OBJECTIVE:To explore optimal regime and less ADR for lupus nephritis(LN) by adopting self-made TCM prescription.METHODS:109 LN patients were assigned into control group(n=48) and therapy group(n=61).Both groups were treated with metacortandracin and cyclophosphamide.Control group were additionally given self-made TCM prescription.The clinical efficacy and adverse drug reaction(ADR) of 2 groups were compared.RESULTS:The curative rate and total response rate of therapy group were 77.05%(47/61) and 95.08%(58/61) respectively and those of control group 58.33%(28/48) and 77.08%(37/48).The curative rate and total response rate of therapy group were all higher than those of control group.There were significant difference between 2 groups(P

Pulmonary arterial hypertension (PAH) is an incurable fatal disease, with the mechanism of imbalance of vascular contraction and relaxation which start as a series of pathophysiological reactions. Angiotensin-converting enzyme 2-angiotensin (1-7)-G protein-coupled receptor axis [ACE2-Ang(1-7)-Mas], an expanded vascular axis which is considered as new axis of renin-angiotensin system (RAS) is a negative regulator of the classical angiotensin-converting enzyme-angiotensin Ⅱ-angiotensin Ⅱ type 1 receptor axis (ACE-AngⅡ-AT1R), a contraction vessel, proliferation axis. A retrospective study concerning the pathophysiological mechanism of ACE2 in the treatment of PAH was done and found that ACE2-Ang-(1-7)-Mas axis could reduce PAH induced by monocrotaline (MCT) through declining the inflammatory cascade, improving endothelial dysfunction and regulating autonomic nerve, reduce PAH induced by hypoxia through inhibiting the proliferation of pulmonary artery smooth muscle cells and reduce PAH induced by congenital heart disease. Therefore, ACE2 may become a future drug of preventing and treating PAH.
4.Antibiotic Susceptibility of Haemophilus Species Isolated from 195 Adults

OBJECTIVE To investigate the ?-lactamases producing rate in Haemophilus influenzae(HIN) and the antibiotics sensitivity,to guide the treatment of Haemophilus species infection.METHODS The strains from respiratory tract samples,were isolated and cultivated in chocolate cultures 2006 to 2007.Drug sensitivity test and beta-lactam enzyme test of Haemophilus species were taken.The results were analyzed by software SPSS11.5.RESULTS Totally 195 isolates of HIN were isolated 32 produced ?-lactamases(16.4%).The sensitivity of H.influenzae to cefuroxime,cefotaxime and ampicillin/sulbactam was 90.8%,93.8% and 95.4%,respectively.CONCLUSIONS Cefotaxime,ampicillin/sulbactam and cefuroxime are the most effective antibiotics for H.influenzae infection.
5.Significance of Serum Lipoprotein(a) in Patients with Acute Ischemic Stroke

Objective To investigate the serum lipoprotein (Lp) (a) level in patients with acute ischemic stroke and the prediction for short-term functional outcome. Methods The levels of serum Lp(a) were measured in 210 patients with acute ischemic stroke (within 24 h after onset) and 100 healthy people matched the gender and age. The patients were assessed with National Institutes of Health Stroke Scale (NIHSS) as admission, and modified Rankin Scale (mRS) as discharge. Results The median of Lp(a) was 325 mg/L (IQR: 180~545) in patients,significantly more than 148 mg/L (IQR: 69~265) in the normal controls (P<0.001). Level of serum Lp(a) was an independent variable for functional outcome (OR=1.004, 95%CI: 1.001~1.007, P<0.001) and death (OR=1.006, 95%CI: 1.001~1.008, P<0.001) in patients. In the receiver operating characteristic curve, the area under curve was more in the combined model [Lp(a) and NIHSS] than the NIHSS alone.Conclusion Serum Lp(a) increased in the in patients with acute ischemic stroke, and may predict the short-term outcome independently.
6.Optimization of Extraction Technology of Hyperici Perforate Herba, Coptidis Rhizoma, Schisandrae Chinensis Fructus, and Ziziphi Spinosae Semen forJinzao Yuanzhi Capsule by Orthogonal Test

