2.INFLUENCE OF DIETARY RESTRICTION ON THE EXPRESSION OF INSULIN SIGNALING GENES IN MOUSE EPIDIDYMIS FAT PAD

Objective:To study the influence of dietary restriction on the expression of insulin signaling pathway genes in mouse adipose tissue. Method:Twenty four C57BL/6 male mice of 2 w age were subjected to either AL (ad libitum) or DR (dietary restriction, 40% from AL) randomly until sacrificed.Then the epididymis fat pads were weighed, and their insulin signaling gene expression was detected by Oligo Insulin Signaling Pathway Microarray (SuperArray), and two of the differentially expressed genes, NPY and AKT3 were validated by Real Time PCR. Results:Dietary restriction significantly reduced the weight of epididymis fat pads and 31 (20 up, 11 down regulation) of 96 insulin signaling pathway genes were differentially expressed. The results of Microarray coincided with that of Real Time PCR. Conclusion:The differential regulation on the expressions of insulin signaling pathway gene may be the important molecular basis of dietary restriction.
3.Prevention of complication of allogeneic segmental small intestinal transplantation in rats

To reduce the surgical complications and raise the successful rate of small intestinaltransplantation(SIT),32 rats received segmental isogenic SIT heterotopieally using modifiedMonehik and Russels'method with the successful rate being 96.9%.The survival rate of the ratsover 10 days was 81.25%with the longest survival over 500 days.It was indicated that those arethe keys of success of SIT such as adequate liquid supplement,excellent anastomosis of blood ves-sels and effective protection to the isograft,as well as the sterile principle and washing isograftregularly.
4.Application of case method teaching to emergency talent training in environmental hygiene

5.Study of Effective Ingredient in Huanglianjiedu Decoction under Different Decocting Conditions

Objective To analysis the effective ingredients in Huanglianjiedu decoction under different decocting conditions. Methods The detecting method of HPLC was established and the ingredients in Huanglianjiedu decoction under different decocting time and times were analyzed. Result Water- soluble glycoside of gardenoside and baicalin reached balance of dissolution within 20 min and their dissolution rate were about 82% and 60% respectively. Water-soluble alkaloids of berberine and palmatine reached balance of dissolution within 30 min and their dissolution rate were about 45%. The dissolution rate of wogonin was 31% at 60 min. The dissolution rate of Crocin-Ⅰ reached top at 20 min and fell down after 20 min. Decocting times for fat-soluble constitutes was more important than water-soluble constitutes. Conclusion The decocting condition has effect on the dissolution rate and stability of the ingredients in Huanglianjiedu decoction. The decocting condition is very important to the quality control of decoction.
6.A comparison of therapeutic effect of retrograde interlocking intramedullary nail and condylar plate fixation in the treatment of comminuted fracture of femoral condyle

Objective To compare the treatment effect of internal fixation with retrograde interlocking intramedullary nails or condylar plates in the treatment of comminuted fracture of the femoral condyle. Methods 36 cases of comminuted fracture of femoral condyle admitted from 2000 to 2005 were treated with retrograde interlocking intramedullary nails (n=19) or condylar plates (n=17). Union time of fracture and the recovery of knee function were observed respectively. Results The average union time of fracture of the cases treated with retrograde interlocking intramedullary nails was 102 days, and the score of knee function was 84.59. Whereas in the cases treated with condylar plates the union time of fracture was 91.35 days, and the score of knee function was 91.10. 7.Effects of calcitonin gene-related peptide on calcification of human dental pulp cells in vitro

砄bjective: To investigate the effect of calcitonin gene related peptide(CGRP) on calcification function of human dental pulp cells(HDPCs) in vitro . Methods: HDPCs cells were exposed to calcitonin gene related peptide at the dosages(mol/L) of 10 -12 , 10 -10 and 10 -8 for 3,6,9, 12 and 15 days respectively. Cell growth was studied by cell counting , alkaline phosphatase(AL) by a AL kit, osteocalin by immunoradiological tecnique and type I collagen mRNA expression by RT PCR. Results: CGRP did not show any effect on the growth of HDPCs, but it increased the expression of AL, osteocalin and type I collagen mRNA in HDPCs in a dose dependent and not time dependent manner. Conclusion: CGRP plays an important role in promoting differentiation and calcification function of HDPCs
8.Clinical effects of docetaxel combined with compound tegafur capsule in the treatment of 38 patients with anthracycline-refractory recurrent metastatic breast cancer

