1.Intergrated infraofacial plasty with intraoral incision under local anaesthesia

Objective To explore an easy,safe and effective method for correction of the hypertrophy of mandibulae angle and inferior facial enlargement.Methods Based on the complexion of mandibulae angle hypertrophy,32 patients were treated through an intraoral incision with complete separation of masseter,polish of the diagonal site,camber gonion osteotomy and mandibular marginal plasty under local anaesthesia.Meanwhile,19 patients with masseter hypertrophy were injected with botulinal toxin A three months after the operation,and 2 patients with facio-buccal fullness were treated with removal of the buddal fat pad.Results 12 patients were satisfied with the results after following-up for one to six months.Conclusion This method is easy-doing,safe,reliable and accords with the principle of aesthetics.Osteotomy and grinding of the bone are convenient,which have little damage to patients with less complications and quick recovery.
2.Study on preparation of mucopolysaccharide from Dasyatis akajei cartilage

The technique of extracting and refining mucopolysaccharide from Dasyatis akajei cartilage obtained was studied and the content of mucopolysaccharide was measured.The results showed that when the content of NaOH was 6%,temperature 40℃, extracting time 6h,followed neutral protein enzyme dissolution for 7h under 40℃,protein precipitation with trichloroacetic acid and 4 times volume 95% C_(2)H_(5)OH for polysaccharide precipitation.the extraction rate was over 21%, the product was white powder and its purity was about (84.53%).Physical and chemical properties of the mucopolysaccharide were identified.

Abstract Objective:To observe and compare the effects of immune responses of HBV DNA caccine by insertion IL-2 gene.Try to ex-Plore the efficient way to enhance the immune effect of DNA vaccines. Methods:The recombinant expression plasmid vector peDNA3.1-S/IL-2with fusion gene of HBsAg and IL-2 cloned was constructed by PCR method and DNA recombinant technique.Then, it was transfered into Cos-7 cells by cation lyposomes and was detected its transient expressing product in vitro.At last,observed and compared the cellular and humoralimmune responses to HBsAg in C57BL/6 mice by intramuscular injection.Results:The pcDNA3. 1-S/IL-2 was constructed successfully by themethod of restriction endonucleases digestion, RCR and DNA sequencing. It can increase the titers of anti-HBs antibody. The bioactivity of IL-2and lymphocyte proliferation responses to HBsAg induced by the mouse spleen immunized with psDNA3.1-S/IL-2 were improved also. Conclu-sion:These results showed the expression of HBsAg-IL-2 fused gene can enhance the immune responses and suggested the insertion of IL-2 maybe a practicable way to enhance the DNA vaccine efficacy.

OBJECTIVEThe non-specific antitumor immunity effect of 4-1BBL-B7-H3 gene was investigated by establishing an oral squamous cell carcinoma human peripheral blood lymphocyte-severe combined immunodeficient (SCID) mice chimeric model.
METHODSForty mice were randomly divided into five groups. All groups, except the non-immune reconstitution group (group D), had reconstructed human partial immune system. The control group (group A) was injected with Tca8113 cells. The Ad4-1BBL-B7-H3 group (group B) was injected with Tca8113 cells transfected by adenovirus containing 4-1BBL-B7-H3 gene. The empty vector group (group C) was injected with Tca8113 cells transfected by adenovirus containing an empty vector. The non-immune reconstitution group (group D) was injected with Tca8113 cells. The non-tumor group (group E) was injected with PBS. The tumor volumes in each group were measured weekly. Human IgG in blood was obtained through the tail vein and was determined by enzyme-linked immunosorbent assay. Human CD3+ and D56 lymphocytes were assessed by flow cytometry. Model animals were killed on the ninth week. Differences in the expression of the natural killer group 2 member D (NKG2D) and toll-like receptor 2 (TLR2) in tumor tissues of each group were observed by immunohistochemical method. 4-1BBL-B7-H3 gene expression in mice tumor tissues was detected by reverse transcription polymerase chain reaction (PCR) and the expressions of major histocompatibility complex 1 class related molecule (M1C) A, M1CB, and TLR2 were detected by real-time quantitative PCR.
RESULTSThe tumor volumes of group B were remarkably lower than those in the other groups (P < 0.05). Human IgG and CD3+ and CD56+ lymphocytes were detected in the peripheral blood of immune-reconstituted mice. These lymphocytes were remarkably higher in group B than those in groups A, C, and E (P < 0.05). Higher NKG2D and TLR2 expression were observed in group B tumor than those in the other groups. The stable expression of 4-1BBL-B7-H3 gene in group B was proven. The expression of M1CA, M1CB, and TLR2 were significantly higher in the group B tumor than those in groups A, C, and D (P < 0.05).
CONCLUSIONThe high 4-1BBL-B7-H3 gene expression in tumor tissues could successfully induce the proliferation of CD3+ and CD56+ lymphocytes. This expression can also directly or indirectly activate TLR2 and up-regulate the expression of NKG2D and its ligands (M1CA and M1CB), which result in an effective antitumor immune response.