Objective To optimize the extraction conditions of Hyperici Perforate Herba, Coptidis Rhizoma, Schisandrae Chinensis Fructus, and Ziziphi Spinosae Semen forJinzao Yuanzhi Capsule.Methods Hyperin extraction rate was set as index, and HPLC was used for determination. The extraction conditions of ethanol concentration, ethanol quantity, extraction time and other factors were studied with orthogonal test and comparative test.Results Ethanol concentration and extracting times were remarkable factors in the test. The optimum extraction technology forJinzao Yuanzhi Capsule was to reflux and extract the materials with 8 times of 70% ethanol for 2 times, reflux extraction for 1.5 hours, filtration. Then 6 times of 70% ethanol was added for 1.5 hours’ reflux, filtering, and merging extracted liquid.Conclusion The optimum extraction technology is reasonable and feasible.
7.Hypothermia combined with chemoradiotherapy in treatment of local advanced cervical cancer

Objective To evaluate the efficacy and toxicity of hyperthermia combined with chemoradiotherapy for local advanced cervical cancer. Methods Sixty-eight patients with local advanced cervical cancer were treated. Each patient was given cisplatin at the dose of 20 mg ivgtt dl -2 and 5 -fluorouracil 0.75 ivgtt dl -2, repeating every week, total 3 weeks. Thirty -eight patients received chemoradiotherapy alone. Thirty patients received hyperthermia with deeply focus technology, the temperature was increased and maintained 42.5 ℃ for 30 min at least, within 2 hours following irridiation. Hyperthermia was given twice per week with 7 or 12 sessions, as the same chemoradiation methods as the reference group.After 40 Gy therapy to estimate by cervical examination and ultrasonography, the patients with down stage were for operation, and the others continued to irridiation and additional brachytherapy with californium 252until radical dose. Results All patients were tolerated, fifteen patients of hyperthermia combined group were operated and ten patients of the reference group were operated, the difference was significance (50.0 % vs.26.3 %, P <0.05). The response rate of the combined group was 90.0 %, and the reference group was 84.2 %.The incidence of radiotherapy toxicity of the combined group was not higher than the reference group.Conclusion Hyperthermia combined with chemoradiotherapy can elevate the resection rate and the local control rate, not increase the radiotherapy toxicity and increase quality of life for cervical cancer patients.
8.Vitro antitumor activity and synthesis of the key intermediate of bakuchiol.

The in vitro antitumor activity of bakuchiol was exploited, compared with tamoxifen. The result of biological activities showed that bakuchiol could inhibit human breast cancer and the IC50 values were 2.89 x 10(-5) mol L(-1) and 8.29 x 10(-3) mol L(-1) against the cells line T-47D and MDA-MB-231 respectively. On the other hand, the key intermediate to synthesize bakuchiol was obtained by the method of Ireland-Claisen rearrangement. Comparing with traditional Claisen rearrangement, the reaction conditions are milder and the reaction reagents are safer.
9.Study of primary cells of breast cancer with special immune response induced by CIK co-cultured with self-dendritic cells

Objective To investigate effect to kill primary breast cancer cells by dendritic cell (DC)with self-freeze thawing antigens of primary breast cancer cells co-cultured with cytokine induced killer cell (CIK). Methods Self-neoplasm antigen by primary cells of breast cancer in log phase growth was prepared and DC and CIK cells was cultured from peripheral blood. The CIK of co-cultured DC loaded self-neoplasm antigen was compared to PBMC, CIK cells with self-neoplasm antigen and the single CIK cells. Results Killing efficiency of PBMC, CIK, Ag-CIK and Ag-DC-CIK were (34.35±3.28) %, (45.91±2.78) %, (50.88±3.22) %, (62.10±5.94) %. There were significant difference between Ag-DC-CIK group and CIK-Ag and CIK group to primary cells of breast cancer (P <0.01). Conclusion The CIK induced from cocultured DC loaded with self-neoplasm antigen show a special killing activity to self-primary tumor cells.
10.The Effect of changes in the Temporomandibular joint blood supply on glyco saminoglycans of the rabbit condylar cartilage and development of bone and joint disease

Objective The purpose of this study was to investigate the effect of blood supply decrease on glycosaminoglycans(GAG)of the cartilage in the temporomandibular joint(TMJ) . Method Four rabbits were used as controls, and 16 rabbits were injected 5% sodirm morrhumate through carotid artery unilaterlally. All specimens were examimined histochemically. Result A reduction in alcian(AB) blue staining was observed after operation, a gradual increase in AB staining with blood supply restoration of TMJ tissue was observed. Conclusion The result suggests that the decrease in blood supply of the temporomnandibular jorint leads to the loss of GAG in the condylar cartilage, which may play an important role in pathogenesis of osteoarthrosis.
