AIM:To investigate the changes of apoptosis and activation of I??-? induced by vinblastine via the blockage of caspase-3 signal transduction pathway, and to explore the possible mechanism of signal transduction pathway involving in the vinblastine-induced apoptosis. METHODS: The breast cancer cell lines Bcap37 were treated with different concentrations of vinblatine dissolved in dimethyl sulphoxide (DMSO) or caspase-3 inhibitor (DEVD-CHO, 100 ?mol/L) for 3 h. The changes of the proliferation were detected by MTT methods. The apoptosis was determined by observing the internucleosomal DNA cleavage and PI staining, and the proteins of pro-caspase-3 and I??-? were detected by Western blotting methods. RESULTS: The results showed that vinblastine induced the pro-caspase-3 degradation. The significantly attenuation of vinblastine-induced apoptosis in breast cancer cell line by caspase-3 inhibitor DEVD-CHO was verified by MTT assay, internucleosomal DNA cleavage and flow cytometry PI staining analysis. The IC50 was 56.8 ?mol/L and 87.4 ?mol/L respectively for two groups. The inhibition of vinblastine-induced phosphorylated degradation of I??-? was also observed by DEVD-CHO. CONCLUSION: Based on these finding, vinblastine induces apoptosis in breast cancer cells via NF-??/I?? signal transduction pathway, which is co-operated by caspase signal pathway. Through the blockage of caspase pathway with caspase-3 inhibitor, vinblastine-induced apoptosis and the phosphorylated degradation of I??-? in breast cancer cells are suppressed greatly.

AIM: To evaluate the influence of radiofrequency (RF) hyperthermia on immunity function in mice. METHODS: The expression pattern of T helper type 1 (Th1) and T helper type 2 (Th2) cytokines in splenocyte culture supernatants, mainly the expression levels of IFN-?, IL-2, IL-4 and IL-10 in splenocyte culture supernatants of mice in tumor-bearing group, surgical resection group, RF therapy group and normal control group were detected by enzyme-linked immunoadsordent assay (ELISA). RESULTS: IL-2 concentration in two weeks after RF therapy group was higher than that in two weeks after surgical resection and normal control groups (P0.05). CONCLUSION: RF hyperthermia may activate the transformation from Th2 to Th1 and facilitate the excretion of Th1 type cytokines that play an important role in the anti-tumor immunity.

OBJECTIVE:
To explore the value of high-resolution temporal bone CT (HRCT) multi-planar reconstruction (MPR) technique for measuring cochlear morpholog in the preoperative evaluation of cochlear implant.
METHOD:
We retrospectively analyzed the clinical data of 80 cochlear implant recipients. The groups were investigated in our research according to the patient's sex, age and whether the inner ear is malformed or not. Temporal bone HRCT MPR was applied to analyze the measurement parameter in the standard orientation of the cochlea image: the largest distance from the round window to the lateral wall (distance A), the perpendicular distance (B) and angle of the basal turn of the cochlea relative to the midsagittal plane (γ). Reproducibility of the measured data was evaluated and the results between the different groups were compared.
RESULT:
Measurement of parameter values between the intraobserver and interobserver showed good reproducibility. In normal group, distance A [(8.79±0.34) mm] and perpendicular distance B [(6.58±0.28) mm] of males' cochleae were significantly larger than A [(8.53±0.47) mm] and B [(6.22±0.43) mm] of females' (P<0.05), but the angle γ was not significantly different between the two groups (P>0.05). The A and B values didn't change with age (P>0.05), but the angle γ tended to decrease with increasing age (r=-0.25, P<0.05). In the malformation group, the value B was significantly smaller than that of the normal group (P<0.01), but for value A and angle γ, there was no significant difference between the two groups (P>0.05).
CONCLUSION
HRCT MPR technique is simple and reliable in measurement of cochlear morphology. It can be used to guide the surgery and provide an objective basis for designing the personalized electrode. More clinical application of this technology is recommended.

Chinese Journal of Postgraduates of Medicine 2017;40(8):693-696
Objective To compare sinus tarsi approach(STA) and extensile L shape approach (ELA) in the surgical treatment of calcaneal fractures. Methods A total of 51 cases of calcaneal fractures treated from May 2014 to December 2016 were enrolled. Twenty of them were performed ORIF via the sinus tarsi approach, while the other 31 cases were performed ORIF via lateral extensile L shape approach. X-ray films were taken postoperatively to record the changes of B?hler angle and Gissane angle. The AOFAS, VAS and SF-36 scores were adopted to assess the therapeutic effect. Results The average follow-up period of STA group was 15.5 months. And patients in ELA group were followed up for an average period of 18.4 months. All the fractures in the series had a boney union at or before the final follow-up. The final correction of B?hler angle (34.60 ± 2.91)° , and Gissane angle (114.45 ± 2.91)° was significantly increased in the STA group; and the final correction of B?hler angle (33.55 ± 2.73)° , and Gissane angle (112.81 ± 4.15)° was also significantly increased in the ELA group; while there was no significant difference between the two groups. The final AOFAS scores , VAS scores and SF-36 scores of the STA were (83.56 ± 7.13), (1.78 ± 0.89), (88.95 ± 4.31) scores, while those in ELA group were (81.91 ± 9.89), (2.01 ± 1.33) and (86.65 ± 4.95) scores. There was no significant difference between the two groups (P>0.05). No wound problem happened in STA group, but there were 2 cases who had wound complications in ELA group. Conclusions No significant difference is found between the two groups in the short-term efficacy of the treatment for calaneal fracture. However, STA has the advantages of lower soft tissue complication rate.

Objective To investigate the mechanism of changes in skeletal muscle proteolytic rate after escharectomy during early stage of severe burn in rats. Methods One hundred and fifty six Wistar rats were subjected to 30% total body surface area (TBSA) full thickness burn on the back, and they were randomly divided into normal control group (group C), burn injury group (group B), escharectomy during early stage (1 PBD) group (group S)and escharectomy after early stage (4 PBD) group (group N). The extensor digitorium longus (EDL) muscle was collected from each rats using a technique that injury to the muscle was avoided, and the proteolytic rate of EDL was determined after in vitro muscle incubation with sufficient oxygen supply with amino acid automatic analyzer. Transcriptional expression of ubiquitin was assessed by Northern Blot. Results The myofibrillar proteolytic rate (MPR) was markedly increased at each time points after scald, and the MPR in group S was significantly decreased compared with group N. The expression of ubiquitin mRNA markedly increased in the three groups compared with group C, and was decreased in Group S. There was a significant positive correlation between MPR and expression of ubiquitin mRNA. Conclusion Escharectomy during early stage can alleviate protein catabolism in skeletal muscle, and may be related to decreased activity of ubiquitin proteasome pathway

Medical Journal of Chinese People's Liberation Army 2001;0(11):-
Objective To investigate the influence of different stimulating factors on protein metabolism of rat skeletal muscle. Methods Three kinds of animal models were used in this study as follows: burn injury group (group B, n =40), endotoxin challenge group (group E, n =40), and burn injury followed by endotoxin challenge group (group B+E, n =40). The proteolytic rate of extensor digitorium longus (EDL) muscle was determined by amino acid auto analyzer after incubation in a muscle incubation system. Results The proteolytic rate of EDL muscle of rats was increased markedly in those three groups, especially the myofibrillar proteolytic rate. The protein breakdown of muscle in group E was higher than that of group B, and it was even much higher in group B+E. Conclusions As compared among burn injury, endotoxin challenge and burn injury followed by endotoxin challenge, the change in muscle proteolytic rate is correlated to seriousness of the injuries
