1.Expressions of STAT3 and phosphorylated STAT3 in cutaneous malignant melanoma lesions

Objective To investigate expressions of STAT3 and phosphorylated STAT3(p-STAT3)in cutaneous malignant melanoma(CMM)lesions, and to explore their significance. Methods The expressions of STAT3 and p-STAT3 were detected with an immunohistochemical method using polymer enzyme-labeled secondary antibodies in skin specimens from 22 CMM lesions and 23 pigmented nevus lesions. The effects of STAT3 and p-STAT3 expressions on lymph node metastasis of CMM were evaluated. Results The expression rate of STAT3 was significantly higher in CMM lesions than in pigmented nevus lesions (95.5% vs. 56.5%, χ2 = 9.23, P < 0.05), so was that of p-STAT3 (90.9% vs. 43.5%, χ2 =11.38, P < 0.05). STAT3 expression in CMM lesions had no obvious effects on lymph node metastasis of CMM (P > 0.05), while the elevated expression of p-STAT3 in CMM lesions was a predictive factor for lymph node metastasis in CMM (OR = 1.88, 95% CI: 1.05 - 3.38, P < 0.05). Conclusion Increased expression of STAT3 and p-STAT3 in CMM may play important roles in the pathogenesis of CMM, and elevated p-STAT3 expression may contribute to invasion and metastasis of CMM.

The new cataloguing rules, RDA, have been implemented in NLM and more RDA bibliographic data can thus be accessed in domestic medical libraries when the Western documents are catalogued by offloading their bibliographic data.The differences in RDA and AACR2 were compared, the common modified fields and descrip-tive contents were described when the RDA bibliographic data were changed into AACR2 bibliographic data in order to provide certain reference for cataloguing of Western documents using the RDA bibliographic data.
3.Effects of all-trans retinoic acid, acitretin and tazarotene on apoptosis and active caspase-3 in human melanoma cells A375

Objective To investigate the effects of all -trans retinoic acid (ATRA),acitretin and tazarotene on apoptosis and active caspase-3 of human melanoma cells A375 and to discuss the relevant significance.Methods The effects of retinoids on apoptosis and active caspase-3 of A375 cells were examined in vitro.Early apoptosis analysis by double staining with annexin V-FITC and PI,and active caspase 3 analysis by the staining of FITC conjugated monoclonal rabbit anti active caspase-3 antibody were measured by flow cytometer.Results ATRA and acitretin induced apoptosis of A375 cells (both P<0.05),but tazarotene was not effective (P>0.05).ATRA and acitretin elevated the cell population with detectable active caspase-3 (both P<0.05),but tazarotene was not effective either (P>0.05).Acitretin played a most prominent role among the retinoids. Early apoptosis ratio was significantly and positively correlated with active caspase-3 percentage in both ATRA and acitretin (P<0.05).Conclusions Acitretin and ATRA induce apoptosis of A375 cells in which caspase is critical.Retinoids are prospectively applied as an assisting alternative for treating melanoma.
4.Changes of the Cytokines and Endothelium Function in Patients with Chronic Pulmonary Heart Failure Disease at High Altitude Area

Objective To investigate the changes and clinical significance of the plasma cytokines and endothelium function in patients with chronic pulmonary heart failure disease at high altitude area.Methods Plasma levels of tumor necrosisfactor-alpha(TNF-?),interleukin-6(IL-6),endothelin-1(ET-1) were measured in 278 chronic pulmonary heart patients with various degrees of heart failure and in 48 healthy controls.Left ventricular ejection fraction(LVEF),Right ventricular ejection fraction(RVEF) and Pulmonary(PASP) were examined with echocardiography.Results Plasma levels of TNF-?,IL-6,ET-1 were elevated in patients of functional classes Ⅱto Ⅳ as compared with age-matched control subjects and were progressively elvated with the decreasing functional status of the patients.The levels of TNF-?,IL-6,ET-1 respectively were positively correlated with PASP but negtively correlated with LVEF and RVEF(P

Objective To investigate the role of oxidized LDL and coronary endothelial dysfunction in the development and progress of coronary heart disease(CHD).Methods 29 patients with unstable angina(UA),25 patients with stable angina(SA) and 20 patients without coronary heart disease(control) were studied.Oxidized low-density lipoprotein(ox-LDL)、 Nitric oxide(NO),and circulating endothelial cells(CEC) were measured in both coronary sinus and peripheral blood during percutaneous coronary intervention(PCI).Results The level of NO in coronary sinus in patients with CHD was lower,while the level of ox-LDL and CEC was markedly higher than those in the control group.In CHD patients,the level of NO in coronary sinus blood was lower,while the level of ox-LDL and CEC was higher than that in peripheral blood.It was no siginificant difference between coronary sinus and peripheral blood was observed in the control group.Conclusion Oxidized LDL not only plays an important role in coronary endothelial dysfunction,but also contributes to the progress in the cornornary heart disease.It may be one of the pathophysiological basis of acute coronary incidents.
6.Changes of plasma ox-LDL and endothelium-dependent vasodilation in patients with acute coronary syndrome.

