Objective To investigate the inhibitory effects of Snail—— a zinc finger transcription factor, anti-sense plasmid on the invasion of ovary carcinoma cell lines in vitro. Methods Four human ovarian carcinoma cell lines ES-2,OVCAR3,HO8910,HO8910PM were analyzed for the expression of Snail and E-cadherin mRNA by RT-PCR.Then anti-Snail plasmid transfection was introduced into HO8910 cells using lipofectin 2000 reagent to investigate the inverse correlation between E-cadherin and Snail expression, and the inhibitory effects of anti-sense Snail were also detected by Transwell motility assay and Matrigel invasion assay. Results The results demonstrated the presence of Snail mRNA in ES-2,HO8910,HO8910PM detected by RT-PCR. By contrast, E-cadherin mRNA was only detected in OVCAR3 by RT-PCR.The inverse relationships were further observed by transient transfection of anti-sense Snail into HO8910 cells, which showed the down-regulated expression of Snail and the re-expression of E-cadherin. Objective To investigate the influence of rosuvastatin on inflammatory factors and immune func-tion in chronic obstructive pulmonary disease(COPD)with pulmonary heart disease.Methods 80 COPD patients with pulmonary heart disease were randomly divided into two groups,40 cases in each group.The control group was treated with conventional medicine comprehensive treatment,the observation group was given rosuvastatin based on the control group.The inflammatory cytokines (TNF -α,IL -1 and hs -CRP)changes,immunoglobulin IgM,IgG and IgA changes situation,and heart,lung function changes related indicators were compared between the two groups before and after the treatment.Results After treatment,the TNF -α,IL -1 and hs CRP levels of the observation group were significantly lower than those before treatment(t =145.5,35.35,57.5,all P <0.05)and those of the control group(t =160.75,11.45,32.99,all P <0.05).After treatment,the IgM,IgG and IgA levels of the observa-tion group were significantly lower than those before treatment (t =4.89 of 18.29,3.42,all P <0.05)and those of the control group (t =2.55 9.67,2.342,all P <0.05 ).After treatment,the FEV1 ,FEV1 /FVC and LVEF of the observation group were significantly higher than those before treatment(t =40.44 33.64,33.79,all P <0.05),while the PAP level was significantly lower than that before treatment (t =24.12,P <0.05),and the FEV1 ,FEV1 /FVC and LVEF of the observation group were significantly higher than those of the control group after treatment(t =16.30, 15.82,18.72,all P <0.05),while the PAP level was significantly lower than that of the control group after treatment (t =13.43,P <0.05).Conclusion Combined rosuvastatin therapy for COPD patients with pulmonary heart disease can effectively reduce body inflammatory response,improve immune function and myocardial contractility,so it can promote functional recovery of heart and lung in patients.

