OBJECTIVE: To establish a CGC method for the determination of pyridine in Potassium Dehydroandrograpol-ide Succinate. METHODS: The determination was performed on HP-INNOWAX capillary with column temperature at 100℃. The FID detector was used and the carrier gas was nitrogen. RESULTS:The linear range of Pyridine was 2.46～39.3?g?mL-1 and the average recovery was 98.07%,RSD=4.00(n=9).Its detection limit was 0.2?g?mL-1.CONCLUSION: The method is accurate and rapid, and applicable for the determination of the residual level of Pyridine in Potassium dehydroandrograpolide succinate.

OBJECTIVE: To establish the GC method for content determination of psoralen and isopsoralen in Buguzhi tincture.METHODS: The determination was performed on HP-1(30 m?0.53 mm,1 ?m) column with nitrogen as carrier gas and FID as detector,the inlet temperature was 260 ℃.The injector temperature was kept at 280 ℃ with diphenhydramine as internal standard.RESULTS:The linear range of psoralen were 4.82～48.22 mg?L-1(r=0.999 9) with an average recovery 103.11% (RSD=2.20%,n=6).The linear range of isopsoralen were 5.09～50.9 mg?L-1(r=0.999 9) with an average recovery of 100.98% (RSD=3.70%,n=6).CONCLUSION: This method is simple,rapid,accurate and reliable for the quality control of Buguzhi tincture.

Objective To study the detection and significance of human papillomaviral DNA(HPV-DNA) in cervical disease. Methods The express of HPV-DNA were detected by hybrid capture 2 in the patients with chronic cervicitis ( n = 166), cervical intraepithelial neolasia ( GIN ) ( n = 89 ) and uterine cervix cancer ( UCC ) ( n = 23 ). Results The positive expression rate of HPV-DNA in GIN Ⅰ , Ⅱ , Ⅲ grade and UCC were 70.8% (17/24) ,79. 3% (23/29) ,88.9% (32/36) ,91. 3% (21/23). They were significantly higher than those in chronic cervicitis 30.7% (51/166) (P ＜ 0. 01 );As the development of cervical disease, HPV-DNA positive expression had increasing trend.Conclusion The detection of HPV-DNA had an important role for the early UCC screening,prevention and treatment.

Objective:To observe the curative effect of modified amniotic membrane transplantation for agricultural pterygium.Methods: A total of 276 cases (276 eyes) with early pterygium were randomized divided into traditional group (138 cases) and modified group (138 cases) as random table. The traditional group was treated with pterygium excision in combination with autologous conjunctival transplantation; the modified group was treated with modified amniotic membrane transplantation; to observe the curative effect of the two groups.Results: After all of patients were followed up for 1 year, 122 cases were cured and 16 cases were recurrence, and the recovery rate was 88.41% in traditional group; 131 cases were cured and only 7 cases were recurrence, and the recovery rate was 94.93% in modified group and was better than traditional group; the difference between the two groups was statistical significant (x2=5.02,P<0.05).Conclusion: The modified amniotic membrane transplantation method is simple in operation, mini invasive for eye tissue, and can retain conjunctival tissue in utmost; it also can improve success rate of surgery and be an effective mean for treatment of pterygium.

The purpose of this study was to observe whether HBV can infect HSC in vitro. LX-2 cells, the human activated HSC cell line, were incubated with human serum containing HBVDNA at final concen-trations from 0.01 to 10 copies/cell and harvested after 24,48 and 72 hours. Hepatitis B surface antigen (HBsAg) and hepatitis B core antigen(HBcAg) were detected by lmmunohistochemistry dyed by DAB. By Immunohistochemistry, no positive particles in LX-2 cells, a few of dark brown particles (HBsAg and HB-cAg) were found in cytoplasm and nucleoli of HepG2.2. 15 cells,a lot of clark brown particles were found in liver biopsies of patients with chronic hepatitis B. In vitro, there was no evidence that HBV can infect and express antigens in LX-2 cells.

Objective To explore the correlation of metabolic disorder of newly diagnosed type 2 metabolism(T2DM) and nonalcoholic fatty liver disease (NAFLD).Methods A total of 117 patients with newly diagnosed T2DM in Beijing Ditan Hospital Affiliated to Capital Medical University from June 2014 to June 2015 were enrolled.Patients were divided into two groups:T2DM with NAFLD of 57 cases and T2DM without NAFLD of 60 cases.Body mass index (BMI),waist circumference (WC),liver and kidney function,serum lipid,glycosylated hemoglobin A1c (HbA1c),fasting glucose,fasting insulin and C-peptide (FCP) were detected.The insulin resistance and β-cell function were assessed by homeostasis model assessment insulin resistance (HOMA-IR) and HOMA-β.Results The incidence of NAFLD in T2DM patients was 51％ (that was,60 cases of T2DM patients with NAFLD),compared with 2TDM group,the metabolic index of T2DM without NAFLD was significantly increased (blood triglyceride (TG):(2.58 + 1.8) mmol/1 vs.(1.22 + 0.4) mmol/l,fasting blood glucose:(11.5+6.1) mmol/l vs.(9.2+4.3) mmol/l,serum uric acid:(465.3+65) umol/l vs.(325.3+72) umoL/l;P =0.03,0.03,0.02);HOMA-IR was more serious ((4.9 ± 2.8) vs.(3.8 ± 2.7);P =0.03);insulin β-cell function was compensatory more obvious ((49.2 ± 27) vs.(29.5 ± 18);P =0.02);prevalence rate of obesity,hypertension and diabetic ketoacidosis (DKD),diabetic retinopathy (DR),diabetic peripheral neuropathy(DPN) were significantly increased(P＜0.05).Logistic regression analysis showed that,smoking(OR=1.405,95％CI:1.262-1.567),male(OR=1.037,95％CI:1.015-1.063),BMI(OR=1.113,95％CI:1.278-2.531),WC (OR =1.624,95％ CI:1.162-1.761) and TG (OR =1.823,95％ CI:1.2822.563) were risk factors of NAFLD (P ＜ 0.05).Conclusion T2DM patients with NAFLD have severer insulin resistance than those without NAFLD.The β-cell function of those patients is compensatory increased.NAFLD prevalence rate significantly increase in patients with obesity,especially in male diabetic patients who smoking.

