1.Protective effects of lipoic acid on cardiomyocytes undergoing hypoxia/reoxygenation injury

Objective To study the protective effect of lipoic acid (LA) on H9c2 cardiomyocytes hypoxia/reoxygenation injury model, and explore its relevant mechanism. Methods Eight strains of H9c2 cardiomyocytes, passaged after cultured to a full view, were divided into 3 groups:normoxia group, hypoxia/reoxygenation group and LA group. The cell survival rate, lactate dehydrogenase (LDH) activity, malondialdehyde (MDA) and heme oxygenase-1 (HO-1) levels were detected and compared. Results The cell survival rates of H9c2 cardiomyocytes in hypoxia/reoxygenation group and LA group were significantly lower than those in normoxia group:(52.86 ± 6.39)%, (69.25 ± 7.63)%vs. (92.31 ± 7.82)%, while the cell survival rate of H9c2 cardiomyocytes in LA group was significantly higher than that in hypoxia/reoxygenation group, and there were statistical differences (P<0.01). The LDH activity and MDA in hypoxia/reoxygenation group and LA group were significantly higher than those in normoxia group:(286.37 ± 27.49), (209.72 ± 25.63) U/L vs. (126.32 ± 18.94) U/L, and (1.72 ± 0.06), (1.13 ± 0.07)μmol/L vs. (0.68 ± 0.06) μmol/L, while those data in LA group were significantly lower than those in hypoxia/reoxygenation group, and there were statistical differences (P<0.01). The HO-1 in hypoxia/reoxygenation group and LA group were significantly higher than that in normoxia group:(213.71 ± 18.94)%, (367.26 ± 23.07)%vs. (87.92 ± 19.23)%, and HO-1 in LA group was significantly higher than that in hypoxia/reoxygenation group, and there were statistical differences (P<0.01). Conclusions The LA plays a protective role on myocardial cell with hypoxia/reoxygenation injurythough increasing the level of HO-1 against oxidative stress.

OBJECTIVE:To establish a HS-GC method for determining methanol and ethanol in human plasma METHOD_S:An external standard method was used The stationary phase was a glass column packed with GDX-102 The carrier gas was nitrogen The column temperature was 140℃ and both the injector temperature and the detector temperature were 200℃ RE_SULTS:A good linearity was obtained in the range of 0 2~7 5g/L of methanol and ethanol The within-day RSD was less than 4% and the between-day RSD less than 5 5% The recoveries were (100 9?3 8)% for methanol,(101 5?3 4)% for et_hanol,respectively The limits of detection for both methanol and ethanol were less than 0 01g/L CONCLUSION:This method is simple,rapid and precise
3.Reports of life experience in chronic obstructive pulmonary disease patients with exercise rehabilita-tion for over five years

Objective To explore life experience in chronic obstructive pulmonary disease (COPD) patients with the habit of exercise for more than 5 years,to enhance the confidence of participating in the rehabilitation in other patients, to help the medical staff pay more attention to guide the patients with COPD to take up the rehabilitation exercise. Methods The narrative research method in qualitative research was used to analyze the collected data. Results The study found that the coping approach of the contradiction between exercise and dyspnea, the ability to live together with the disease peacefully and the enthusiasm of persuing life were deserved to be learned. Conclusions Full of perseverance and persisted rehabilitation exercise may alleviate the symptoms, reduce the number of acute exacerbation of COPD and improve the quality of life for COPD patients. Medical staff should encourage COPD patients to participate in the rehabilitation in the right time with appropriate methods.
4.The preliminary application of five repetition sit-to-stand test for evaluating lower limb function in the patients with chronic obstructive pulmonary disease

