Objective:To explore the correlation between serum protein factor level and clinical symptoms and cognitive impairment in patients with schizophrenia and to predict the degree of cognitive impairment, so as to provide an auxiliary method for clinical evaluation of cognitive impairment severity and prognosis of schizophrenia. Methods:From September 2017 to April 2019, 71 schizophrenic patients diagnosed in the First Affiliated Hospital of Kunming Medical University were selected as the patient group, and 65 healthy volunteers from the physical examination center of the same hospital were selected as the control group. The concentrations of tumor necrosis factor α(TNF-α), brain-derived neurotrophic factor(BDNF) and calcium-binding protein β(S100β) in peripheral blood were detected by ELISA method.Cognitive function was evaluated by MATRICS consensus cognitive battery(MCCB) cognitive assessment. The clinical symptoms of patients were evaluated by positive and negative syndrome scale(PANSS) scale.SPSS 20.0 software was used for statistical analysis, independent sample t-test was used for comparison between groups, and Pearson correlation analysis was used for the relationship between serum protein factor level and cognitive function and clinical symptoms. In order to objectively predict, evaluate and verify the severity of cognitive impairment in schizophrenia, Bayes discriminant function was established with serum protein factor concentration and PANSS total score as independent variables and the defect degree of cognitive factors in MCCB as dependent variables. Results:The serum TNF-α((63.2±25.2)pg/L vs (31.4±14.3)pg/L) and S100β((68.0±26.4)pg/L vs (47.3±20.2)pg/L) concentrations in the patient group were higher than those in the control group. The concentration of serum BDNF in the patient group was lower than that in the control group ((2 517.8±1 140.2)pg/L vs (5 202.2±447.2)pg/L), and the difference was statistically significant ( P=0.000). In the retrospective test of cognitive impairment severity in Bayes discriminant function model, the correct discrimination rates of four cognitive factors were speed of processing(SoP) 69.0%, Verbal learning(VeL) 63.4%, reasoning and problem solving(RPS) 76.1% and visual learning(ViL) 73.2%.The correct discrimination rates of cross-examination were SoP 66.2%, VeL 60.6%, RPS 73.2%, ViL 66.2. Conclusion:The levels of serum protein factors TNF-α, BDNF and S100β and clinical symptom scores of schizophrenia have different degrees of correlation with the severity score of cognitive impairment.Bayes discriminant function model has higher correct discrimination rate for the severity of cognitive impairment of schizophrenia. It is found that the levels of schizophrenia-related protein factors and clinical symptom scores may have predictive effect on the severity of cognitive impairment, providing a more objective basis for the clinical efficacy evaluation of schizophrenia patients.

The aim of this study was to fabricate biomatrix/polymer hybrid scaffolds using an electrospinning technique. Then tissue engineered heart valves were engineered by seeding mesenchymal stromal cells (MSCs) onto the scaffolds. The effects of the hybrid scaffolds on the proliferation of seed cells, formation of extracellular matrix and mechanical properties of tissue engineered heart valves were investigated. MSCs were obtained from rats. Porcine aortic heart valves were decellularized, coated with poly(3-hydroxybutyrate-co-4-hydroxybutyrate) using an electrospinning technique, and reseeded and cultured over a time period of 14 days. In control group, the decellularized valve scaffolds were reseeded and cultured over an equivalent time period. Specimens of each group were examined histologically (hematoxylin-eosin [HE] staining, immunohistostaining, and scanning electron microscopy), biochemically (DNA and 4-hydroxyproline) and mechanically. The results showed that recellularization was comparable to the specimens of hybrid scaffolds and controls. The specimens of hybrid scaffolds and controls revealed comparable amounts of cell mass and 4-hydroxyproline (P>0.05). However, the specimens of hybrid scaffolds showed a significant increase in mechanical strength, compared to the controls (P<0.05). This study demonstrated the superiority of the hybrid scaffolds to increase the mechanical strength of tissue engineered heart valves. And compared to the decellularized valve scaffolds, the hybrid scaffolds showed similar effects on the proliferation of MSCs and formation of extracellular matrix. It was believed that the hybrid scaffolds could be used for the construction of tissue engineered heart valves.

Objective To analyze the immunological characteristics of Trichinella spiralis secretory antigen P53 and to evaluate its value in diagnosis of trichinellosis. Methods An open read frame of secretory antigen P53 was cloned from Trichinella spiralis by reverse transcriptasepolymerase chain reaction (RT-PCR) and then sequenced. Bioinformatics analysis was performed to search for its homologues in other helminths and predict its potential linear B cell epitopes and T cell epitopes. The sequence coding mature peptide was inserted into prokaryotic expression vector pET28a(+) and the purified recombinant product was identified by Western blot using serum samples of patients infected with Trichinella spiralis or other helminth. Results Bioinformaties analysis results showed that there was no P53 homologue in other helminths, which indicated that there were many linear B cell epitopes and T cell epitopes in TsP53. The recombinant P53 antigen only reacted with the serum samples of patients infected with Trichinella spiralis without any cross-reaction with the serum of patients infected with other helminths. Conclusion P53 has strong immunogenicity and immunoreactivity, which may be a promising candidate for developing Trichinella spiralis specific diagnostic method.

This study was aimed to investigate the effects of Shen-Y uan Y i-Qi Huo-Xue (SYYQHX) capsule com-bined with early percutaneous coronary intervention (PCI) on near-term quality of life (QOL) in unstable angina (UA) patients. Seventy-eight patients diagnosed with UA were selected and randomly divided into the treatment group and control group, with 39 patients in each group. Early PCI was undergone after coronary angiography. Before PCI, the control group was given routine western medication. The treatment group was given routine western medication plus SYYQHX capsule, three pills once, three times daily. The treatment lasted for 30 days. The QOL scores were evalu-ated among patients from both groups before and after treatment in order to determine the efficacy on angina, electro-cardiogram (ECG) and traditional Chinese medicine (TCM) main symptom. The results showed that compared to pre-treatment, the scores of physical limitation (PL), angina stability (AS), angina frequency (AF), and treatment satisfac-tion ( TS ) were significantly increased after treatment ( P < 0 . 05 ) . Compared with the control group , after treat-ment with SYYQHX capsule, the AF, duration time and symptoms of palpitation, fatigue and shortness of breath were obviously improved (P< 0.05). The scores of AS, AF and TS in the treatment group were significantly increased (P < 0.05). It was concluded that SYYQHX capsule combined with early PCI can improve the near-term QOL and TCM main symptoms among UA patients.
