Objective To observe the effect of aloe on intestine motility in the old costive mice and investigate the mechanism for aloe promoting an intestinal motility. Methods The content of aloin in aloe powder was determined by high performance liquid chromatography(HPLC).The old mice aged 15 months were randomly divided into 6 groups(10 in each group):blank control, positive control, constipation model,low-dose aloe,middle-dose aloe,high-dose aloe plus model. The mice of with equivalent volume of distilled water. On the eighth day,the mice except control group were given Compound Diphenoxylate to establish constipation model. With the black Indian ink as marker, the first time of black stool discharge, the character and weight of the stool, and the ink propulsion rate by intestines in mice were observed respectively. The serum level of nitric oxide(NO)was determined by spectrophotometry. Results The content of aloin in aloe powder was 0.266%.Compared with constipation model group, aloe groups in different dose decreased the first black stool time and increased stool grains and weight in 6 hours of constipated mice. The ink propulsion rates of intestines in the aloe groups were significantly higher than that of model group as well. The NO level in high-dose aloe group decreased more significantly compared with model group(P<0.05),and there was a negative correlation between the serum NO level and propulsion rate of intestines(r=-0.346.P<O.05). Conclusions Aloe could promote the mobility of intestine and ameliorate the constipation of mice, which might attribute to the decrease of the serum NO level.

DNA barcoding technology is widely opplied for species identification and discovery by using short and standard fragments of DNA sequences. In this study, the cytochrome c oxidase subunit 1 (COI) sequence as a DNA barcode was used to identify the Gekkogecko Linnaeus medicinal materials and adulterants in order to provide a new identification method of G. gecko Linnaeus. Genomic DNA was extracted from the experimental samples. The COI regions of nrDNA were amplified using polymerase chain reaction and sequenced bidirectionally. The alignment and NJ tree construction were performed in MEGA6.0. COI sequences can be obtained from all samples. The average intra-specific K2P distance of G. gecko Linnaeus was 0.005 and the maximum intra-specific distance was 0.013. All the G. gecko Linnaeus medicinal samples clustered into a clade in the NJ tree and can be distinguished from the adulterants. In a conclusion, COI can be used to correctly identify G. gecko Linnaeus, and it will be a potential DNA barcode for identification of other animal medicinal materials.

