OBJECTIVE: To study the immunomodulatory effects of Astragalus polysaccharide (APS) in type 1 diabetic mice. METHODS: A mouse model of type 1 diabetes mellitus was established by intraperitoneal injection of multiple low dose streptozotocin (MLD-STZ). The diabetic mice were intraperitoneally administered 100, 200, 400 mg/kg APS or 1 ml normal saline (NS) every day respectively, then the diabetic mice were sacrificed after 15 or 30 days of treatment. The effect of APS on insulitis was determined via pancreatic histological analysis. Serum insulin autoantibody (IAA) levels were measured by radio-immunoassay (RIA). Proliferation ability of splenocytes to concanavalin A was tested by using [(3)H] thymidine incorporation assay. The levels of cytokine interferon-gamma (IFN-gamma) and interleukin-4 (IL-4) secreted by splenocytes were determined by enzyme linked immunosorbent assay (ELISA) method, and the expression of peroxisome proliferator-activated receptor gamma (PPARgamma) in spleens was characterized using Western-blot analysis. RESULTS: Attenuated insulitis, down-regulation of the serum IAA levels and Th1/Th2 cytokine ratio, decrease of the proliferation ability of splenocytes to concanavalin A, and up-regulation of the PPARgamma levels in spleens showed a significant time- and dose-dependent response to APS treatment as compared with the NS-treated group. CONCLUSION: APS possesses immunotherapeutic effects on mice with type 1 diabetes mellitus through improving the cell- and humoral-mediated immunity.

Objective To study deletion mutations and polymorphism of PINK1 gene exons in patients with Parkinson' s disease (PD) in the littoral of Zhejiang Province, and analyze the association between these changes and the etiology of PD.MethodsAll exons of PINK1 gene in 200 PD patients( 112early-onset PD and 88 late-onset PD) and 220 controls (68 young controls and 152 old controls ) were amplified by polymerase chain reaction (PCR). All the positive PCR products were sequenced, and genotypes were detected by dot blot allele and genotype frequencies of PINK1 were compared by the Chisquare test.ResultsNo deletion mutations of the exons were found in all patients and controls. However,a known heterozygote point mutation G12169A in exon 5 was identified in 2 patients with early-onset PD.And G12164A polymorphism and G12101A polymorphism were located on PINK1 gene of exon 5. There were G/G and G/A genotypes in G12164A polymorphism,no A/A genotype. There were G/A and A/A genotypes in G12101A polymorphism,no G/G genotype. The chain relation polymorphism A/A genotype frequency was significantly higher in the PD group 84/200 (42.0% ) than the control group (52/220,23.6%,x2 =4.034,P =0.045 ).The frequency was also significantly higher in the late-onset PD (40/88,45.5% ) than the old control (32/152,21.1%,x2 =3.951,P =0.047 ).There were no significant differences in alleles frequencies of other groups.ConclusionThe deletion mutation and point mutation are rare in PD patients in littoral of Zhejiang Province.Chain relation polymorphism at G12164A and G12101A in PINK1 gene might be a susceptible factor for PD patients.

Objective To investigate the diagnosis and treatment of Waldenstrom's macrogiobulinemia(WM).Methods The clinical information of 15 patients with WM were analyzed retrospectively. Results 3 cases had achieved complete response(CR).9 cases had achieved partial response(PR).7 cases remained alive, one of them lived beyond 79 months. Conclusion The traditional diagnosis of WM is not enough, and immunophenotype described for WM diagnosis is more important.WM is an incurable disease.Treatment with plasmapheresis and chemotherapy appears highly effective and safe for WM.

DNA computer is a new research field which combines bot h the computer science and molecular biology. DNA computer is proposed to solve a class of hard problems of mathematical complexity by using a set of DNA sequen ces encoding all candidate solutions to the computational problem of interest an d find out the correct answers by serial manipulations of biochemical reactions. DNA computer is exactly a biomolecular computer which stores a vast quantity of information with high density. DNA computer, by means of its huge parallel comp utation and brute force search strategy, can solve the NP complete problems with polynomial time. The recent advances and principle of DNA computer are introduc ed. The future development and the bioinformatical significance of DNA computer are also analyzed and discussed.

Objective To observe the positive rate of serum anti-cardiac troponin Ⅰ autoantibodies (anti-cTnI) in the patients with coronary atherosclerotic heart disease(CHD) and its clinical significance. Methods A total of 200 CHD patients and 200 healthy controls from the Department of Cardiology,Drum Tower Hospital of Nanjing University Medical School during September 2015 and May 2016 were collected in the case-control study.Serum anti-cTnI was detected with an indirect ELISA method.Meanwhile, the general data, chnical biochemical parameters and coronary vascular lesion from the two groups were analyzed. Results The positive rate of anti-cTnI in CHD patients(21/200,10.5%) was significantly higher than that in healthy controls(4/200,2.0%,P < 0.01).According to the detection results of serum anti-cTnI,the participants were divided into three groups, including anti-cTnI positive group with CHD,anti-cTnd negative group with CHD and anti-cTnI positive group in healthy controls. The median age and serum Glu level in the anti-cTnI positive group with CHD were significantly higher than those in the anti-cTnI positive group in healthy controls (P < 0.05),while the levels of serum HDL-C and apoA I were inverse (P < 0.01).The median age and Gensini score in the anti-cTnⅠ positive group with CHD were significantly higher than those in the anti-cTnI negative group with CHD,while the level of serum HDL-C was inverse (P < 0.05).Conclusion Anti-cTnI may be more likely to occur in old CHD patients with severe coronary artery lesions, and the low levels of serum HDL-C and apoA Ⅰ may correlate to the production of anti-cTnⅠ.

Objective To study the correlation between histology and serological index changes in the patients with HBeAg-neg-ative chronic hepatitis B(CHB).Methods 135 cases of HBeAg-negative CHB underwent the liver biopsy,HBV-DNA copy and de-tection of serum alanine aminotransferase(ALT),total bilirubin(TBiL),aspartate aminotransferase(AST),albumin(ALB),pro-thrombin activity(PTA),hyaluronic acid(HA)and laminin(LN).The relationship between the histopathological findings and the se-rological examination results were analyzed. Results There were statistically significant differences between serum HBV-DNA copy levels and the liver tissue inflammation grades(χ2 =23.64,P <0.05);serum ALT,TBiL,AST,ALB,PTA and HA concentrations in the high grade of inflammation were higher than those in the low grade of inflammation, the difference was statistically significant (P <0.05);in which the concentrations of serum ALT,TBiL,AST and HA were positively correlated with the inflammation grades (P <0.05);while ALB and PTA and HA were negatively correlated with the inflammation grades(P <0.05).Conclusion The ser-um level of HBV-DNA copy,TbiL,ALT,AST,ALB and PTA concentrations are related with the inflammatory grade of liver tissue in the patients with HBeAg negative CHB,the serum HA level is related with the fibrosis level of liver tissue. These indicators could provide the reference value for judging the disease condition and prognosis of HBeAg negative CHB patients.
