1.Pathologic quantitative analysis of atherosclerrosis in rabits to verify introvensional ultrasonographic results

To verify the role of introvascular ultrasound(IVUS) in diagnosis of atherosclerosis. MethodsINUS wasused to measure the position, quality and the thickness of the plaque in the elastic artery and biopsy materials. After using HE andHart stains, a micrometer was used to measuse the thickness of plaques and arterious walls. And electron microscopy was used to ob serve the pathological changs of the plaque. ResultsBy histological observation compared with IVUS, the conformable rate of plaque anmounted to 94%. The plaque could be seen in 12 sites of control group. Among 33 sites, 32 plaques were found pathologic exmination while IVUS showed 30 plaques in the experimental group. The two methods had a gcod correlation on the thick measurement of plaque and the media( r = 0.835, P<0.05 and r = 0.814, P< 0.05). But they had a poor correlation on the measurement of the intirna( r = 0.055). ConclusionsBy pathological examination and the quantitative analysis, IVUS can obtain the common purpose in diagnosing atherosclerosis and measuring the media thickness. But it can not be used in measurement of the intima.
2.Joint action of CXCL16 and TNF-α genes polymorphisms in patients with atherosclerotic cerebral infarction

Objective To investigate the joint action of CXCL16 G1850A and TNF-α T1031C genes polymorphisms in patients with atherosclerotic cerebral infarction (ACI). Methods CXCL16 gene, G1850A and TNF-α gene T1031C mononucleotide polymorphism were tested with polymerase chain reaction followed by restriction fragment length polymorphism (PCR-RFLP) in 120 ACI patients and 75 healthy controls. Results The CXCL16 gene 1850 site AA genotype (35.8% vs 20.0%), A allele frequency (59.6% vs 44.0%), the TNF-αgene 1031 site CC genotype(2.5% vs 1.3%), C allele frequency(21.3% vs 11.3%)in ACI group were significantly higher than in the control group(P < 0.05). There was a positive correlation between the joint action of CXCL16 G1850A and TNF-α T1031C genes polymorphisms and ACI (χ2= 7.502,df = 2,P = 0.023). Conclusion The CXCL16 gene 1850, A allele and TNF-α gene 1031 C allele were risk factors for ACI. There is a positive correlation between the joint action of CXCL16 G1850A and TNF-α T1031C genes polymorphisms on ACI.
3.Effect of dexmedetomidine on proliferation and differentiation of rat embryonic neural stem cells in vitro

Objective To investigate the effect of dexmedetomidine (DEX) on proliferation and differentiation of rat embryonic neural stem cells in vitro. Methods Embryonic neuraI stem cells of fetal SD rats were separated from rats with gestational age of 14-16 days , and underwent primary culture for nestin expression. Cells were divided into control group, low-dose DEX group, and high-dose DEX group, which were cultured with 0,1 and 10 ng/mL DEX respectively. Cell viability was detected by MTT assay;the proliferation rate was estimated by BrdU incorporation; the phosphorylation of ERK1/2 in the proliferation phases was detected by western blot; the differentiation of neuronal cells,astrocytes,oligodendrocyte and neural stem cells were assessed by immunocytochemistry of cell specific markers. Results More than 95% of the embryonic neural stem cells in primary culture were Nestin-positive. Cell viability , BrdU-positive cells and phosphorylation of ERK1/2 in low-dose DEX group increased significantly (P<0.05), and decreased significantly in high-dose DEX group compared with control group and low-dose DEX group (P < 0.05). NeuN-, GFAP- and MBP-positive cells increased (P < 0.05), while Nestin-positive cell decreased (P<0.05) in both low and high dose DEX group compared with control group. However, no significant difference was found in low and high dose DEX group (P > 0.05). Conclusions The proliferation of neuraI stem cells can be promoted by low-dose DEX and depressed by high-dose DEX , as well as regulated by DEX of the phosphorylation of ERK1/2. DEX induces neural stem cells non-selective differentiation.
4.Effects of infrasound with low sound pressure level on the content of IGF-1 in brain of rats with focal cerebral ischemia-refusion injury

