1.Expression and clinical slgnificance of CerbB——2 and p53 protein in cervical squamous carcinoma

Objective To explore the expression and clinical significance of two oncogenes,CerbB-2 and p53,on cervical squamous carcinoma.Methods The immunohistochemical S-P process was performed on 45 cases of cervical squamous carcinoma.The expressions of CerbB-2 and p53 on 15 cases of CIN(cervical intraepithelial neoplasia)and on 10 cases of normal cervical tissues were chosen and the relationship between the expressions and the related clinicopathological factors were analyzed.Results The positive expression rate of CerbB-2 on normal cervical tissues, CIN and cervical squamous carcinoma was 0,26.7% and 66.7%, respectively; that of p53 was 0,13.3% and 68.9% ,respectively.From the normal cervical epithelium to the CIN and to the cervical squamous carcinoma,the positive rate of CerbB-2 and p53 was increased significantly(P<0.05);the positive rate of CerbB-2 was associated with the pathological grade and the metastasis of lymph node(P <0.05) and that showed no statistic significance with the clinical stage ,the diameter of tumour,the infiltration of vessel and infiltrative depth(P > 0.05) ;the positive rate of p53 is only associated with the pathological grade(P <0.05) ;the difference between the CerbB-2 and p53 expressions on cervical squamous carcinoma were not statistically significant(P> 0.05).Conclusion The expressions of CerbB-2 and p53 have some practical values on the occurrence, the development and the clinical prognostic evaluation for the cervical squamons carcinoma.
2.Comprison of surgical reconstruction of digestive tract after gastric resection

Objective To compare total gastrectomy jejunal loop P-type esophagus jejunum Roux-en-Y anastomosis(PRY) and non-type esophageal transection of the jejunum improved Roux-en-Y anastomosis(URY) two different digestion Road reconstruction on the nutritional status of patients and gastrointestinal symptoms. Methods 152patients with total gastrectomy required of gastric cancer patients immediately divided into two groups ,76 patients in each group ,respectively PRY and URY surgical reconstruction of digestive tract, were followed up for 12 months, two groups were compared on nutritional status and gastrointestinal symptoms. Results PRY operation time and postoperative complication rate we re more than URY group(all P <0.05) ;two groups 12 months after the mortality and weight changes, hemoglobin, total protein, albumin, and all reflux esophagitis the incidence rate was no significant difference (all P > 0.05); after 3 months and 6 months in both groups food intake < 300ml/second person, eating frequency >5 times/d and the difference in the incidence of RSS had statistical significance (all P < 0.05). Conclusion URY surgical reconstruction of digestive tract and maintain the continuity of muscle conduction,and the surgical procedure was simple,a good prognosis and relatively PRY more reasonable in terms of surgical procedures.
3.Design and application of cardiac stimulator for rapid atrial pacing animal model.

A cardiac stimulator for rapid atrial pacing animal model was designed in this paper. According to the needs of clinical research, output pulse parameters of the cardiac stimulator can be designed. The cardiac stimulator will be controlled through magnet. Efficiency of the cardiac stimulator was validated by animal experiments.

4.Effect of cell passage on differentiation of bone marrow mesenchymal stem cells into neural stem cells

BACKGROUND:It is unclear whether serial cel passage in vitro influences the differentiation of bone marrow mesenchymal stem cel s into neural stem cel s.
OBJECTIVE:To investigate the effect of cel passage on the differentiation of bone marrow mesenchymal stem cel s into neural stem cel s.
METHODS:Rat bone marrow mesenchymal stem cel s were isolated and cultured by the whole bone marrow adherence method. Bone marrow mesenchymal stem cel s at passages 3, 6, 9, 12 were incubated in serum-free medium. After culture for 7 and 14 days, cel biological characterization was observed and differenitaiton ability into neural stem cel s was observed by detecting Nestin expression in cel s using flow cytometry. Then, the cel s were further induced to differentiate and cel multipotential differentiation capacity was detected by measurement of nerve enolase and glial acidic protein expression.
RESULTS AND CONCLUSION:Under induction, bone marrow mesenchymal stem cel s at different passages were al differentiated into Nestin-positive neural stem cel s. However, there was a significant difference in differentiation proportion of cel s at different passages (P<0.05). Strongest differentiation ability was found in the passage 6 cel s, with the Nestin expression up to (93.7±2.3)%at 7 days of induction and (96.2±1.8)%at 14 days of induction. The proportion of differentiated cel s at passages 6 and 9 was signfi cantly higher than that at passages 3 and 12. Moreover, adherent cel s were positive for nerve enolase and glial acidic protein. Al these findings indicate that the differentiation of bone marrow mesenchymal stem cel s into neural stem cel s is correlated with cel passage. Cel s at lower or higher passages are both detrimental to cel differentiation.
5.Detection of Langerhans cells and plasmacytoid dendritic cells in condyloma acuminatum lesions of patients

