3.Effect of Skp2 antisense oligodeoxynucleotide on growth and proliferation of colorectal cancer SW 480 cells

Objective To investigate the effect of S-phase kinase-associated protein 2 antisense oligodeoxynucleotide (Skp2 ASODN) on the growth and proliferation of colorectal cancer cells SW480 and its mechanism .Methods The liposome-mediated dif-ferent concentrations of Skp2 ASODN was adopted to transfect SW480 cells ,the final concentrations of 0 .050 ,0 .125 ,0 .250 ,0 .500 , 1 .000 ,2 .000 ,4 .000 μmol/L were respectively set up with Skp2 nonsense oligodeoxynucleotides(NSODN) and blank group as con-trol .Then the inhibited effect of growth and proliferation of colorectal cancer cells were measured by the inverted microscope obser-vationandmethylthiazolyltetrazolium(MTT),thecellcyclewassurveyedbytheflowcytometry(FCM).TheexpressionofmRNA and protein of Skp2 and P27kip1 were inspected by reverse transcription-polymerase chain reaction(RT-PCR) and immunocytochem-istry methods .Results The inverted microscope observed that the SW480 cells had no change in form and grew at a slower speed . When the Skp2 ASODN concentration reached 0 .125 μmol/L ,the inhibition rate was 14 .48% (P<0 .05) .As the Skp2 ASODN concentration increase ,its inhibition action on the growth and proliferation of SW480 cells was enhanced in a dose-dependent man-ner(P<0 .01) .The SW480 cells were increased in G0/G1 phase(P<0 .01) and decreased in S phase(P<0 .05) ,the progression of cell cycle was blocked .RT-PCR and the immunocytochemistry showed that both Skp2 mRNA and protein levels were down-regula-ted(P<0 .05) .Although the P27kip1 mRNA level had no change(P>0 .05) ,but its protein expression was elevates obviously (P<0 .01) .Conclusion Skp2 ASODN can inhibit the growth and proliferation of SW480 cells .Its possible mechanism is that the ubiq-uitin degradation of the Skp2 to P27kip1 is decreased by the gene occlusion effect of Skp2 ASODN ,thus the cell cycle progression is arrested .
4.Clinical Observation of Ulinastatin in the Treatment of Acute Pancreatitis

OBJECTIVE:To observe therapeutic efficacy of ulinastatin in the treatment of acute pancreatitis. METHODS:A to-tal of 80 acute pancreatitis patients were randomly divided into control group and observation group,with 40 cases in each group. Control group was given conventional treatment as gastrointestinal decompression,analgesia and spasmolysis,anti-infective treat-ment,fluid replacement,electrolyte disturbances balance,in vivo microcirculation improvement;observation group was additional given ulinastatin 100 000 U added into 5% Glucose injection 250 ml,ivgtt,bid,on the basis of control group,and then given medicine qd after improving symptoms;finally,stopped taking medicine immediately the symptom disappeared. Both groups was given 8d of treatment. Serum levels of IL-6 and TNF-α were observed in 2 groups before and after treatment,and clinical symp-tom,the time of sign and lab index improvement were also observed. Clinical efficacy was evaluated. RESULTS:After treatment, serum levels of IL-6 and TNF-α in observation group was significantly lower than in control group;the time of clinical symptom and sign,lab indicator improvement were significantly shorter than in control group,with statistical significance(P<0.01). The ef-fective rate (95.00%) of observation group was significantly higher than that (70.00%) of control group,with statistical signifi-cance(P<0.05). CONCLUSIONS:Ulinastatin can significantly reduce acute pancreatitis significantly,improve clinical symptom, promote disease recovery and clinical efficacy.
5.Cell morphology and micronuclei in oral exfoliative cells

砄bjective: To evaluate the diagnostic significance of exfoliative cytological examination and micronucleus assay. Methods: 26 cases of leukoplasia (LK) and 18 cases of oral squamous cell carcinoma (SCC) were examined. Cells were scrapped from the lesions, stained with Papaniceolaou. The keratinization types and the configuration of the cells were studied. Smears were also stained using the Feulgen plus fast green method. The frequency of micronucleated cells was counted. Results: Mixed keratinization was observed in all 15 cases of LK without dysplasia and in 7 out of 11 cases of LK with dysplasia. Parakeratosis was found in 17 out of 18 cases of SCC. There was a significant difference of cell keratinization types and cytologic diagnosis between the cases of LK and SCC( P 0.05).Conclusion: Parakeratosis in oral exfoliative cells may indicate a high risk of malignance.

