2.The Protective Effect of the Serum Containing Tongmai on the Anoxic Myocardial Cells

Objective To observe the influence of serum containing Tongmai on the anoxic myocardial cell. Methods The serum containing Tongmai effective components was extracted and used to interfere the anoxic myocardial cell, then the concentration of the LDH and troponin I in the culture medium of the cardiac myocyte was measured. Results The concentration of LDH and troponin I increased significantly in the culture mediun of the anoxic cardiac myocyte. The intervention of the serum containing Tongmai can significantly decreased the concentration of LDH and troponin I in the culture medium of the anoxic cardiac myocyte. Conclusion Tongmai can protect the cardiac myocyte from anoxic damage.
3.Arsenic trioxide induced p15INK4B gene expression in myelodysplastic syndrome cell line MUTZ-1.

OBJECTIVETo investigate the mechanisms of arsenic trioxide (As(2)O(3)) induced demethylation.
METHODMethylation of p15INK4B gene in MUTZ-1 cell was detected by PCR using a methylation specific primer (MSP), the expression of P15, DNA methyltransferase (DNMT) 1, DNMT3A and DNMT3B gene by RT-PCR, the As(2)O(3) induced growth inhibition of MUTZ-1 cell by MTT method.
RESULTSP15 gene failed to express in MUTZ-1 cells after methylation. The expression was recovered after the cells exposed to As(2)O(3). As(2)O(3) could significantly down-regulate DNMT3A and DNMT3B but not DNMT1 gene on mRNA level in a dose dependent manner.
CONCLUSIONAs(2)O(3) could activate the expression of p15 gene by demethylation or/and by inhibiting DNMT3A and DNMT3B gene.

4.Establishment and pathophysiological changes of a rat model of increased blood flow-induced pulmonary arterial hypertension by anastomosis of the left common carotid artery to left external jugular vein

ObjectiveToestablisharatmodelofincreasedbloodflow-inducedpulmonaryarterialhypertension generatedbyanastomosisoftheleftcommoncarotidarterytoleftexternaljugularvein.Methods 45maleSDratswere divided into three groups:the shunt group , the ligation group and the sham group .At twelve weeks after the procedure , the general status of the rats was observed . Heart conditions , cardiac output and shunt patency were measured by echocardiography .Right ventricular systolic pressure ( RVSP ) and Qp/Qs were checked by catheterization . Right ventricular hypertrophy index ( RVHI) was calculated and lung tissues were examined by pathology using hematoxylin -eosin and elastin Van Gieson staining .All data were analyzed statistically by one-way ANOVA test using SPSS 16.0.Results There was no significant difference in body weight gains between the groups .The patency rate of shunt was 84.6%.The heart was enlarged in the group shunt .Cardiac output increased significantly in the shunt group than that in the other two groups [(309.8 ±33.1) mL/min?kg vs.(245.6 ±31.9) mL/min?kg, (240.8 ±30.9)mL/min?kg, respectively, P<0.05].In the shunt group Qp/Qs was 2.16 ±0.38 and RVSP increased to (35.8 ±4.9) mmHg, RVHI was 0.3263 ± 0.0342, significantly higher than that of the other groups .The pulmonary arteriolar wall was evidently thickened in contrast to that in the sham group [ ( 22.3 ±1.7 )% vs.( 10.6 ±1.7 )%, P <0.05 ) .Conclusions Anastomosis of the left common carotid artery to left external jugular vein can successfully establish pulmonary arterial hypertension model induced by high blood flow in rats .
5.Influence of serum contained Tongmai herbal medicine on intracellular calcium concentration and L- type calcium channel in anoxic myocardial cells

AIM: To observe the influence of serum contained Tongmai herbal medicine on calcium overload of the anoxic myocardial cells. METHODS: Serum contained Tongmai herbal medicine was prepared with the serum pharmacological method and the model of anoxic myocardial cells was established. The intracellular free calcium concentration and the expression of the L-type calcium channel were determined with Fura-2/AM and RT-PCR, respectively. RESULTS: The serum contained Tongmai herbal medicine and levo-anlodipine decreased the intracellular free calcium concentration in the anoxic myocardial cells. The serum contained Tongmai herbal medicine also suppressed the expression of L-type calcium channel in the anoxic myocardial cells, but levo-anlodipine had no effect on the expression of L-type calcium channel. CONCLUSION: Tongmai herbal medicine can attenuate calcium overload in anoxic myocardial cells.

7.The efficacy and safety of HAA regimen as induction chemotherapy in 150 newly diagnosed acute myeloid leukemia

Objective To explore the efficacy and safety of HAA regimen (homoharringtonine,cytarabine and aclarubicin) in the treatment of 150 newly diagnosed adult acute myeloid leukemia (AML).Methods All patients entered the study from May 1999 to June 2008 were treated with HAA regimen. Coxsurvival analysis was used to estimate the survival rate and differences between M1/M2 and M4/M5 were compared with 2-sided log-rank test. Results Out of the 150 patients, 121 (81%) achieved complete remission (CR). After the first course, CR rate was 68%. The CR rates of 97%, 84% and 38% were achieved in patients with favorable, intermediate and unfavorable cytogenetics, respectively. For the patients with CR, the median follow-up time was 16.5 ( 1.5-100.5 ) months, and the estimated 3-year survival rate was 45%. The estimated 3-year relapse free survival rate was 52% for the 121 patients with CR.Conclusions HAA regimen may be an efficacious and safe regimen with a good toleration in the induction therapy for newly diagnosed AML, and a high CR rate could be achieved with only one or two courses.
8.Relation between Beta-2-glycoprotein I and hepatitis B virus surface antigen.

OBJECTIVETo clarify the binding character between Beta-2-glycoprotein I (Beta-2-GPI) and HBsAg.
METHODSBeta-2-GPI was purified from human plasma and labelled with biotin. Solid phase enzyme linked absorbance assay was used to investigate its binding with HBsAg.
RESULTSBiotinylated Beta-2-GPI was found to bind HBsAg and the reaction could be inhibited by excess unlabelled Beta-2-GPI.
CONCLUSIONSBeta-2-GPI may play a role in hepatitis B virus infection.

9.Assessment of Technology and Clinical Application of Hydrosurgery System.

This paper introduces the technical features of hydrosurgery system and evaluates it from three aspects:safety and effectiveness, economy and technology maturity. The evaluation results show that the safety and effectiveness are high, and the safety mainly involves the misuse of the system, which cause the adverse events. A large number of literatures have proved that the system is more suitable for the debridement of fine parts. It is estimated that hydrosurgery system has better economic performance, but the cost and patients' cost is also increased correspondingly. In the aspect of technology maturity assessment, this technology is a patented technology, and the number of published papers both at home and abroad is on the rise, indicating that the initial application of the technology is better and the technology is in infancy stage. Therefore, hydrosurgery system has certain advantages in all aspects. Hospitals need to allocate the equipment according to their own needs and clinical application, while a wider range of clinical trials is needed to provide evidence-based evidence for clinical applications and promotion.
