1.Intergrated infraofacial plasty with intraoral incision under local anaesthesia

Objective To explore an easy,safe and effective method for correction of the hypertrophy of mandibulae angle and inferior facial enlargement.Methods Based on the complexion of mandibulae angle hypertrophy,32 patients were treated through an intraoral incision with complete separation of masseter,polish of the diagonal site,camber gonion osteotomy and mandibular marginal plasty under local anaesthesia.Meanwhile,19 patients with masseter hypertrophy were injected with botulinal toxin A three months after the operation,and 2 patients with facio-buccal fullness were treated with removal of the buddal fat pad.Results 12 patients were satisfied with the results after following-up for one to six months.Conclusion This method is easy-doing,safe,reliable and accords with the principle of aesthetics.Osteotomy and grinding of the bone are convenient,which have little damage to patients with less complications and quick recovery.
2.Experimental research of dose distribution and protection for mobile intra-operative radiotherapy accelerator in operating room

Objective To study the dose distribution characteristics formobile intra-operative radiotherapy accelerator (Mobetron) in an operating room,and to provide basic data for developing appropriate radiation protection measures and protection standard.Methods For most commonly used electron energy 9 MeV,TLD dosimeters were placed at 50,100,150 and 200 cm high plane,respectively.For each plane,the measurement points were selected at every 50 cm from the central axis at every 45° at eight different directions.Also different electron energies,such as 4,6,9 and 12 MeV,were taken into consideration at the plane at 100 cm height.After 10 Gy with a dose rate of 10 Gy/min were delivered,the TLD dosimeters were used to read out the data.Results For 9 MeV,at the phantom plane(100 cm high plane),the average doses were 169,756,395 and 241 μSv at 50,100,150 and 200 cm from the central axis,respectively.Themaximum deviation between the doses at 50 cm from the central axis in different angles and their average values were 9.1%.In the identical angle,the average doses of 50,100,150 and 200 cm high planes at the distance of 100 cm from central axis were 527,756,570 and 141 μSv,respecitvely.For the energies of 4,6,9 and 12 MeV,the average doses were 573,486,689 and 781 μSv at 100 cm from the central axis at 90° of 100 cm high plane.Conclusions For the same energy,the dose values at different directions were decreased by the minus exponential function law with the distance.The doses were uniformly distributed at different directions at the same distance from the central axis.The doses on the plane of 100 cm height were much higher than those at other heights,and the dose values were increased with the election energy.
3.Construction of MHC class Ⅱ transactivator recombinant adenovirus vector

Objective To construct a recombinant adenovirus vector containing the gene of major histocompatibility complex(MHC)class Ⅱ transactivator(C Ⅱ TA).Methods The restriction fragment of CIITA was inserted into pUC57 vector with EeoR Ⅰ and Xho Ⅰ.Then,recombinant plasmid pShutde-GFP-CMV CⅡTA was constructed with EcoR Ⅰ and Sal Ⅰ,and was confirmed by restriction enzyme digestion and sequeneing.After the treatment with Ⅰ-Ceu Ⅰ and Ⅰ-See Ⅰ,the fragment C Ⅱ TA from recombinant plasmid DShuttle-GFP-CMV.CⅡTA Was inserted into vector pAdxsi.And the pAdxsi-GFP-C Ⅱ TA wag packed into liposome,and was transfected to 293 cens.Results Recombinant plasmid pShuttle-GFP-CMV-C Ⅱ TA Was constructed successfully. After packed into vector pAdxsi, and transfected to 293 cells, significant virus Dlaques were observed,which showed the successful homologous recombination.The titer of the purified AdC Ⅱ TA was 2.0×10~(11) PFU/ml.Conclusions Recombinant adenovirus AdC Ⅱ TA containing gene of MHC class Ⅱ transactivator was established successfully.

Objective To evaluate the toxicity and efficacy of thermotherapy used with a streptococcus A preparation (OK-432) and cisplatin for treating malignant pleural effusion (MPE),and to assess patients' tolerance.Methods A total of 158 MPE patients were randomized into experimental and control groups.Closed drainage of the thoracic cavity was conducted with all the patients using a central venous catheter,and systematic chemotherapy was administered individually on the basis of each patient's condition.Patients in the control group were treated with intrapleural administration of OK-432 (0.5 mg) and cisplatin (40-60 mg) weekly for a maximum of 4 weeks,while those in the experimental group were given 60min of high frequency electrical thermotherapy 30-60 min after the administration of the drugs.The thermotherapy was at 40-43.5 ℃ and administered twice a week.Efficacy,quality of life and toxicity were compared between the two groups.Results The overall response rate in the experimental and control groups was 90.6% and 79.5% respectively;and grade I-II toxicity was similar.No grade Ⅲ or more serious toxicity was observed.The life quality scores in both groups were significantly improved after treatment.Conclusions Thermotherapy combined with intrapleural OK-432 and cisplatin can improve the effectiveness of therapy and life quality in MPE patients without increased toxicity.
7.Regulation of MHC class Ⅱ transactivator on MHC class Ⅱ expression of pancreatic cancer cell line CaPanC-2 and PanC-02 in vitro