Objective To investigate the possible roles of Langerhans cells(LCs) and plasmacytoid dendritic cells(pDCs)in the pathogenesis of condyloma acuminatum(CA). Methods Tissue specimens were obtained from the lesions of 23 patients with CA and from perilesional normal skin of 13 of the 23 patients. The expressions and distribution of CD1a, CD2AP and CD123 were analyzed using the streptavidin-peroxidase (SP) immunohistochemical method. Statistical analysis was carried out to compare the density and percentage of CD1a + LCs, CD2AP + pDCs and CD123 +pDCs between lesional skin and perilesional normal skin. Results Most CD1a + LCs were observed in the prickle cell layer of the epidermis, and a few in the papillary layer of the dermis. There was no significant difference in the density or percentage of CD1a+ LCs in the epidermis or dermis between the lesional and perilesional skin specimens (all P > 0.05). CD2AP + pDCs and CD123 + pDCs were distributed mainly in the papillary layer of the dermis, and their density and percentage were significantly higher in the lesional than in the perilesional skin specimens (all P < 0.05). Conclusion Human papillomavirus infection may affect the quantity of pDCs but not LCs in local mucous membranes.
6.Application of HRCT multi-planar reconstruction for measuring cochlear morphology in cochlear implantation.

OBJECTIVE:
To explore the value of high-resolution temporal bone CT (HRCT) multi-planar reconstruction (MPR) technique for measuring cochlear morpholog in the preoperative evaluation of cochlear implant.
METHOD:
We retrospectively analyzed the clinical data of 80 cochlear implant recipients. The groups were investigated in our research according to the patient's sex, age and whether the inner ear is malformed or not. Temporal bone HRCT MPR was applied to analyze the measurement parameter in the standard orientation of the cochlea image: the largest distance from the round window to the lateral wall (distance A), the perpendicular distance (B) and angle of the basal turn of the cochlea relative to the midsagittal plane (γ). Reproducibility of the measured data was evaluated and the results between the different groups were compared.
RESULT:
Measurement of parameter values between the intraobserver and interobserver showed good reproducibility. In normal group, distance A [(8.79±0.34) mm] and perpendicular distance B [(6.58±0.28) mm] of males' cochleae were significantly larger than A [(8.53±0.47) mm] and B [(6.22±0.43) mm] of females' (P<0.05), but the angle γ was not significantly different between the two groups (P>0.05). The A and B values didn't change with age (P>0.05), but the angle γ tended to decrease with increasing age (r=-0.25, P<0.05). In the malformation group, the value B was significantly smaller than that of the normal group (P<0.01), but for value A and angle γ, there was no significant difference between the two groups (P>0.05).
CONCLUSION
HRCT MPR technique is simple and reliable in measurement of cochlear morphology. It can be used to guide the surgery and provide an objective basis for designing the personalized electrode. More clinical application of this technology is recommended.

7.Argon plasma coagulation combined with bifurcated metal stents for treatment of airway stenosis in the carinal region A 16-case analysis

A total of 16 patients with airway stenosis including benign lesion (n=4) and malignant disease (n=12) were treated with argon plasma coagulation (APC) and Z-type covered retrievable metallic stent.L-and I-type stents were placed by guidance of brenchoscope,while Y-type stent was placed by the guidance of both bronchoscope and fluoroscope.Airway stenosis was from (58.8 +9.1)% before APC to (7.5±2.4)% after APC in tracheal,from (67.4±7.4)% to (19.4±4.1)% in left main bronchus,from (69.6±8.9)% to (27.6±5.4)% in right main bronchus.Symptoms of chest distress and breathlessness were improved remarkably,tachypnea indexes were decreased,but Kamofsky performance scope were increased.Twelve stents were successfully installed by the first time,including 9 Y-shape;otherwise,4 stents failed to be installed because the carinal was too wide to insert the Y-shape stent in two patients with lung cancer,finally,L+I type stents were used.Median survival time after successful stenting was 10 months,and mean time was 13 months.APC combined with bifurcated metal stants relieved obstruction and improved quality of life.
8.Clinical epidemioiogical analysis of hospital-associated pneumonia in senile patiens in Guangzhou strict