ObjectiveTo investigate feasibility of use of glucose injection instead of glucose powder in oral glucose tolerance test (OGTT). MethodsSixty healthy adult volunteers without history of diabetes were recruited for a standard OGTT with 75 g anhydrous glucose powder first. One week later, they were randomly divided into two groups, each of the one group (30 volunteers) orally took seven ampoules (20 ml/ampoule) and each of the other group (30 volunteer) took 7.5 ampoules of 50％ glucose injection for OGTT again, as compared to those with standard OGTT.Plasma levels of glucose and insulin were examined to evaluate whether different forms and dosage of glucose had similar results in OGTT. ResultsIn 23 volunteers with normal glucose tolerance, their plasma levels of glucose were ( 4. 8 ± 0. 4 ), ( 6. 7 ±0. 6), (5.9 ±0. 8), (5.5 ±0. 9) and (4. 8 ±0. 9) mmol/L at 0, 30, 60, 120 and 180 min after oral load with 75 g anhydrous glucose powder, respectively. These values changed to (4. 8 ± 0. 3 ), (7.5 ± 1.1 ),(6.8±1.8), (6.3 ±1.0) and (4.6 ±1.2) mmol/Lor (4.7 ±0.3), (7.2±1.3), (6.1 ±1.1),(5.6 ± 0. 9 ) and (4. 3 ± 0. 9) mmol/L after oral load with seven ampoules ( 15 volunteers) or 7. 5 ampoules of (8 volunteers ) of 50％ glucose injection, respectively.With standard OGTT, 37 cases of impaired glucose tolerance were found from 60 volunteers tested, and their plasma levels of glucose were (5. 2 ±0.6), (9. 1 ±1.4), (8.9 ±2.6), (6.7±2.0) and (4.7 ±1.0) mmol/L at 0, 30, 60, 120 and 180 min after oral load with 75 g anhydrous glucose powder, respectively; (5. 1 ± 0. 7 ), ( 8. 8 ± 1. 7 ), (9. 0 ±3.0), (7.3±2.2) and (5.1 ±1.1) mmol/L (15 volunteers) or (5.3 ±0.6), (8.8 ±1.9), (8.5 ±2. 4), (6. 6 ± 1.4) and (4. 8 ± 1.6) mmol/L (22 volunteers) at 0, 30, 60, 120 and 180 min after oral load with seven or 7.5 ampoules of 50％ glucose injection, respectively, with no statistically significant difference between varied methods.Normal serum level of insulin was found in 38 of 60 volunteers, with their logarithmic transformation of serum insulin levels of 1.5 ± 0. 3, 3.9 ± 0. 3, 3.7 ± 0. 4, 3.2 ± 0. 6 and 2.2 ±0. 8 at 0, 30, 60, 120 and 180 min, respectively after glucose load in standard OGTT, and 20 of 38 volunteers with normal serum insulin of 1.7 ± 0. 4, 3.9 ± 0.4, 3.4 ± 0. 7, 3.3 ± 0. 8 and 2. 4 ± 0. 7 at 0,30, 60, 120 and 180 min after oral load with seven ampoules of 50％ glucose injection, respectively, or 18 of 38 with normal serum insulin of 1.7 ± 0. 4, 3.9 ± 0. 4, 3.8 ± 0. 5, 3. 3 ± 0. 7 and 2. 3 ± 1.0 at 0, 30,60, 120 and 180 min after oral load with 7. 5 ampoules of 50％ glucose injection, respectively, with no statistically significant difference between varied methods. Twenty-two cases of high serum level of insulin were found from 60 volunteers with standard OGTT, with their logarithmic transformation of serum insulin of 2.2±0.6, 4.7 ±0.5, 4.9±0.7, 4.2 t 1.0 and 2. 8 ±0.9 at0, 30, 60, 120 and 180 min after oral load with 75 g anhydrous glucose powder, respectively; 10 of 22 volunteers were found with high serum insulin level of its logarithmic transformation of 2. 4 ± 0. 6, 4. 7 ± 0. 5, 4. 7 ± 0. 3, 4. 1 ± 0. 8 and 2. 8 ± 1.1 at 0,30, 60, 120 and 180 min after oral load with seven ampoules of 50％ glucose injection, respectively ; and 12 of 22 volunteers were found with high serum insulin level of its logarithmic transformation of 1.9 ± 0. 5,4. 5 ± 0. 6, 4. 6 ± 0. 6, 3. 7 ± 1.0 and 2. 4 ± 0. 9 at 0, 30, 60, 120 and 180 min after oral load with 7. 5ampoules of 50％ glucose injection, respectively; with no significant difference between varied methods.There also was no statistically significant difference in occurrence of adverse effects between these three OGTT methods. ConclusionsEither seven or 7. 5 ampoules of 50％ glucose injection can substitute 75 g anhydrous glucose powder in OGTT, with similar test results and safety.

