1.The Role of Recombinant p53 Adenovirus Vector in the Growth of Lung Adenocarcinoma Cell Line GLC-82

Wild-type p53 has been shown to be involved in several aspects of cell growth control. In order to study the role of wild-type p53 in the growth of lung adenocarcinoma cell line GLC-82, recombinant p53 adenovirus vector and the control LacZ gene were constructed. Infection conditions were detected by biochemistry staining for LacZ s expression product ?-gal, immunohistochemical analysis for p53 protein expression, and PCR technique for the infected cells. The cell growth rate and cell cycle were analysed with flow cytometry. The results showed that the constructed recombinant adenovirus vector could infect cells with high level expression of (?-gal and p53 protein and p53 cDNA could be found in the infected cells. The wild-type p53 could inhibit GLC-82 cell proliferation and cause cellular death. These results indicated that wild-type p53 gene mediated by adenovirus vector might be used in the treatment of lung adenocarcinoma.
2.Inhibition of Foreign Retinoblastoma Gene Mediated by Recombinant Adenovirus Vector on Growth of Bladder Carcinoma Cells

In order to study the bladder carcinoma cell growth suppression by introduction of foreign retinoblastoma (Rb) gene and explore a gene therapy approach for bladder cancer, a replication-deficient adenovirus vector encoding a wild-type Rb, AdCMVRb, was constructed and transfected into the cultured human bladder carcinoma cell line EJ. The efficiency of gene transfection and expression was detected by immunochemical staining, Western blotting and polymerase chain reaction. The role of Rb in suppressing EJ growth was observed by cell-counting, [3H]thymidine incorporation and flow cytometry. The results showed that wild-type Rb gene could be transfected effectively into cultured EJ with Ad-CMVRb and could arrest the cells at GO/Gl phases of the cell cycle, leading to inhibition of DNA synthesis. The results demonstrated the potential of adenovirus-mediated Rb gene therapy for bladder cancer.
3.Detection of HPV DNA by PCR in Lesions of Condylomate Acuminata Treated with CO_2 Laser and Microwave In Vitro

Twelve specimens of genital warts were excised from 12 patients and divided into three parts. One part was untreated, the second and the third part were treated with CO_2 laser and microwave respectively. HPV DNA was amplified and detected in 100% of untreated specimens (HPV16 and HPV11 in six patients each), in 83% and 50% of specimens treated with CO_2 laser and microwave respectively. To explore the efficacy of using gonadotropin-releasing hormone agonist (GnRH-a) with different cycles after conservative laparoscopic surgery in the treatment of endometriosis.Research was conducted on 108 patients with endometriosis Ⅲ or Ⅳ diagnosed by conservative laparoscopy from January 2010 to June 2011.Among them,57 patients (group A) were managed with GnRH-a postoperatively for 3 months and the others (group B) 6 months.Clinical efficacy,relapse rate and pregnancy rate after withdrawal were compared between two groups.The clinical efficacy rate of groups A and B were 73.7% and 90.2% respectively.And there were significant statistical differences between them.However,the relapse rate of groups A and B were 14.0% and 5.9% and the pregnancy rate of two groups 45.0% and 4/12 respectively.And there were no statistical differences.A 6-month regimen of GnRH-a can reduce and delay the recurrence of endometriosis and improve the clinical efficacy.A short-term dosing of GnRH-a is indicated for those patients with fertility requirements.
5.Pathological characteristics of lupus-like renal damages induced by exogenous double stranded DNA

Objective To investigate the pathological characteristics of lupus-like renal damages induced by double stranded DNA (dsDNA) derived from Trypanosoma Equiperdum (TE). Methods The TEs were propagated in normal rats and isolated from fresh rat blood by DEAE cellulose-chromatography. Their kinetoplast dsDNA (kDNA) was purified with Gibson's method. The emulsive mixture of kDNA and incomplete Freund's adjuvant (IFA) was injected into normal BALB/c mice subcutaneously. Eight weeks Later some parameters were examined, including sera titers of ANA and anti-dsDNA antibodies, 24h urine protein concentration, ESR, BUN, Scr and renal histological active index (AI). The pathological characteristics of renal tissues were observed under optical and electron microscopes, and then compared with that of BXSB mice and lupfis nephritis (LN) patients with positive anti-dsDNA antibodies in the sera. Results The results of all immunological parameters of TE kDNA-immunized mice corresponded with that of LN. Their renal damages mainly represented nephropathy syndrome. The pathological characteristics of these mice were similar to that of BXSB mice and LN patients, but Ⅱ (mesangial proliferative) and Ⅳ (diffuse proliferative) subtypes were more common in the former. Conclusion The pathological characteristics of renal damages in the mice immunized with TE dsDNA are similar to that of human LN induced by anti-dsDNA antibodies. This mice model could be used as a tool for invostigating the pathogenesis of LN.
6.Sulfentanil combined with bupivacaine for subarachnoid block on cesarean section patients

