1.The change and significance of lung function in 71 children with atypical asthma

Objective To investigate the change of lung function in children with atypical asthma and different lung function index to evaluate the difference of bronchial dilation test positive rate.Methods Seventy-one cases of atypical asthma children routine monitoring lung function,underwent bronchial dilation test,the airway index changes were observed.Results Seventy-one cases of atypical asthma children with big airways function index of forced expiratory volume in first second FEV1,percentage of FEV1 to forced vital capacity (FVC) were basic normal,and the reaction of small airways function index of forced expiratory flow at 25% of FVC,forced expiratory flow at 50% of FVC,forced expiratory flow at 75% of FVC,forced expiratory flow at 25%-75% of FVC were decreased in different degrees,and mainly for medium and severe damage.FEV1 as evaluation index,the positive rate of bronchial dilation test diastolic was 47.9% (34/71),and forced expiratory flow rate at 25%-75% in FVC as evaluation index,the positive rate of bronchial dilation test was 69.0% (49/71),there was significant difference (x2 =8.086,P =0.004).Conclusions Lung function changes in atypical asthma children are mainly on small airways,and mainly for medium and severe damage,the bronchial dilation test positive rate in such children using the evaluation,forced expiratory flow rate at 25%-75% in FVC can assist FEV1,decrease the rate of misdiagnosis and missed diagnosis.
2.Effects of lithium on CDK5 and PP2A expressions in chronic aluminum exposure rats

Objective To explore the effects of lithium on expressions of cyclin-dependent kinase 5 (CDK5) and protein phosphatase 2A (PP2A) in the brains of chronic aluminum exposure rats so as to further understand the mechanism of lithium's inhibition on tau phosphorylation. Methods We divided 12 chronic aluminum chloride exposure rats into treatment group and non-treatment group (6 rats in each). Treatment group was given lithium chloride (200 mg/kg?d) via gastric perfusion daily for 6 weeks,while the non-treatment group sodium chloride at the same dosage and the normal group (6 non-aluminum exposure rats of the same month old) without intervention. Six weeks later,all the rats received Morris water maze test for learning memory function; CDK5 and PP2A expressions and phosphorylated tau protein level in rat hippocampus were determined by Western blotting. Results Compared with normal group,non-treatment of chronic aluminum exposure group showed higher phosphorylated tau protein level and CDK5 expression in the brain (P0.05). Conclusion Lithium may reduce tau phosphorylation and neurofibrillary tangle formation by inhibiting the expression of CDK5.
3.Synonymous mutations in the second and the fourth codons stimulate the expression of human cardiac troponin I gene in E.coli

Objective To induce the site-directed mutation of human cardiac troponin I (cTnI) gene, express the mutant in E. coli, and to study the effects of the mutation on the prokaryotic expression of cTnI. Methods The cDNA encoding cTnI was cloned with RT-PCR from the total RNA extracted from human myocardium tissues. A pair of primers was designed and, after the mutations were induced at the second and the fourth codons, inserted into prokaryotic vector pET-28c (+) and transform the recombinant to BL21 (DE3) bacteria. After purified with Ni-NTA resin, the histidine-tagged fusion protein expressed by IPTG-induced was identified by Western blotting and the expression yield of cTnI protein was investigated. Results The expression of the recombinant carrying processed cTnI cDNA was stronger than that in control group. Conclusion cDNA encoding cTnI was successfully cloned. The recombinant with mutations can be more efficient expressed in E. coli. The cTnI protein can be purified to near homogeneity.
4.Induced differentiation of rat bone marrow mesenchymal stem cells into mesangial-like cells in vitro

Objective To reconfirm the mesangial cell differentiation potential of BMSCs and to study the mechanism of cell induction.Methods Rat BMSCs were isolated and incubated with platelet-derived growth factor(PDGF)-BB to induce the mesangial cell phenotype.At day 7,the cell morphology,mesangial cell-specific markers and cell contraction function were examined.MAPK/ERK kinase inhibitor(PD98059) was added to examine its impact on cell growth and mesangial cells marker expression.Results After cultivation under the differentiation condition for 7 days,the induced cells expressed ?-actin and Desmin,which highly resembled cultured mesangial cells in rat.The induced cells with increasing level of PDGF receptors mRNA expression changed into a wide range of shapes from spindle to stellate and were contracted in response to angiotensin Ⅱ.PD98059 perturbed the induced cells mesangial cell-specific markers mRNA expression.Conclusion BMSCs can differentiate into mesangial-like cells with PDGF-BB induction in vitro.The mechanism involved in the differentiation was mediated through MAPK/ERK1/2 signal transduction pathway.
5.Relationship between gene polymorphism of serum amyloid A protein 1 and coronary heart disease

6.Reversal effect of recombinant human Endostatin on cisplatin resistance line A549/DDP human lung adenocarcinoma cells

Objective To evaluate reverse effect of recombinant human Endostatin on drug-resistance of A549/DDP cells to cisplatin (DDP). Methods Lung adenocarcinoma cell line A549 and its DDP-resistant cell line A549/DDP were treated with DDP and recombinant human Endostatin. Difference in drug resistance was analyzed between different regimens ( DDP, Endostatin and combination) and between different cell lines ( human lung adenocarcinoma A549 and drug resistant A549/DDP), after a 72h-treatment in vitro. Reverse effect of recombinant human Endostatin on drug-resistance of A549/DDP to DDP was tested by MTT assay. Results The observed 50% inhibitory concentration ( IC50 ) was (0.72 ± 0.05 ) ug/ml against A549 and ( 11.54 ± 0.64)against A549/DDP in DDP, and (2.0 ± 0.1 ) μg/ml against A549/DDP in rh-Endostatin- DDP combination respectively, with a reversal fold (RF) of 5.77 and a relative reversal rate of 88. 2%. Conclusion rh-Endostatin may reverse drug-resistance of A549/DDP cells to DDP.
7.Protection of Shexiang Antithrombosis Pill on experimental cerebral ischemia in rats

