AIM:To investigate myosin heavy chain(MHC)gene expression and the effects of captopril and betaloc at an early stage of hypertension. METHODS:Model of hypertension was made by partly narrowing two bilateral renal arteries(2K2C). The rats were divided into four groups at random. (1) control group; (2)2K2C group;(3)captopril group;(4)betaloc group.Levels of ?-MHC and ?-MHC mRNA were determined by dot-blot. RESULTS:?-MHC mRNA expression were gradual1y reduced in 2K2C group,while ?-MHC mRNA expression were increased,and the marked changes were observed at 72h postoperation. Captopril could inhibit the changes in MHC gene expression; but betaloc could not. CONCLUSION: The expression of MHC gene has changed at an early stage of renal hypertensive rat,and renin-angiotensin system may play an important role in this change.

Objective To study the expression of P16 ,Ki‐67 in cervical squamous epithelial lesions and the significance of their detection combined with human papillomavirus(HPV) .Methods 188 cases of cervical tissues from surgical excision were collected , including 48 cases of cervical cancer ,100 cases of cervical intraepithelial neoplasia(CIN) ,and 40 cases of chronic cervicitis ,which were set as cervical cancer group ,CIN group and cervicitis group ,respectively .Expression of P16 ,Ki‐67 in cervical tissue were de‐tected by using immunohistochemistry ,and the HPV infection were detected by using PCR technology .Results The positive ex‐pression rates of P16 and Ki‐67 in cervical cancer ,CIN group was significantly higher than those in cervicitis group(P<0 .05) ,and the positive expression rates of P16 and Ki‐67 in cervical cancer group were higher than those in CINⅠ and CINⅡ group (P<0 . 05) ,but between the cervical cancer group and CIN Ⅲ group there were no statistically significant difference(P>0 .05) .With the increase of CIN level ,the positive expression rates of P16 and Ki‐67 increased ,among the three CIN groups there were significant difference(P<0 .05) .In cervical cancer group and CINⅢ group ,the expression rates of P16 and Ki‐67 were both significantly high‐er than the other 3 groups(P<0 .05) .In CINⅠ - Ⅲ groups ,the over expression rates of P16 and Ki‐67 were statistically different (P<0 .05) .The expression of P16 positively correlated with the positive rates of HPV infection (rs =0 .706 ,P=0 .011);the ex‐pression of Ki‐67 positively correlated with the positive rates of HPV infection(rs=0 .695 ,P=0 .021) .Conclusion CIN and cervi‐cal cancer of early stage could be diagnosed and the pathological progress of CIN could be predicted by using the combined detection of P16 ,Ki‐67 and HPV .

The course of functional laboratory science is the bridge between classroom and clinical practice.In functional laboratory science teaching,we should pay attention to features of nursing care,focus on training compassion,clinical awareness,collaboration ability,practical skills for nursing students and combine the reform of functional laboratory science teaching with cultivation of professional quality for nursing students.

Objective To investigate the effect of MR imaging in rats with series coil. Methods MR scanning was performed on the self-made aqueous solution phantom with the series coil and other three coils respectively, using the same FRFSE- XL pulse sequence. SNR of the same image was obtained in the same position slice for all four coils. Twelve SD rats were divided into three model groups and one normal group. FRFSE-XL FSPGR and Probe-SI pulse sequence were respectively used to show the brain injury,1H-MRS, spinal cord injury (SCI) and abdomen with series coils. Results The SNR of series coil (39. 7)was over six times higher than the SNR of knee joint coil (6. 41 ), which was the best in other three coils.Grey matter and white matte; the brain structure and hematomas were clearly depicted in the rats'brain on T1WI and T2WI. From the second week after injection of 6-OHDA into the rats brain, the ratio of NAA/Cr decreased (before injection 1.24 ,two weeks after injection 0. 781 ). The spinal cord and the configuration of the white matter were clearly showed on T2WI image, and the location and degree of SCI in the rat thoracic spinal cord were also well defined on T2 WI image. The pulse sequence of FSPGR T1 WI was used for rat abdomen scanning. The scan time was only eight seconds. It is helpful to overcome the breathing movement effect and show the structure of rat abdomen. Conclusion It provided a good means for the rat MR in vivo to make use of 3.0T MR combined with series coils.

Functional experiment plays an important role in understanding the mechanisms of diseases and principles of diagnosis and treatment. It requires students to master basic skills of func-tional operation, and complicated disciplines of functional alteration as well. Thus functional experi-ment needs to be an open course. The use of the virtual reality (VR) and real-time recording tech-niques provides potential for this exploration. By the way of real-time recording system, HD video of experiment operation is played, which helps guiding students to learn basic experiment skills; while based on the development status of VR technology, it is more applicable for learning by oneself, such as preview before the class and review for the test, and furthermore, the advantage of VR technique will be more apparent, if the key development focuses on experiment extensions to disclose more compli-cated functional alterations. This new technique helps to improve teaching effects of functional experi-ment.

