Lower extremity deep venous thrombosis (LEDVT) is one of the common complications for postoperative patients with gynecologic oncology,which affect the prognosis of surgical patients seriously.LEDVT mainly caused by vein wall damage of pelvic surgical procedures and lower extremity infusion,slow venous blood flowing leading by anesthesia and bedridden,the hypercoagulability due to fasting before and after surgery and lack of fluid amount.
2.Assessment of coronary stenosis by 256-slice spiral CT

Objective:To assess coronary stenosis using 256-slice spiral CT (CTA).Methods:The data of 126 pa-tients diagnosed as coronary heart disease (CHD)using CTA and coronary angiography (CAG)Were retrospectively analyzed.The CAG results for CHD Were regarded as “gold standard”diagnosing CHD.The CTA diagnostic value for CHD and consistency of CAG and CTA diagnosing coronary stenosis Were analyzed.Results:A total of 393 cor-onary segments Were assessed using CTA.According to CAG results for CHD ,the CTA diagnostic accuracy Was 89.3%,sensitivity Was 91.4%,specificity Was 84.8%,negative predictive value Was 82.2% and positive predictive value Was 92.8%;the consistency of CAG and CTA diagnosing coronary stenosis Was good (Kappa value =0.755, P<0.01).Conclusion:The 256-slice spiral CT can diagnosing coronary stenosis,it possesses high sensitivity in di-agnosing vascular stenosis;as a simple and noninvasive examination it can be used as a reliable method for early screening subhealthy population and reexamine of putients With coronary heart disease.
3.Application of bioinformatics analysis in identifying a novel annexin subfamily-anx32

Objective: To investigate the structural characteristics and the primary functions of antigen gene cC1. Meth-ods: The primary and the secondary structures were analyzed using bioinformatics programs provided by Internet servers.The predicted 3D structure of cC1 was established by homology protein modeling method. Anticoagulant activity of GST-anx32 was assayed by modified kaolin partial thromboplastin time (KPTT). Results: cC1 had high homology to annexinsgenes both at nucleic acid and at amino acid level. It contained 4 homologous regions, and each region included the typical an-nexin motif "G-X-G-T (38 residues)-D/E". The results of KPTT assay showed that the recombinant protein GST-anx32 hadhigh anticoagulant activity. Conclusion: cC1 has the common structures of annexins but the homology to the extant annexinsis no more than 48%, cC1 is a member of a novel annexin subfamily and designed as annexin32.

AIM: To determine whether chronic hypercholesterolemia affects ionic currents on cardiac ventricular myocytes of rats. METHODS: Whole - cell patch - clamp technique was used to record the ionic currents in single cardiac myocytes isolated from normal cholesterolemia and hypercholesterolemia rats. 7.Augmentation index's change and its influencing factor in patients on continuous ambulatory peritoneal dialysis

Objective To analyze the factors influencing augmentation index (AI) by examining the AI changes in patients who received continuous ambulatory peritoneal dialysis (CAPD) therapy, and evaluate the artery elastic function situation. Methods The height, weight, blood pressure, blood indicators were detected in 46 CAPD patients (CAPD group) and 42 controls (control group). AI was managed by OMRON HEM-90OOAI, and the influencing factor was analyzed. Results Systolic blood pressure(SBP),diastolic blood pressure(DBP), pulse pressure, central aortic pressure and AIp75 (AI value, when pulse was 75 beats/min) in CAPD group were significantly higher than those in control group. Within CAPD patients,the AIp75, in female was significantly higher than that in nude [(96.67±8.95 )% vs (89.89±6.46)% ,P =0.022]. Simple correlation showed positive relationship in gender, age, SBP, DBP, pulse pressure and central aortic pressure with AI; and negative relationship in height, weight and weight index with AI; while there was no relationship between AI and all kinds of chemical examination indicators or dialysis years.Multiple linear regression analysis evidenced that gender,age, height, DBP and central aortic pressure of CAPD patients had linear regression relation with AI. Conclusions It indicates a decreased artery elasticity and an increased stiffness occurs in CAPD patients. AI has tight correlation with age, height, DBP and central aortic pressure, and is an non-invasive factor thai partially reflects the arterial elasticity. Central aortic pressure is relevant with aortic elasticity and rigidity closely. Measuring central aortic pressure is very imporrant for preventing cardiovascular accident.
8.Expression of tissue inhibitor of metalloproteinases 1 in chronic renal failure rats with arterial calcification

