3.Effects of trichostatin A on HDAC8 expression, proliferation and cell cycle of molt-4 cells.

The effects of Trichostatin A (TSA) on histone deacetylase 8 (HDAC8) expression, proliferation and cell cycle arrest in T-lymphoblastic leukemia cell line Molt-4 cells in vitro were investigated. The effect of TSA on the growth of Molt-4 cells was studied by MTT assay. Flow cytometry was used to examine the cell cycle. The expression of HDAC8 was detected by using immunocytochemistry and Western blot. The results showed that proliferation of Molt-4 cells was inhibited in TSA-treated group in a time- and dose-dependent manner. The IC50 of TSA exposures for 24 h and 36 h were 254.3236 and 199.257 microg/L respectively. The cell cycle analysis revealed that Molt-4 was mostly in G0/G1 phase, and after treatment with TSA from 50 to 400 microg/L for 24 h, the percents of G0/G1 cells were decreased and cells were arrested in G2/M phase. Treatment of TSA for 24 h could significantly inhibit the expression of HDAC8 protein in Molt-4 cells (P<0.01). It was concluded that TSA could decrease the expression of HDAC8 in Molt-4 cells, which contributed to the inhibition of proliferation and induction of cell cycle arrest in Molt-4 cells.
4.Study on the molecular mechanism of two new mutations causing haemophilia B

Objective To study two new factor Ⅸ mutations Cys82Ser and Ile288Ser in vitro and research the molecular mechanism of haemophilia B. Methods PcDNA3. 1 ( - ) FⅨwt expression plasmid was prepared. The mutated FⅨcDNA expression plasmids, PcDNA3.1 ( - ) FⅨM1 (Cys82Ser) and PcDNA3. 1 ( - ) F Ⅸ M2 (Ile288Ser) were constructed by megaprimer method respectively. Transient expression experiments were performed using HEK293 cells transfected with the expression vectors containing the wild-type or the mutation recombinant cDNA. PcDNA3. 1 ( - ) was used as a blank control. The expression proteins were detected by ELISA, factor activity assay and flourescence stain. Results The results suggested that the two FⅨ gene mutations did not induce the reduction of the mutant FⅨ mRNA compared with the wild-type FⅨ mRNA. The FⅨ:Ag in culture media and cell lysate of wild type conduct were assigned as 100. 0%. The results of PcDNA3.1 ( - ) FⅨ M1 mutation protein were (27. 1 ± 5. 2)% and (99.4 ±4. 1)% respectively. For PcDNA3. 1( - )FⅨM2, the results were (5.3 ± 1.8)% and (31.7 ±2. 5)% respectively. The FⅨ: C in culture media of wild type conduct was also assigned as 100. 0%. Then the two types of mutant protein were ( 8. 5 ± 3.2 ) % and < 1%, respectively. Immunofluorescence microscopy result suggested that the intensity of perinuclear spot was reduced in cells transfected with PcDNA3.1 ( - ) FⅨM2 while staining for PcDNA3. 1 ( - ) FⅨM1 was predominantely diffuse without perinnclear enhancement. Conclusions These results strongly suggest that the FⅨ Cys82Ser mutation protein is not been correctly folded, by any possibility. The mutation protein has secretion defect. The secretion dysfunction and the protein degradation intracellular are possiblely the molecular pathology of Ile288Ser mutant protein.
5.Study on quality assurance system of grade 3 laminar operation room

ObjectiveTo explore the features of the cleanliness change with time during the use of laminar operating rooms in order to establish reliable quality assurance system. MethodsTwo grade 3 lami-nar operation rooms were chosen to be tested after one and a half year and two and a half years, laser counter was used to detect the static particulates after cleaning for 30 minutes and 45 minutes. ResultsAfter the laminar operation room was used for one and a half years the amount of 0.5 μm particulates in indoor air was less than 350 per liter, and after two and a half years it's above 350 per liter after cleaning for 30 minutes, an obvious difference than the previous one. The anaount of 1 μm and 2 pan particulates also increase obviously, but ff cleaning for 45 minutes the amount of 0.5 μm particulates in indoor air will fulfill the qualification. Al-though the amount of 0.5 μm, 1 pan and 2 μm particulates after 2 years using was higher than the one and a half yesrs'using after cleaning for 45 minutes, but the difference had no statistic meaning. CondusionsThe elesnliness of laminar operation rooms will drop as time prolonged, Amount of particulates in indoor air will in-crease too but the cleanliness can be ensured by extending cleaning time. After the quality assurance sys-tem was established the amount of smile particulates is still within the standard level
6.Clinical Efficacy of Acupuncture and Moxibustion Therapy Combined with Flames in 30 Cases of Nerve Root Type Cervical Spondylosis

