1.β-arrestin1 promotes leukemia cell K562 migration and invasion by activating STAT3 signaling pathway

Objective:To investigate the function of β-arrestin1 and its related mechanisms in migration and invasion capacity of leukemia cell K562. Methods:β-arrestin1-siRNA or negative siRNA was transfected into K562 cells of experiment group or control group. The migration and invasion capacity of K562 cells was detected by transwell tests;and the protein expression of β-arrestin1 and pSTAT3 were detected by Western blot. Results:As compared to control group and negative siRNA group,the migration and invasion capacity of transfected β-arrestin1-siRNA group were attenuated about 43% or 52% ( P<0. 05 );Western blot showed that the expression of phosphorylation of STAT3 was decreased inβ-arrestin1-siRNA group. And the STAT3 inhibitors obviously suppressed the migration and invasion capacity of K562 cells. Conclusion:In leukemia cells K562,β-arrestin1 activates STAT3 signaling pathway to promote the cell migration and invasion.
2.Expression of S100A4 and hypoxia-induced factor 1α protein in type I endometrial carcinoma and their significances

Objective To investigate the expression of S100A4 and hypoxia-induced factor 1α (HIF-1α) protein in type Ⅰ endometrial carcinoma,and to analyze their relationship.Methods The expressions of S100A4 and HIF-1α protein were detected in 65 cases of type Ⅰ endometrial carcinoma and 40 normal endometrial tissues.Results The positive rate of S100A4 in endometrial carcinoma was 47.7 % (31/65),the normal tissues was 0 (0/40),and there was significant difference (x2 =27.069,P =0.001).The positive rate of HIF-lα was significantly higher in endometrial carcinoma [63.1% (41/65)] than that in the normal tissues [7.5 % (3/40)] (x2 =31.417,P =0.001).The expressions of S100A4 and HIF-1α were positively related with FIGO staging,pathological grades,myometrial invasion and lymphatic metastasis (all P < 0.05).The expressions of S100A4 and HIF-1α were positively correlated in type Ⅰ endometrial carcinoma (r =0.348,P =0.005).Conclusion The expression levels of S100A4 and HIF-1α are closely related with the oncogenesis and progression of type Ⅰ endometrial carcinoma,and their co-detection may be useful in prognosis assessment.
3.An investigation of human brucellosis infection in Qi County of Shanxi Province in 2010-2012

Objective To analyze the status of human brucellosis infection in Qi County.Methods From 2010 to 2012,by using Rose Bengal plate agglutination test (RBPT) and standard tube agglutination test(SAT),serological detection was conducted in suspected brucellosis cases and risk populations occupational exposed to Brucella,including livestock traders,slaughters,raisers,milkmaids and veterinary in Qi County.The sex distribution,time distribution,occupation and age distribution were analyzed.Brucella were isolated from patients in acute stage by blood culture,and serotypos were determined by traditional identification method.Results From 2010 to 2012,977 people were examined; 273 people were serological positive(27.9%) ; male[31.6%(232/735)] was higher than female[16.9%(41/242),P < 0.05].March to August were the peak seasons in every yeas.Most of the patients were engaged in farming,aged from 21 to 60 years old.Eleven strains of Brucella Melitensis biotype 3 were isolated in 30 cases of blood culture samples.Conclusions The incidence of human brucellosis is increasing year by year in Qi County.Infected people are mostly young men.It is necessary to strengthen health education,increase the awareness of self protection in order to reduce the occurrence of brucellosis.
4.Technical characteristics and clinical feasibility of 2 implant supported immediate loading mandibular implant complete denture by linear occlusion

Objective To investigate the technical characteristics and clinical feasibility of 2 implant supported immediate loading mandibular implant complete denture by linear occlusion.Methods40 cases of edentulous patients in the fifth people's hospital of yuhang district hangzhou from January 2011 to January 2017 were selected, these patients were divided into 2 implants supported immediate loading implant supported mandibular complete overdenture group (immediate loading implant group, n=20) and conventional complete denture repair group (n=20) two groups according to the treatment methods, the planting denture satisfactions, body marginal bone resorption, complications of the two groups were statistically analyzed.ResultsThe planting retention function, masticatory function, language function, comfort scores of the immediate loading implant group were significantly higher,there was significant difference between two groups(P<0.05), the bone resorption was significantly lower,there was significant difference between two groups(P<0.05), the incidence of complications 10% (2/20) was significantly lower than the conventional complete denture repair group 40% (8/20).there was significant difference between two groups(P<0.05).ConclusionThe clinical feasibility of 2 implant supported immediate loading mandibular implant complete denture by linear occlusion is higher than conventional complete denture repair.
5.Status quo of elder abuse and its countermeasures in nursing

