Objective:To investigate the expression pattern of MMP-11 and MMP-14 in breast carcinoma, and the effect of MMP-11 on breast carcinoma cell migration and invasion. Methods:MMP-11 and MMP-14 expression were examined in 161 invasive breast carcinoma tissue samples and 10 normal breast tissue samples. siRNA was used to knockout MMP-11 in breast carcinoma cell line MB-231 and Transwells were used to evaluate changes in migration ability and invasion ability. Results:Both MMP-11 and MMP-14 were highly expressed in breast carcinoma samples,122 and 149 samples out of 161,respectively. The expression of both proteins were correlated with lymph node metastasis and TNM staging. After knockout of MMP-11,the expression of both proteins decreased in MB-231 cell line and experiments show that the cell′s migration and invasion abilities were significantly weakened. Conclusion:MMP-11 and MMP-14 could promote invasion and metastasis of breast carcinoma. Knockout of MMP-11 results in the downregulated expression of MMP-14,and the inhibition of breast carcinoma cell′s migration and invasion. They could be potential prognostic markers and treatment targets for of breast carcinoma.
3.Influence of White Prepared Lateral Root of Aconite on Blood Biochemical Indicators in Normal and Kidney Yang Deficient Rats

Objective To explore the differences of blood biochemical indicators between normal and kidney Yang deficient rats administered orally different doses of white prepared lateral root of aconite.Method Eighty Sprague-Dawley(SD) male rats were divided randomly into normal group and kidney Yang deficient model group,forty rats in each group.After producing kidney Yang deficient model by injecting hydrocortisoni natrii succinas in intramuscular,normal and model rats were respectively and randomly divided into little,low,medium,great dose group and blank group,eight rats in each group.Those rats were administered orally with different concentrations of white prepared lateral root of aconite every day for two weeks.Twenty-four hours after the last oral administration,the blood were obtained and used for testing adrenocorticotrophic hormone(ACTH),cyclic adenosine monophosphate(cAMP),cyclic guanosine monophosphate(cGMP),glucose(GLU),cholesterol(CHO),alanine aminotransferase(ALT),urea nitrogen(BUN).Results Before taking medicine,model rats' cAMP/cGMP and ACTH were lower than normal rats',and they showed a trend of rise with adding dose.Before taking medicine,model rats' GLU was lower than normal rats'.With adding dose,model rats' GLU increased,however,normal rats' GLU reduced.When taken medium dose white prepared lateral root of aconite,model rats' CHO was higher than normal rats'.Taking medicine made both groups ALT rise,but normal rats' went up more quickly than model rats'.Model rats' BUN was higher than normal rats',and taking low dose or little dose medicine could lead to incline of reduction.Conclusion White prepared lateral root of aconite caused different blood biochemical effects on normal and kidney Yang deficient rats,and more remarkable toxicity effect on normal rats.
4.Analysis of the outcomes of multi-spot medical practice trial in Kunming

Objective To accurately understand multi-spot medical practice in China and evaluate its effect.Methods Operation data of 2008 and 2009 were gathered from all medical institutions in Kunming to analyze the effect of the practice trial using the fixed effect model.Results Under control of such factors as the scale and type of the hospitals,clinic visits of the second certified hospital benefited by the practice increased by 14%than before,whereas the medical revenue of the institution increased by 29%.The effect proves more significant in terms of total clinic visits and medical revenues at level-1 and level-2 medical institutions.Conclusion Multi-spot medical practice in Kunming proves successful as it increased medical service supply and medical revenue,especially for level-1 and level-2 medical institutions.This effect possibly results from releasing high-level physicians down to lower-level institutions,helping these institutions to attract patients.
5.Cloning and characterization of BmBrat in silkworm, Bombyx mori.

NHL proteins, which play important roles in regulation of cell proliferation and differentiation, have been extensively studied on mammals. Here, we cloned a member of NHL protein family namely BmBrat in silkworm. The full-length cDNA sequence of BmB rat was obtained by means of the rapid amplification of cDNA ends (RACE), including 3 614 bp. The ORF is 2 580 bp long, encoding a protein with 859 amino acid residues. The molecular weight is 94.3 kDa and the isoeledtric point (pI) is 6.65. The BmBrat expression profile was detected by RT-PCR at L5D3 larval stage, and it was expressed in all tissues, including silk gland, midgut, fat body and malpighian tubule. However, it was highly expressed in ovary and head. The expression profile was also detected at different stage of embryo development, and reached a peak at the 4th and 5th days of the embryonic period. Anti-BmBrat polyclonal antibody was generated f6llowing prokaryotic expression, protein purification and mice immunization, which is highly specific and effective for recognizing BmBrat protein through Western blotting and immunofluorescence staining. Subcellular localization of BmBrat in hemocytes revealed that it was specifically expressed in cytoplasm. This study provides a foundation for further research of the biological function of BmBrat gene.

6.The inhibitory effects of chlorhexidine in the development of rat periodontitis models

Objective:To investigate the inhibitory effects of different concentrations of chlorhexidine in the development of peri-odontitis models in rats.Methods:periodontitis models were established by the ligation of bilateral first molars and orally challenge with P.gingivalis W83.0.05%,0.1%,0.2% and 0.5% chlorhexidine were used to wash the periodontal pocket and oral mucosa of the rats.4 weeks later,absolute real time quantitative PCR was used to count the copy of P.gingivalis W83 in rat periodontal pockets.Scanning electron microscopy was used to observe the distribution of P.gingivalis W83 on rat teeth surface.Immunohisto-chemical technique was used to detect the expression of TNF-αin gingival tissue of the rats.Results:0.2% and 0.5% chlorhexi-dine reduced the copy of P.gingivalis W83 on teeth surface and in periodontal pockets (P <0.05);0.1% -0.5% chlorhexidine reduced the expression of TNF-αin gingival tissue (P <0.05).Conclusion:0.1% -0.2% chlorhexidine can inhibit the develop-ment of chronic periodontitis in rats.

Cerebral microbleeds (CMBs) are a key biomarker of cerebral small vessel disease on magnetic resonance imaging,They have potential clinical relevance to future stroke risk.Therefore,the detection of CMBs has important clinical significance for various cerebrovascular disease phenotypes.This article briefly summarizes the detection method of CMBs,mainly investigating the clinical significance of CMBs in general population and in patients with ischemic stroke,cerebral hemorrhage,vascular cognitive impairment,cerebral amyloid angiopathy,and leukoaraiosis.

10.Screening of Mimic Epitopes of Toxoplasma gondii Antigen by Phage-displayed Peptide Library

Specific antibodies purified from sera of rabbit infected with Toxoplasma gondii were used to immun-oscreen the 12-mer and 7-mer phage random peptide libraries. By 3 rounds of screening, 70 clones were picked out ra-ndomly to test the specificity and 43 were found positive by ELISA. 23 of the 43 positive clones showing strong reaction in ELISA were picked out for sequencing. Clone A7 was selected as mimic antigen in ELISA test after analysis of the 23 short peptide sequences. 23 out of 47 sera from rabbits infected with Toxoplasma gondii were ELISA positive by A7 mimic antigen with a positive rate of 68.1%, and 10 of 155 sera from healthy persons showed false positive with a sp-ecificity of 93.5%. The primary result suggests that mimic antigen may have a potential use in diagnose of toxoplasmosis.