Objective To study the effects and mechanisms of infrasound with low sound pressure level on focal cerebral ischemia-refusion injury. Methods Focal cerebral ischemia was produced by 2 hours of occlusion of the middle cerebral artery in rats. lnfrasound generated by infrasound 8TM device was used as treatment factor. Thirty-two Sprague-Dawley rats were divided into three groups: sham group (n=8), model group (n=8) and in-frasound group (n=16) , and the infrasound group was subdivided into 20- and 120-min infrasound groups, with 8 rats in each group. Neurological symptoms was assessed at 2 h, 1 d, 3 d and 7 d, respectively. These rats were sacrificed after 7 days of infrasound treatment and their brains were harvested. The number of IGF-1 posive cells of ischemia cortex was counted by using immunohistochemical technique. Results Compared with model group, neurological symptoms of rats in 120-min infrasound group was significantly improved (P<0.05); the number of IGF-1 positive cells of ischemia cortex in 120-min infrasound group increased significantly as compared with that in model group (P<0.01). Conclusion Infrasound with low sound pressure level (120 min/d, 7 d) could exert neuroprotective effect in focal cerebral ischemia injury by increasing expression of IGF-1 in brain.
5.Protective effect of pulsed magnetic field against cerebral ischemia-reperfusion injury in rats

Objective To examine the neuroprotective effect of pulsed magnetic field in a animal focal cere-bral ischemica-reperfusion injury model. Methods Forty-eight Sprague-Dawley (SD) rats were randomized into 3 groups, a sham-operation group, a model group and a pulsed magnetic field group, with rats 16 in each group. Mid-dle cerebral artery occlusion (MCAO) method was employed to establish the focal cerebral ischemia-reperfusion inju-ry model in rats of model group and pulsed magnetic field group. Rats in sham-operation group was subject to the same operation procedure but not underwent ischemia-reperfusion. The infarction volume, histopathological damage and expressions of IGF-1 in ischemic brain tissue were investigated to evaluate the effect of pulsed magnetic field. Results The infarction volume was reduced, histopathological damage alleviated and expressions of IGF-1 in ische-mic brain tissue elevated in the pulsed magnetic field group as compared against the model group (P<0.05). Con-clusions Pulsed magnetic field might provide neuro-protection against cerebral ischemia-reperfusion injury.
6.Changes in Insulin-like growth factor-1 levels in the brains and livers of rats after cerebral ischemic injury

Objective To observe dynamically the expression of insulin-like growth factor(IGF-1)in the brain and liver,and to explore changes in IGF-1 levels.Methods The thread method waa used to establish middle cerebral artery occlusion in rats.The expression of IGF-1 on the ischemic side of the cerebral cortex and in the liver wag observed dynamically using immunohistochemical techniques. Results The number of IGF-1-positive cells increased significantly,peaking on the 3rd day in the cortex and the 7th day in the liver.Conclusion The expression of IGF-1 Wag upregulated in the brain after the experimental cerebral ischemia;peripheral humoral regulation of IGF-1 responded more slowly and recovered to some extent at the 7th day after cerebral ischemia.
8.Sonographic characteristics of chronic lymphocytic thyroiditis with or without nodules