The article reviews the current status of elder abuse from the perspective of epidemiology, characteristics of abuse, risk factors and nursing assessment and management, presenting the idea that nurses are playing a major role in the prevention,detection and resolution of elder abuse. It also introduces that nurse can screen out the situation of elder abuse timely by applying the proper assessment tool flexible and respond to the occurrence of elder abuse as well as avoid the elder abuse again by comprehensive application of nursing knowledge and management skills. Some advices about future research and mismanagement in elder abuse being given.
6.The effects of recombinant human resistin on differentiation of orbital preadipocytes of TAO patients in vitro

Background The enhance of differentiation of orbital preadipocyte is a main factor for pathogenesis of thyroid associated ophthalmopathy(TAO).Experiment has proved that orbital preadipocyte can differentiate into mature adipocyte expressing resistin,and resistin is a adipocyte factor with biological activity.Objective The present study is to investigate the effects of recombinant human resistin on differentiation of orbital preadipocytes derived from TAO patients orbital tissues.Methods Orbital adipose tissue was obtained from 10 eyes with TAO during the orbital decompression surgery.Orbital adipose tissue of the normal group was obtained from 5 removed eyes during the surgery due to different reasons.Preadipocytes were isolated and cultured using expand culture method.The 2-5 generations of preadipocytes were identified by oil red O staining under the light microscope.0,10,25,50,100ng/mL of recombinant human resistin were added into medium in the first day after differentiation of cells.The expression of PPARγ mRNA in differentiated adipocytes were detected by reverse trancripaton PCR(RT PCR).Results The differentiation of preadipocyte presented the similar procedure and outcome between the TAO group and normal group.Differentiated adipocyte was stained into the jacinth by oil red O.The expression of PPARγ mRNA in differentiated cells was gradually declined with the increase of concentration of recombinant human resistin (F=241.341,P<0.01).A dose dependent inhibitory effect was detected in the expression of PPARγ mRNA.The weakest expression of PPARγ mRNA in differentiated cells was 100ng/mL resistin group.Conclusion The recombinant human resistin has an inhibitory effect on the differentiation of orbital preadipocytes by reducing the expression of adipose related genes.Resistin is probably a new research approach to treatment of TAO.
7.Situation Changes of DNA Demethylation Analysis of FOXP3 TSDR at the Beginning of Patients with Secondary Pulmonary Tuberculosis

Objective To analyze changes of DNA demethylation analysis of FOXP3 TSDR at the beginning of the secondary pulmonary tuberculosis patientsby utilizing real-time PCR technology.Methods To select 47 patients of secondary pulmona-ry tuberculosis as a research group from June 2014 to May 2015 and 40 healthy donors as a control group.The peripheral blood mononuclear cells (PBMC)of research group and control group were isolated.CD4+CD25+T cells were isolated from PBMC.Genomic DNA was isolated from CD4+CD25+T cells.PCR was performed in a final reaction volume containing dem-ethylation-specific primers.Plasmid standard was generated by PCR products were enzyme digestion,TOPO TA cloning,and recycling and purification.A real-time PCR system was established by quantitatively analyzing the specificity of FOXP3 TS-DR demethylation to treg (regulatory T-cell).Treg numbers of control group at week 0 and research group treated at week 0,week 2,week 4 and week 8 by using real-time PCR assay of the FOXP3 TSDR demethylation.The experimental data was analyzed by using SPSS 1 6.0 software.Results The M.tuberculosis in sputum of research group were positive by smear mi-croscopy,however the results of control group were negative.The treg frequency of control group,2+ group and 3+ group respectively was 1.63%±0.70%,1.96%±0.10% and 0.86%±0.21%,respectively.The difference between the treg fre-quency of control group and that of 2+ group by smear microscopy had not statistical significance,however which of 3+group was opposite.The average treg frequency of research group treated at week 0,2,4 and 8 respectively was at 1.05%, 2.04%,3.44% and 2.79%,range of which respectively was 0.32%~2.03%,0.95%~3.95%,2.35%~4.95% and 1.02%~4.27%,95% confidence interval of which respectively was (0.93%,1.18%),(1.85%,2.24%),(3.27%,3.61%) and (2.60%,2.98%).The treg frequency of difference between control group and research group at week 0 had statistical significance (t=4.669,P<0.05).The treg frequency was influenced by time of therapy,using One-Way ANOVA analysis (F=347.2,P<0.001,df=3,within-subjects Contrasts:F=407.4,P<0.001,df=3).Test of the treatment time and group interaction effect was linear (F=678.2,P<0.001,df=1).Conclusion DNA demethylation analysis of FOXP3 TSDR was high sensitivity and specificity in monitoring changes of treg at the beginning of the secondary pulmonary tuberculosis pa-tients.
8.Expression and clinical significance of estrogen receptor and progesterone receptor in endometrial carcinoma in aged patients