Objective To determine the distribution and antibiotic resistance of hospital - associated pneu-monia(HAP) in senile patient,and the high risk factors of HAP,pathogenic bacterium and prognosis,so as to instruct the clinic prevention and treatment. Methods Patients with age over 60 years old,who were diagnosed of HAP with confirmed pathogens. Pathogens were identified, then the antibiotic resistance was determined by Kirty-Baucer disk dif-fusion assay. High risk factors of HAP, pathogenic bacterium and prognosis were analyzed by Logistic regression analy-sis. Results The first 10th pathogens of HAP patients were pseudomonas aeruginosa( 16.1% ),staphylococcus au-reus( 14.6% ) ,klebsiella pneumonia( 10.2% ), escherichia coil (8.8%) ,staphylococcus hemolyticus (7.3%). MR-SA accounted for 90% in staphylococcus aureus. The drug resistance of pseudomonas aeruginosa to imipenem was 29.5%. The high risk factors of HAP were acute brain accident, hypoalbuminemia, tracheal intubation or mechanical ventilation, rennal failure,COPD, gastric tube, anaemia,liver disfunction,long length of stay ( all P < 0.05 ). The mor-tality of senile HAP was 29.2%. Conclusion There are high antibiotic resistance and high mortality of HAP in se-nile patients. Measures should be taken to modify the risk factors.
9.Study on the expression of Glut1, HIF-1α and Ki-67 in hepatocellular carcinoma and their relationship

ObjectiveTo explore the expression of Glut1, HIF-1α and Ki-67 in hepatocellular carcinoma (HCC) and their relationship between their expression and clinicopathological features.MethodsImmunohistochemical study (EnVision method) for Glut1, HIF-1a and Ki-67 were performed on tissue microarray which consisted of 171 cases of HCC, 55 cases of adjacent non-neoplastic liver tissues, and 22cases of normal liver tissues.ResultsThe expression rate of Glut1 ,HIF-1α and ki-67 in 171 cases of HCC was 15.2%, 19.9% and 66.1%, respectively, which was much higher than that in the adjacent non-neoplastic liver tissues (1.8% ,1.8%and 5.5%) and normal liver tissues(all negative).The expressions of Glut1 and HIF-1α were positively correlated with the differentiation degree of HCC and TNM stage(P <0.01, P <0.05).The expression of ki-67 was positively correlated with the differentiation degree of HCC.There was a significant positive correlation between the expressions of Glut1 and HIF-1α in HCC (r1 =0.553, P <0.05), the expressions of Glut1 and HIF-1α were positively correlated with ki-67(r2 =0.560,r3 =0.613, P <0.05).ConclusionsGlut1, HIF-1α and ki-67 may play a role in the tumorigenesis and progression of HCC in some degree.Combined detection of Glut1, HIF-1α and Ki-67 may be helpful to judge the degree of malignancy and potential metastasis and evaluate the prognosis.
10.INFLUENCES OF KONJAC-POLYSACCHARIDE ON THE LEVELS OF TISSUE LIPIDS AND FOUR INORGANIC ELEMENTS IN RATS

This paper reports a study regarding the verity of the hypocholestero-lemic effect of konjac-polysaccharide. The konnyaku powder (KP) used in this study was prepared and refined from the tubers of Amorphophallus ko-njac K. Koch and contained 84.8% of glucomannan. Male and female Spra-ue-Dawley rats aged 5 weeks were divided into 5 groups and fed on normal basal diet, hypercholesterolemic diet (control diet) and 3 test diets (i.e. KP was added to the control diet at a dosage of 2.5%, 5% or 10%) respe- ctively, for 12 weeks.The results obtained from this study showed that KP could markedly lower the level of the cholesterol in sera and livers of rats feeding hyper-cholesterolemic diets. At the end of the 4th week of the feeding experiment, the serum cholesterol level of the 5% and the 10% KP groups, and the liver cholesterol level of the 10% KP group were shown to be significantly lower than those of the control group. At the end of the 12th week, serum cholesterol levels of all the 3 KP groups were found to be lowered to the level of the normal group and so did the liver cholesterol level of the 10% KP group. The lipotropic (anticholesteatosis) effect of KP was also confirmed by the hepatic histopathological examination. Besides the hypocholest-eroletmic eftect, KP diets can also increase the bulk of stool. Finally, there were not any harmful effects on the absorption and utilization of Ca, Fe, Zn and Cu being found.
