1.Protective effects of polydatin on mouse hepatocyte injury induced by hydrogen peroxide in vitro

AIM To investigate the effects of polydatin on mouse hepatocyte injury induced by hydrogen peroxide(H 2O 2). METHODS Mouse hepatocytes were prepared. SOD activity, MDA content, GSH level, GSH-px activity, glutamic-pyruvic transaminase (ALT), nitric oxide(NO) and nitric oxide synthase(NOS) were determined with different method, respectively. RESULTS After 1 h incubation, the mouse hepatocytes with various concentrations of polydatin (0 05, 0 1, 0 5, 1, 2, 4 mmol?L -1 ), significantly decreased the contents of NO and MDA, inhibited the NOS activity, increased the activities of SOD and GSH-px, suppressed the consumption of GSH, and reduced the elevated ALT level in the supernatant induced by H 2O 2. CONCLUSION Polydatin can protect hepatocytes from oxidative injury induced by H 2O 2.
2.Effects of sinomenine on the psychic dependence on morphine and the brain cyclic AMP level in mice

AIM To explore the effects of sinomenine (Sin) on psychic dependence induced by morphine (Mor) in mice and its relation to the cAMP content of the cerebral cortex. METHODS A biased procedure induced the strong place preference. The effect of Sin (10, 30, 60 mg?kg -1 , ip) on the rewarding property of morphine was observed in a conditioned place preference(CPP) paradigm in mice,at the same time, the change of cAMP content in cerebral cortex was measured by the radioimmunoassay(RIA). RESULTS Sin significantly shortened the time of staying in morphine-pair compartment and lowered the cAMP level of mice. CONCLUSION Sin could attenuate effects of psychical dependence induced by Mor, its mechanism may be related to decrease of cAMP level in the brain, and Sin did not display psychical dependence in the experiment.
3.Relationship between vitamin D and abdominal obesity in non-diabetic population in Taiyuan community

Objective To evaluate the serum 25-hydroxyvitamin D [25 (OH)D3] levels in non-diabetic people in Taiyuan community,and its relationship with obesity and abdominal obesity.Methods This crosssectional study enrolled 417 non-diabetic people in Tainyuan community from July to September in 2011 [225 males and 192 females,mean age (47.6 ±7.8) years].For all the enrolled subjects,we collected data about anthropometric indexes,medical history,blood glucose,lipid profile,25 (OH) D3,fasting insulin (Fins) level,homeostasis model assessement for insulin resistance (HOMA-IR),body fat content (%) measured with dualenergy X-ray absorptiometry (DEXA),and intra-abdominal adipose tissue (IAAT) and abdominal subcutaneous adipose tissue (SCAT) calculated based on magnetic resonance imaging at the 4th to 5th lumbar disc level (IAAT≥80 cm2 was considered abdominal obesity).Correlation analysis was performed to assess the relationships of serum 25 (OH) D3 levels with biochemical and body fat indexes.Results The average serum level of 25 (OH) D3 was 44.5 nmol/L in the 417 people,with vitamin D deficiency in 261 cases (62.6%) and vitamin D insufficiency in 109 cases (26.1%).Of the vitamin D deficient and insufficient people,77.6% had abdominal obesity according to IAAT (IAAT ≥ 80 cm2).According to Pearson analysis result,lg 25 (OH)D3 was negatively correlated with lg Fins,lg HOMA-IR,waist circumference,systolic blood pressure,diastolic blood pressure,total abdominal adipose tissue (TAAT),IAAT (all P <0.05).It was also negatively correlated with waist-to-hip-ratio,body mass index (BMI),and body fat content (%),but without statistical significance (all P >0.05).In stepwise multiple linear regression model,lg HOMA-IR (t =-4.278,P =0.001) and TAAT (t =-5.146,P =0.002) were independently correlated with lg 25 (OH) D3.Conclusions About 90% of all non-diabetic population in Taiyuan community have vitamin D deficiency or insufficiency,of whom 4/5 have abdominal obesity.Serum 25 (OH)D3 level is negatively correlated with abdominal adipose tissue,but not significantly associated with BMI.HOMA-IR and TAAT are independent risk factors for vitamin D deficiency.
4.Vibrio vulnificus: cultivation, identification and antimicrobial susceptibility

Objective To investigate the cultivation, biochemical features and drug susceptibilities of Vibrio vulnificus. Methods Three strains of Vibrio vulnificus were isolated from fatal patients in the Second Hospital of Jiaxing, Zhejiang province. Cultivation, identification and antibacterial susceptibility test were performed. Results Vibrio vulnificus grew on blood agar as dull-gray, opaque colonies with β-hemolysis. The organism presented positive in lactose, cellobiose fermentation and O/129 (10 μg) tests, but lack of inositol and rhamnus. The antibacterial susceptibility tests showed that Vibrio vulnificus strains were sensitive to ciprofloxacin, levofloxacin, imipenem, cefepime, ceftazidime, ceftriaxone, compound sulfamethoxazole and nitrofurantoin, however, resistant to gentamicin, tobramycin, aztreonam and cefazolin. Conclusions Vibrio vulnificus can be isolated from blood, bubbles fluid, and stool. Rapid identification, early diagnosis, and prompt empirical antibacterial therapy are important for reducing the mortality.
5.Comparative study on bacteriostasis among compositions of Gegen Qinlian Decoction against Streptococcus pneumoniae