7.Study on the relationship between serum retinal binding protein 4 level and acute cerebral infarction or its risk factors

Objective To study the relationship between serum retinal binding protein 4 (RBP4) level and acute cerebral infarction,and to analyze the correlation between RBP4 level and the related risk factors of acute cerebral infarction.Methods One hundred and sixty patients with acute cerebral infarction were chosen as observation group,and 80 cases of contemporaneous healthy examined people were selected as control group.The levels of serum RBP4,body mass index (BMI),fasting blood glucose (FBG),fasting serum insulin (FINS),glycosylated hemoglobin (HbA1c),total cholesterol (TC),triglyceride (TG),high density lipoprotein cholesterol (HDL-C),low density lipoprotein cholesterol (LDL-C) and blood pressure after admission of each group were measured.Statistic analysis was processed between the level of serum RBP4 and acute cerebral infarction or related risk factors.Results The level of serum RBP4 in observation group was increased compared with that in control group [(41.68 ± 5.12) mg/L vs.(23.84 ± 3.26) mg/L],and there was significant difference (P < 0.01).The Pearson relative analysis showed positive correlation between the level of serum RBP4 and related risk factors of acute cerebral infarction FBG,FINS,LDL-C,BMI,systolic pressure.The Logistic stepwise regression analysis suggested that BMI,LDL-C and age were independently associated with the level of serum RBP4.Conclusion RBP4 plays an important role in the incidence of acute cerebral infarction,high RBP4 level may be one of the dangerous factors of acute cerebral infarction patients and could provide reference to prevents the cerebral infarction.

Objective To develop an HPLC analytic method for determination of cyromazine residue in water.Methods Chromatographic column of YMC C18(150 mm?4.6 mm,5 ?m)was used,mobile phase was methanol+0.02 mol/L NH4Ac=15+85(V/V),flowing rate was 1.0 ml/min,DAD detection wavelength was 210 nm.Results Regression equation was y=243.8x-4.2,r=0.999 96.The minimal detectable concentration was 0.010 mg/L;the rate of recovery was 94%-100% and the relative standard deviation was less than 3.0%.Conclusion The method is satisfactory in precision,accuracy and sensitivity.
9.Diagnosis and treatment for ipsilateral fractures of femoral neck and shaft

[Objective]To investigate the clinical features,reasons of missed diagnosis and treatment for ipsilateral fractures of femoral neck and shaft.[Method]Twelve cases of ipsilateral fractures of femoral neck and shaft treated from 1999 to 2005 were retrospectively reviewed.Seven cases' neck fractures were diagnosed separately before operation,2 cases' were diagnosed during operation and 3 were diagnosed after operation.Among the 12 cases,3 cases were treated with reconstructive intramedullary nail to fix both shaft and neck fractures;2 cases were treated with DHS to fix both shaft and neck fractures;1 case was treated with retrograde interlocking intramedullary nail to fix shaft fracture and with canulated screws to fix neck fracture;2 cases were treated with interlocking intramedullary nails and canulated screws;1 case combined supracondylar fracture of femur,was treated with LISS-DF to fix shaft and supracondylar fracture,canulated screws to fix the neck fracture;the neck fractures were found in 3 cases after shaft fixation with plate,then fixed with canulated screws.[Result]All cases were followed up for 1~6 years,with an average of 3.4 years.All femoral shaft fractures were united and 11 neck fractures were united,1 neck fracture was malunion,1 case was osteonecrosis of femoral head.[Conclusion]The ipsilateral fracture of femoral neck and shaft is rare,and the neck fracture is easy to miss diagnosis.The orthopaedic surgeons should consider the femoral neck fractures by analyzing the mechanism of high energy injury patient with femoral shaft fracture.Pelvic X-ray should be taken conventionally or CT scan when necessary.The surgical treatment for ipsilateral fractures of femoral neck and shaft depends on the position of femoral shaft,and the situation of femoral neck.
10.Determination of Deltamethrin Residue in Water by Rephase High Performance Liquid Chromatogrophy

Objective An analytic method with rephase high pressure liquid chromatogrophy(RP-HPLC) was developed to determine the residue of deltamethrin in water.Methods Water sample was filtrated and determined by RP-HPLC.Chromatographic column was YMC C_(18)(250 mm?4.6 mm,5?m),mobile phase was methanol:water=90:10(V/V),the flowing rate was 0.8 ml/min, DAD detecting wavelength was 205 nm.Results The regression equation was y=655.6 x-1.8,r=0.999 99.The lowest detectable concentration was 2.9?g/L.The recovery rate was 98.0%-102.0%and the relative standard deviation was 0.29%-0.48%. Conclusion The method is proved to be satisfactory in precision,accuracy and sensitivity and can be used for the rapid determination of deltamethrin residue in water.