Objective To observe the anaesthetic effect of sulfentanil combined with bupivacaine for subarachnoid block on cesarean section patients and the influence on the patient and fetus.Methods Eighty patients who were scheduled for elective cesarean section under subarachnoid block,were distributed into two groups:bupivacaine with no sulfenlanil(group B)and bupivacaine with sulfentanil(group BS).Detected SBP,DBP,MAP,HR,SpO2 before and after anesthesia.The time of drug onset,motor recovery and pain recovery were recorded,the effect of muscle relaxation was evaluated,and the drag reaction,neonatal Apgar score and anesthesia complication were recorded.Results There was no significant difference in the time of drug onset and motor recovery,muscle relaxation,neonatal Apgar score between the two groups.MAP was lower after 3-10 min of anesthesia than before anesthesia,but there was no significant difference after 30-120 min of anesthesia in the two groups.The rate of drag reaction was 27.5% in group B,but no case in group BS.The time of pain recovery was longer in group BS than that in group B(P<0.01).The rate of temporary itch of skin after operation was 17.5% in group BS.Conclusion The addition of sulfentanil to hyperbaric bupivacaine provides adequate anesthesia for cesarean section and good postoperative analgesia.

This article summarized perioperative nursing experience of 6 patients underwent percutaneous microwave or radiofrequency ablation liver partition and portal vein embolization for planned hepatectomy(PALPP).The key points of nursing included:psychological counseling applied throughout the perioperative treatment;personalized preoperative biliary drainage;nursing intervention targeting at Enhanced Recovery After Surgery(ERAS);complication-directed prevention and nursing after microwave or radiofrequency ablation,portal vein embolization,and radical hepatectomy.All 6 patients were recovered and discharged successfully.
8.Sedative and Anti-stress Action of Shenqiwuweizi Capsule

OBJECTIVE: To study the sedative and antistress action of Shenqiwuweizi capsule. METHODS: The sedative and hypnotic experiments were made to observe the action of Shenqiwuweizi capsule on the mice's sleep induced by pentobarbital sodium at subthreshold dosage or hypnotic dosage. The swimming and hypoxia tolerance experiment were performed to observe the effects of Shenqiwuweizi capsule on the swimming time and hypoxia tolerance time in mice. RESULTS: Shenqiwuweizi capsule could obviously prolong the sleeping time of mice induced by pentobarbital sodium, increase the number of sleeping animals caused by pentobarbital sodium at subthreshold dosage, and significantly prolong the time of swimming and hypoxia tolerance in mice. CONCLUSION: Shenqiwuweizi capsule had sedative and anti-stress action.
9.Correlations between cell proliferation and recurrence of benign meningiomas

Objective:To study the relationship between the recurrence of benign meningiomas and cell proliferation, and to find some parameters that can predict the recurrence of benign meningiomas. Methods:49 samples from 15 recurrent benign meningiomas and 22 non recurrent benign meningimas were immunohistochemically stained with PCNA antibody and Ki 67 antibody.The ratios of positive tumor cells were calculated,then PCNA LI and Ki 67 LI of different groups were compared. Results:PCNA LI or Ki 67 LI in recurrent benign meningiomas was significantly higher than non recurent benign meningiomas individually( P 0.05). Conclusion:The proliferative potential of recurrent benign meningiomas was higher than that of non recurrent benign meningiomas.When the PCNA LI was greater than 2.0%,the recurrent risk was high.The proliferative potential did not increase when benign meningiomas recurred.
10.Comparative study on the effects of automatic and semi-automatic hemodialyzer reuse

Objective To explore the influence of automatic and semi-automatic hemodialyzer reuse method on hemodialyzer reuse effect. Methods 1728 dialyzers were randomly divided into automatichemodialyzer reuse group and semi- automatic hemodialyzer reuse group with 864 dialyzers in each group. Thetime of douching and testing, the cost of sterilization,the frequency of the pyrphgen reaction,the broken dialyzer membrane and re-examined dialyzer between the two groups were measured. Results The time of douching dialyzer, testing of total cell volume and pressure in the semi- automatic hemodialyzer reuse group was (26.443±3.237), (2.172±0.128) and (2.157±0.090) minutes respectively,while the automatic hemodialyzer reuse group was (5.793±0.193), (1.257±0.118) and (1.110±0.076) minutes respectively. The frequency of re-examined dialyzer in testing total cell volume and pressure was 499(57.755%), 243(28.125%) respectively. At the same time, all dialyzer in semi-automatic hemodialyzer reuse group could be examined successfully at a time. The cost of sterilization in automatic henmdialyzer reuse group was (9.330±0.138)yuan. No pyrogen reaction and broken dialyzer membrane happened. Conclusions The semi-automatic bemodialyzer reuse group can retrench cost during perfusion,but consumes long douching time, lacks matching detection equipment, difficult to detect, and is not easy to read data and has high re- examination rate. while in the automatic hemodialyzer group, it is convenient of douching and detection, but the cost of sterilization and equipment is high, and clinical demand can be fulfilled only when the dialysis center can allocate reasonable number of the machines.