Objective To explore the relationship of the plasma ox-LDL level and endothelium flow mediated vasodilation(FMD)in acute coronary syndrome patients(ACS).Methods From March to October 2004,in the Department of Cardiology,the Fourth PLA Hospital,51 patients with ACS were divided into 2 groups:AMI group(22patients)and UAP group(29 patients).20 subjects with normal coronary angiograms served as the control group.Nitrir oxide(NO)and plasma ox-LDL was measured by spectrophotometer and Sandwich ELISE method,respectively.The brachial arterial hyperemia-induced flow mediated vasodilation(FMD)and sublingual nitroglycerin(NTG)mediated vasodilation were measured by high resolution ultrasound.Results (1)The FMD in ACS group was significantly decreased compared with the control group[(7.2?1.42)、(7.6?1.12)vs(15.46?1.2),P0.05).(2)The No in ACS group was sigificantly lower than that in control group;the NO in AMI group was even lower[(48.46?12.44)、(60.42?10.30)vs(94.72?12.34)?mol/L,P1,which meant ox-LDL was a risk factor;the OR of FMD was
7.Application of the PDCA cycle in the standardized residency training in hematology department

Objective:To explore the role of PDCA cycle in the standardized residency training in the department of hematology.Methods:Thirty residents trained in the department of hematology of Peking University Third Hospital from January 2019 to September 2019 were enrolled into experimental group, and 36 residents trained from January 2018 to December 2018 were enrolled into control group. Experimental group received the PDCA cycle teaching method and control group did the traditional one. At the end of the training, the results of theory examination and practical operation skills examination of the two groups were compared. The statistical significance was analyzed on the assessment scores and teaching satisfaction between the two groups. SPSS 20.0 was used for t test. Results:Compared with the control group, the examination scores and teaching satisfaction of the residents in the experimental group were higher than those in the control group ( P < 0.05). Conclusion:The teaching mode of PDCA cycle is conducive to improving the effect of standardized residency training in the department of hematology.
8.Clinical value of thyroid-stimulating antibody measurement using CHO-hTSHR cell line

Objective To evaluate the clinical value of CHO-hTSHR cells in detecting thyroid stimulating antibody (TSAb). Methods The cAMP production and TSAb activity were measured and cal-culated by stimulating CHO-hTSHR cell line with IgGs of normal control group and Graves" disease ( GD )group. TSAb positive standard was set to more than the mean + 2SD of TSAb activities in control subjects.The positive percentage of TSAb activity in GD group was calculated. Results The cAMP production and TSAb activities of GD group were higher than those of normal group[( 353. 65±126. 34 ) pmol/L vs (237.21±77. 15)pmol/L, ( 149. 08±53. 26)% vs ( 100±32. 52)%, P <0. 05] . The value that higher than 165% was set to be positive for TSAb. The positive percentage of TSAb in GD group was 50% ( 14/28). Conclusion CHO-hTSHR cell line constructed by our group is suitable for detecting TSAb activity in the sera of patients with GD.

10.Protective effect of curcumin on rat hippocampal neuron damaged by LPS-activated microglia conditioned medium

AIM: To establish the cell model of hippocampal neurons impaired by the supernatant of microglia conditioned medium (MCM) induced by LPS, and to study the mechanisms of neuron damage and the protection effect of curcumin. METHODS: The microglia were activated and stimulated by LPS at concentration of 5 μg/L. The supernatant of activated MCM was used to stimulate the hippocampal neurons. The morphology of the neurons was observed under light microscope and the survival rate was detected by MTT. The availability of respiration in the hippocampal neurons was determined by detecting the ratio of lactic acid/pyruvic acid, and the expression of 3 types of 1,4,5-inositol triphosphate (IP3R1, IP3R2, IP3R3) was detected by RT-PCR. RESULTS: The hippocampal neurons were damaged severely by the supernatant of activated MCM, and the morphology of the neurons changed significantly. Compared to control group, the ratio of lactic acid/pyruvic acid in MCM group was elevated and the expression levels of IP3R1 and IP3R3 in the hippocampal neurons were significantly increased. The ratio of lactic acid/pyruvic acid and the expression of IP3R1 in curcumin+MCM group were evidently lower than those in MCM group. However, no effect of curcumin on IP3R3 expression was observed, the changes of neural morphology was also ameliorated in curcumin+MCM group. CONCLUSION: Curcumin protects rat hippocampal neurons from damage induced by LPS-induced microglia condition medium.