Objective To determine the value of five repetition sit-to-stand test (FTSST) to evaluate lower limb function in the patients with chronic obstructive pulmonary disease (COPD).Methods Sixty-one patients with COPD were tested for FTSST,isokinetic testing muscle strength for lower limb,6-minute walking test.The relationship between FTSST time and muscle strength,6-minute walking test distance (6MWD) were analyzed.Results FTSST time was (7.86±2.21) s,extensor peak torque was (67.58± 24.83) Nm,extensor relative peak torque was 0.95(0.46) Nm/kg,6MWD was (444.05±112.53) m.FTSST time related to extensor peak torque,extensor relative peak torque,6MWD and types of complication,correlation coefficient values were-0.303,-0.393,-0.428,F =2.813,P < 0.05.Multivariate linear regression analysis showed that when the dependent variable was FTSST time,the variables of the equation were 6MWD and types of complication.Conclusions FTSST can be used for the rapid evaluation of lower extremity function in patients with COPD.The lower limb function gets worse,the FTSST time will be longer.Nursing staff should pay more attention to the lower limb function of patients with COPD,along with complications,using FTSST to evaluate lower extremity function.
5.Experiences of patients with chronic obstructive pulmonary disease to take up the home- based resistance training

Objective To explore experiences of patients with chronic obstructive pulmonary disease (COPD) to take up the home- based resistance training. Methods Twenty- two patients with COPD who completed the 12 weeks home- based resistance training were interviewed deeply, the phenomenological analysis method was used. Results Though reading, analysis, reflection, separation, theme refining, two themes were drawn: self- perceived benefits; pyhcological, social and healthy status barriers on taking up and persisting in the exercises. Conclusions The study finds that self- perceived benefits, psychological changes and confusion in COPD patients during home- based resistance training.As the medical staff, to seize the COPD patients for the purpose of health to do the health education and strengthen the relevant training, to help the patient perceive benefits on resistance training, to enhance the confidence of participating in the pulmonary rehabilitation, to promote the behavior change. Additionally, we should identify obstacles early and take up the intervention, to stimulate the enthusiasm of home- based resistance training in patients with COPD.
6.Expression and significance of SIRT1 and p53 in endometrial carcinoma with diabetes mellitus

Objective To investigate the expressions and significances of SIRT1 and p53 in endometrial carcinoma with diabetes mellitus.Methods Immunohistochemistry was carried out to examine the expressions of SIRT1 and p53 in 39 cases of endometrial carcinoma with diabetes mellitus and 40 cases of endometrial carcinoma without diabetes mellitus and 25 cases of normal endometrium,and the correlations between these indexs were analysed.Results The positive rate of SIRT1 and p53 in experimental group was 64.1%,59.0% respectively,and 85.0%,52.5% in control group.The positive rate of SIRT1 in experimental group was lower than that of control group (x2 =4.561,P =0.033).The positive rates of SIRT1 and p53 in control group were higher than that of normal endometrium (x2 =21.462,P< 0.001; x2 =6.771,P =0.009).Expression of SIRT1 was positively correlated with p53 (r =0.360,P =0.024).Conclusions Hyperglycemia deregulates SIRT1 in endometrial carcinoma.SIRT1 may promote tumorigenesis by silence of p53.
7.Study on the relationship of serum HCG levels and serum KI-67 expression with the depth of trophoblast invasion in tubal pregnancy

Objective To explore the relationship of depth of trophoblastic invasion with trophoblast cell activity and serum β-hCG according to the expression of proliferation antigen Ki-67 which viewed as an indicator of cell proliferation activity.Methods Fallopian tube specimens collected from 108 patients who underwent operation treatment for fallopian tubal pregnancy in our hospital were investigated by light microscopic examination.They were divided into three groups according to the depth of trophoblastic infiltration: Ⅰ group (stage): trophoblastic invasion of tubal mucosa,Ⅱ group(stage): trophoblastic invasion of the muscularis,Ⅲ group(stage): trophoblast invasion of serosa layer(muscularis penetration).The expression of Ki-67 was detected by SP method and blood β-hCG was detected within 2 hours of preoperative.The level of β-hCG,the expression of Ki-67 and the depth of trophoblast invasion were analyzed.Results Mean level of serumβ-hCG in Group Ⅰ,Ⅱ and Ⅲ were(1416.64 ± 859.94)U/L,(3380.33 ± 2392.36)U/L and(6999.33 ± 4949.90)U/L respectively.Positive expression rate of cell proliferation antigen Ki-67 in Group Ⅰ,Ⅱ and Ⅲ were 21.95%,53.66% and 6.40% respectively.There were significant difference on the expression of Ki-67 between group Ⅰ and group Ⅱ,group Ⅱ and Ⅲ group,group Ⅰ and group Ⅲ(x2 =3.94,4.07,4.35,respectively,P < 0.05).The serumβ-hCG level also displayed statistics difference in the three groups(F =9.914,P < 0.01).The positive expression of Ki-67 and serum β-hCG level were positively correlated with each other(r =0.678,P < 0.05)Conclusion The high level of the serum β-hCG indicates high expression of Ki-67 and deeper trophoblast invasion of tubal wall.
8.Clear cell sarcoma of gastrointestinal tract:clinicopathologic analyses and review of litera-tures

