[ ABSTRACT] AIM: To discuss the relevance between the pathogenesis of diabetic gastroparesis and the large-conductance calcium-activated potassium channels ( BKCa ) in gastric smooth muscle cells.METHODS:The SD rats were randomly divided into control group and model group. The gastric smooth muscle cells of the SD rats were enzymatically iso-lated in a low calcium solution containing papain. The current was recorded by patch clamp single channel recording tech-nique.The expression of KCNMA and KCNMB1 were observed by the method of immunohistochemistry.RESULTS:The value of BKCa single channel conductance was (220.10 ±10.90) pS;the channels had distinct voltage dependent and cal-cium dependent characteristics. In outside-out patch (Vm =+30 mV), the activation of BKCa was blocked by 200 nmol/L IbTX completely.Compared with control group, the open probability and amplitude of current in model group significant-ly increased, while the mean open time and mean close time significantly decreased.Compared with control group, the ex-pression of KCNMB1 in model group was significantly increased.CONCLUSION: Up-regulation of β1-subunit and in-crease in BKCa functional activities may be associated with diabetes gastroparesis in rats.

Objective To investigate early diagnosis, treatment and prev ention of tibia fatigue fracture. Methods From August 1988 to July 1999, 66 case s of tibia fatigue fracture were treated with exposure to the irradiation of Zho u Lin Spectrum Treatment Device, plaster splintage, according to their different clinic characteristics and results of X-gram.Results The 66 cases were followe d up for an average of 6 months. The result was excellent in 50 cases, good in 1 6, with the excellent and good rates of 100%. Conclusions As a result of a chro nic course of accumulative injury, the tibia fatigue fracture develops from quan titative changes to qualitative changes. It is essential for medical workers to have a good knowledge of the classification and features of the condition. It is also important for the army to make scientific and reasonable training plans fo r the soldiers.
3.Dry solid staging fermentation.
Chinese Journal of Biotechnology 2009;25(2):200-206
Very high gravity (VHG) ethanolic fermentation is a new perspective technology for the fuel ethanol production. Compared with traditional hot cook process in most ethanol plants, uncooked process using milled grain slurry in combination with granular starch hydrolyzing enzymes makes high gravity fermentation much easier to control. In this study, dry solids staging technique was first time reported in uncooked process for fermentation ethanol. We further studied the difference between the new process and the batch fermentation, including different initial fermentation concentrations and different starting times. The results showed that at the same dry solid concentration of 30% and the same enzyme dose at 0.22% (W/W), the final ethanol output of new process was increased to 18.50% (V/V) from 17.06% (V/V) of the conventional process. This study demonstrated the new application of uncooked fermentation technology.

Objective:To explore the current situation of unmet needs and family environment among gynecology malignant tumor patients and to explore the correlations between them. Methods:From March 2018 to March 2019, this study selected 222 gynecology malignant tumor patients at three Class Ⅲ Grade A hospitals in Hainan Province with the cross-sectional investigation as subjects by convenience sampling. All patients were investigated with the General Information Scale, the Chinese version of Short-Form Survivor Unmet Needs Survey (SF-SUNS) and the Chinese version of Family Environment Scale (FES-CV) . A total of 222 questionnaires were sent out and 216 valid questionnaires were collected with 97.30% for the recovery rate. Results:Among 216 gynecology malignant tumor patients, the total scores of the Chinese version of SF-SUNS and the FES-CV were (73.79±13.82) and (49.12±8.14) respectively.Single factor analysis showed that there were statistical differences in the scores of the Chinese version of SF-SUNS among gynecology malignant tumor patients with different ages, family monthly incomes, education levels and clinical stages of International Federation of Gynecology and Obstetrics. Correlation analysis showed that the total score of FES-CV and scores of the cohesion, expressiveness, independence, knowledge, entertaining and organization dimension were negatively correlated with the dimensions of the Chinese version of SF-SUNS with statistical differences ( P<0.05) . The conflict dimension was positively correlated with the Chinese version of SF-SUNS with statistical differences ( P<0.05) . Hierarchical regression analysis showed that the main influencing factor of unmet needs of gynecology malignant tumor patients was family environment after controlling demographic variables. Conclusions:Unmet needs of gynecology malignant tumor patients are in the intermediate level. Family environment has close correlation with the unmet needs of gynecology malignant tumor patients. Medical staff and their family members should meet needs of patients by improving the family environment so as to improve their quality of life.
5.Effect of temperature on the no cook, very high gravity ethanol fermentation process.
Chinese Journal of Biotechnology 2010;26(3):330-334
The effect of temperature on a very high gravity ethanol fermentation using no cook process was investigated. We found that a gradient temperature control strategy could improve the fermentation efficiency significantly, With the assistance of a new raw starch hydrolyzing enzyme and a gradient temperature control strategy, the ethanol concentration could reach up to 20% (V/V) within 90 h using commercially available dry yeast, when sorghum was used as the raw material and the dry substrate concentration was controlled at 35%.

OBJECTIVE The N-methyl-D-aspartate(NMDA)receptor has been shown to be strongly associ-ated with rapid antidepressant effects.GW043 is a com-pound with a novel structure that we designed and syn-thesized to act on the NMDA receptor(NMDAR).METH-ODS In this study,we first confirmed the target of GW043 using a receptor binding assay. We observed the effect of GW043 on NMDAR currents in vivo and in vitro assays using a membrane clamp technique with a view to characterizing the function of GW043.We investi-gated the antidepressant effect of GW043 in rodent behavioral models such as FST,TST and CUMS.Fur-thermore,we explored the mechanism of GW043 onset using Western blotting,BrdU staining,Golgi staining and electrophysiological techniques.RESULTS GW043 interacts with high affinity only at the NMDAR.Electro-physiological studies have indicated that GW043 is a par-tial agonist of NMDAR.Meanwhile, behavioral experi-ments were conducted to confirm the antidepressant effect of GW043 in rodents. The mechanism study found that GW043 regulate synaptic plasticity through LTP and BDNF/mTOR pathways and increase the number of new-born neurons to cause antidepressant effects.GW043,a partial agonist of NMDAR,reversed depression-like behav-ior in rats by modulating synaptic plasticity, suggesting an antidepressant effect.CONCLUSION The results suggest that GW043 is a partial agonist of NMDA recep-tors and has significant antidepressant effects.

Squamosa promoter binding protein-like (SPL) family is a group of important transcription factors involved in the regulation of plant growth and development and the response to environmental stress, but there are few studies in perennial fruit trees such as citrus. In this study, Ziyang Xiangcheng (Citrus junos Sib.ex Tanaka), an important rootstock of Citrus, was used as the material for analysis. Based on plantTFDB transcription factor database and sweet orange genome database, 15 SPL family members were genome-widely identified and cloned from Ziyang Xiangcheng, and named CjSPL1-CjSPL15. Sequence analysis showed that the open reading frame (ORF) length of CjSPLs ranged from 393 bp to 2 865 bp, encoding 130-954 amino acids. Phylogenetic tree divided 15 CjSPLs into 9 subfamilies. Gene structure and conserved domain analysis predicted 20 different conserved motifs and SBP basic domains. Analysis of cis-acting promoter elements predicted 20 different promoter elements, including those related to plant growth and development, abiotic stress and secondary metabolites. The expression patterns of CjSPLs under drought, salt and low temperature stresses were analyzed by real-time fluorescence quantitative PCR (qRT-PCR), and many CjSPLs were significantly up-regulated after stress treatment. This study provides a reference for further study on the function of SPL family transcription factors in citrus and other fruit trees.