Objective To study and distinguish the sonographic characteristics of chronic lymphocytic thyroiditis (CTL) and its nodules.Methods Ultrasonographic characteristics of 107 patients with histologically confirmed CTL was observed with a 7.5-12 MHz transducer and confirmed with the operation and pathology.Seventy four cases of CTL were examined by color Doppler flow image (CDFI),serum thyroid hormone levels were determined in 38 cases of CTL.Results In 57 cases of CTL,the pattern of diffuse low echoes was found in 14 cases (24.6 %),and nodulose pattern in 43 cases (75.4 %) in which diffuse numerous hypoecoie microdules in 16 (37.2%),and sporadic small nodules in 27 (62.8 %).All cases showed degeneration and disappearance of thyroid follicles,lymph cells invasion in varying degrees on pathology.Nodular goiter was commonly detected in 40 cases CTL (37.4 %),adenoma in 4 cases (3.7 %),hyperthyroidism in one case,non-Hodgkin lymphoma in one case,thyroid carcinoma in 4 cases (3.7 %).Color dopple flow imaging showed in 74 cases,0-Ⅰ 34 cases,Ⅱ 38 cases,Ⅲ 2 cases.The increased flow were observed in 38 cases in both Anti-Tpo and Anti-TG,and in 26 cases in TSH.Conclusion Ultrasonography is complex in CTL,with more complications.Ultrasonography are distinguished between the nodule of CTL and the nodule in the other thyroid diseases.Ultrasonography could help to avoid unnecessary surgical intervention,and provide strong evidence for CTL and its complication in clinic treatment.
9.Protective effects of resveratrol on sepsis and its involved mechanisms

Aim To investigate the effect of resveratrol (Res)on septic mice and LPS-insulted H9c2 cells,as well as its involved mechanisms.Methods By use of a mouse cecal ligation and puncture-induced septic model (CLP),the survival of septic mice was evalua-ted after resveratrol treatments.H9c2 cells were insul-ted by LPS and then treated with resveratrol,the mR-NA expressions of TNF-α,SIRT1 and other class III HDAC members were detected using RT-PCR and real-time PCR,Finally,the protein levels of nuclear p65, an important subunit of NF-κB,were measured in H9c2 cells using Western blot assay,to reveal the effect of resveratrol on LPS-induced nuclear transloca-tion of NF-κB.Results Compared with the control septic animals,intraperitoneal injection of resveratrol (1 or 5 mg·kg -1 ·d -1 )significantly increased the survival of septic mice.Furthermore,resveratrol signif-icantly increased mRNA expressions of SIRT1,SIRT2, SIRT6 and SIRT7 in LPS-insulted H9c2 cells.Res-veratrol also remarkably inhibited LPS-induced nuclear translocation of NF-κB.Conclusion An appropriate dose of resveratrol protects septic mouse hearts from the injury induced by LPS through the activation of SIRT family members and the inhibition of NF-κB pathway.
10.The bronchoscopic features of endobronchial tuberculosis induced by lymphatic fistula and the efficacy of interventional treatment

Objective To explore the bronchoscopic features of endobronchial tuberculosis induced by lymphatic fistula and the efficacy of interventional treatment. Methods The data on 31 patients with endobronchial tuberculosis induced by lymphatic fistula who had received bronchoscopic diagnosis and treatment in our hospital during the period of January 2010 to June 2013 were reviewed. The bronchoscopic features , along with the frequency of interventional therapies and duration of the therapies , were retrospectively analyzed; and the efficacy of the therapies and the related complications were assessed. Rusults The endoscopic appearance showed: granuloma (19.4%), necrosis (51.6%), granuloma with necrosis (25.8%), and fistulous opening (3.2%). Dark gray matter or carbon deposition inside the lesions was the characteristics of endobronchial tuberculosis induced by lymphatic fistula. There were 76 orificium fistulae in 31 patients , mostly in the right side. The median frequency of treatment was five in patients with single orificium fistulae and the median treatment duration was 65 days; whereas the median frequency of treatment was nine in patients with multiple fistulae and the median treatment duration was 108 days. There were significant differences between the two groups (P < 0.05 for both comparisons). The effectiveness rate of treatment was 98.7%. The therapy-related complication was of a small amount of bleeding, with a rate of 2.1%. Conlusions Endobronchial tuberculosis induced by lymphatic fistula shows certain characteristics under bronchoscopic examination. Bronchoscopic clamping combined with infusions with antituberculosis agents is a safe, simple, effective therapeutic method.