Purpose To study the clinicopathologic and genetic features of clear cell sarcoma of the gastrointestinal tract. Methods One case of clear cell sarcoma of the gastrointestinal tract was analyzed by histology, immunohistochemistry, fluorescence in situ hy-bridization ( FISH) , with review of relevant literatures. Results A female patient was admitted to the hospital with a history of 1-week abdominal pain. Computed tomography revealed a mass in right hemicolon. A grey-white ulcerated mass was observed in the colon, in-volving the entire thickness of the colon wall extending into the subserosa. Microscopically, medium-size oval or round cells were ar-ranged in sheets and scattered osteoclast-like multinucleated giant cells were also presented. Immunohistochemically, the tumor cells were strongly positive for S-100 protein, but negative for HMB-45, Melan-A, CD117, CD1a and PCK. FISH showed the rearrangement of EWSR1 in 74% of the neoplasm cells. Conclusions Clear cell sarcoma of the gastrointestinal tract is an extraordinarily rare gastro-intestinal neoplasm that shows peculiar histopathological, immunohistochemical, ultrastructural, and genetic features. Whether clear cell sarcoma-like gastrointestinal tumor represents a distinct entity awaits larger series, including cytogenetic or molecular biological in-vestigation.
9.Clinicopathological analysis of seven cases of pemphigoid nodularis

Objective To analyze clinical characteristics of pemphigoid nodularis. Methods Seven cases of pemphigoid nodularis were included in this retrospective study. The clinicopathological features of pemphigoid nodularis were retrospectively analyzed, including patients′gender, age at onset, clinical manifestations, treatment and follow?up. Results Of the 7 patients, 4 were female and 3 were male, with the median age at onset being 59 years. Clinical manifestations mainly included markedly pruritic prurigo?like lesions and nodular lesions with or without blisters. The most common misdiagnoses were nodular prurigo and eczema. Pathology showed epidermal hyperplasia and subepidermal clefts, collagen hyperplasia in the papillary dermis, and perivascular infiltration of lymphocytes and eosinophils in the superficial dermis in all the patients. Direct immunofluorescence assay showed linear deposition of IgG and C3 in the basement membrane zone, and indirect immunofluorescence was positive in 2 patients. Glucocorticoids combined with immunosuppressants were effective in all the patients. Conclusions Pemphigoid nodularis is easy to be misdiagnosed, and immunopathology is helpful for its diagnosis. Glucocorticoids combined with immunosuppressants appear to be effective for its treatment.
10.Application of handgrip strength test for evaluating mobility in patients with stable chronic obstructive pulmonary disease(COPD)

Objective To explore the reliability of handgrip strength test for evaluating mobility in patients with stable chronic obstructive pulmonary disease.Methods Sixty-one COPD patients in stable stage were measured for handgrip strength and 6-minute walking test(6MWT).The receiver operating characteristic curve(ROC) was calculated to determine the best cutoff points of handgrip strength.Results Handgrip strength was (33.72-±7.47) kgf,6MWD was (437.06±97.96) m,handgrip strength was moderately correlated with 6MWD (r=0.404,P=0.001).6MWD≥350 m was used to classify two groups,and there was significant difference between two groups(P<0.05).Area under the curv e was 0.722,and the best cutoff points was 32.8 kgf.Conclusion Handgrip strength test can be a useful tool to quickly identify mobility in patients with stable COPD.