Objective To supply anatomic foundation for TRAM and DIEP flaps breast reconstruction with both sensory innervation and rectus abdominis muscle function preserved. Methods Dissections of T8～T12 nerves in anterior abdominal wall were performed on 9 embalmed female cadavers (18 sides). By reconstructing DIEP flaps in 15 cases, the natures of the intercostal nerves and the relations with deep inferior epigastric perforators were investigated. Results Most intercostal nerves pierced the rectus abdominis muscle in the lateral one third. There were interconnections between muscular branches of segmental nerves. Sensory branches traveling with vascular perforators were divided into medial branch and lateral branch. The mean length of pure sensory nerve was (27.6?12.2)mm. Conclusion It is possible to preserve sensory innervation by coapting lower intercostal nerves to the 4th thoracic nerve in TRAM and DIEP breast reconstruction. Sparing lateral one third of rectus abdominis muscle does not cause denervated atrophy in the rectus abdominis muscle after TRAM breast reconstruction.

Objective To study the correlation of p53,ki-67 and bcl-2 expression in breast cancer with the clinical response to neoadjuvant chemotherapy,and search for biological markers to guide therapy and predict prognosis.Methods The expression of the p53,ki67 and bcl-2 was examined by immunohistochemical(assay) in samples of breast cancer tissue which taken from 118 cases of operable breast cancer patients.The relationship between p53,ki-67 and bcl-2 and the effect of neoadjuvant chemotherapy was analyzed by(chi-square) test.Results The overall response rate to neoadjuvant chemotherapy was 68.6%.Patients with expression of ki-67 were more likely to respond to treatment.The effect of neoadjuvant chemotherapy had a significantly negative correlation with p53 expression.There was no significant difference between the response to neoadjuvant chemotherapy and bcl-2 expression.Expression of p53 protein was correlated with low(expression) of bet-2.Expression of p53 and ki-67 protein was significantly related.Conclusions The(results) indicated that p53 and Ki-67 expressions are strong prognostic molecular markers that can be a guide for neoadjuvant chemotherapy and prediction of prognosis for patients with breast cancer.

Objective To evaluate the reliability and sensitivity of detection of thyroid carcinoma lymph node micrometastasis with MUC1.Methods In 488 cases of thyroid disease,1% methylene blue 1.0-2.0mL was injected into the thyroid nodule or surrounding thyroid tissue 24hours before operation.The dye-stained lymph nodes were detected at operation and RT-PCR method was used to determine MCUI in the clinically blue-stained lymph nodes.Results The detection rate of the blue-stained lymph nodes in thyroid carcinoma was 93%,and in benign lesions was 0%.In the research group,80 cases were found to have blue-stained lymph nodes,the detection rate was 95% and was higher than the pathologr diagnostic rate which was 86%(P

Objective:To observe the effect of Xingnaojing injection in the treatment of severe traumatic brain injury .Methods:From August 2013 to December 2014 , 110 patients with severe traumatic brain injury were randomly divided into the control group and the research group with 55 ones in each .The control group was treated with the conventional treatment , including removal of intracrani-al hematoma, bone disc decompression according to surgical indications , anti-infection, dehydration intracranial pressure reduction and nutritional support treatment .The research group was treated with Xingnaojing injection additionally .After the one-week, two-week and 4-week treatment , the GCS score of the two groups were compared .The rate of lung infection and stress ulcer in 4 weeks were com-pared.The clinical outcome of the two groups after the 4-week treatment was also evaluated .Results:The GCS score of the research group was higher than that of the control group after the 2-and 4-week treatment (P<0.05).There was no significant difference in the incidence of stress ulcer between the two groups , while the incidence of pulmonary infection in the research group was lower than that in the control group (P<0.05).The total effective rate of clinical prognosis in the research group (90.9%) was significantly higher than that in the control group (72.7%, P<0.05).Conclusion:Xingnaojing injection combined with the conventional treatment for the patients with severe traumatic brain injury can decrease the clinical complications and improve the clinical symptoms of patients , which is worthy of clinical promotion .