Objective To examine whether tissue inhibitor of metalloproteinases-1 (TIMP-1) is involved in arterial calcification of chronic renal failure (CRF) rats. Methods CRF model was induced in male Wistar rats by garage daily with 2% adenine 250 mg/kg. The calcification of aorta, femoral artery, renal artery and coronary artery was evaluated histomorphometrically by van Kossa-stained sections at 2-, 4-, 6- and 8-week respectively. RT-PCR and Western blot were used to observe the expressive levels of TIMP-1 mRNA and protein. Expressions of TIMP-1, osteopentin (OPN) and core binding factor α1 (Cbfα-1) protein were analyzed by immunhistochemistry. Results Serum urea nitrogen, creatinine, inorganic phosphate, calcium-phosphorus product and intact parathyroid hormone (iPTH) increased significantly in the model animals compared with control group after 2 weeks (P<0.01). Medial calcification was found in above four arteries of model groups after 6 weeks. RT-PCR and Western blot showed that TIMP-1 expression of model group was significantly higher than that of control group (P< 0.05), and obviously elevated in a time-dependent manner. The expression of TIMP-1 and OPN in calcified aortic smooth muscle cells increased obviously (P<0.05), and positive immunostaining of Cbfα-1 was found. The expression of TIMP-1 was positively correlated with OPN and Cbfα-1 (r=0.317, P=0.000; r=0.485, P=0.000). Conclusions The pathology of arterial calcification in CRF rats induced by adenine is similar to CRF patients, which may serve as a useful model of CRF with arterial calcification. The up-regulation of TIMP-1 seems to participate in the formation and development of vascular calcification in CRF.
9.Decline of learning and memory in senility and expression of cyclin-dependent kinase-5 in brain development of rats

BACKGROUND: Cyclin-dependent kinase-5 (CDK-5) is one of the members in cyclin-dependent protein kinase family. The attention has being drawn by researchers on the relationship between the expression and distribution of CDK-5 mRNA and its protein in the brain during brain development and neural degeneration in thought-cognition.OBJECTIVE: To probe into the influence of CDK-5 on neurogeny and neural degeneration during cerebral development.DESIGN: Single factor analysis of variance.SETTING: Histological and Embryological Department and Neurobiological Department in Third Military Medical University of Chinese PLA.MATERIALS: The experiment was performed in Histological and Embryological Department and Neurobiological Department in Third Military Medical University of Chinese PLA. Twenty-five Wistar rats of 5 phases were employed, named embryonicphase (E8-E21), neonatal phase (P0-P15),childhood (P16-2 months), grown-up phase (> 2 months) and senile phase (> 8 months), 5 rats in each group.METHODS: In situ hybridization histochemistry (ISH) and immunohistochemistry (IHC) staining was adopted in brain sections from embryonic phase to senile phase.MAIN OUTCOME MEASURFS: Distribution and expression of positive cells of CDK-5 mRNA and protein in various brain areas.RESULTS: Twenty-five rats entered result analysis for all. ① The expression of CDK-5 mRNA presented in entire development from E14 to P350and was in tendency of stability after growth-up. CDK-5 mRNA localized mainly in neurons and positive regions distributed mainly in cerebral cortex, hippocampus, thalamus, hypothalamus, cerebellum and a part of nerve nuclei. ② The expression of CDK-5 was strong after birth and it was weaker in embryonic and senile rats. Positive regions concentrated mainly in peripheral ventricle, hippocampus, cerebellum and a part of nerve nuclei.The expression only presented in hippocampus and Purkinje cellular layer of cerebellum in senile rats.CONCLUSION: CDK-5 in brain runs through entire phases of neural development, it expresses more significantly in neonatal phase and childhood and declines after growth-up, especially in senile phase. The declined expression of CDK-5 in hippocampus of senile rats is closely associated with decline of learning and memory in senility probably.
10.Expression of Bovine Interleukin-2 Gene in Pichia pastoris

The Interleukin-2 gene cDNA was cloned into the Pichia pastoris expression vector pPICZB,which is under the control of the alcohol oxidase promoter AOX1. The linearized recombinant plasmid of BoIL2-pPICZB,digested by Sac I ,was transformed into X-33 strains by electroporation. The multi-copy insert transformants were screened by Zeocin-resistance and induced by 1% methanol. The intracellular expression products were tested by SDS-PAGE analysis and Western blotting. Purified recombinant BoIL2 was gained by metal-chelating affinity chromatographic (MCAC). Assay with murine CTLL-2 cells showed that the recombinant BoIL2 exhibited the biological activity.