Objective To investigate the effect of MHC class Ⅱ transactivator( MHC Ⅱ TA ) on MHC Ⅱ expression of human and rat pancreatic cancer cell line CaPanC-2 and PanC-02. Methods The recombinant adenovirus (Ad-C Ⅱ TA) was transfected into the CaPanC-2 cell and the PanC-02 cell, then it was cultured for 24, 48, 72 h. The C Ⅱ TA mRNA expressions were assayed by Real Time PCR and CⅡ TA protein expressions were determined by Western blotting. The percentage of positive expression cells of MHC Ⅱ were measured by flow cytometry. Results The C Ⅱ TA mRNA expressions of CaPanC-2 cell at 24, 48, 72 h were 16769 ± 6455, 261568 ±348850 and 834816 ±97783 folds higher than that of control group (P <0.05). The CⅡTA mRNA expressions of PanC-02 was 548546 ± 87755, 1242684 ± 624888 and 1401647 ± 726145 folds higher than that of control group ( P <0.05). CⅡ TA protein was not expressed in CaPanC-2 cell and PanC-02 cell. After transfection for 48 h, the CⅡ TA protein expressions of CaPanC-2 and PanC-02 were 0.746 ± 0.499 and 0.631 ±0.244. The percentage of positive expression cells of MHC Ⅱ in CaPanC-2 cells after 24, 48, 72 h were (8.1±0.3)%, (18.9±0.3)%, (78.5±0.90%, which were significantly higher than that in control group [ ( 7.0 ± 0.1)% , P < 0.01 . The percentage of positive expression cells of MHC Ⅱ in PanC - 0 2 were (5.1 ±0.2)% , (37.3 ±2.0)% , (68.8 ±2.2)%, which were significantly higher than that in control group [ (2.2 ±0.2)% , P <0.01). Conclusions Transfection of Ad-C Ⅱ TA could increase the expression of CⅡTA and MHC Ⅱ molecules of pancreatic tumor cells in vitro.
8.Influence and significance of DHA on expression of angiogene-sis-related genes in SGC7901 cells

Objective: To investigate the influence and significance of DHA on expression of angiogenesis-related genes in SGC7901 cells. Methods:SGC7901 were treated with DHA (5, 10, 20, 40, and 80μmol/l) for different times (24, 48, and 72 h), and the growth inhibition was detected by MTT. The expression of vascular endothelial growth factor (VEGF-C), cyclooxygenase-2 (COX-2) vascular cell adhesion molecule-1 (VCAM-1), and PTEN mRNA were detected by fluorescence-based quantitative poly-merase chain reaction (qRT-PCR). Their corresponding protein levels were tested by Western blot. Results:DHA significantly inhibited the growth of SGC7901 cells in a dose-and time-dependent manner (P<0.05). The expression of the angiogenesis-related genes signifi-cantly changed, as shown by RT-PCR and Western blot analyses. Compared with the control group, the expressions of VEGF-C, COX-2, and VCAM-1 were down-regulated, whereas the expressions of PTEN were up-regulated, after DHA treatment (P<0.05). Con-clusion:DHA inhibits cell growth in gastric cancer SGC7901 cells. The effect may be due to its reduction of VEGF-C, COX-2, and VCAM-1 gene expression, as well as its promotion of PTEN expression in gastric cancer cells.
9.Diagnostic:value of 16-SCT manifestation in Acute appendicitis

Objective To evaluate 16-SCT in diagnosing aeute appendicitis.Methods The CT findings of 34 cases were retrospectively analyzed.These cases were all proved acute appendicitis by surgical-pathology.Results 30 cases were confirmed by CT,with accuracy 88% (30/34).The diameter of 28 appendix was more than 6mm;2 collapsed with thickening wall.Periappendiceal fat stranding could be seen in 25 cases.Perforated appendicitis happened in 4 cases.Abscess performed in 2 cases.3 cases were missed,1 misdiagnosed.Conclusion 16-SCT could confirm acute appendicitis and its complication timely and accurately.
10.Correlation study of tissue inhibitor of matrix metalloproteinase-1 gene polymorphisms and chronic periodontitis

Objective: To investigate the relationship between TIMP-1 gene polymorphisms and susceptibility of chronic periodontitis(CP) in Han Cantonese. Methods: Buccal swabs from 70 CP patients and 74 periodontal healthy controls were collected. DNA was extracted from these buccal swabs by using Chelex-100. TIMP-1 +372T/C (rs4898)、TIMP-1+533C/T (rs1062849) polymorphisms were tested by polymerase chain reaction - restriction fragment length polymorphism (PCR-RFLP). Allele distribution and genotypes frequencies in the patients and controls were analyzed. Results: Frequency variance of allele T and C at site of TIMP-1 +372 in patients and controls showed no statistical difference. TIMP-1+533C/T polymorphism of TIMP-1 hadn't been found in the present study. Conclusion: There is no relationship between TIMP-1 +372T/C polymorphism and susceptibility of chronic periodontitis, and TIMP-1 +533C/T polymorphism doesn' t exist among Han Cantonese.