Objective To investigate epidemic status and explore the influential factors of pulmonary tuberculosis (PTB) in the servicemen of armed forces and to provide reference basis for control and prevention. Methods According to the questionnaire, the test of PPD (purified protein derivative), X-ray checking and sputum bacteria culture method had been used. A 1∶1 matched case-control study was carried out in the servicemen with 162 cases and 162 controls. Odds ratios (ORs) were calculated for the relevant factors. Results The prevalence rate of activated tuberculosis, the bacteriological positive prevalence rate, the smear positive prevalence rate and PPD positive reaction rate respectively were 176.66/105, 33.12/105, 22.08/105 and 34.31%. The prevalence rate of officers was obviously higher than that of soldiers, and the army resident in the countryside was higher than that in the city. The prevalence rate of high training intensity was higher than that of low training intensity. With the increasing of the age and time duration in army, the prevalence rate of pulmonary tuberculosis and the positive rate of PPD reaction presented a ascending tendency. Single factor conditional logistic regression analysis showed that the occurrence of PTB was closely associated with training intensity, body mass index, the number of scar, housing condition, the contact history of infection source, area of enrollment and character, their ORs value were 5.12, 0.37, 0.42, 0.51, 1.82, 0.57 and 1.63 respectively (P

Objective: To construct and identify a lentiviral vector harboring RNAi sequence targeting the human high mobility group A1(HMGA1) gene.Methods: The effective sequence of siRNA targeting the HMGA1 gene confirmed in our previous study,the complementary DNA containing both sense and antisense Oligo DNA of the targeting sequence was designed,synthesized and cloned into the pGCL-GFP vector diced by the restriction enzyme of HpaⅠ and XhoⅠ,which contained the U6 promoter and green fluorescent protein(GFP).The resulting lentiviral vector containing HMGA1 shRNA was named LV-sh HMGA1 and confirmed by PCR and DNA sequencing.A total of 293T cells were cotransfected with LV-sh HMGA1,pHelper 1.0 and pHelper 2.0.All the virus stocks were produced by Lipofectamine2000-mediated transfection.The titer of the virus was tested according to the expression level of GFP.Results: PCR analysis and DNA sequencing demonstrated that the RNAi sequence targeting the human HMGA1 gene was successfully inserted into the lentiviral vector.The titer of the recombinant lentiviral vector was 5?107 TU/ml.Conclusion: The successful construction of the lentiviral vector of HMGA1 has prepared the ground for further studies on the functions of the HMGA1 gene with the RNAi technique.

Objective To explore the correlation of helicobacter pylori(HP) infection in liver cirrhosis(LC) of different syndrome patterns with blood ammonia level.Methods Two hundred and seventeen LC patients were divided into Hp-positive group and Hp-negative group.The patients were classified into 6 syndrome patterns: liver-Qi stagnation,internal accumulation of water-dampness,stagnation of damp-heat,deficiency of liver and kidney yin,deficiency of spleen and kidney yang,and blood-stasis blocking collaterals.Parameters of blood ammonia level,hepatic function and blood coagulation function were observed in LC patients with different syndrome patterns.Results The Hp infection rate in 217 LC patients was 40.1%,the difference being insignificant in the patients with different syndrome patterns and hepatic function grading.The percentage was in a decreasing sequence in syndrome patterns of stagnation of damp-heat,internal accumulation of water-dampness,blood-stasis blocking collaterals,deficiency of liver and kidney yin,liver-Qi stagnation,deficiency of spleen and kidney yang.The difference of blood ammonia level was significant between Hp-positive group and Hp-negative group(P0.05).Conclusion Hp infection is an important factor of inducing the increase of blood ammonia level in LC.Blood ammonia level increases in LC patients after Hp infection,especially in patients with blood-stasis blocking collaterals.The hepatic function grading is corrected with blood ammonia level,the worse the hepatic function,the higher the blood ammonia level.