6.Effects of therapy with small volume of different fluids on renal blood flow in endotoxemia rats

Objective To evaluate the effects of therapy with small volume of different fluids on renal blood flow in endotoxemia rats.Methods Thirty parthogen-free SD rats weighing 180-250 g were randomly divided into 5 groups (n = 6 each):group Ⅰ control; group Ⅱ LPS; group Ⅲ LPS + 7.5 % hypertonic saline (HS);group Ⅳ LPS + hydrozyethly starch (HES) 130/0.4 and groupⅤ LPS + hypertonic saline plus hydroxyethly starch (HS-HES) 40. The animals were anesthetized with intraperitoneal 3% pentobarbital 40 mg/kg. Left carotid artery was cannulated for BP and HR monitoring and fluid administration. In groupⅡ-Ⅴ LPS 1 mg/kg was administered via arterial cannula. In group Ⅲ, Ⅳ and V 4 ml/kg of 7.5% HS, HES 130/0.4 AND HS-HES 40 were administered via arterial cannula respectively at 30 min after LPS administration.In groupⅠ and Ⅱ normal saline 4 ml/kg was given insteadt. Renal blood flow was measured with Doppler ultrasound before LPS (T1 ,baseline), at 30 min after LPS (T2), 10, 30 and 60 min after fluid therapy (T3, T4, T5). The animals were then sacrificed and both kidneys were removed for microscopic examination with light microscope. Results Renal blood flow was significantly decreased and was significantly recovered to some extent by therapy with different fluids especially with HS-HES 40 in group Ⅴ. Conclusion Therapy with small volume of HS,HES or HS-HES could increase renal blood flow and inprove renal microcirculation especially HS-HES.

Cancer malnutrition observed in 31%~87% patients with cancer can reduce the tolerance and compliance to radiotherapy ,interrupt or delay the treatment ,and significantly affect the prognosis .The malnu-trition may be aggravated when the duration of radiotherapy is prolonged and chemotherapy is applied concurrent -ly.PG-SGA is the most suitable method for nutritional risk screening in patients with malignant tumor .According to the result of nutrition assessment ,giving the adaptive nutrition support can improve the nutritional status of pa-tients and the curative effect in the early stage of radiotherapy ,as wells as post radiotherapy .

The triple negative breast cancer is one of importance in clinical subtypes of breast cancer, which is easy to recur and metastasis, and its prognosis is very poor.Radiotherapy, as an effective method for breast cancer,can reduce the risk of local recurrence.This article elaborates its characteristics,the progress of ra-diotherapy in the breast conserving surgery and modified radical mastectomy in triple negative breast cancer,when is the appropriate time for radiotherapy and radiotherapy sensitization,and hope that it will be helpful to the treat-ments.
9.Optimization of Microwave-assisted Extraction of Polyphenols from Enteromorpha prolifra by Orthogonal Test

Objective To optimize microwave-assisted extraction of polyphenols from Enteromorpha prolifra.Methods Based on single-factor tests,an efficient microwave-assisted extraction(MAE)technique was developed to extract bioactive polyphenols from E.prolifra through orthogonal L16(4)5 test.Results The highest yield(0.923±0.013)mg/g was obtained when microwave power,solvent to raw material ratio,irradiation time,ethanol concentration,and extraction cycles were 500 W,25 mL/g,25 min,40%,and 3,respectively,which was higher than that of Soxhlet extraction with methanol for 6 h,ultrasound-assisted extraction with 40% ethanol for 1 h twice and heat reflux extraction with 40%ethanol for 2 h twice.Conclusion This finding indicates that MAE is a superior technique for the extraction of polyphenols due to less impurity,higher time efficiency and yield.
10.Association study of clusterin polymorphism rs11136000 with late-onset Alzheimer's disease in Bai population

Objective To explore the association between clusterin(CLU) gene rs11136000 poly-morphism and late-onset Alzheimer's disease ( LOAD) in Bai population from Dali Bai Autonomous Prefec-ture of Yunnan Province.Methods A case-control study including 109 LOAD patients and 120 normal con-trols matched for age,sex and level of education was taken in Dali Bai population.Polymerase chain reaction (PCR) and single nucleotide polymorphism (SNP) site testing were used to detect genotype and allele fre-quency of CLU SNP rs11136000.SPSS 17.0 was applied to analyze the data.Result ①The different fre-quency ( C:65.60%,T:34.40%,CC:38.53%,CT:54.13%,TT:7.34%) of CLU SNP rs11136000 genotypes and alleles distribution in Bai between LOAD patients and healthy controls showed no statistical significance (χ2=1.529, P=0.216;χ2=2.805, P=0.246) .②The serum total cholesterol ( TC) of LOAD patients was significantly higher than that in that of control group( t=2.508, P=0.013) .Conclusion The results suggest that CLU rs11136000 polymorphism may not be the susceptible gene of LOAD,and high serum total choles-terol is more common in LOAD patients.