Objective To explore the characteristics and its clinical significance of troponin I(cTnI),myo-cardial enzymes and intraoperative hemodynamic changes in the pediatric patients undergoing living donor liver trans-plantation. Methods Liver transplantation was performed in 50 congenital biliary atresia children who were ranged from grade Ⅲ or Ⅳ in Tianjin First Central Hospital from January 2013 to December 2014 according to the American Society of Anesthesiologists(ASA),meanwhile,the method of the combined intravenous - inhalation anesthesia was ap-plied during operation. Blood samples were drawn from central vein before skin incision(T0 baseline),at 30 min of an-hepatic phase(T1),30 min of neohepatic phase(T2),and 12 h,36 h after operation(T3,T4). Levels of cTnI,crea-tine kinase(CK),lactate dehydrogenase(LDH)and α - hydroxy butyric acid dehydrogenase(α - HBDH)were mear-sured,respectively. Furthermore,heart rate(HR),mean arterial blood pressure(MAP),central venous pressure(CVP) and arterial blood gas analysis[pH value,pa(O2 ),pa(CO2 ),and base excess(BE)]were monitored at the moment of T0,T1,T2 as well as the end of surgery. Results The levels of cTnI,CK,LDH and α - HBDH in T1 - T3 were in-creased,and there was a peak at the T2 compared with the baseline at T0(all P ﹤ 0. 05). At T3 and T4,cTnI,CK, LDH and α - HBDH levels significantly decreased compared with those at T2(all P ﹤ 0. 05),the levels of cTnI were (0. 06 ± 0. 02)μg/ L,(0. 37 ± 0. 52)μg/ L,(0. 05 ± 0. 02)μg/ L,CK levels were(344. 6 ± 209. 5)U/ L,(466. 1 ± 116. 4)U/ L,(219. 3 ± 111. 5)U/ L,LDH levels were(552. 3 ± 414. 9)U/ L,(966. 4 ± 454. 1)U/ L,(322. 8 ± 108. 8) U/ L,and α - HBDH levels were(301. 6 ± 124. 0)U/ L,(456. 4 ± 168. 4)U/ L,(146. 2 ± 80. 2)U/ L,respectively. The levels of hemodynamics significantly changed in anhepatic phase and neohepatic phase. Compared with T0:T1,HR ac-celerated,MAP,CVP decreased,BE value increased,and the differences were statistically significant(all P ﹤ 0. 05);T2,open vena cava and back to the blood volume surge,CVP,MAP increased,HR decreased but still higher than T0, BE value further increased,and the differences were statistically significant(all P ﹤ 0. 05). After the surgery,various hemodynamic indexes fell to preoperative levels,the levels of HR were(103. 1 ± 5. 9)times/ min,(128. 8 ± 8. 5) times/ min,(115. 1 ± 0. 3)times/ min,(103. 5 ± 5. 9)times/ min,MAP levels were(59. 7 ± 9. 1)kPa,(48. 7 ± 5. 4) kPa,(58. 6 ± 7. 1)kPa,(59. 1 ± 8. 6)kPa,CVP levels were(7. 5 ± 4. 3)kPa,(3. 9 ± 4. 6)kPa,(5. 8 ± 3. 5)kPa, (7. 2 ± 4. 1)kPa,BE levels were( - 1. 5 ± 5. 0)mmol/ L,( - 0. 4 ± 5. 7)mmol/ L,(1. 0 ± 3. 8)mmol/ L,(2. 4 ± 2. 2)mmol/ L,respectively. Conclusions The myocardial injury may appear during the perioperation of pediatric living donor liver transplantation and gradually aggravated during the anhepatic phase. The worst injury peaks at 12h and it gradually returns to the preoperative level 36 h postoperativelly.

Objective: To investigate the role of indoleamine 2, 3-dioxygenase in the development of uterine cervical squamous carcinoma. Methods: From January 2008 to December 2008, 116 uterine cervical carcinoma specimens and 18 metastatic lymph node specimens from patients with CIN Ⅰ-Ⅲ and uterine cervical squamous carcinoma were evaluated for iDO expression by immunohistochemistry. Twenty normal cervical specimens and 20 normal lymph node specimens were used as the controls. Results: The expression of IDO was not found in normal cervix and CIN Ⅰ. In CIN Ⅱ, IDO expres-sion was weakly positive in 2 cases (2/10, 20%) and negative in 8 cases (8/10, 80%). In CIN Ⅲ, IDO expression was weak-ly positive in 8 cases (8/13, 61.5%), positive in 1 case (1/13, 7.7%) and negative in 4 cases (4/13, 30.8%). The positive ex-pression rate of IDO in cervical cancer stage Ⅰ -Ⅳ was 100% (83/83). In cervical cancer stage Ⅰ A and Ⅰ B, the positive ex-pression rate of IDO was significantly higher than that in CIN Ⅱ and CIN Ⅲ (P<0.01). The positive expression rate of IDO in cervical cancer stage Ⅱ A-Ⅳ B was significantly higher than that in Ⅰ A and Ⅰ B. IDO expression was associated with cervi-cal cancer progression (OR=0.807, P<0.01). IDO expression in primary lesions with lymph node metastasis was significant-ly higher than that in those without lymph node metastasis. IDO expression rate was 100% in metastatic lymph nodes. The IDO expression was not associated with cervical squamous carcinoma differentiation degree (OR=-0.139,P>0.05). Conclu-sion: In CIN Ⅱ, escape mechanisms that stimulate cervical squamous carcinoma progression is gradually developed. IDO expression in metastatic lymph nodes is possibly associated with immune tolerance. IDO expression is not associated with differentiation degree of cervical squamous carcinoma. IDO may be a prognostic factor for uterine cervical squamous carci-noma and a therapeutic target for treatment.