Object To observe the protective effect of Shexiang Antithrombosis Pill (SATP) on experimental cerebral ischemia in rats. Methods In cerebral ischemia model induced by bilateral common carotid artery clamping and controlled hemorrhage to a mean arterial pressure of 6.7 kPa (50 mmHg), cerebral water content, SOD activity, MDA, Ca 2+ , and LA contents were measured and the ischemic tissue pathology was observed. Results SATP can decrease cerebral water content, lessen pathological change, increase SOD activity and decrease MDA, Ca 2+ , and LA contents. Effect of water pill is better than that of honey pill. Conclusion SATP can protect the cerebral tissue from ischemia injury by suppressing lipid peroxidation. This effect may be related to increasing antioxidase activity and decreasing acidosis of LA and overload of calcium in cell.
8.Evaluation of point-of-care testing for D-dimer in elderly patients

Objective To evaluate the clinical performance of AQT90 FLEX,a novel time-resolved fluorescence based point-of-care test (POCT) for quantification of D-dimer in elderly patients.Methods The method from Quantitative D-dimer assay (WS/T 477-2015) for testing equipment performance was used as a reference to evaluate the clinical performance of AQT90 FLEX.The correlation was compared between testing results of D-dimer using the AQT90 immune-assay analyzer and those using the ACL TOP coagulation analyzer.Results At high concentration of D-dimer,the within-run precision coefficient of variation (CV)was 2.619%,and at low concentration of D-dimer,the within-run precision CV was 2.767%.The pollution-carrying rate was 0.12%.The measured data from AQT90 and ACL TOP had a correlation coefficient of r =0.9491 (P < 0.01).The equation of the line of best fit for D-dimer with which all AQT90 results can be adapted to the ACL TOP was:AQT90 =2.52 ACL TOP + 0.15.The number from the equation was slightly greater in female than that in male,and it was also increased in elderly.Conclusions The AQT90 FLEX had rational precision and linearity in determination of concentration.There was a high agreement between the testing results from AQT90 and those from ACL TOP.It was recommended to use a slope of 2.52 and an intercept of 0.15 to adjust the D-dimer values of the ACL TOP to the AQT90 FLEX assay systems.POCT for D-dimer by AQT90 FLEX raises feasibility for use in elderly patients.
9.Characteristics of cognitive dysfunction in essential tremor patients

Objective To explore the characteristics and influencing factors of cognitive dysfunction in patients with essential tremor (ET).Methods We recruited ET patients diagnosed by the Department of Neurology of the First Affiliated Hospital of Xi`an Jiaotong University and healthy volunteers who matched the ET patients in age, gender and education level for the study.We recorded all the patients` demographic information, tremor degree, and family history based on the family tree.All the participants were tested by MMSE, MoCA, ADL, HAMD and HAMA.Results There were 88 ET patients and 63 normal subjects included in the study.According to MMSE, 31.82% of the patients had cognitive dysfunctions, with orientation, short-term memory, calculation ability, language skills, retelling, reading comprehension, three-level command and drawing being significantly lower than those of the healthy volunteers (P<0.01);orientation was the most serious damage in cognitive function domain (K=0.624, S=0.726);three-level command was the least serious damage (K=0.274, S=0.319).According to MoCA, 86.36% of the ET patients had cognitive dysfunction higher than normal people (P<0.05);visual space and execution, clock drawing task, naming, attention, 100-7, language skills, abstract thinking and orientation were significantly lower than normal people (P<0.01);the most serious damage in cognitive function domain was visual space and execution (K=0.651, S=0.786); the least serious damage cognitive function domain was “100-7” (K=0.406, S=0.484). Education level and age affected cognitive dysfunction (P<0.05). ADL scores showed negative correlation with cognitive function (correlation =-0.375 and -0.383, respectively; P<0.001). After the effects of anxiety and depression were excluded, onset age and tremor grading were correlated with cognitive dysfunction (P<0.05). When the above factors were put into binary Logistic regression model, education level was found to be contributed to the model (P<0.05).Conclusion Patients with ET widely suffer from cognitive impairment. Age, education level, daily life disability, age of onset, and tremor degree classification can affect cognitive dysfunction.
10.Mechanism of Escherichia coli injury under UV disinfection

Objective To study the inactivation kinetics and injury mechanism of Escherichia coli under UV disinfection in drinking water.Methods The inactivation kinetics of E.coli ATCC 25922 was determined by plate count methods in the UV disinfection experiment.The morphology,cell membrane permeabilization and injury of biological macromolecules were observed under a transmission electron microscope (TEM).The rate of ONPG hydrolysis and changes in activities of antioxidant enzymes were observed Raman spectroscopy.Results the changes of damaged cells involved morphological damage such as loss of the structural integrity of the wall and membrane,condensation of cellular material and leakage of significant amounts of cytoplasmic material,a more than four-fold increase of cell membrane permeabilization and damage to the structure of protein,nucleic acids and phospholipid.Conclusion UV disinfection can lead to a multi-target damage.