Objective To explore the correlation between axon guidance factor Semaphorin 5A and clinicopathological features and its role in the invasion and metastasis of gastric cancer.Methods The expression of Semaphorin 5A in gastric cancer tissues of 171 patients with different gender,age,histological type and TNM stage was detected with immunohistochemistry assay.The expression of Semaphorin 5A was determined by Western blotting assay in gastric cancer cell lines SGC7901 and MKN-45 with metastatic ability and gastric cancer cell lines SNU-1 and AGS without metastatic ability.With RNA interfere technique(RNAi),Semaphorin 5A siRNA expression vector was constructed and transfected into gastric cancer cell line SGC7901.The stable gastric cancer cell line down-expressing Semaphorin 5A was established.The effect of Semaphorin 5A gene silencing on the adhesion,migration and invasion of gastric cancer cell was examined by cell adhesion test,wound healing test and transwell chamber assays.Results The expression level of Semaphorin 5A was correlated with the differentiation degree of gastric cancer(x2 =6.32,P =0.01),lymphnode metastasis(x2 =7.68,P=0.01)and distant metastasis of gastric cancer(x2 =13.67,P =0.00),not correlated with age(x2 =0.21,P=0.79),gender(x2=1.79,P=0.15)and the depth of gastric cancer invasion(x2=1.34,P=0.55).The expression of Semaphorin 5A in cell lines SGC7901 and MKN-45 was significantly higher than that of cell lines SNU-1 and AGS(P＜0.01).Semaphorin 5A gene silencing significantly suppressed the adhesion,migration and invasion abilities of gastric cancer cells.Conclusion Semaphorin 5A may play a catalytic role in the invasion and metastasis of gastric cancer through increasing the adhesion,migration and invasion abilities of gastric cancer cell.

Objective To investigate the antitumor effect and its mechanism of microbubble contrast combined with Mitoxantrone exposed to low-frequency ultrasound on human breast cancer cell MCF-7. Methods MTT method was applied to examine the growth inhibition of MCF-7 treated with Mitoxantrone. MCF-7 cells were randomly divided into 4 groups:Mitoxantrone group (group D), ultrasound+Mitoxantrone group (group U+D), ultrasound+microbuble +Mitoxantrone group (group U+M+D) and control group (group C). The cytoactive of each group was examined with MTT. The intracellular drug content in each group was measured with high performance liquid chromatography. The morphology of MCF-7 cells apoptosis was observed with transmission electron microscopy (TEM). Results The IC_(50) of Mitoxantrone was 2.87 μg/ml. The differences of cytoactive among all groups were significant (P<0.05). The intracellular drug content of group U+M+D was higher than that of group U+D, and the latter was higher than that of group D. The morphological changes of apoptosis were observed with TEM. Conclusion Low-frequency ultrasound can promote intracellular drug content as to enhance the sensitivity of chemotherapy drugs on tumor cells, and this effect can be enhanced by microbubble contrast exposure to low-frequency ultrasound.

Objective To determine the evaluating prognostic value of plasma Nt-proBNP in patients with congestive heart failure.Methods 107 patients with congestive heart failure were included in this study,followed the degree of rehospitalization and mortality rate of the cured patients after left hospital.The plasma concentrations of Nt-proBNP were measured by enzymelinked immunozorbent assay(ELISA).Results Concentrations of Nt-proB-NP of Ⅱ,Ⅲ,Ⅳ classincation were(1129.8±383.2),(5849.7±567.6),(9506.3±492.4)ng/L in NYHA functional classification respectively(P＜0.01);the plasma level of Nt-proBNP had significant positive correlation with NYHA functional classification(r=0.759,P＜0.01).The degree of rehospitalization were(0.85±0.96),(1.98±0.86),(2.58±0.91)respectively(P＜0.01);mortality rate were 8.3%,12.5%,45.2%respectively(P＜0.01).Conclusion Plasma Nt-proBNP level may be the more important clinic value in evaluating prognosis in patients with congestive heart failure.

Objective To compare the efficacy of intralesional glucocorticoid injection with a needle-free injector versus an ordinary injector for the treatment of keloid. Methods A total of 60 patients with keloid were enrolled and randomly divided into two groups by using a random number table to receive intralesional injection of compound betamethasone with a needle-free injector(n=31)or an ordinary injector(n=29). The injection was given at a dose of 0.2 ml/cm3 once every 3 weeks for 3 sessions. Parameters for therapeutic efficacy were assessed, adverse reactions were recorded, and clinical pictures were taken before and after each treatment. Statistical analysis was carried out by the Mann-Whitney U test and chi-square test with the SPSS 19.0 software. Results Compared with the ordinary injector group, the needle-free injector group showed significantly different injection time during the first and second treatment (U=299.000, 773.500, respectively, both P=0.000), as well as duration of pain after the first injection(U=730.000, P=0.003). After three sessions of treatment, there was a significant difference in the volume, height, hardness of keloid, scores for pain, itching and appearance, and number of injection points between the needle-free injector group and ordinary injector group (U=295.000, 336.500, 264.000, 464.000, 451.500, 308.000, 233.500, P=0.001, 0.007, 0.000, 0.041, 0.043, 0.003, 0.001, respectively). No significant differences were observed in the incidence of adverse reactions between the two groups(all P>0.05). Moreover, the interval for lesion recurrence was significantly shorter in the needle-free injector group(11.8 days, 95%CI:10.96-12.6 days)than in the ordinary injector group(21.2 days, 95%CI:13.96-28.45 days). Conclusion Compared with the ordinary injector, the needle-free injector shows better efficiency for the treatment of keloid with decreased difficulty in injection and improved compliance in patients.