Object To explore the regularity of recipe composition by observing different bacteriostasis among compositions of Gegen Qinlian Decoction (GQD) against Streptococcus pneumoniae. MethodsTo evaluate the minimal inhibitory concentration (MIC) of 15 decoctions through double fold dilution method. Results Not all of the decoctions are of the same MIC. Conclusion Not all of the 15 decoctions have the same bacteriostasis against S. pneumoniae. Among the four herbs of GQD Coptis chinensis Franch. has the strongest bacteriostasis and the next Scutellaria baicalensis Georgi. While Glycyrrhina wralensis Fisch and Pueraria lobata (Willd.) Ohwi have no bacteriostasis, but they can influence the bacteriostasis of C. chinensis and S. baicalensis when combinating with them.
6.Study on pharmacology of Wunzhonganwei Soft Capsule

AIM: To evaluate the main pharmacological effect of Wunzhonganwei Soft Capsule. METHODS: Analgesic effect was investigated by hot plate test and writhing test, anti-inflammation effect by auricular swelling test in mice and toe swelling test in rat, anti-diarrhea effect by diarrhea test induced by rhubarb, effect on gastric emptying by phenolsulfonphthalein empty test in mice and effect on small intestine propulsive test by charcol powder propulsive rate test. RESULTS: Wunzhonganwei Soft Capsule enhanced thermal stimulation threshold in mice, decreased the occurrence of writhing caused by glacial acetic acid in mice, inhabited xylene-induced auricular swelling in mice and carrageenan-induced toe swelling in rat, decreased the number of loose stools induced by rhubarb in mice., inhabited the function of gastric emptying induced by metoclopramide or in normal mice, antagonized the inhibitory effect of gastric emptying induced by atropine and inhabited small intestine propulsive movement induced by neostigmineor or in normal mice. CONCLUSION: Wunzhonganwei Soft Capsule has the effect of anti-inflammation, analgesia, antidiarrhea and adjusting the function of gastrointestinal movement.

This study aimed to explore an ideal teaching model for medical humanistic education and comprehensively improve the humanistic accomplishment in medical students.
9.Screening and Expression of Recombinant Human Monoclonal Antibody Fab Fragments Specific to Entamoeba histolytica

Objective To prepare recombinant human monoclonal antibody Fab fragments specific to the surface antigen of Entamoeba histolytica. Methods Total RNA was isolated from lymphocytes which were separated from an asymptomatic E. histolytica cyst carrier. The genes of IgG light chain and Fd region of heavy chain were amplified by a reverse transcriptase PCR and ligated with a plasmid vector. After the genes were introduced into Escherichia coli, the clones expressing Fab fragments specific to the surface antigen of E. histolytica were screened and the product was purified. Results Thirty thousand clones were screened and one of them was proved positive to the surface antigen of E. histolytica. Conclusion This study demonstrated that the bacterial system can be used to produce recombinant human monoclonal antibody Fab fragments specific to the surface antigen of E. histolytica and they may be applicable for the future diagnosis and treatment of the infection.
10.Effect of tumor length on clinicopathologied and prognosis of node-negative esophageal carcinoma patients

Background and purpose:At present, the relationship between tumor length and prognosis of esophageal carcinoma patients has been a controversial topic, and there have been few studies describing the effect of tumor length on clinicopathology and prognosis of node-negative esophageal carcinoma patients. The purpose of this study was to investigate the effect of the tumor length on clinicopathology and prognosis of node-negative esophageal carcinoma patients.Methods:The clinicopathological characteristics and survival time of 686 node-negative esophageal carcinoma patients, conifrmed by surgical pathology specimens in the First Afifliated Hospital of Nanjing Medical University from Jan. 2008 to Dec. 2010, were retrospectively analyzed. The optimal cut-off value was determined by decision tree model. Univariate and multivariate methods were used to analyze the prognostic factors of node-negative esophageal carcinoma patients.Results:In decision tree analysis, esophageal tumor length was correlated with an increasing hazard ratio for death with a cut-off value at 3 cm. No signiifcant differences were found in gender, onset age, lesion site and pathological type between 2 groups which were patients with tumor length≤3 cm and tumor length >3 cm (P>0.05). The only 1 difference between 2 groups was T stage (P<0.001), and tumor length had a positive correlation with T stage (r=0.373). The 1-, 3- and 5-year survival rates of patients with tumor length≤3 cm and tumor length >3 cm were 95.7%, 84.4%, 76.1% and 88.3%, 57.8%, 46.5% respectively, and the difference was statistically signiifcant (P<0.001). Tumor length was signiifcantly associated with overall survival in univariate and multivariate analyses.Conclusion:Tumor length has close relationship with T stage in node-negative esophageal carcinoma patients. Tumor length≤3 cm could be considered as T1-2 and >3 cm as T3. Tumor length is an important prognostic factor for esophageal carcinoma patients without lymphatic metastasis.