ObjectiveTo investigate the efficacy and toxicity of docetaxel combined with compound tegafur capsule(S-1)on anthracycline-refractory recurrent metastatic breast cancer (ARMBC).Methods Thirty-eight ARMBC patients were given intravenous 70 mg/m2 docetaxel at day 1,and oral 60 mg/m2 S-1twice every day at day 1 to 14.Every 3 weeks was one cycle and each patient received at least two cycles.ResultsAfter treatment,among these 38 patients,there was 2 complete response (CR) (5.3 %),20 partial response (PR) (52.6 %),10 stable disease (SD) (26.3 %),and 6 progressive disease (PD) (15.8 %).Overall objective response rate was 57.9 % (95 % confidence intervaal: 42.6 %-74.2 %) while clinical benefit response rate was 73.7 % (95% confidence interval: 58.4 %-89.1%).The median time to progression (TTP) was 7.8 months(95 % confidence interval:6.7-8.9 months),and median overall survival time(OS)was 15.7 months (95 % confidence interval: 12.9-18.8 months).The main toxic reaction was myelosuppression,and grade Ⅲ and Ⅳ adverse events including leucopenia occurred in 21.1% of all cases.Most common grade Ⅰ and Ⅱ adverse events,such as hand-foot syndrome,nausea,vomiting,diarrhea,liver dysfunction,and oral mucositis,were tolerable.ConclusionGood clinical efficacy is achieved in the therapy of metastatic breast cancer with docetaxel and S-1 combination regimen and toxic reaction is tolerable.
9.Detection of Th1/Th2Cytokines in the Peripheral Blood Lymphocytes of Patients with Condyloma Acuminatum

Objective To detect cytokine levels secreted by CD3 + T cells and monitor the Th cell im-balance in the patients with condyloma acuminatum(CA).Methods The peripheral blood lymphocytes were isolated and stimulated prior to analysis.Cytokine detection was performed using monoclonal antibodies,which were fluorochrome-conjugated for easier direct manipulation in flow cytometry analysis.The results were pre-sented as cytograms showing dual staining of lymphocytes.Results The levels of IL-2,IL-12,IFN-?and TNF-?produced by Th1cells decreased,while the level of IL-10produced by Th2cells had no significant change in CA patients in comparison with healthy controls.Conclusions There is Th1/Th2imbalance in CA patients with a decline of Th1type cytokines.The Th1/Th2imbalance might be the mechanism that HPVs es-cape from immunosurveillance in CA patients.
10.Fungal Infection among Patients with Maintenance Hemodialysis:A Clinical Analysis

OBJECTIVE To investigate the risk factors,characteristics,measures of prevention and treatment of hospital fungal infection among maintenance hemodialysis patients.METHODS Clinical data of 128 maintenance hemodialysis patients between Jan 2002 and Dec 2006 were enrolled in this retrospective study of hospitalized infection cases.RESULTS During above period,36 infection episodes of 27 cases were occurred.The major infection site was lungs(69.4%).the Candida albicans was the main pathogen species(61.1%).The underlying and complicating diseases such as diabetes,chronic obstructive pulmonary disease and tumor,long antibiotics using,long hospitalization,poor nutrition status,inadequate hemodialysis,and venous indwelling catheter were the risk factors which led to fungal infection among maintenance hemodialysis patients.CONCLUSIONS The maintenance hemodialysis patients are easy to induce fungal infection.The C.albicans is the main pathogen species.The incidence of underlying diseases is rather high.Treatment of original diseases and complicating diseases,the standard using of antibiotics,adequate hemodialysis,the nutrition status and the reduction of the indwelling time of venous catheter are all the keys to decrease the fungal infection.