Objective To investigate expression of elderly patients with endometrial cancer of estrogen receptor and progesterone receptor and its clinical significance.Methods 45 elderly patients with endometrial carcinoma from March 2012 to June 2014 were selected as the observation group, over the same period healthy women with normal endometrium as control group, using S-P immunohistochemical method, to detect the expression of receptor and progesterone receptor in two groups.Results In observation group, estrogen and progesterone receptor positive expression rates were 60.00%, 46.67%, compared with control group(93.33%, 86.67%), was significantly lower (P<0.05).Patients with low differentiation of estrogen and progesterone receptor, positive expression rate was 37.50%, 25.00%, was lower than other two groups, but which had no difference.Estrogen, progesterone receptor positive expression rates in progress stage were 35.71%, compared with patients ( 80.65%, 74.19%) in early stage, was significantly lower (P<0.05).The positive expression rate of estrogen receptor in patients with lymph node metastasis was 66.67%, compared with the patients with no metastasis, the difference was not statistically significant.The positive expression rate of progesterone receptor was 76.67%, compared with the non metastasis group (46.67%), and was significantly lower (P<0.05).Conclusion The expression of estrogen and progesterone can reflect the biological behavior and prognosis of endometrial carcinoma in aged patients, and have important significance for clinical diagnosis and treatment.
9.Effects of a novel thiazolidinedione ANY_2 on glycometabolism in HepG_2 cells

AIM:To investigate the effects of a novel thiazolidinedione ANY2 on glycometabolism in HepG2 cells.METHODS:HepG2 cells are exposed to high concentrations of insulin for 24 h to build the model of insulin-resistance(IR-HepG2).The effects of ANY2,including glucose consumption(GC),gluconeogenesis and glucolysis were tested in this model.RESULTS:ANY2 could dose-dependently enhance GC in IR-HepG2 cells at high,medium and low levels of glucose.Compared with Modle Group,ANY2 increased IR-HepG2 cells glycogen synthesis significantly only in high dose group.It inhibited gluconeogenesis from lactic acid,but increased the production of lactic acid and the activity of pyruvate kinase(PK)in glucolysis.CONCLUSION:Compound ANY2 has remarkable effects on improving glycometabolism in IR-HepG2 cells.
10.The correlation between chemotherapeutic efficacy and breast cancer susceptibility gene 1 and class Ⅲ β-tubulin protein expression in non-small cell lung cancer patients

Objective To investigate the predictive value of breast cancer susceptibility gene 1 (BRCA1) and class Ⅲβ-tubulin protein expression in tumor tissue for the efficacy of taxol and cisplatin combined chemotherapy (TP) in stage Ⅲβ/Ⅳ non-small cell lung cancer(NSCLC) patients. Methods A total of 92 stage Ⅲβ/Ⅳ NSCLC patients were recruited with 87 patients evaluated. Bronchoscopy or lung puncture tumor biopsy samples were obtained with BRCA1 and class Ⅲβ-tubulin protein expression examined immunohistochemically before chemotherapy. The patients were randomly assigned to be received 4 to 6 cycles of TP chemotherapy regiments and followed up until death or lost. Response rate (RR) , overall survival (OS) and time to tumor progression (TTP) were assessed. Results Among the 87 evaluated patients, the positive expression rates of BRCA1 and class Ⅲβ-tubulin were 57. 5% (50/87) and 48. 3%(42/87) respectively. There was no significant difference in clinical characteristics among patients with different positive expression rate. According to different expression of BRCA1 and class Ⅲβ-tubulin, the patients were divided into four groups: group A (low expression of both BRCA1 and class 1 p-tubulin) ,group B (high expression of both BRCA1 and class Ⅲβ-tubulin) , group C (high expression of only BRCA1) and group D (high expression of only class Ⅲβ-tubulin). The RR was higher in group A than other three groups (60. 7% , 34. 8% , 9/19 and 6/17 respectively). The OS and TTP were longer in group A than other three groups [OS: (539. 4 ± 17. 6) days, (267. 2 ± 20. 5) days, (325. 6 ± 24. 1) days and (283.7±26.2) days respectively ; TTP: (256. 9 ± 28. 4) days, (143.8±17.6) days, (179. 3 ± 19. 8)days and (152. 6 ±23. 5) days respectively]. There were no significant differences among the other three groups. Conclusions The expression level of BRCA1 and class Ⅲβ-tubulin in tumor tissue is probably a predictor for the efficacy of TP chemotherapy in NSCLC patients. TP chemotherapy is more suitable for the NSCLC patients with lower expression of both BRCA1 and class Ⅲβ-tubulin. Our study may provide a new sight for tailored chemotherapy in NSCLC patients.
