1.Effects of a static magnetic field on proliferation, apoptosis and secretion in human umbilical cord endothelial cells

Objective To evaluate the effects of static magnetic fields (SMFs) of different intensity and exposure duration on the proliferation and apoptosis of human umbilical cord endothelial cells (HUVECs),and their release of nitric oxide (NO),6-keto-prostacyclin 1α (6-keto-PGF1α) and endothelin (ET-1).Methods Cultured HUVECs were exposed to a SMF at 5,22,86 or 135 mT for 8,12 or 24 hours.Their proliferation and apoptosis were monitored by flow cytometry (FCM).The medium was collected to test its NO content by optical density.ET-1 and 6-keto-PGF1α were measured by radioimmunization.Results ( 1 ) The proliferation of HUVECs increased when the cells were exposed to a SMF at 5 mT for 8 h,but a SMF at 135 mT for 12 h or 24 h inhibited the proliferation of HUVECs.(2)An SMF had no effect on apoptosis of HUVECs.(3)An SMF at 5 mT for 8 h increased the release of NO and 6-keto-PGF1 a,but the release of NO and 6-keto-PGF1 a decreased when the SMF intensity was 135 mT or the cells were exposed to an SMF for 12 h or 24 h.(4) An SMF at 5 mT or 22 mT for 8 h did not effect the release of ET-1.An SMF at 86 mT or 135 mT increased the release of ET-1.Compared with a control group,an SMF at 5 mT for 12 or 24 h did not affect the release of ET1,but at 22,80 or 135 mT,the release of ET-1 decreased significantly.Conclusions Exposure to a low intensity SMF for a short duration could improve the proliferation of HUVECs and increase the release of vasoactive factors,but if HUVECs are exposed to a strong SMF or exposed for a long duration,the proliferation and the release of vasoactive factors is decreased.
2.Effect of angiotensin Ⅱ and aldosterone in patients with congenital heart disease by preoperative nursing interventions at high altitude area

Objective To study the effect of angiotensin Ⅱ and aldosterone (ALD) in patients with congenital heart disease by preoperative nursing interventions at high altitude area. Methods 86 patients with congenital heart disease were divided into the experimental group and the control group on average by random. The experimental group was treated with preoperative nursing interventions and routine nursing before operation and the control group was treated with routine nursing only. Angiotensin Ⅱ and aldosterone were detected before anesthesia, 30min after operation and after pulling out of endotracheal tube respectively. Results The concentration of Angiotensin Ⅱ and aldosterone was significantly lower in the experimental group than the control group. Conclusions The risk of open surgical operation for congenital heart disease at high altitude can be reduced by preoperative nursing interventions.
3.Determination of Eucalyptol, Camphor and Menthol in Compound Menthol Camphor Eucalyptus Oil Solution by GC

Objective: To establish a method for the determination of eucalyptol, camphor and menthol in compound menthol camphor eucalyptus oil solution by GC.Methods: An HP-INNOWAX 19091N-216 capillary column(60 m× 0.32 mm , 0.50 μm)was used.The carrier gas was nitrogen with the flow rate of 30 ml·min-1 , the gas was hydrogen with the flow rate of 40 ml·min-1 and the oxidant gas was air with the flow rate of 400 ml· min-1.The detector was FID and the inlet temperature was 250℃.The temperature program was as follows: the initial column temperature was 50℃, and then risen to 160℃ with a rate of 10℃·min-1 and kept for 5 min, and finally risen to 220℃ with a rate of 20℃·min-1 and kept for 3 min.The split ratio was 15∶1 and the injection volume was 1 μl.Results: The linear range of eucalyptol, camphor and menthol was 0.031 9-2.550 0 mg·ml-1 (r=1.000 0), 0.041 3-3.305 0 mg·ml-1 (r=1.000 0) and 0.053 7-4.294 0 mg·ml-1 (r=1.000 0), respectively.The average recovery was 98.24% (RSD=0.3% , n =9), 98.97% (RSD=0.4% , n =9) and 98.98% (RSD=0.5% , n =9), respectively.Conclusion: The method is sensitive and accurate with good stability, which can be used to determine the contents of eucalyptol, camphor and menthol in compound menthol camphor eucalyptus oil solution.

5.Efficacy of statin in treatment of patients with chronic obstructive pulmonary disease at stable stage

Objective To explore the therapeutic effects of statin in patients with chronic obstructive pulmonary disease (COPD) in stable period. Methods 83 patients with COPD in stable period were recruited and randomly divided into two groups, treatment group (n = 41) and control group (n = 42).The control group received conventional routine therapy, and treatment group was given rosuvastatin calcium 20 mg once per day on the basis of routine treatment. The levels of sera C-reactive protein (CRP), pulmonary function (FEV1、FEVl%pred), and COPD assessment test (CAT) were observed at the time points of 0 week and 12th week, 24th week and 48th week were compared between two groups. Results At 12th week after the treatment, sera CRP levels, FEV1, FEV1%pred and CAT scores in either control group or treatment group showed some extent of improvements as compared to those at the time point of 0 week, and there was statistical difference of sera CRP levels and CAT scores between two groups (P < 0.05). There was no statistical difference of FEV1, FEV1%pred between two groups (P > 0.05) .24th week and 48th week after the treatment; sera CRP levels, FEV, FEV1%pred and CAT scores in two groups all significantly improved and were statistical different between two groups (P < 0.05). Conclusion Statin can alleviate inflammatory reaction, improve pulmonary function and life quality of patients with COPD.
6.Effect of continuous positive airway pressure treatment on serum levels of hemorheology and c-reactive protein in patients with obstructive sleep apnea hypopnea syndrome