Objective To study the clinical efficacy of both acupuncture combined with flames and simple acupuncture and moxibustion treatment of nerve root cervical spondylosis,so as to provide appropriate basis for the improvement of nerve root cervical spondylosis treatment. Methods A total of 60 cases of nerve root cervical spondylosis were enrolled for the study and randomly divided into two groups with the treatment of acupuncture combined with dragon moxibustion group(treatment group,n=30)and simple acupuncture group(control group,n=30). The clinical symptoms before and after treatment,and signs changes between the two groups were compared. Results Patients of both two groups showed significant im?provement after treatments(P<0.05);CSR simplify McGill Pain Scale scores inquiry(P<0.05). The total effective rate of the treatment group was 96.67%,which was 90.0%in the control group,with statistically significant difference(P<0.05). It indicated that the combined use of acupuncture and moxibustion therapy had better efficacy than simple acupuncture treatment. Conclusion The combined use of acupuncture and moxibustion therapy with flames in nerve root cervical spondylosis showed better clinical effect,which can be used as a preferred method of clinical treatment pro?grams.

9.Effect of long non-coding RNA CCAT1 on metastasis, invasion and epithelial-mesenchymal transition of endometrial carcinoma cell

Objective To investigate the effect of CCAT1 on migration,invasion and epithelial-mesenchymal transition (EMT) in endometrial carcinoma (EC) cell.Methods Using quantitative PCR assay,level of CCAT1 was detected in EC tissues to find its association with the initiation and malignancy degree of EC.Wound heal assay and transwell invasion assay were performed to study the effect of CCAT1 on migration and invasion ability of EC cell,while qPCR and western blot assay were utilized to detect the levels of related genes.Results In EC tissues,level of CCAT1 was significantly upregulated (P < 0.001) and was positively associated with the malignancy degree of EC.After CCAT1 knockout,shorter migration distance was found,fewer cell passed through the chambers,levels of metastasis-related genes (MMP2 and MMP9) and mesenchymal markers (Snail,Zeb1 and Twist1) were up-regulated,and epithelial markers (E-cadherin and ZO-1) were down-regulated.Conclusion CCAT1 was up-regulated in EC tissue and its expression level was positively associated with the malignancy degree of EC.CCAT1 knockdown inhibited the metastasis,invasion and epithelial-mesenchymal transition of EC cell.
10.Anti-cancer effects of deguelin on human leukemia K562 and K562/ADM cells In Vitro.

In order to investigate the anti-cancer effects of deguelin and on K562 and K562/ADM cells in vitro and the underlying molecular mechanism and compare the cytotoxicity of deguelin on K562, K562/ADM cells and human peripheral blood mononuclear cells (PBMCs). The effects of deguelin on cell proliferation were assessed by MTT assay. Apoptosis were detected by Annexin V/PI double-labeled cytometry. The effects of deguelin on the cell cycle were studied by a propidium iodide method. Our study showed that deguelin inhibited the proliferation of K562 cell and K562/ADM cell in a time- and dose-dependent manner and had minimal effects on normal human peripheral blood mononuclear cells. The ratio of IC(50) value of deguelin of 24 h on K562/ADM cells to K562 cells was only 1.27, which was significantly lower than the ratio of IC(50) value of ADM (higher than 20). Deguelin could induce apoptosis of K562 cells and K562/ADM cells. K562 cells were arrested at G(2)/M phase while K562/ADM cells were arrested at G(0)/G(1) phase. Our results suggested that deguelin was a novel anti-leukemia agents with high efficacy and low toxicity and it is also a promising agent for reversing drug resistance.
