1.Expression of C-erbB-2 in sinonasal squamous cell cancer cells and its significance

Objective To analyze the expressions and clinical significance of human epidermal growth factor receptor-2 (C-erbB-2) protein in sinonasal squamous cell cancer cells.Method Immunohistochemical and RT-PCR methods were used to detect the expression of C-erbB-2 in 62 cases of sinonasal squamous carcinoma tissues,30 cases of nasal polyps and 25 cases with normal nasal mucosa.The relationships between the expression of C-erbB-2 in sinonasal squamous carcinoma tissues and clinical pathological characteristics were analyzed.Results There was significant difference in the expression of C-erbB-2 in sinonasal squamous carcinoma tissues, nasal polyps and normal nasal mucosa tissues(P <0.05).The expression of C-erbB-2 was positively correlated with the clinicopathologic stage, tumor classification and lymph node involvement(P<0.05).Conclusion The expression of C-erbB-2 is correlated with tumorigenesis,invasion and metastasis of sinonasal squamous cell carcinoma.It can be used as an auxiliary diagnostic and prediction maker as well as a new therapeutic target of sinonasal squamous cell cancer.

OBJECTIVE:
To explore the expression and significance of programmed cell death 5 (PDCD5) and Bcelllymphoma/lewkmia-2(Bcl-2) in sinonasal squamous cell carcinoma (SNSCC).
METHOD:
Immunohistochemical method and Western Blot method was used to determine the expression of PDCD5 and Bcl-2 in specimen of SNSCC in thirty cases, sinonasal inverted papillomas (SNIP) in thirty-eight cases, and normal nasal mucosa in twenty cases.
RESULT:
(1) The expression of PDCD5 protein in SNSCC significantly decreased compared with SNIP and normal nasal mucosa. (2) The expression of Bcl-2 protein in SNSCC up-regulated obviously compared with SNIP and normal nasal mucosa. (3) Positive rate of PDCD5 protein and Bcl-2 protein in well, moderate and low differentiatied group is respectively 100.00%, 83.33%, 38.89% and 50.00%, 70.83% and 100.00%, the difference was statistically significant (P < 0.05). (4) In the follow-up cases, the survival rate of the patients with higher expression of PDCD5 protein was higher, but that with lower expression of Bcl-2 protein was higher.
CONCLUSION
The inactivation of PDCD5 protein and the activation of Bcl-2 protein may play an important role in the development of SNSCC, and there are a positive correlation between PDCD5 and Bcl-2 protein in SNSCC, which may be identified as a new therapeutic target.

Objective To explore the expression and significance of Dickkopf-1 (Dkk-1) and cell lymphoma/leukemia-2 (Bcl-2) protein in sinonasal squamoucell carcinoma(SNSCC) .MethodThe immunohistochemical SP method and Western blomethod were adopted to determine the expression of Dkk-1 and Bcl-2 in 30 specimenof SNSC(SNSCgroup) ,38 specimenof sinonasal inverted papillomas(SNIP group) and 20 specimenof middle turbinate mucosa(control group) .ResultThe expression of DKK-1 protein in the SNSCgroup wasignificantly down-regulated compared with the SNIP group and the control group ,while the expression of Bcl-2 protein in the SNSCgroup wasignificantly up-regulated compared with the SNIP group and the control group;in the SNSCgroup ,the positive rateof DKK-1 protein and Bcl-2 protein in the high and middle differentiation group and the low differentiation group were 100 .00% ,68 .75% ,33 .33% and 50 .00% ,62 .50% ,100 .00% respectively ,the differencewere statistically significan(P<0 .05) .Conclusion Dkk-1 protein may play an importanpromoting role in the developmenand pro-gression procesof SNSC,the expression of Dkk-1 protein hanegative correlation with the expression of Bcl-2 protein in SNSC,which may become new targespoof SNSCgene therapy .
4.Clinical significance of PRL-3 expression in sinonasal squamous cell cancer cells

Objective To investigate the clinical significance of PRL-3 expression in sinonasal squamous cell cancer(SNSCC).Meth-ods The immunohistochemical method and RT-PCR were adopted to detect the PRL-3 protein expression level in 62 cases of SNSCC tissue (SNSCC group),30 cases of nasal polyps(NP group)tissue and 25 cases of normal nasal mucosa tissue(control group).The obtained results were compared.Results Both in the protein level and gene level detection,the expression of PRL-3 in the SNSCC group was higher than that in the NP group and control group,the difference was statistically significant (P <0.05).The expression of PRL-3 had no significant differences among different ages and between different genders(P >0.05),but with TNM stage increasing,differentiation degree decreasing and complicating lymph node metastasis,the expression of PRL-3 was significantly increased(P <0.05).Conclusion The PRL-3 expression can serve as good reference for the proliferation activity of SNSCC,its expressing intensity can obviously reflect the SNSCC cell proliferation activity,PRL-3 probably is an independent prognostic index of SNSCC,indicating poor prognosis.
5.Hydroxyapatite-bone morphogenetic protein ossicular prosthesis versus allogeneic cartilage ossicular prosthesis for ossicular chain damage

BACKGROUND:There is a great dispute on the choice of repair materials for ossicular chain damage.
OBJECTIVE:To explore the effects of new hydroxyapatite-bone morphogenetic protein ossicular prosthesis versus al ogeneic cartilage ossicular prosthesis in patients with ossicular chain damage.
METHODS:Sixty patients with chronic otitis media were equal y assigned into a control group and a test group. Two groups of patients underwent tympanoplasty and ossicular chain reconstruction with al ogeneic cartilage ossicular prosthesis or hydroxyapatite-bone morphogenetic protein ossicular prosthesis, respectively. Twelve months after surgery, fol ow-up results were compared between two groups.
RESULTS AND CONCLUSION:Air conduction value and air-bone gap value were both improved significantly in the two groups after surgery (P<0.05), and especial y, the postoperative air conduction value was better in the test group than the control group (P<0.05). Implant detachment was found in three cases of the control group, whereas did not occur in the test group. Improved hearing was found in 28 cases in the test group with an improvement rate of 93%and 25 cases in the control group with an improvement rate of 83%. There was a significant difference in the hearing improvement between the two groups (P<0.05). Overal , two kinds of prostheses can share similar effects on the ossicular chain repair.

6.The expression of EMS1 and DcR3 protein in laryngeal carcinoma and the relation between EMS1 and DcR3.

OBJECTIVE:
To investigate the expression of EMS1 and DcR3 in laryngeal carcinoma and analyze the relation of EMS1 and DcR3.
METHOD:
The expression of EMS1 and DcR3 protein in 41 laryngeal carcinoma fresh samples and 41 para-carcinoma tissues (to cutting margin > 0.5 cm) were measured by flow cytometry, and 15 normal laryngeal mucosa samples were also studied as controls.
RESULT:
(1) The quantitative and qualitative expression of EMS1 and DcR3 protein in laryngeal carcinoma tissues was obviously higher than those in para-carcinoma and in normal laryngeal mucosa tissues respectively (P < 0.05). There was no significant difference between the expression of para-carcinoma and normal laryngeal mucosa tissues. (2) In laryngeal carcinoma, the expression of EMS1 and DcR3 protein was independent of patients' clinical classification, tumor size, smoking history, patients' age and sex but associated with tumor metastasis, pathological grade and clinical stage. (3) In laryngeal carcinoma, the expression of EMS1 was positively correlated with that of DcR3.
CONCLUSION
EMS1 was positively related to DcR3, which might play an important role in the carcinogenesis and development of laryngeal carcinoma by synergic effect.