Objective To investigate the effects of continuous positive airway pressure (CPAP) on the levels of hemorheology and C-reactive protein in patients with obstructive sleep hypopnea apnea syndrome (OSAHS). Methods 58 moderate to severe OSAHS subjects were selected as treatment group and 32 healthyadults were selected as control group. Hemorheology and C-reactive protein in all subjects were examined and compared. The levels of hemorheology and C-reactive protein were also examined and compared before and after 3 months CPAP treatment in treatment group. Results In treatment group, the levels of the whole blood viscosity and hs-CRP were significantly higher than those in control group (P < 0.01). After treatment of CPAP for three months, the levels of the whole blood viscosity and hs-CRP obviously decreased (P < 0.01). Conclusion Levels of hemorheology and hs-CRP are elevated in patients with OSAHS and CPAP therapy could effectively decrease serum levels of hemorheology and hs-CRP in patients with OSAHS.
7.Content Determination of Chlorogenic Acid, Quercetin and Kaempferol inPyrrosia Lingua (Thumb.) Farwell by HPLC

Objective To establish an HPLC method for simultaneous separation and determination of chlorogenic acid, quercetin and kaempferol inPyrrosia lingua (Thumb.) Farwell. Methods The separation was performed on Phenomenex C18 column (250 mm×4.6 mm, 5μm) with the mobile phase of methanol-acetic acid (pH=3.0) solution and gradient elution. Flow rate was 1.0 mL/min;the UV detection wavelength was 254 nm;column temperature was 35℃.Results The calibration curves for chlorogenic acid, quercetin, and kaempferol were in good linearity in the range of 0.000 24-3.00μg (r=0.999 9), 0.000 16-2.00μg (r=0.999 9), and 0.000 16-2.00μg (r=0.999 9), respectively. The limits of detection (S/N=3) were 3.29, 0.43 and 0.33 ng/mL, respectively. The average recovery rates were 97.73%, 98.07% and 96.92%, respectively. ConclusionThe method is simple, precise and sensitive. It provides scientific proof for separation and determination of chlorogenic acid, quercetin and kaempferol inPyrrosia lingua (Thumb.) Farwell.
8.Expression and significance of SIRT1 and p53 in endometrial carcinoma with diabetes mellitus

Objective To investigate the expressions and significances of SIRT1 and p53 in endometrial carcinoma with diabetes mellitus.Methods Immunohistochemistry was carried out to examine the expressions of SIRT1 and p53 in 39 cases of endometrial carcinoma with diabetes mellitus and 40 cases of endometrial carcinoma without diabetes mellitus and 25 cases of normal endometrium,and the correlations between these indexs were analysed.Results The positive rate of SIRT1 and p53 in experimental group was 64.1%,59.0% respectively,and 85.0%,52.5% in control group.The positive rate of SIRT1 in experimental group was lower than that of control group (x2 =4.561,P =0.033).The positive rates of SIRT1 and p53 in control group were higher than that of normal endometrium (x2 =21.462,P< 0.001; x2 =6.771,P =0.009).Expression of SIRT1 was positively correlated with p53 (r =0.360,P =0.024).Conclusions Hyperglycemia deregulates SIRT1 in endometrial carcinoma.SIRT1 may promote tumorigenesis by silence of p53.

10.Determination of Uric Acid Based on Active Copper-Containing Carbon Nanodots-modified Electrochemical Biosensor

The active copper-containing carbon nanodots were prepared by hydrothermal method, and then characterized by fluorescence spectroscopy and UV-visible absorption spectroscopy.Subsequently, a highly sensitive and selective electrochemical biosensor was fabricated on the basis of this synthesized carbon nanodots with electro-deposition technique.The electrode behavior was investigated by cyclic voltammetry, electrochemical impedance spectroscopy, and differential pulse voltammetry.Furthermore, the catalysis mechanism was studied.The experimental results indicated that the biosensor exhibited a strong electrocatalytic activity toward the oxidation of uric acid (UA).What′s more, the interference from ascorbic acid and dopamine was eliminated effectively.Under the optimum conditions, there were linear relationships between the anodic peak current and the concentration of UA (1.00-300.0 μmol/L), and the limit detection was 0.30 μmol/L (S/N=3).The prepared biosensor had advantages such as easy fabrication, strong anti-interference ability, high sensitivity, and wide detection range, and could be used for real sample detection.
