples.The isolated strains were sensitive to ceftriaxone,cefepime,ciprofloxacin,ofloxacin,tigecycline and compound sulfamethoxazole (SMZ-TMP),but resistant to the other 11 tested antibiotics.Results of PCR amplification by using the primers of common genes encoding β-lactamase of Enterobacteriaceae strains were all negative.Fragments of genes encoding the β-lactamase of the isolated Ralstonia mannitolilytica strain were successfully amplified,which were TK49_09850,TK49_12955,TK49_14470,TK49_14495and TK49_18990.The sequences of the amplified gene fragments were not similar to those of the common β-lactamase-encoding genes in Enterobacteriaceae strains.Conclusion The patient was infected with Ralstonia mannitolilytica.The isolated Ralstonia mannitolilytica strain showed a high-level drug resistance with a noticeable diversity against different β-lactam antibiotics.The genes encoding β-lactamase of the isolated Ralstonia mannitolilytica strain were completely different to those of Enterobacteriaceae strains.
8.Pradefovir:a new drug that targets to the liver for treatment of hepatitis B

Objective To review pharmacological mechanism, pharmacokinetics, clinical research progress and prospects of pradefovir, a liver targeted medicine for hepatitis B.Methods The studies of pradefovir were summarized by searching literature databases of Web of Science,Elsevier ScienceDirect,Springer Link,Wiley Online Library, Pubmed, CNKI, Wanfang and VIP datebase.Results Pradefovir is a prodrug that targets to the liver, which absorbs rapidly by oral administration.Pradefovir could be quickly converted to adefovir with hepatic drug metabolizing enzyme CYP3A4. Compared with adefovir dipivoxil, it has shown smaller nephrotoxicity and larger liver targeting.Conclusion Pradefovir has shown favorable safety and effectiveness in the clinical study and has no durg resistance to be found.The approval Ⅲ clinical trial has been acquired of pradefovir in USA and has enteredⅠ clinical trial currently in our country, which has good prospects for clinical application in future.
9.Psychological stress affects the expressions of two kinds of cytokines in rat temporomandibular joint cartilage

BACKGROUND:Animal modeling has reported that psychological stress can lead temporomandibular joint disease in rats, and there wil be pathological changes.
OBJECTIVE:To observe the effects of psychological stress on the expressions of vascular endothelial growth factor and transforming growth factor-β1 in rat temporomandibular joint condylar chondrocytes
METHODS:Forty-eight adult male Wistar rats were randomly divided into the experimental group and the control group, 6 rats in the experimental group and 12 rats in the control group. Thirty-six Wister male adult rats in the experimental group were subjected to the fol owing stimulating factors, such as electroshock, keeping stil ness, climate change and food tempting in order to make them under psychological stress. Then, the experimental group rats were executed at 3 weeks after stimulation (3 weeks stimulation group), 6 weeks after stimulation (6 weeks stimulation group) and 6 weeks normal feeding after 6 week stimulation (recovery group), 12 rats were sacrificed at each time point. The 12 rats in the control group were executed at the same time as the last experimental group. The temporomandibular joint specimens were taken to produce slices.
RESULTS AND CONCLUSION:The vascular endothelial growth factor-positive rate of recovery group was significantly larger than that of the 3 weeks stimulation group and the control group (P<0.05);transforming growth factor-β1 positive rate in the 6 weeks stimulation group was significantly greater than that in the control group (P<0.05). It indicated that vascular endothelial growth factor was related with the new blood capil aries repairing tissue, and transforming growth factor-β1 might play a role that stimulates bone resorption in psychological stress.
10.Effect of different doses of monocrotaline in combination with isopropylarterenol on the hemodynamic response, heart index and right heart hypertrophy index in rats

Objective To study the effect of different doses of monocrotaline in combination with isopropylarterenol on the hemodynamic resonse, heart index and right heart hypertrophy index in rats.Methods Sixty-four healthy adult SD rats, male:female=1∶1, body weight 200-250 g, were randomly divided into blank control group (n=16) and three model groups (n=16 in each group).The high dose model group (n=16) received i.p.injection of monocrotaline 80 mg/kg once and epinephrine 10 mg/kg once daily for one week.The moderate dose group received i.p. injection of monocrotaline 55 mg/kg once and epinephrine 8 mg/kg once daily for one week.The low dose group received i. p.injection of monocrotaline 30 mg/kg once and epinephrine 3 mg/kg once daily for one week.The rats were fed for 6 weeks, and then pulmonary artery pressure and right ventricular pressure were tested and heart index and right ventricular hypertrophy index were determined.Results Compared with the control group, the mean pulmonary artery pressure and right ventricular systolic blood pressure in the low dose monocrotaline group were not significantly changed, but significantly changed in the moderate dose monocrotaline group ( P<0.05) .The heart index and right ventricular hypertrophy index in the low dose monocrotaline group were not significantly changed, but in the moderate dose monocrotaline group, the heart index was significantly reduced ( P<0.01 ) and the right ventricular hypertrophy index was significantly increased ( P<0.05 ) .Conclusions The use of a single injection of 55 mg/kg monocrotaline in combination with continuous injection of 8 mg/kg isopropylarterenol once daily for one week can ensure the survival rate of rats, and the successful formation of pulmonary artery hypertension, leading to heart weakness.