Objective To explore the clinical manifestations,diagnosis,misdiagnosis and treatment of primary hyperparathyroidism.Methods The clinical data of 36 patients with primary hyperparathyroidism admitted into the First Affiliated Hospital of Chongqing Medical University from January 2009 to October 2014 were reviewed and analyzed retrospectively.Results The manifestations of primary hyperparathyroidism were various and atypical.Primary hyperparathyroidism was often misdiagnosed as other diseases by 25.0％ (9/36).Among the patients,the ratio of male to female was 1:1,and median age was 58 years old.Of all patients,there were 9 persons with joint pain,digestive system symptoms in 7 cases,cervical mass discovered in 6 cases(3 cases were found when B ultrasonic examination),urinary calculus in 5 cases,fatigue symptoms in 4 cases,osteoporosis and urinary calculus in 2 cases,1 case with fracture,2 cases without any symptoms.Their calcium and parathyroid hormone level were increased by various degree.The accuracy rates of level diagnosis of Color Doppler ultrasound combine with CT or 99Tcm-MIBI were improved obviously,85.2％ and 93.1％ respectively.Twenty-eight patients received surgical treatment and pathology examination confirmed the accurate pathological types after surgery.There were 23 cases of parathyroid adenoma,1 case parathyroid hyperplasia,1 case parathyroid carcinoma,2 cases of parathyroid adenoma with nodular goiter,parathyroid adenoma merged thyroid carcinoma in 1 case.Serum calcium and parathyroid hormone level declined obviously after operation.The other 8 patients received non-surgical treatment,7 cases were alleviated and 1 case died due to irreformable high blood calcium crisis.Conclusion There should be pay attention to recognize primary hyperparathyroidism,check the blood calcium,and about suspected cases should be performed as early as parathyroid tested,neck color Doppler ultrasound.Early surgical treatment can significantly improve the prognosis and quality of life.

Objective To investigate the distribution and drug resistance of clinical bacteria infection in the Second Affiliated Hospital of Kunming Medical University,so as to provide reference for the treatment of bacterial infections.Methods 4 802 strains of bacteria isolated from this hospital,from January 2013 to December 2013,were retrospectively analysed.The isolates were identi-fied by using VITEK-2 Compact bacterial identification system.Drug resistance was measured by using disc diffusion test,and its results were interpreted according to the breakpoints of Clinical and Laboratory Standards Institute(CLSI)2013.WHONET 5.6 was applied for analysis.Results These pathogens were mainly isolated from urine,sputum,blood,secretions and pus,accounted for 31.7%,21.4%,1 9.7%,1 1.7% and 7.0%,respectively.In the clinical isolates,gram negative bacilli accounted for 55.8%, which was mainly Escherichia coli(26.3%).Gram positive cocci accounted for 31.7%,,which was mainly coagulase negative staph-ylococcus(1 5.0%).Fungi accounted for 3.1%,which was mainly Candida albicans.Escherichia coli and Klebsiella pneumonia were most sensitive to carbapenem,resistance rate was less than 10.0%.The detection rates of Escherichia coli and Klebsiella pneumoni-ae producing extended-spectrumβ-lactamases(ESBLs)was 61.1% and 49.1%,respectively.Among non-fermentative gram nega-tive bacilli,excepting Pseudomonas aeruginosa had good sensitivity to Amikacin,Pseudomonas aeruginosa and Acinetobacter bau-mannii showed high resistance to most antibiotics(resistance rate was more than 50.0%).Among gram positive bacteria,the detec-tion rates of meticillin-resistant Staphylococcus aureus and methicillin-resistant coagulase negative Staphylococcus were 42.3% and 65.6%,respectively.The resistance rates of Enterococcus faecium to most of antibacterials were higher.Except for linezolid and teicoplanin,the resistances of Enterococcus faecium to other antibacterials were higher than those of Enterococcus faecalis.Only a strain of Enterococcus faecium resistant to vancomycin was isolated.Conclusion Resistance monitoring might have significance in guiding the clinical rational use of antimicrobial agents,and reducing the spread of antibiotic resistance in bacteria.
